Studies on Genetic Stability of Micropropagated Plants and, Reintroduction in an Endemic and Endangered Taxon: Syzygium travancoricum Gamble (Myrtacae)

  • Tissue culture techniques arguably are an important approach for ex situ conservation of rare and endangered plant species. However, there is utmost importance on maintaining the genetic integrity of the introduced plants especially in tree species. To examine the genetic integrity of the micropropagated plants, we randomly screened few hardened plants of Syzygium travancoricum, a critically endangered tree taxon, using Randomly Amplified Polymorphic DNA (RAPD) markers. Twenty-three random. primers were tried and twenty-five polymorphic loci were identified. The dendrogram based on the Unweighted Pair-Group Method Arithmetic Average and Nei's similarity index depicted about 97% homology between the mother plants and micropropagated plants. Further, an attempt was made to reintroduce the micropropagated plants in the wild. Over three hundred small trees could be successfully established.

  • To improve device yield and throughput, faults in plasma processing equipment should be quickly and accurately diagnosed. Despite many useful information of ex-situ sensor measurements, their applications to recognize plasma faultshave not been investigated. In this study, a new technique to identify fault causes by recognizing X-ray photoelectron spectroscopy (XPS) using neural network and continuous wavelet transformation (CWT). The presented technique was evaluated with the plasma etch data. A totalof 17 experiments were conducted for model construction. Model performance was investigated from the perspectives of training error, testing error, and recognition accuracy with respect to various thresholds. CWT-based BPNN models demonstrated a higher prediction accuracy of about 26%. Their advantages over pure XPS-based models were conspicuous in all three measures at small networks.

  • Although the bulk composition of materials is one of the major considerations in extractive metallurgy and environmental science, surface composition and topography (edges and dislocations are preferred sites for physicochemical reactions) control surface reactivity, and consequently play a major role in determining metallurgical phenomena and pollution by heavy metals and organics. An understanding of interaction mechanisms of different chemical species with the mineral surface in an aqueous media is very important in natural environment and metallurgical processing. X-ray photoelectron spectroscopy (XPS) has been used as an ex-situ analytical technique, but the material to be analyzed can be any size from $100{\mu}m$ up to about 1 cm. It can also measure mixed solids powders, but it is impossible to ascertain the original source of resulting x-ray signals where they were emitted from, since it radiates and scans the macro sample surface area.

  • Ferroelectric thin film capacitors with high dielectric constant are important for the application of memory devices. In this work, We have systematically investigated the variation of grain sizes depending on the process condition of two-step process. Both in-situ annealing and ex-annealing have been compared depending on the annealing time. C-V measurement, ferroelectric properties, leakage current, XRD and SEM were performed to investigate the electircal properties and microstructural properties of Pb(La, Ti)O$_3$ films.

  • Potassium doped $BaFe_2As_2$ superconducting thin films by using an ex situ pulsed laser deposition technique were fabricated in various conditions to find out an optimal growth condition. Controlled conditions were annealing temperature, annealing time, and mass of potassium. The $Ba_{1-x}K_xFe_2As_2$ thin films which has most good quality is fabricated at a condition of annealing temperature at $700^{\circ}C$, annealing time of 60 minutes, and 0.6 g of potassium lumps. In this condition we were able to fabricate good quality films with high transition temperature of ~ 39 K.

Efficient Screening of Surfactant for Soil Washing (토양세척을 위한 계면활성제의 효과적 선정)

  • Soil washing process being operated in ex-situ mode using surfactants could be appropriate one of the most effective one for remediation. The choice of surfactants has been considered most significantly to accomplish tile reduction of expenditure and the increase of efficiency. This study was carried out screening test and solubility, washing experiment, and surfactant sorption experiments for 18 kinds of surfactant obtained. Results from each surfactant's PSR obtained by the slope indicated that nonionic surfactants have much higher solubility for HOCs than anion surfactants for that. The washing experiment to find out a removal efficiency of each surfactant's TPH, LE1017 and LE1019 showed high removal efficiency. Through on the result of estimating the extent of adsorption of surfactants for soils, nonion surfactants showed higher adsorption to soils than anion surfactants.

  • 본 발표는 국가 멸종위기종의 보전을 위해 환경부에서 지정하는 서식지외보전기관의 그간 성과와 향후 운영방향에 대한 것이다. 서식지외보전기관은 서식지내에서 보전이 어려운 야생동 식물을 서식지외에서 체계적으로 증식 보전할 수 있도록 연구경험, 인력, 시설 등이 갖추어진 연구기관을 심사해 환경부에서 지정하고 지정대상기관이 운영하는 제도이다. 국립낙동강생물자원관은 최근 3년간 멸종위기식물 4종(섬개현삼, 분홍장구채, 대청부채, 큰바늘꽃)과 일반식물 1종(고란초)의 현장관리 및 증식연구 등을 통해 보전기반을 마련하여 현장평가를 거쳐 환경부로부터 2018년 1월 30일 지정서를 교부받았다. 이에 지금까지의 연구결과와 향후 연구방향에 대해 발표하여 관련분야 전문가의 고견을 받아 서식지외보전기관의 관리와 운영에 반영하고자 한다.

  • High-Tc YBa2Cu3O7-x superconducting thin films have been prepared by single-target off-axis RF magnetron sputtering. Optimal ratio of Y : Ba : Cu of the single-target was determined as 1 : 1.65 : 3.35 in order to obtain the stoichiometric films. Tc, crystalline phase, and microstructures of the surface and cross-section of the ex-situ YBa2Cu3O7-x thin films on MgO(100) had a Tc, zero of 80K, and the films on LaAlO3/Si had a Tc, on-set of 90 K and a Tc, zero of 70 K.

A Study on the Cleanup Process of HOCs-Contaminated Soil by Ex-situ Soil Washing Technology (Ex-situ 토양세척기법에 의한 소수성 유기오염물질로 오염된 토양의 정화에 관한 연구)

  • In this study, a series of batch-scale tests were conducted to optimize the design parameters for the application of soil washing techniques to the hydrophobic organic compounds(HOCs)-contaminated soil and to find the effective methods for the recovery of surfactants from washing effluent by using solvent. Several nonionic surfactants (polyoxyethylene oleyl ester) and sophorolipid were applied to the artificially contaminated soil (4,000 mg n-dodecane/kg dry soil). The effects of washing time, concentration of surfactant solution, dilution ratio, and temperature on washing efficiencies were examined. Hydrophile-liphophile balance (HLB) number was proven to be one of the important parameters for soil washing. The HLB numbers of OA-5 and sophorolipid are too low to form a stable soil-water emulsion. They showed very low washing efficiencies less than 10e1o. If HLB number is in the proper range to form a stable soil-water emulsion, surfactant having higher solubility for HOCs shows higher washing efficiency. OA-14 having higher HLB number than OA-9 formed more stable soil-water emulsion. But its washing efficiency was about 20% due to a lower molar solubility ratio (MSR) than OA-9. OA-9, which forms a stable soil-water emulsion and has comparatively high sotubility for HOCs, showed about 60% washing efficiency by itself. To recover anthracene effectively from OA-9 washing effluent by using benzene as an organic solvent, desirable temperature and pH were $30^{\circ}C$ and 2, respectively.

Genetic Diversity of Quercus gilva in Je-ju Island (제주도 개가시나무의 유전구조와 유전적 다양성)

  • This study was to analyze the genetic diversity of Quercus gilva Blume growing in Jeju Island for developing a preservation strategy. We examined the genetic diversity and structure using 6 ISSR primers and investigated 67 polymorphic ISSR amplicons in 80 trees distributed among five populations. The average of proportion of polymorphic loci were 93%, the average level of Shannon's information index was 0.237, and Nei's genetic diversity was 0.156. According to the analysis of the molecular variance (AMOVA), $F_{st}$ was 0.169 indicating there was a genetic variation among five populations. 17% of the total variation was allocated among the five populations, while the other 83% of the total variation was in individual trees in each population. The result could be due to the uneven number of trees among the five populations. Based on these results, the preservation strategy could be developed, for examples, considering for designation as "forest genetic resources conservation area" about the habitat, monitoring continuously, fostering the growth of seedling, ex situ preservation of genetic resources, and comparing the differences of environmental and genetic characteristic with population in ex situ.
