The Effects of Sunbanghwalmyung-eum Extract on Acute Sinusitis-induced Mouse (선방활명음(仙方活命飮)이 알레르기 비염을 동반한 급성 비부비동염 마우스에 미치는 영향)

  • Background and Objectives: Sinusitis, referring to an inflammatory disease that occurs on the mucus membrane inside the sinus, is one of the most common diseases in the otorhinolaryngology area. In oriental clinic, Sunbanghwalmyungeum (SHE) has been used as a primary prescription to treat inflammatory diseases and intumescence and to treat sinusitis. The aim of this study was to investigate the anti-inflammatory and anti-allergic effects of SHE on acute sinusitis induced mice. Materials and Methods: BALB/c mice were divided into three groups: the normal group, the group inoculated with S. pneumoniae which caused them allergic rhinitis (control group), and the group treated with the SHE extract after it was treated the same as the control group (sample group). We investigated the inhibition of Th 2 cell differentiation by SHE and the suppression of NF-${\kappa}B$ activation. Results: NF-${\kappa}B$ activation was suppressed, and iNOS & COX-2 production were inhibited by SHE in acute sinusitis. IL-4 and STAT 6 also appeared to be suppressed. The number of eosinophils in the sample group noticeably decreased when compared to the control group. In the general morphologic change, the increase of damaged respiratory ciliated epithelium & eosinophil's infiltration were decreased in the sample group. Goblet cells were maintained in the sample group. MIP-2 and HSP-70 decreased in the sample group. Apocrine secretion decreased in the sample group. Conclusion: The results suggest that SHE is significantly effective in the treatment of inflammation caused by acute sinusitis through the suppression of NF-${\kappa}B$ activation and the inhibition of Th 2 cell differentiation.

Changes in Ceramide in Stratum Corneum and Anti-inflamatory Effects of Sopungdojeok-tang on Atopic Dermatitis (소풍도적탕(消風導赤湯)이 아토피 피부염의 항염증효과와 각질층 ceramide변화에 미치는 영향)

  • Objective : This study was designed to identify lipid protection formation in stratum corneum and anti-inflammatory effects of Sopungdojeok-tang(SD) on atopic dermatitis(AD). Materials and Methods : In Vivo, SD extract was orally administered to BALB/c mice at $2.5m{\ell}/kg/day$ for 2 days after 5% sodium dodecyl sulfate evoked atopic dermatitis in abdominal skin. Morphological changes were observed by immunohistochemical stain using monoclonal antibodies(BrdU, ceramide, MIP-2, $NF-{\kappa}B$ p50, IL-4, and STAT6) and TUNEL method. In vitro, the alterations of IL-4 mRNA expression were detected by RT-PCT in SD extract treated EL4 cells after phorbol-12-myristate-13-acetate and 4-tert-Octylphenol induce Th2 skewed condition. Results : SD is used in Oriental Medicine for its potential curative for atopic dermatitis. In this study, we have investigated the anti-inflammatory and lipid lamella repair effects of SD were investigated. SD decreased the number of eosinophil in atopic dermatitis induced mice. In the histological properties, the hyperplasia, edema, infiltration of lymphocytes, damage of intercellular space of stratum corneum, BrdU positive reacted cells in stratum basal, and degranulated mast cells and capillaries in dermal papillae decreased in mice with SD. Treatment of SD also decreased MIP-2, STAT6 and IL-4 in dermal papillae. The IL-4 mRNA expression decreased in a dose-dependant manner in SD treated EL4 cells. In addition, decrease of $NF-{\kappa}B$ p50 and increase of apoptotic cells in dermis were observed in SD treated mice. These data suggest that SD may beneficial for atopic dermatitis. Conclusions : These data suggest that SD is beneficial in treatment of atopic dermatitis, and that SD provides lipid protection in stratum corneum and anti-inflammatory effects on atopic dermatitis.

The Effects of Kamichungsangboha-tang on the Respiratory Patterns and Tracheal Tissues in Allergic Asthma (가미청상보하탕이 Allergy성 기관지천식에 미치는 실험적 연구)

  • This study was aimed to find out Kami-Chungsangboha-tang's effects on allergic asthma using Egg-White Implant(EWI) method model recommended by Siqueira et al. Kami-Chungsangboha-tang has widely been prescribed and used in Oriental Medicine for controlling and curing of the symptoms of Asthma(dyspnea, wheezing, cough, sputum, chest discomfort). The Previous studies have demonstrated that Kami-Chungsangboha-tang had analgesic, sedative, anticonvulsive and antihistamine effects. But it was uncertain whether Kami-Chungsangboha-tang could inhibit the allergic reaction, so alleviate the symptoms of allergic asthma. With such aim, the respiratory patterns and eosinophil infiltration in the tracheal mucosa of rats with experimentally induced allergic asthma (using EWI method) were mainly observed. The results are as follows ; 1. Kami-Chungsangboha-tang decreased the number of malformed respiration with respect to control group (p<0.02). 2. Kami-Chungsangboha-tang decreased the number of eosinophils infiltrated in tracheal mucosa with respect to control group (p<0.05). Depending upon above results, it is considered that Kami-Chungsangboha-tang has the inhibitory effects on the process of allergic asthma and suggested that it could be used in relieving patients of the symptoms caused by allergic asthma.

Anti-asthmatic Effects of Samjajihwang-tang in OVA-induced Mice (삼자지황탕(三子地黃湯)의 생쥐 모델에 대한 항천식 효과)

  • This study aimed to evaluate the anti-asthmatic effects of Samjajihwang-tang (SJT) using OVA-induced asthmatic mice model. Asthmatic mice model was conducted by repeated challenge of OVA using C57BL/6 mice. Each group was treated with distilled water, SJT (400 mg/kg and 200 mg/kg) extract or cyclosporin A (10 mg/kg) for the later 8 weeks, Penh (plethysmography and enhanced pause), immune cells subpopulation, eotaxin, IL-5, TNF-${\alpha}$, Anti-OVA-lgE in BALF (bronchoalveolar lavage), and lung tissue was analyzed, No cytotoxicity of SJT was shown on hFCs (human fibroblast cells). Administration of SJT significantly decreased Penh levels comparing to control group. SJT treatment significantly ameliorated the increase of total cells number and eosinophil including of immune cell subpopulation of $CD3^+/CD69^+$, $CCR3^+$, $B220^+/CD22^+$, $B220^+/CD45^+$, and $B220^+/lgE^+$ cells in BALF comparing to control group. Eotaxin, IL-5, TNF-${\alpha}$, and Anti-OVA-lgE level in BALF were significantly decreased by SJT treatment too. Histopathological finding verified the improvement of infiltration of inflammatory cells and collagen tissue in the SJT groups comparing to control group. These results strongly suggest that SJT would be a effective candidate for herbal-originated anti-asthmatic drug. However, this drug should be further studied for characterization of the accurate action and underlying mechanisms using variant disease model in the future.

The Effects of Probiotics-Fermented Magnolia Denudata in Ovalbumin Induced Allergic Rhinitis Animal Model (알레르기 비염 동물 모델에서 유산균 발효 신이(辛夷)의 효과)

  • Objectives : We aimed to determine therapeutic effects of probiotics-fermented Magnolia denudata(MD) in the allergic rhinitis model mice. Methods : Polyphenol production, DPPH radical scavenging activity and NO inhibition of fermented MD by different bacterial strains were evaluated to select the one that is most suitable for fermentation. Thirty C57BL/6 mice were divided randomly into 5 groups as follows: normal group, ovalbumin(OVA)-treated plus water fed(CON group), OVA-treated plus unfermented MD fed(UMD group), OVA-treated plus fermented autoclaved MD fed(A-FMD group) and OVA-treated plus fermented unautoclaved MD fed(FMD group). After 9 weeks, we observed changes in the blood cell count, OVA-specific IgE level, nasal rubbing, nasal mucosal tissue and body weight. Results : Extract of MD fermented by Bifidobacterium breve(BB) for 48 hours showed the highest anti-oxidant activity and anti-inflammatory activity out of all the other bacterial strains. The number of eosinophil count in A-FMD, FMD group and platelet count in FMD group showed statistically significant decrease(p<0.05). OVA-specific IgE level decreased in all 3 experimental groups, significantly in UMD and A-FMD group. Nasal symptoms were attenuated in all 3 experimental groups, statistically significant in A-FMD and FMD group (p<0.05). Histologically, infiltration of eosinophils into the nasal mucosa decreased in all 3 experimental groups, especially marked decrease in FMD group. Conclusions : According to the above results, it is considered that probiotics-fermented Magnolia denudata has inhibitory effects on the allergic rhinitis animal models.

  • Allergic asthma is a worldwide public health problem and a major socioeconomic burden disease. It is a chronic inflammatory disease marked by airway eosinophilia and goblet cell hyperplasia with mucus hypersecretion. Mouse models have proven as a valuable tool for studying human asthma. In the present report we describe a comparison of mouse asthma models. The experiments were designed as follows: Group I was injected with ovalbumin (OVA, i.p.) on day 1 and challenged with 1% OVA (aerosol exposure) on days $14{\sim}21$. Group II was injected on day 1, 14 and aerosol-immunized on days $14{\sim}21$. Group III was injected on day 1, 14 and immunized by 1% OVA aerosol on days $18{\sim}21$. We assessed asthma induction by determining the total number of white blood cells (WBC) and eosinophils as well as by measuring cytokine levels in bronchoalveolar lavage fluid (BALF). In addition, we evaluated the histopathological changes of the lungs and determined the concentration of immunoglobulin E (IgE) in serum. Total WBC, eosinophils, Th2 cytokines (IL-4, IL-13) and IgE were significantly increased in group I relative to the other groups. Moreover, histopathological studies show that group I mice show an increase in the infiltration of inflammatory cell-in peribronchial and perivascular areas as well as an overall increase in the number of mucus-containing goblet cells relative to other groups. These data suggest that group I can be a useful model for the study of human asthma pathobiology and the evaluation of existing and novel therapeutic agents.

Secretion of MCP-1, IL-8 and IL-6 Induced by House Dust Mite, Dermatophagoides pteronissinus in Human Eosinophilic EoL-1 Cells

  • The house dust mite (Dermatophagoides pteronissinus) is an important factor in triggering allergic diseases. The function of eosinophils, particularly in the production of cytokine or chemokine, is critical in understanding the pathogenesis of inflammatory diseases. In this study, we examined whether D. pteronissinus extract (DpE) induces the expression of monocyte chemotactic protein 1 (MCP-1)/CCL2, IL-8/CXCL8, and IL-6 that mediate in the infiltration and activation of immune cells and in its signaling mechanism in the human eosinophilic cell line, EoL-1. DpE increased the mRNA and protein expression of MCP-1, IL-8, and IL-6 in a time- and dose-dependent course in EoL-1 cells. In our experiments using signal-specific inhibitors, we found that the increased expression of MCP-1, IL-8, and IL-6 due to DpE is associated with Src family tyrosine kinase and protein kinase C $\delta$ (PKC $\delta$). In addition, the activation of extracellular signal-regulated kinase (ERK) is required for MCP-1 and IL-8 expression while p38 mitogen-activated protein kinase (MAPK) is involved in IL-6 expression. DpE induced the phosphorylation of ERK and p38 MAPK. PP2, an inhibitor of Src family tyrosine kinase, and rottlerin, an inhibitor of PKC $\delta$, blocked the activation of ERK and p38 MAPK. DpE induces the activation of ERK and p38 MAPK via Src family tyrosine kinase and PKC $\delta$ for MCP-1, IL-8, or IL-6 production. Increased cytokine release due to the house dust mite and the characterization of its signal transduction may be valuable in understanding the eosinophil-related pathogenic mechanism of inflammatory diseases.

Epidemiological study on prevalence of fascioliasis in slaughtered cattle (도축 소의 간질 감염실태에 관한 역학적 고찰)

  • This study was carried out to investigate the prevalence of fascioliasis of slaughtered and farmed cattle in Gwangju area from February to November in 2007. A total of 1,000 cattle fecal samples were collected from slaughter houses (n=805) and farms (n=185). Twelve (1.2%) were found as positive cases with excretion of the egg of Fasciola spp in the fecal specimens, and 128 (12.8%) were positive in intestinal parasitism using the flotation and sedimentation procedures. The infection rate of fluke larvae from the slaughtered cattle at abattoirs in Gwangju was 0.75% (6 out of 805 heads). In histopathology, there were several liver lesions such as inflammation with infiltration of eosinophil, polymorphonuclear cells, mononuclear cells and multinucleated giant cells, proliferation of connective tissue, calcification and abscess formation.

  • Transient peripheral eosinophilia occurs in several disorders, such as allergic diseases, cancer, and parasitic infections. However, in most cases, their presence is not accompanied by tissue destruction or organ dysfunction. In certain disease states, eosinophils can accumulate in any organ in the body and cause tissue destruction as a result of the eosinophil infiltration or the toxic effects of the degranulated proinflammatory products. Idiopathic hypereosinophilic syndrome is a rare disorder characterized by persistent eosinophilia of an unknown origin, usually associated with a dysfunction of organs such as the heart, lung, skin, and nervous system. Idiopathic hypereosinophilic syndrome usually has an indolent course over a period of several months. However, in some cases, they have grave symptoms if vital organs such as heart and lung are infiltrated. Here we report a case of idiopathic hypereosinophilic syndrome presenting acute pulmonary edema involving the heart, bone marrow, and lung with a review of the relevant literatures.

The effects of Gamisipjeontang-gamibang on injured tissue after burn elicitation (가미십전탕(加味十全湯) 가미방(加味方)이 화상(火傷)의 조직 손상에 미치는 영향)

  • Objective : The purpose of this study is to examine the effects of Gamisipjeontang-gamibang(GT) on an experimental bum elicitation. Method : The absorbance of the photo cell mixed with GT at the Abs 560 nm was measured after irradiation for 1 min. In order to know the antioxidant effect on skin cell of mice after burn elicitation, superoxide dismutase(SOD) activity was measured. Also, in order to know effects on the skin regeneration in the burned mice, we counted the eosinophil in blood from animals via cardiac puncture and observed the histological structure in the epidermal basal layer and the dermal section on the 3rd, 7th and 14th day after burn elicitation. We also studied changes in angiogenesis in the capillary surrounding the basal layer and dermal papilla. The changes of HSP70 distribution and changes of p53 positive reaction were observed to investigate the changes of the stress in the skin as well. Result : The results indicated that GT has a significant impact on the antioxidant effect on skin cells of mice after bum elicitation by increasing SOD activity in vitro test. GT seems to decrease MIP-2 which induces neutrophil infiltration and promotes the angiogenesis dose-dependently. Furthermore, GT decreased HSP70, the expression of which was induced by elevated temperatures, and p53 which induce a apoptosis after stress. Conclusion : GT can be applied to burned skin through its antioxidant effect and skin regeneration property.
