Studies on the Immobilized Whole-cell Enzyme of Arthrobacter simplamide Polymer

  • Arthrobacter simplex (ATCC 6946) was cultured, induced and immobilized in acrylamide polymer. The characteristics of the immobilized whole-cell enayme were studied using hydrocortisone as the substrate. The enzyme activity was increased during the incubation of the gel particle in 0.5% peptone media. The ennzyme reaction kinetics of the Δ'-dehydrogenase (3-oxosteroid Δ'-oxydo reductase, E. C. 1.3.99.4) foliowed the Michaelis-Menten type. Km and Vm values were different significantly after immobilization of the cell. The optimum pH and temperature were changed, too. Nitrogen sources such as casitone, peptone or tryptone were good media for the enzyme reaction. And there was no need to add cofactors of the enzyme in the pre-sence of energy sources used in the test. The effect of metal ions on the enzyme activity was insignificant. Organic solvents were used increase the substrate concentration and there was no optimum solvent concentration depending on the substrate concentration.

Optimization of the Optical Resolution of Racemic $\alpha$-Methylbenzylamine Catalyzed by Enzymatic Reaction in Organic Media (유기용매에서 효소반응을 통한 라세믹 $\alpha$-Methylbenzylamine 광학적 분할의 최적화)

  • Optical resolution of racemic ${\alpha}$-methylbenzylamine was carried out by using Bacillus licheniformis protease in organic media. Enantioselective amidation of racemic amino with an ester as an acyl donor was successfully employed to resolve the racemate. To enhance reaction rate and enantioselectivity, pH-adjustment by lyophilization of enzyme dissolved in buffer, colyophilization with salts or lyoprotectants, selection of solvents and molecular design of esters were investigated. The optimization of the resolution reaction achieved about 30-fold increase in initial reaction rate and about 12-fold increase in enantioselectivity, respectively.

  • Cyclodextrin glucanotransferase [CGTase, E.C.2.4.1.19] is an extracellular enzyme, which catalyzes he formation of ${\alpha}$-, ${\beta}$-, ${\gamma}$- CDs from starch. Their proportions of formations depend on enzyme sources and reaction conditions. To understand what determines the product specificity of CGTases, we examined the alteration of product specificity of CGTase from Bacillus macerans by organic solvent sand pH. At acidic pH range less than pH 6 where the enzyme was unstable, the ratio of ${\alpha}$-/ ${\beta}$-CD production was increased 4 times more than that at neutral pH range. As we increased the concentration of 2-butanol, ${\alpha}$-/ ${\beta}$-CD ratio was proportionally increased but / ratio remained constant. The ${\alpha}$-/ ${\beta}$-CD ratio of products was increased in the reaction media which yielded low products.

  • Tyrosinases catalyze the hydroxylation of a monophenol (monophenolase activity) and the conversion of an o-diphenol to o-quinone (diphenolase activity), which are mainly involved in the modification of tyrosine residues into 3,4-dihydroxyphenyl-alanine (DOPA) and DOPA/DOPAquinone-derived intermolecular cross-linking. Previously, we obtained a slightly acidic and cold-active tyrosinase, tyrosinase-CNK, by our recombinant protein approach. The enzyme showed optimal activity at pH 6.0 and 20 ℃ with an abnormally high monophenolase/diphenolase activity ratio and still had approximately 50% activity compared with the highest activity even in ice water. Here, we investigated reaction stability of the recombinant tyrosinase-CNK as a psychrophilic enzyme. The enzyme showed remarkable thermal stability at 0 ℃ and the activity was well conserved in repeated freeze-thaw cycles. Although water-miscible organic solvent as reaction media caused the activity decrease of tyrosinase-CNK as expected, the enzyme activity was not additionally decreased with increased concentration in organic solvents such as ethanol and acetonitrile. Also, the enzyme showed high salt tolerance in chaotropic salts. It was remarkably considered that 2+ metal ions might inhibit the incorporation of Cu2+ into the active site. We expect that these results could be used to design tyrosinase-mediated enzymatic reaction at low temperature for the production of catechols through minimizing unwanted self-oxidation and enzyme inactivation.

Molecular Cloning, Characterization, and Application of Organic Solvent-Stable and Detergent-Compatible Thermostable Alkaline Protease from Geobacillus thermoglucosidasius SKF4

  • Several thermostable proteases have been identified, yet only a handful have undergone the processes of cloning, comprehensive characterization, and full exploitation in various industrial applications. Our primary aim in this study was to clone a thermostable alkaline protease from a thermophilic bacterium and assess its potential for use in various industries. The research involved the amplification of the SpSKF4 protease gene, a thermostable alkaline serine protease obtained from the Geobacillus thermoglucosidasius SKF4 bacterium through polymerase chain reaction (PCR). The purified recombinant SpSKF4 protease was characterized, followed by evaluation of its possible industrial applications. The analysis of the gene sequence revealed an open reading frame (ORF) consisting of 1,206 bp, coding for a protein containing 401 amino acids. The cloned gene was expressed in Escherichia coli. The molecular weight of the enzyme was measured at 28 kDa using sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The partially purified enzyme has its highest activity at a pH of 10 and a temperature of 80℃. In addition, the enzyme showed a half-life of 15 h at 80℃, and there was a 60% increase in its activity at 10 mM Ca2+ concentration. The activity of the protease was completely inhibited (100%) by phenylmethylsulfonyl fluoride (PMSF); however, the addition of sodium dodecyl sulfate (SDS) resulted in a 20% increase in activity. The enzyme was also stable in various organic solvents and in certain commercial detergents. Furthermore, the enzyme exhibited strong potential for industrial use, particularly as a detergent additive and for facilitating the recovery of silver from X-ray film.

  • Chitinase (EC 3.2.1.14) from culture fluid of Bacillus licheniformis KFB-C14 was purified 66-folds to homogenity in overall yield of 21% by ammonium sulfate fractionation, DEAE-Toyopearl, Butyl-Toyopearl and TSK-Gel HW-55F column chromatography. The enzyme protein had a molecular weight of about 86,000 and was composed of one subunit. The enzyme was significantly stable not only at high temperature but also on treatment with organic solvents and protein denaturants such as SDS, urea and guanidine-HC1. The optimum temperature and pH for reaction was 60$\circ $C and 6.0, respectively. The enzyme activity was inhibited by only Mn$^{2+}$ ion, but not inhibited by EDTA, N- ethylmaleimide and pCMB. The enzyme had high activity with colloidal chitin (V$_{max}$: 421) and commercial chitin (V$_{max}$: 480), but not with typical substrates of exo type chitinase. The thermostable chitinase had an useful reactivity for producing functional chitooligosaccharide, showing the production of (GlcNAc)$_{1}, (GlcNAc)$_{3}$, and (GlcNAc)$_{2}$ as major product.

  • The temperature dependency and the stability of enzyme systems for $11{\alpha}-hydroxyla­tion$ of progesterone were investigated using Aspergillus phoenicis. Though A. phoenicis conserves high enzyme activities for lactose hydrolysis even at high temperatures, the bioconversion reaction of progesterone by this strain was found to be very temperature sensitive. The compositions of reaction mediums of inside and outside of cells were analyzed using sonication technique. At early stage of reaction, the concentration of $11{\alpha}-hydroxyprogesterone$ of cell inside was higher than that of outside. But as the reaction proceeded further, the $11{\alpha}-hydroxyprogesterone$ existing inside of cells being converted into another products, its concentration was lower within the cells that in the bulk medium. Even in the reaction mediums containing organic solvents, A. phoenicis was founded to be able to metabolite, so that $11{\alpha}-hydroxyprogesterone$ can be produced continuously from fixed bed reactions packed with immobilized A phoenicis in vivo.

  • We examined some properties of yeast cell wall lytic enzyme produced by Dicyma sp. YCH-37. Several metal ions, reducing reagents, and chemical modifiers have little effects on the lytic activity, except guanidine-HCl. Yeast cells of early log phase were more susceptible to the enzyme than those of stationary phase, and heat-treated cells were more easily lysed than intact living ones. Yeast cells pretreated with organic solvents such as butanol and acetone were more susceptible to the enzyme than intact living ones. Yeast cells cultured in Yeast extract-Malt extract medium containing 0.5 M ammonium sulfate were easily lysed by the lytic enzyme, and yeast cells cultured without shaking were more easily lysed by the enzyme than those with shaking. When SDS, ${\beta}-mercaptoethanol$, Triton X-100, sodium sulfite, and KCl were added to enzyme reaction mixture each, lysis of yeast cells was more effective.

Preparation of Aliphatic Polyester by Lipase Catalyzed Transesterificatoin in Anhydrous Organic Solvents (유기용매에서 Lipase에 의한 지방족 폴리에스터의 합성)

  • Enzyme-catalyzed polycondensatlon reaction of aliphatic polyesters with several repeating units was studied using the biocatalytic activities of lipases from different sources. Porcine pancreatic lipase (PPL) was found to be best in utilizing bls(2,2,2-trichloroethyl) glutarate and 1,4-butanediol as substrafes. The reaction was also catalyzed to some extent by the lipases from Humicola lanuginos and Psudomonas sp. In the series of short-chain diols(C2-C4), bis(2,2,2-trichloroethyl) glutarate was iransesterified fastest with 1,4-butanediol and for the long-chain diols (PEG-300-PEG-1000), the reaction was fastest with PEG-400. With PEGs, only monoesterification product was obtained. PPL functioned well in relatively hydrophilic organic solvents such as tetrahydrofuran(THF), ether and acetonitrile. The reaction rate was accelerated as the reaction temperature was raised from $20^{\circ}C$ to $60^{\circ}C$ while Mn values of the reaction products were not affected by the reaction temperature. End group analysis by NMR showed that Mn values of the polymer were in the range of 1500-4000 daltons.

Overproduction, Purification, and Characterization of Heat Stable Aldolase from Methanococcus jannaschii, a Hyperthermophic Archaea

  • An aldolase gene has been cloned from Methanococcus jannaschii. The coding region of the gene has been expressed in E. coli using a pET system to a level of 30% of total cellular proteins. The protein was purified to more than 95 % homogeneity by heat treatment and ion exchange chromatography. The protein performed an aldol condensation reaction with glyceraldehyde as substrate and dihydroxyacetone phosphate as a carboxyl donor. The protein was determined to be a type II aldolase which requires the $Zn^{2+}$ ion as a metal cofactor. This enzyme has a broad range of optimum pH (7-9) and temperature ($50-80^{\circ}C$). It shows strong stability against heat, chemical denaturants, as well as a high percentage' of organic solvents. The half-life of this enzyme at $85^{\circ}C$ is more than 24 h and it maintains more than 90% of aldolase activity in the presence of 6 M urea, 50% acetonitrile, or 15% isopropyl alcohol.
