Relations between the Micorfloral Composition and the Environmental Factors Affecting Korean Infants during Lactation (수유기 영아의 장내균총 조성과 환경인자와의 관계)

  • The relations between micorfloral compositions and environmental factors of 32 Korean infants were sought through statistical analysis after examination of fecal bacterial and questions to their mothers about feeding experiences at three different times for each subjects, about one week after birth, before weaning (2-3 months after birth), and after weaning 95-7 months after birth). The majority of mothers fed their infants cereal foods after the age of 4 months and began weaning with fruit juice and commercial weaning foods. Defection frequencies and fecal pH of infants decreased significantly during the examination period and 37.5% of total infants in fecal samples increased significantly during the examination period, which means that the kinds of bacterial genera increase with aging of infants. Frequencies of streptococci were significantly higher in infants fed delivered by Caesarian section than infants delivered naturally. Frequencies of clostridia were significantly higher in infants fed with cereal food before 4 months of age than after 5 months. The infants fed with probiotics showed significantly higher frequencies of veillonella at about 1 week old. They also showed significantly higher frequencies of clostridia before weaning than the infant fed with no probiotics, but significantly lower frequencies of C. perfringens before weaning. The infants fed with probiotics showed significantly higher number of streptococci at the age of about 1 week and significantly higher numbers of total aerobes before weaning, but significantly lower numbers of bacteroides after weaning than their counter parts. The fecal pH was directly proportional to the number of clostridia, klebsiella, and total aerobes at about 1 week after birth, to the number of E. coli before weaning, and to the number of streptococci and clostridia after weaning. Fecal pH had a negative relationship to the total number of anaerobes in 1-week-old infants. The infants that had diarrhea during lactation showed higher frequencies of bacteroides before weaning than those that didn't.

  • A sediment control dam is an artificial structure built to prolong sedimentation in the main dam by reducing the inflow of suspended solids. These dams can affect changes in dissolved organic matter (DOM) in the water body by changing the river flow regime. The main DOM component for Yeongju Dam sediment control of the Naeseongcheon River was analyzed through 3D excitation-emission matrix (EEM) and parallel factor (PARAFAC) analyses. As a result, four humic-like components (C1~C3, C5), and three proteins, tryptophan-like components (C2, C6~C7) were detected. Among DOM components, humic-like components (autochthonous: C1, allochthonous: C2~C3) were found to be dominant during the sampling period. The total amount of DOM components and the composition ratio of each component did not show a difference for each depth according to the amount of available light (100%, 12%, and 1%). Throughout the study period, the allochthonous organic matter was continuously decomposing and converting into autochthonous organic matter; the DOM indices (fluorescence index, humification index, and freshness index) indicated the dominance of autochthonous organic matter in the river. Considering the relative abundance of cyanobacteria and that the number of bacteria cells and rotifers increased as autochthonous organic matter increased, it was suggested that the algal bloom and consequent activation of the microbial food web was affected by the composition of DOM in the water body. Research on DOM characteristics is important not only for water quality management but also for understanding the cycling of matter through microbial food web activity.

Mineral Distribution of the Southeastern Yellow Sea and South Sea of Korea using Quantitative XRD Analysis (정량X선회절분석법을 이용한 황해 남동부, 한국남해 및 제주도 남단 표층퇴적물의 광물분포 연구)

  • We studied the mineral composition and mineral distribution pattern of 131 surface sediments collected at the cruise in 2000 and 2007 from Southeastern Yellow Sea, South Sea of Korea and Southern part of Jeju Island. Mineral compositions of surface sediments were determined using the quantitative X-ray diffraction analysis. Surface sediments were composed of rock forming minerals (quartz 37.4%, plagioclase 11.7%, alkali feldspar 5.5%, hornblende 3.1%), clay minerals (illite 19.2%, chlorite 4.7%, kaolinite 1.8%) and carbonate minerals (calcite 10.7%, aragonite 3.4%). Distribution of clay minerals is very similar with fine-grained sediments, and especially same as the distribution of HSMD (Hucksan Mudbelt Deposit), SSKMD (South Sea of Korea Mudbelt Deposit) and JJMD (Jeju Mudbelt Deposit). The coarse sediment seemed to be relic sediment during the last glacial maximum and mainly consisted of rock forming minerals. Whereas the fine sediments mainly composed of clay minerals. Based on the clay mineral composition, main ocean current and geographical factor, HSMD and SSKMD might have derived from the rivers around the Korean Peninsula. However, JJMD is complex mudbelt deposit, which formed by Korean rivers and oceanic sediments.

Effect of Aminoethoxyvinylglycine Dipping Treatment on Ethylene Production and Cell Wall Composition of 'Tsugaru' Apple Fruits during Cold Storage (Aminoethoxyvinylglycine 침지처리가 '쓰가루' 사과의 저온저장중 에틸렌발생과 세포벽성분들의 변화에 미치는 영향)

  • This study was conducted to determine the influence of postharvest dipping treatment with aminoethoxyvinylglycine (AVG) on ethylene production and composition of non-cellulosic neutral sugars in cell walls of 'Tsugaru' apple fruits during storage. Fruits were harvested on August 20, soaked in AVG 50 and 75 $mg L^{-1}$ solution for 5 minutes, and stored in cold storage chamber at $0{\pm}1^{\circ}C$ for 60 days. Fruit quality factor, ethylene productions, and cell wall component changes were investigated at 20 days interval. As a result, the fruit firmness and acid content were much higher in AVG treated fruits than those of untreated one during 60 days of cold storage. Ethylene production of AVG treated fruits was reduced to the level of 1/10 compared with untreated one. As to the change of non-cellulosic neutral sugars in the cell walls of 'Tsu- garu' fruits, the major sugar was arabinose and galactose in water, CDTA and $Na_2CO_3$ soluble fractions. The content of arabinose and galactose in untreated fruits increased as the softening of fruits was in progress, but the fruits treated with AVG showed a little change during storage, so it is predicted that these two cell wall compositional sugars were not solubilized by the treatment of AVG. Accordingly, the marketability of 'Tsu- garu' fruits could remarkably increase when soaking the fruits in AVG solution after harvest.

Chemical Composition Characteristics of Dustfall in Nakdong River Area (낙동강 하류역 강하먼지의 화학적 조성 특성)

  • Dustfall samples were collected by the modified American dust jar (bulk type) at 5 sampling sites in the Nakdong river area from lune 2002 to May 2003. Nineteen chemical species (Al, Ca, Cd, Cr, Cu, Fe, K, Mg, Mn, Na, Ni, Pb, Si, V, Zn, $Cl^-$, $NO_3^-$, $SO_4^{2-}$, and $NH_4^+$) were analyzed via the combination of ICP/AES, AAS, IC and UV. The purposes of this study were to qualitatively evaluate the chemical composition of dustfalls by examining their regional and seasonal distribution patterns. Computation of the enrichment factor showed that well-defined anthropogenic sources, particularly in Pb were found in the order Gamjeondong (industrial area), Wondong, Silla University, Samrangiin and Mulgum. The seasonal mean of soil contribution showed its highest value (16.3%) during the winter with an annual mean of 11.2%. The concentration ratio of [$SO_4^{2-}/NO_3^-$] was found to be highest (5.12) during the winter, while the lowest ratio value (3.30) was seen during the all. fall, Also regional equivalent ratios of [$SO_4^{2-}/NO_3^-$] were found in the order: Silla University (6.78), Gamjeondong (4.98), Mulgum (3.95), Wondong (3.85), and Samrangjin (2.87). Seasonal distribution of water soluble components for total dustfall were found in the order: spring (71.6%), summer (61.2%), fall (49.2%) and winter (48.6%) with a mean ratio of 57.6%. Regional contribution of sea salts of water soluble ions were found in the order: Silla university (34.5%), Gamjeondong (28.3%), Wondong (17.3%), Samrangiin (17.2%) and Mulgum (13.8%), the total mean contribution rate was 22.1%. As for the chemical composition of dustftll on the lower Nakdong river, there is a decreased influence of sea salt and artificial anthropogenic sources and increased influence of soil particle inland. Also, the total amount of deposition on the lower Nakdong river has decreased, with the river's surface serving as a confounding factor in resuspending dusts.

Effect of Co-existence of Carbofuran and Chlorothalonil on the Short-term Bioconcentration Factor in Brachydanio rerio(zebrafish) (Carbofuran과 Chiorothalonil의 공존이 Brachydanio rerio(zebrafish)를 이용한 단기간 생물농축계수의 측정에 미치는 영향)

  • This study was performed to investigate the effect of co-existence of carbofuran and chlorothalonil on the short-term bioconcentration factor in Brachydanio rerio(zebrafish). The fishes were exposed to the single and combined treatment of carbofuran and chlorothalonil for 1, 3 and 5 days. Experimental concentrations of carbofuran were 0.05 and 0.10 ppm under the single treatment. And those of chlorothalonil were 0.005 and 0.010 ppm. Experimental concentrations of the combined treatment of carbofuran and chlorothalonil were 0.05 ppm+0.005 ppm, 0.05 ppm+0.010 ppm, 0.10 ppm+0.005 ppm for 1, 3 and 5 days, respectively. Carbofuran and chlorothalonil in fish and in test water were extracted with n-hexane and acetonitrile. GC-ECD was used to detect and quantitate carbofuran and chlorothalonil. 1-day, 3-day and 5-day bioconcentration factors(BCF$_1$, BCF$_3$ and BCF$_5$) of each pesticide were obtained from the quantitation results. The depuration rate of each pesticide was determined over the 24-h period after combined treatment. The results were as follows: Carbofuran did not bioaccumulate in zebrafish under the single and combined treatment for testing periods. BCF$_1$ values of chlorothalonil in concentration of 0.005 and 0.010 ppm under the single treatment were 0.508, 0.621, BCF$_3$ were 1.327, 1.511 and BCF$_5$ were 1.331, 1.597, respectively. BCF$_1$ values of chlorothalonil were 0.512, 0.520 and 0.619, respectively, when the concentration of carbofuran and chlorothalonil in combined treatment were 0.05+0.005, 0.05+0.010 and 0.10+0.005 ppm. BCF$_3$ values of chlorothalonil 1.341, 1.338 and 1.513, respectively, and BCF$_5$ values of chlorothalonil were 1.332, 1.327 and 1.521, respectively, under the above combined treatment. Depuration rate constants of chlorothalonil in concentration of 0.005 and 0.010 ppm under the single treatment were 0.011 and 0.012. Depuration rate constants of chlorothalonil were 0.011, 0.010 and 0.011, when the concentration of carbofuran and chlorothalonil in combined treatment were 0.05+0.005, 0.05+0.010 and 0.10+0.005 ppm. It was observed that no significant difference of carbofuran and chlorothalonil concentration in fish extracts, test water, BCFs and depuration rate constants of carbofuran and chlorothalonil between combined treatment and single treatment. It was considered that no appreciable interaction at experimental concentrations due to lower concentrations than LC$_{50}$. It is suggested that the difference of BCFs between carbofuran and chlorothalonil due to those of fat composition of fish and solubility of carbofuran and chlorothaionil.

Effect of Cultural Conditions on Polysaccharide Production and its Physicochemical Properties in Cordyceps militaris

  • This study was concerned with the optimization of liquid culture conditions for mycelial growth and polysaccharide production and its physicochemical properties in Cordyceps militaris. The one factor at a time method was adopted to investigate the effects of medium composition, environmental factors and C/N ratio. Among the these varialbles, glucose 80g/L, yeast extract 10g/L, $MgSO_4{\cdot}7H_{2}O\;0.5g/L$, $KH_{2}PO_4\;0.5g/L$ were proved to be the most suitable carbon, nitrogen, and mineral sources, respectively. The optimal temperature, initial pH, working volume were identified to be $24^{\circ}C$, 7.0 and 100ml, respectively. Under the optimal conditions, the strategies in shake flask culture and 5L jar fermentor led to mycelial growth of 29.43 g/L, 28.88g/L and polysaccharide production of 2.53g/L, 6.38 g/L, respectively. Among the phisicochemical properties, relative concentrations(w/v) of total sugar, uronic acid, protein and hexoseamine were identified to be 74.07%, 1.13%, 0.91%, and 0.46%, respectively. The fraction of neutral and acidic polysaccharide were identified to be 81.9% and 18.1%, respectively.

Inhibitory Activity of Bumblebee Worker (Bombus terrestris L.) Venoms on Nitric Oxide, TNF-${\alpha}$ and IL-6 Production in Lipopolysaccharide-Activated Macrophages

  • To elucidate the composition of bumblebee (Bomb us terrestris) venom (BBV) and the anti-inflammatory effect of BBV. The major components of BBV by LC chromatography and SDS-PAGE were identified. The production of nitric oxide (NO) and proinflammatory cytokines was examined by lipopolysaccharide (LPS) in a macrophage cell line, RAW 264.7 cells, with BBV. BBV inhibits LPS-induced NO in a dose dependent manner. We also found that BBV inhibits proinflammatory cytokine, tumor necrosis factor (TNF)-${\alpha}$ and interleukin (IL)-6 production. These findings mean that BBV can be used in controlling macrophages mediated inflammation related disease. Additional studies on the pharmacological aspects of the individual components of BBV are recommended for future trials.

Performance of a submerged membrane bioreactor for wastewater mimicking fish meal processing effluent

  • The objective of this work was to analyze organic matter removal, nitrification, biomass growth and membrane fouling in a submerged flat-sheet membrane bioreactor, fed with synthetic wastewater, of similar composition to the effluents generated in a fish meal industry. After biomass acclimatization with saline conditions of 12 gNaCl/L and COD/N ratio of 15 in the bioreactor, results showed that the organic matter removal was higher than 90%, for all organic loading rates (0.8, 1, 1.33 and $2gCOD/L{\cdot}d$) and nitrogen loading rates (0.053, 0.067, 0.089 and $0.133gN/L{\cdot}d$) tested during the study. However, nitrification was only carried out with the lowest OLR ($0.8gCOD/L{\cdot}d$) and NLR ($0.053gN/L{\cdot}d$). An excessive concentration of organic matter in the wastewater appears as a limiting factor to this process' operating conditions, where nitrification values of 65% were reached, including nitrogen assimilation to produce biomass. The analysis of membrane fouling showed that the bio-cake formation at the membrane surface is the most impacting mechanism responsible of this phenomenon and it was demonstrated that organic and nitrogen loading rates variations affected membrane fouling rate.
