Numerical Study on the Flow Field about Multi-element Airfoils and the Effect of the Lift-enhancing Tabs (다중-익형 주위 유동장 및 양력-향상 탭의 영향에 대한 수치적 연구)

  • The flow fields over multi-element airfoils with lift-enhancing flat-plate tabs were numerically investigated. Common choice of the height of the lift-enhancing tabs usually ranges from 0.25% to 1.25% of the reference airfoil chord, and in this study the effect of the position of the tab with l%-chord height was studied by varying the distance of the tab from the trailing edge ranging from 0.5% to 2% of the reference chord. In this paper, the effects of lift-enhancing tabs with various position were studied at a constant Reynolds number on a two-element airfoil with a slotted flap. Computed streamlines show that the additional turning caused by the tab reduces the amount of separated flow on the flap.

Memory Enhancing and Antioxidant Properties of Fermented Chongmyung-tang

  • The Chongmyung-tang (CMT) has been used as an oriental herbal medicine for the purpose of enhanced learning and memory. Recently, since fermentation may give a positive effect on pharmacological actions of herbal medicine, many studies are focused to find fermented medicinal herbs with improved bioactivity. In the present study, memory enhancing, antioxidant and reducing power activity of CMT and fermented CMT with Aspergillus oryzae (FCMT-A) or Saccharomyces cerevisiae (FCMT-S) were determined. To evaluate the memory enhancing activities of CMT, FCMT-A and FCMT-S, we performed passive avoidance test using scopolamine induced amnesia model. Administration of CMT, FCMT-A and FCMT-S showed a significant memory enhancing effect about 72.5, 78.3, 71.8% of the normal group respectively. CMT, FCMT-A and FCMT-S also exhibited strong $DPPH{\bullet}$, ${\bullet}{O_2}^-$, $NO^{\codt}$, $ABTS^{{\cdot}+}$ scavenging activities and reducing ower. It was also found that fermented CMT has slightly higher scavenging activities on $DPPH{\bullet}$, $ABTS^{{\cdot}+}$ radicals compared to CMT. These results revealed that CMT, FCMT-A and FCMT-S had memory enhancing and radical scavenging activities. In addition, the fermentation of CMT was more or less important for elevated memory enhancing and antioxidant activities of CMT.

Cognitive Enhancing Activity of the Steamed and Fermented Extracts of Codonopsis lanceolata Radix (양유(洋乳)의 증숙 및 발효 추출물의 인지능 개선 활성)

  • This study was designed to determine and compare the cognitive enhancing effect of various Codonopsis lanceolata extracts by steaming, fermentation and high hydrostatic pressure process. We prepared water extract of C. lanceolata, steamed C. lanceolata, steamed and fermented C. lanceolata and C.lanceolata by high hydrostatic pressure process and fermentation. Cognitive enhancing effect of extracts was evaluated in scopolamine-induced memory impairment mice using by passive avoidance test and Morris water maze tests. MTT assay was conducted to investigate neuroprotective effect on glutamate induced cell death in HT22 cells. Steamed and fermented C. lanceolata water extract decreased escape latency in Morris water maze test and increased the latency time of the passive avoidance test compared to other extracts. Furthermore, the steamed and fermented C. lanceolata water extract showed neuroprotective effect. These results suggest that steaming and fermentation process more improve cognitive enhancing effect of C. lanceolata than other process.

Antibacterial and Antibiotic Activity Enhancing Effect of Extract and Fractions from the Root of Rumex japonicus Houtt (참소리쟁이 뿌리 추출물 및 분획의 항균 활성과 항생제 증강 활성)

  • Background: The objective of this study were to determine the antibacterial activity and antibiotic activity-enhancing effect of 70% ethanol extract of the root of Rumex japonicus Houtt. and its fractions when used in combination with gentamicin against aerobic skin flora. Methods and Results: The antibacterial activity and antibiotic (gentamicin) activity enhancing effect against aerobic skin flora were determined using the disc diffusion assay. Chloroform fraction (CF) and ethyl acetate fraction (EF) showed higher activities against Staphylococcus aureus and Staphylococcus epidermidis than those shown by other fractions. Regarding the antibiotic (gentamicin) activity-enhancing effect against aerobic skin flora, the n-hexane fraction (HF) and CF showed strong activity. The combination of HF and CF with gentamicin was evaluated using the broth dilution assay to determine the inhibitory effect on the growth of aerobic skin flora. The combination of CF with gentamicin exhibited the highest inhibitory effect on the growth of S. aureus and S. epdermidis. MTT assay performed to determine the viability of L929 cells revealed that EF treatment resulted in viability of 33.96 - 116.76% at the tested concentration. The combination of 70% ethanol extract and its other fractions with gentamicin showed low cell toxicity. Conclusions: Appropriate use of antimicrobial agents is important prior to the development of new antibiotics. The 70% ethanol extract of the root of R. japonicus Houtt. and its fractions showed significant synergism with gentamicin when used in combination against S. aureus and S. epdermidis. Thus, R. japonicus Houtt. could be used as a functional materials in antimicrobial-related fields.

Immune Enhancing Effect of Boummyunyuck-dan (보음면역단의 면역 증강 효과)

  • Objective : To investigate immune enhancing effects of Boummyunyuck-dan (BMD) Methods : In this study I investigated the effect of BMD on cell proliferation and viability. In addition, I investigated production of cytokines (IL-2, IL-4 and $IFN-{\gamma}$), NO, and $TNF-{\alpha}$ in human T-cell leukemia, MOLT-4 cells. The cells were cultured for 24h in the presence or absence of BMD. Result : BMD increased the cell viability by 15% (P<0.05) and enhanced IL-2, IL-4 and $IFN-{\gamma}$ production compared with media control in a dose-dependent manner (P<0.01) at 24h. BMD also increased mRNA and protein expression levels of $IFN-{\gamma}$ in MOLT-4 cells. In addition, I also assessed the effects of BMD on production of NO and $TNF-{\alpha}$ from the peritoneal macrophages because NO and $TNF-{\alpha}$ as a potent macrophage-derived immune reaction regulatory molecule has received increasing attention. However, BMD had no effect on NO and $TNF-{\alpha}$ production in the cells. Conclusion : These data indicate that BMD has some immune-enhancing effect, and that its action may be due to the proliferation and cytokine production of T cells.

The effect on gene expression profile of rat hippocampus caused by administration of memory enhancing herbal extract (육미지황탕가미방(六味地黃湯加味方)이 흰쥐의 기억능력과 중추신경계 유전자 발현에 미치는 영향)

  • The herbal extract (YMT_02) is a modified herbal extracts from Yukmijihwangtang (YMJ) to promote memory-enhancing. The YMJ extracts has been widely used as an anti-aging herbal medicine for hundred years in Asian countries. The purpose of this study is to; 1) quantitatively evaluate the memory-enhancing effect of YMT_02 by hehavior task, 2) identify candidate genes responsible for enhancing memory by cDNA microarray and 3) assess the anti-oxidant effect of YMT_02 on PC12 cell. Memory retention abilities are addressed by passive avoidance task with Sprague-Dawley (SD) male rat. Before the training session, the rats are subdivided into four groups and administrated with YMT_02, Ginkgo biloba, Soya lecithin and normal saline for 10 days. The retention test was performed. 24 hours after the training session. The retention time of the YMT_02 group was significantly (p<0.05) delayed $({\sim}100%)$, whereas Ginkgo biloba and Soya lecithin treatment delayed 20% and 10% respectively. The hippocampi of YMT_02 and control group were dissected and mRNA was further purified. After synthesizing cDNA using oligo-dT primer, the cDNA were applied and mRNA was further purified. After synthesizing cDNA using oligo-dT primer, the cDNA were applied to Incyte rat GEMTM 2 cDNA microarray. The microarray results show that prealbumin(transthyretin), phosphotidy lethanolamine N-methyltransferase, and PEP-19 are expressed abundantly in the YMT_02 treated group. Especially, PEP-19 is a neuron-specific protein, which inhibits apoptotic processes in neuronal cell. On the other hand, transcripts of RAB15, glutamate receptor subunit 2 and CDK 108 are abundant in control group. Besides, neuronal genes involved in neuronal death or neurodegeneration such as neuronal-pentraxin and spectrin are abundantly expressed in control group. Additionally, the YMT_02 shows an anti oxidative effect in the PC12 cell. The list of differentially expressed genes may implicate further insight on the action and mechanism behind the memory-enhancing effect of herbal extracts YMT_02, for example, anti-apoptotic, anti-oxidative, and neuroprotective effects.

  • Protaetia Brevitarsis Seulensis(white grub) has been traditionally used as medicinal stuff to treat blood stasis, occlusion of menstruation, tetanus and liver cancer in Asian countries (Korea, Japan, China, Taiwan, India and Myanmar). Especially, Donguibogam, which is traditional korean medicinal book, described the white grub as traditional medicine to treat hepatic diseases and vascular disorders. The white grub has been considered as highly nutritional food. The major constituents of white grub are rich in protein, healthy fats, iron, calcium. Recent studies announced that white grub has hepatoprotective effect and anti-microbacterial effect. However, the immuno-enhancing effect of white grub extracts in RAW 264.7 macrophage cells has not been studied yet. In this study, the various concentrations of white grub extract were examined to find immuno-enhancing effects on RAW 264.7 cells. Cytotoxicity was determined by MTT assay and immuno-enhancing effect of white grub extract was investigated by measuring nitric oxide (NO) production compared with only lipopolysaccharide (LPS) treatment. White grub extracts (0.001 - 10 mg/ml) did not show cytotoxicity. Additionally, white grub extracts (0.001 - 1mg/ml) had Immuno-enhancing effect on RAW 264.7 cells compared with only LPS treated group. These results might be provided proof to develop beneficial immuno-enhancing material for human health.

  • Absorption enhancing effect of 5 MSA-Na at the mucous memberanes of the rectum and duodenum was studied via in vitro sac test. There were no significant differences between two membranes. Effect of 5MSA-Na on the transfer of CMZ to the tissue was also studied via in situ loop method. There was no significant difference at the duodenum, compared to the control when CMZ was administered intravenously. Transfer of CMZ to the loop of the rectum was increased in the presence of 5MSA-Na compared to the control, which might be attributed to the enhanced permeability of the mucous memberane of the rectum.

Antibacterial Activity and Enhancing Antibiotic Effect of Extract and Fractions from Curcuma longa against MRSA Strain (MRSA에 대한 울금 추출 및 분획물의 항균활성과 항생제 증강 효과)

  • Curcumin content of butanol fraction from C. longa was found to be 22.4942% of the highest content. However, in DPPH radical scavenging ability and antibacterial activity against methicillin resistance Staphylococcus aureus(MRSA, CCARM3696), ethylacetate fraction contained 2.5791% of curcumin was exhibited highest activity. In comparison of enhancing antibiotic(ampicillin) effect against MRSA, ethanol extract contained 1.7838% of curcumin showed more strong activity. This indicates that the ethanol extract and some fractions from C. longa can have antibacterial activity and enhancing antibiotic effect possibly without curcumin. Appropriate use of antimicrobial agent was important point prior to the development of new antibiotics. And in that sence, extract and fractions of C. longa were worth using as synergist of antibiotics and natural antimicrobial agent.

Studies on the Enhancing Effect of Polymyxin B on the Antibodies Response of Enterobacterial Antigens (Pomyxin B의 장계세균항원(腸系細菌抗原)에 대(對)한 항체산생(抗體産生) 증강작용(增强作用)에 관(關)한 연구(硏究))

  • Various kinds of antibiotics are generally believed to have inhibitory effects on the antibody response. However, as polymyxin B which belongs to the cyclic polypeptide group of antibiotic was found to have some enhancing effects on the antibody response of rabbits to enterobacterial common antigen(CA) under specified conditions, experiments were carried out on this problem with the following results. 1. When mixture of polymyxin B and CA derived from Salmonella typhimurium(STM) was treated 30 minutes at $37^{\circ}C$ and injected three times into rabbits by intravenous route, the antibody response to CA was weaker than rabbits injected CA only. 2. Mixture of polymyxin B and CA showed a marked antibody production when injected into rabbits primed with small amounts of heat-extracted antigen of STM, while the injection of CA alone showed low titers of response. 3. Mixture of polymyxin B and heat-extracted CA-containing antigen of Escherichia coli 014 also showed a increased antibody production than CA alone in rabbits primed with antigen of STM. 4. The effect of polymyxin B appeared in different ways. This antibiotic did not enhance the CA antibody response in rabbits primed with small amounts of E. coli 0111 and 055, but enhance in rabbits primed with Shigella flexneri. 5. No enhancing effect on the antibody response was observed by polymyxin B in rabbits primed with CA. 6. No enhancing effect on the antibody response was also noted in rabbits primed with STM antigen in case polymyxin B and CA were administered simultaneously but in veins of different places. 7. Bacitracin did not enhance the CA antibody response in primed rabbits with STM antigen, but neomycin slightly enhance the response. 8. Lipopolysaccharide showed no priming effect on the CA antibody response, and no enhancement of the CA antibody response in rabbits printed with STM. 9. The priming effect of STM antigen against CA antibody response was very weak as compared with the effect of CA derived from STM antigen.
