Optimization of the Production of a Thermostable Antifungal Antibiotic (내열성 항곰팡이 항생물질의 생산 최적화)

  • The optimum conditions for the production of an antifungal antibiotic from Bacillus sp. YJ-63 were investigated. The oprimumized medium consisted of 1.5% soluble starch, 1% tryptone and 0.5% yeast extract, and temperature and initial medium pH for production were optimal at 35$^{\circ}C$ and pH 6.0, respectively. Production yield was significantly improved by shaking culture using 50 ml medium in 500 ml flasks. Under these conditions, the production of the antifungal antibiotic was growth-dependent, from 35hrs into cultivation to the stationary phase and endospore formation.

Comparison of Bacterial Numbers and Treatment Efficiencies in Bioreactors of Various Advanced Wastewater Treatment Processes (다양한 고도폐수처리공정에서의 생물반응조 세균수와 처리효율과의 비교)

  • Bacterial numbers, such as endospore-formers, and treatment efficiencies were investigated for Rotating Activated Bacillus Contactors (RABC) and other advanced wastewater treatment processes including anaerobic-anoxic-oxic (A2O), sequencing batch reactor (SBR) and biological aerated filter (BAF). Endospore-forming bacterial numbers in the RABC showed 129-fold higher levels than those of the existing advanced systems. RABC process demonstrated the highest bacterial numbers in its bioreactors (paired t-test, p<0.01). RBC biofilms and aeration tanks of the RABC system showed 131- and 476-fold higher than other existing advanced processes, respectively. Mean treatment efficiencies of the existing systems were 83.5% for chemical oxygen demand (COD), 59.1% for total nitrogen (TN) and 76.8% for total phosphorus (TP). However, RABC process removed 96.9% for COD, 96.9% for TN and 91.9% for TP for highly concentrated food wastewater (COD>1,500 mg/L, TN>150 mg/L, TP>50 mg/L). Treatment efficiency was significantly reduced when the numbers of Bacillus genus in the bioreactors decreased below $10^6CFU/mL$. The automated RABC (A-RABC), in which dissolved oxygen concentrations are automatically controlled, showed higher treatment efficiencies compared to the RABC process. The RABC system maintained sufficient bacterial numbers for the effective treatment of highly concentrated food wastewater. Moreover, final effluent was in agreement to water quality standards.

Structure and Regulation of a Complex Promoter Region from an Alkali-tolerent Bacillus sp.

  • A DNA fragment from an alkali-tolerent Bacillus sp., conferring strong promoter activity, was subcloned into the promoter probe plasmid pPL703 and the nucleotide sequence of this promoter region was determined. The sequence analysis suggested that this highly efficient promoter region containing the complex clustered promoters comprised three kinds of promoters (P1, P2 and P3), which are transcribed by $\sigma^B (formerly \sigma^{37}), \sigma^E(formerly \sigma^{29}) and \sigma^A (formerly \sigma^{43})$ RNA polymerase holoenzymes which play major rules at the onset of endospore formation, during sporulation and at the vegetative phase of growth, respectively. S1 nuclease mapping experiments showed that all three promoters had staggered transcription initiation points. The results of chloramphenicol acetyltransferase assay after the subcloning experiments also indicated that the expression of these clustered promoters was correlated with the programs of growth and endospore development. Promoter P1, P2 and P3 were preceded by 75% AT, 79% AT and 81% AT regions, respectively, and a partial deletion of AT-rich region prevented transcription from promoter P1 in vivo. Two sets of 5 -AGTGTT-3 sequences and inverted repeat sequences located around the promoter P1 were speculated as the possible cis acting sites for the catabolite repression in B. subtilis. In vivo transcripts from these sequence regions may be able to form a secondary structure, however, the possibility that a regulatory protein induced by the excess amount of glucose could be bound to such a domain for crucial action remains to be determined.

  • In this study, basic research was conducted to secure microbial resources applicable to autonomous crack healing concrete. To this end, in this experiment, biomineral-forming microorganisms were separated from natural sources, and the ability of survival in cement and calcium carbonate precipitation were compared to secure suitable microbial resources. Bacillus-type bacteria forming endospores were isolated from the sample, and the amount of calcium carbonate produced by the six microorganisms identified by 16S rRNA sequencing was compared. Two types of microorganisms, Bacillus velezensis and Bacillus subtilis, with the highest calcium carbonate precipitation were selected, and the survival of the microorganisms was confirmed through phase contrast microscopy after being cured after being added to the mortar. In addition, it was confirmed that the autonomous crack healing capability by the crack healing material produced by microorganisms was confirmed by artificially generating cracks in the mortar.

Isolation and Identification of a Bacteriolytic Enzyme-producing Bacterial Strain from Pusan Coastal Sea (해양에서 용균효소를 분비하는 균주의 분리와 동정)

  • In order to produce the bacteriolytic enzyme, bacterial strains capable of excreting a large amount of the enzyme were screened from the coastal sea water samples in Pusan. The bacterial strain SH-1, which showed the highest activity among 43 bacteriolytic enzyme producing bacteria, was finally selected for further studies. The strain SH-1 was an endospore-forming grampositive rod, and the position of spore was paracentral. These morphological characteristics assigned the isolated strain to the morphological group I classified by Gordon. The fatty acid composition of the bacterial stain was analyzed to be consisted of branched chains of iso-Cn and anteiso-Cn. Based on the percent content of the branched chain (93.85%), the isolates could be identified as a species of Bacillus. According to the experimental results of the API system (API 50CHB & API 20E) the strain was identified as Bacillus subtilis. Numerical texonomy, in which 82 major characters were examined using several species of Bacillus as the standard bacteria, indicated that the strain SH-1 showed 90% similarity to Bacillus subtilis. Thus, the isolated strain SH-1 could be identified as Bacillus subtilis.

  • Obligate bacterial parasite attacking Heterodera glycines was firstly found from Chungju soybean field in Korea. Diameters of sporangium and central body were 5.6 ${\mu}{\textrm}{m}$ and 1.9 ${\mu}{\textrm}{m}$ under light microscopy (LM), and 3.9 ${\mu}{\textrm}{m}$ and 1.8 ${\mu}{\textrm}{m}$ under transmission electron microscopy (TEM). Endospore showed cup-shaped with smooth-type exosporium without peg-like thickening in polar area under SEM and TEM. Bacteria completed its life cycle in female of soybean cyst nematode after adhering on cuticle of second-stage juvenile. From these results, the Pasteuria found from Chungju was identified with P. nishizawae.

Levan-Producing Bacillus subtilis BS 62 and Its Phylogeny Based on Its 16S rDNA Sequence

  • A viscous substance producer strain BS62, which was isolated from conventional Chungkookjang, was examined for its productivity of levansucrase and levan during soybean fermentation at $37{\circ}C$. After one day of cultivation, the enzyme activity reached the highest level, 8 units $ml^{-1}$. Extracts of fermented soybeans were precipitated by ethanol and hydrolyzed by either 0.1 N HCl or invertase, and the hydrolyzates were analyzed using thin layer and ion chromatographies. Fructose was the only sugar detected. This suggest that fructose was derived from the levan produced by the strain BS62 during soybean fermentation. The aerobic, endospore-forming bacterium BS62 was identified as a Bacillus subtilis sp., based on the composition of its cellular fatty acids and phylogeny, which was determined by its 16S rDNA sequence.

  • To utilize hemicellulosic biomass efficiently, the microorganism producing xylanase was isolated from fermented sawdust. It was a Gram positive, aerobic and rod shape bacterium. It had endospore and secreted strong hydrolases, such as amylase and protease. Through morphological, cultural and physiological tests, it was identified as Bacillus sp. GS. To increase the productivity of xylanase from Bacillus sp. GS, the enzyme production medium was optimized. The composition of the medium and incubation conditions were like follows xylan 1.25%, yeast extract 0.1%, NaN030.2%, K2HP04 0.1%, MgSO4 0.02%, mineral salt 0.005%, pH 6.5, incubation temperature 37$^{\circ}C$.

  • A bactirium prohibiting the growth of fungus Botrytis cineria KT 433 was isolate and identified from soil. The isolated strain was gram positive, aerobic bacteria with cream color, round and mucoid. It showed ord form of 0.45$\times$1.1 ${\mu}{\textrm}{m}$ at the cultivated for 24 hrs and the ellipsoided endospore wer observed after culting for 72 hrs. The optimum growth temperature and pH wer 35$^{\circ}C$ and pH 5.0~8.0, respectively. It could assimilate daxtrin, maltose, glucose, mannose, ribose and collobiose as a sole carbon source. The isolated was confirmed to be a Bacillus sp. strain from the findings. The antibiotic from the isolated strain was stable up to 121$^{\circ}C$. The strain, especially, showed specific activity for mold and yeast. However, there was not significant antibacterial activity.

Effects of Bacillus and Endospore Germinations on Organic Matter Removal (Bacillus와 내생포자 발아가 유기물 제거에 미치는 효과)

  • The Bio Best Bacillus(B3) and Rotating Activated Bacillus Contactor(RABC) processes, in which Bacillus strains are predominating, are reported to remove nitrogen and phosphorus as well as organic matter effectively. Nevertheless the nutrient removal characteristics of the Bacillus strains have not been studied in detail so far. This study investigated the organic and nutrient removal by Bacillus strains, Bacillus megaterium(KCTC 3007), Paenibacillus polymyxa(KCTC 3627), and Bacillus sp. A12, C21, F12, and L1(isolated from a B3 process), by incubating the strains in 0.2% nutrient broth at $30^{\circ}C$. Burkholderia cepacia(KCTC 2966), a common activated sludge organism, was used as a reference species for comparison. Although the degradation rate was affected by the population sire, the specific removal rates of organic matter by Bacillus strains were greater by $2\sim5$ times than that of Burkholderia. In particular, the culture bottles inoculated with the endospores of Bacillus megaterium and Bacillus sp. C21, F12, and N12 showed significantly higher degradation rate than those of vegetative cells.
