A Comparative Study on the Ultrastructures of the Egg Envelope in Fertilized Eggs of Fishes, Characidae, Three Species (카라신과 어류 3종의 수정란 난막 미세구조에 대한 비교 연구)

  • The structures of the egg envelope in fertilized eggs of three species of characidae, head and tail light fish (Hemigrammus ocellifer), black tetra (Gymnocorymbus ternetzi), and buenos aires tetra (Hemigrammus caudovittatus) were studied using light and electron microscopes. The fertilized eggs in all species were colorless, transparent, spherical and non-floted type. The egg envelopes have a single micropyle resembling the pathway of sperm in the area of the animal pole. The micropyle was surrounded by protruded lines of the egg envelope in a radiated form. Egg envelopes of fertilized eggs in both head and tail light fish and buenos aires tetra consisted of three distinct layers; an outer layer, a middle layer and an inner layer. And that of blacktetra consisted of two layers; an outer layer and an inner layer. Also, an outer layers of both head and tail light fish and black tetra were adhesive types but, in that of buenous aires tetra was non-adhesive type. An outer surface of egg envelope in black tetra was arranged by pores regularly. In that of head and tail light fish and buenos aires tetra have a rough side. An inner layer of egg envelope in fertilized eggs consisted of lamellae alternating with interlamellae of lower electron density; an inner layer of fertilized eggs in head and tail light fish consisted of three layers, that of black tetra was four layers, and that of buenos aires tetra was five layers.

  • The structures of the fertilized egg envelope in three species of cichlidae, golden severum (Cichlasoma severum var.), convic cichlid (Cichlasoma nigrofasciatum) and discus (Symphysodon aequifasciatus) were investigated by routine light and electron microscopies. The fertilized eggs of all three species were of the non-transparent, ellipsoidal, adhesive and non-floted type. The egg envelopes have a single micropyle, which is thought to the pathway of sperm in the area of the animal pole. In golden severum, an outer surface of egg envelope was covered by a reticular layer, that of convic cichlid was covered by an amorphous reticular layer and that of discus was covered by a branched reticular layer. The fertilized egg envelopes consisted of two distinct layers, an adhesive outer layer and an inner layer, consisted of lamellae alternating with interlamellae of lower electron density, in all three species; an inner layer of golden severum was $15\sim17$ layers, that of convic cichlid was $14\sim16$ layers, and that of discus was $18\sim19$ layers.

  • The structures of the fertilizedegg envelope in threespecies of cyprinidae, leoparddanio (Brachydanio frankei), cherry barb (Barbustitteya) and white cloudmountain fish (Tanichthys albonubes) were investigated by light and electron microscopy. The fertilized eggs of all three species have colorless, transparent, spherical and non-floted type. The egg envelopes have a single micropylewhich is thought to the pathway of sperm in the area of the animal pole. Also, an outer layersof both cherry barband white cloud mountain fish areadhesive types, but in that of leoparddanio has non-adhesivetype. In leopard danio, an outer surface ofegg envelope is covered by mushroom-likestructure, that of cherry barb is arranged by button-like structure, and that of white cloud mountain fish isarranged by rod-likestructure. Inboth leopard danioand cherrybarb, the eggenvelopes consist of three distinct layers; an outer layer, a middle layer and an inner layer, and that of white cloud mountain fish hastwo layers. An innerlayer of egg envelopein leopard danio consist of 10 layers, cherry barb has 8 layers, and white cloud mountain fish has $5\sim6$ layers.

  • Ultrastructure of fertilized egg envelope in the tomato clown anemonefish (Amphiprion freatus) was examined by mean of light, scanning and transmission electron microscopies. The fertilized egg of tomato clown anemonefish was of non-transparent, ellipsoidal, adhesive and non-floted type, and there were numerous oil droplets in the yolk sac and adhesive filaments in the area of the animal pole. The outer surface of egg envelope was smooth. The fertilized egg envelope consisted of two distinct layers; a non-adhesive outer electron-dense layer and an inner layer, consisting of five horizontal low electron-dense lamellae alternating with the middle electron dense interlamellae.

  • The ultrastructure of the fertilized egg envelope from pale chub , Zacco platypus belong to Cyprinidae was studied using scanning and transmission electron microscopes. The fertilized egg was an adhesive type, have a single micropyle resembling the pathway of sperm in the area of the animal pole. The micropyle was surrounded by 5 peaks of hill. An outer surface of the fertilized egg envelope was arranged by pore canals irregularly. In section of fertilized egg, the egg envelope consistes of three layers, an outer adhesive layer, a middle electron dense layer and an inner lamellae layer consisting of 9 layers. These ultrastructural characters of fertilized egg envelope from pale chub can be utilized in taxonomy of teleost.

  • Ultrastructure of gametes in the three-spine stickleback, Gasterosteus aculeatus aculeatus was observed, utilizing light, scanning and transmission electron microscopes. The egg of three-spine stickleback is spherical and demersal type. The eggs are highly adhesived to each other but not to substrates. There are many oil droplets in vitelline membrane. The outer surface of egg envelope is arranged by mushroom-like structures and pore canals. The egg have a micropyle, sperm entry site, in the area of the animal pole. The egg envelope consists of three layers, an outer layer with high electron density, a middle layer consisting two layers and an inner layer consisting of 16 to 20 layers. In the fertilized egg envelope, the molecular weights of these components ranged from 14 kDa to 205 kDa. The molecular weights of nam protein bands are 19.4 kDa, 36.7 KDa, 39.4 kDa, 42.9 kDa, 46.1 kDa and 53.0 kDa. The head of spermatozoa is spherical shape and the acrosome is absent. The mitochondria in midpiece are arranged from one to three layers and separated from the axoneme by the cytoplasmic canal. The tail has two lateral fins and the axoneme is of the 9+2 structure.

  • The oogenesis and ultrastructure of fertilized egg envelope of false dace were investigated by light and electron microscope. The cytoplasm of false dace oogonia was basophilic and many nucleoli were located at inner side of nuclear membrane. In primary oocytes, yolk vesicles were distributed in marginal area only and egg envelope was not formed on egg outside. In secondary oocyte, the egg envelope was formed and yolk vesicles were increased than that of early stage in cytoplasm. The amount of basophilic substance was decreased. In case of matured egg, thickness of egg envelope and site of egg were increased, basophilic substance was distributed in egg envelope around only. The yolk vesicles were changed to yolk mass in accordance with development. The fertilized egg was of ellipsoidal, adhesive type and yellowish, have a single micropyle in the area of the animal pole. The fertilized egg envelope consisted of three layers, an outer adhesive layer, a middle layer consisting of 6 lamellae alternating layers and an inner electron dense layer. An outer surface of the fertilized egg envelope was arranged by adhesive fibrous structures. In conclusion, it is summarized that the oogenesis of false dace were the increase of cell size, the formation and accumulation of yolk, and decrease of basophilic intensity in cytoplasm. These ultrastructural characteristics of fertilized egg envelope from false dace can be utilized in taxonomy of teleost.

Effects of Stocking Density on Performance and Physiological Responses of Egg-type Breeder Laying Hens in Cages (케이지 내 사육밀도가 산란종계의 생산성 및 생리적 반응에 미치는 영향)

  • This study was conducted to determine the effects of stocking density on performance and physiological responses of egg-type breeder layers in cages. A total of 264 Hy-Line Brown egg-type breeder layers at the age of 32 weeks were divided into 4 groups with 3 replicates of 22 birds (20 females, 2 males) per replicate and assigned to four stocking density treatments with 22 birds per cage with different sizes (980, 735, 640 and 560 $\textrm{cm}^2$/bird). The birds were fed the same experimental diet ad libitum for 12 weeks, and laying performance, egg and eggshell qualities, fertility and hatchability were determined. At the end of the experimental period, 9 birds were selected in each treatment in order to measure the scores of plumage condition. Antibody titers to Newcastle disease and infectious bronchitis were also determined. No significant differences were observed in laying performances, eggshell strength, Haugh unit, fertility and hatchability among the treatments. In birds housed 640 $\textrm{cm}^2$ per bird, eggshell thickness were significantly lower (P<0.05) than those of the other groups from 7 to 12 weeks. There was no significant difference in antibody titers to Newcastle disease. In birds housed 980 $\textrm{cm}^2$ per bird, antibody titers to infectious bronchitis were significantly (P<0.05) higher than those of the other groups. With increase in the level of stocking density, the scores of plumage condition were also significantly decreased (P<0.01). This study demonstrated that moderate increment of stocking density did not resulted in reduction in laying-performances of egg-type breeder layers. However, it seemed that the higher stocking density might give more stress to the birds from physical stimuli and thus were harmful environment causing higher mortalities compared to the lower ones.

Dietary Self-selection and Nutrient Feeding Systems for Egg-type Growing Pullets and Layers (난용계의 선택채식과 영양소 공급체계)

  • From the observations of dietary self-selection by growing pullets, step-up protein or reverse protein and single-stage low protein pullet feeding systems were developed. They offered another pullet feeding concept that appears to control the body weight effectively and to reduce the consumption of feed and nutrients without impairment of subsequent laying performance. It is obvious from the feed and nutrient consumption pattern of layers fed diets for self-selection of energy, protein and calcium that they have a daily cyclic requirement rather than a constant requirement for nutrients. It seems that a practical self-selective feeding system is needed to meet the daily cyclic requirement for nutrients without consuming an excess of energy and protein at certain times of the day as compared to the complete or single diet where layers have to consume extra energy and protein in the afternoon when they have a specific appetite mainly for calcium.
