To compare the efficacy of Stephaniae Tetrandrae Rhizoma and Sinomenii Caulis which are medicinal plants used as Bang Kee(防己), the plants were combined with Astragali Radix and Lycopii Herba into 6 different medicinal decoctions. The specimen decoctions prepared were each decoctions of Stephaniae Tetrandrae Rhizoma(St), Sinomenii Caulis(Si), Stephaniae Tetrandrae Rhizoma-Astragali Radix(StA), Stephaniae Tetrandrae Rhizoma-Lycopii Herba(StL), Sinomenii Caulis-Astragali Radix(SiA) and Sinomenii Caulis-Lycopii Herba(SiL). They were dosed to rats with induced hyperlipidemia. In the groups of rats with hyperlipidemia induced by high cholesterol diet, in the group which St was dosed to, there was no significant effect on the level of Tc, TG, HDLc. And in the group which Si was dosed to, the level of Tc, TG dropped and that of HDLc rose. In the groups of rats with hyperlipidemia induced by high cholesterol diet, in the group which StA was dosed to, there was no significant change in the level of Tc, TG, or HDLc. But in the group which SiA was dosed to, the level of Tc and TG dropped and that of HDLc rose. In the groups of rats with hyperlipidemia induced by high cholesterol diet, in the group which StL was dosed to, the level of TG dropped, but there was no significant change in the level of Tc or HDLc. And in the SiL group, there was no significant change in the level of Tc or TG, but the level of HDLc rose. On diseases such as hyperlipidemia, when using drug couple of Bang Kee-Astragali Radix as a treatment, Sinomenii Caulis is recommended rather than Stephaniae Tetrandrae Rhizoma, and when using drug couple of Bang Kee-Lycopii Herba as a treatment, Sinomenii Caulis and Stephaniae Tetrandrae Rhizoma neither showed significant effect. And as Lycopii Herba interrupts the effect of Sinomenii Caulis, drug couple of Sinomenii Caulis-Lycopii Herba is not recommended.
We have developed a fully automated high throughput drug screening (HTDS) system based on the microfluidic cell culture array to perform combinational chemotherapy. This system has 64 individually addressable cell culture chambers where the sequential combinatorial concentrations of two different drugs can be generated by two microfluidic diffusive mixers. Each diffusive mixer has two integrated micropumps connected to the media and the drug reservoirs respectively for generating the desired combination without the need for any extra equipment to perfuse the solution such as syringe pumps. The cell array is periodically exposed to the drug combination with the programmed LabVIEW system during a couple of days without extra handling after seeding the cells into the microfluidic device and also, this device does not require the continuous generation of solutions compared to the previous systems. Therefore, the total amount of drug being consumed per experiment is less than a few hundred micro liters in each reservoir. The utility of this system is demonstrated through investigating the viability of the prostate cancer PC3 cell line with the combinational treatments of curcumin and tumor necrosis factor-alpha related apoptosis inducing ligand (TRAIL). Our results suggest that the system can be used for screening and optimizing drug combination with a small amount of reagent for combinatorial chemotherapy against cancer cells.
Lamotrigine is a newer anti-epileptic drug for adjunctive treatment of refractory epilepsy, partial seizures, generalized tonic-clonic seizures, and bipolar disorder. Lamotrigine overdose causes serious central nervous and cardiovascular problems, but reports are uncommon. Few lamotrigine overdoses have been described because anti-epileptic drug use is limited and usually used with combination of other anti-epileptic drugs. In addition, most patients visit emergency departments with multi-drug overdoses, so few cases of lamotrigine poisoning alone exist. We had a female patient visit our emergency department a couple of hours after a lamotrigine overdose treated with intravenous hydration and urine alkalization by NaHCO3. She recovered successfully without any evidence of renal injury. However, she developed profound rhabdomyolysis, a previously unreported complication of this medication. We suggest that serial creatine kinase levels should be measured after lamotrigine poisoning.
Sterically stabilized liposomes (SSL) have been introduced for longer circulation in blood than conventional liposomes (CL). However, there are a couple of problems in SSL preparation due to the instability of phospholipid and the degradation of drug in aqueous conditions. To solve these problems, it is necessary to go through lyophilization process. Therefore, in this study, effects of lyophilization on SSL were evaluated for physical characteristics changes upon rehydration of lyophilized SSL such as the particle size, efficiency of drug entrapment, turbidity and drug release. SSL containing streptozocin, a water-soluble anticancer drug as a model compound, were prepared with DSPC and DSPE-PEG 2000. The size was controlled to 100 nm by extrusion with polycarbonate membrane, and sucrose was used as a cryoprotectant for lyophilization at the 1:3 (lipid:sucrose) ratio. Upon rehydration of lyophilized SSL, the average size was in the range of $50{\sim}200\;nm$ which is adequate for longer circulation in blood, and the encapsulation efficiency was kept as its initial state. Rehydrated SSL were not adsorbed to rat plasma protein and revealed a similar drug release profile to that of fresh SSL before lyophilization. Therefore, lyophilization could be introduced efficiently to overcome aqueous instability problems of SSL.
Purpose: This study was carried out to investigate the research of acupuncture and moxibustion treatment for sinusitis. Methods: I mainly investigated 14 different kinds of recent edition of Chinese books and journals as references. Results: In the view of oriental medicine, sinusitis is regarded as Piyon(鼻淵). The loci used in the otopuncture therapies were Naepi(內鼻), Pye(肺), Aeck(額), Shinsangsun(腎上腺), Naebunpi(內分泌), and Weipi(外鼻) according to their frequencies. The drugs used in the drug-injection therapies were Corticoids, Lidocaine, Vitamin B, and herbal drugs. In addition, there were couple therapies mentioned in the references such as the electro-acupuncture therapies, the point penetration therapies, the point through-sinus acupuncture therapies, the point application therapies, the moxibustion therapies, the three-edged needle therapies, the ultra-sono acupuncture therapies, the cupping therapies, and the laser acupuncture therapies. Conclusions: There were various newly developed acupuncture and moxibustion treatment for sinusitis. Among them, the otopuncture therapies, the drug-injection therapies and the electro-acupuncture therapies were more frequently used than others.
Objectives : This study was planned to evaluate the therapeutic effectiveness and possible underlying mechanism of TPE (Tetrapanax papyrifer stem(inner part of the stem Extract) and AQE (Akebiae quinata stem Extract) on osteoarthritis. Methods : Osteoarthritis models were induced through intra-articular injection of MIA (monosodium iodoacetate) 50 μL with 80 mg/ml in rats. Excluding the normal group, Osteoarthritis-induced rats were divided into 4 groups (Control, INDO, TPE, AQE). The drug concentrations were indomethacin 5 mg/kg, TPE 200 mg/kg, and AQE 200 mg/kg, and were orally administered once a day for a couple of weeks. After drug supplementation, the effects of TPE and AQE were measured with serum diagnosis, western blotting, and histopathological staining. Results : It was found that the DPPH and ABTS free radical erasure ability of AQE was better than that of TPE. AQE administration improved rear limb overload and it led to relieving pain. Both PTE and AQE significantly reduced the expression of inflammatory mediators COX-2, iNOS, and inflammatory cytokine IL-1β and IL-6 by inhibiting the phosphorylation of IκBα and deactivating the pathway of NF-κBp65. On the other hand, TNF-α was significantly reduced only by administration of AQE. In addition, histopathological analysis showed that the administration of AQE compared to PTE suppressed cartilage degeneration and effectively suppressed damage to proteoglycan, a component of ECM. Conclusion : Reviewing these experimental results, TPE and AQE possessed the effect of delaying the progress of osteoarthritis and protecting cartilage. In addition, the results of this study show that AQE has a better cartilage protection effect than TPE.

Glycyrrhizin (GA) content in licorice was determined by a couple of methods using HPLC, respectively. In Method(I), GA content itself was determined from the licorice aqueous extract, while in Method (II) glycyrrhetinic acid (GHeA ; the aglicone of GA) content corresponding to the quantity of GA was measured from the chloroform extract of the hydrolyzed product of licorice aqueous extract. A reverse phase column Hibar Lichrosorb RP-18 (E. Merck) was used as the stationary phase. As the mobile phase MeOH: $H_{2}O$(0.05M-$NaH_{2}PO_{4}$)=58 : 42 solution in Method (I), and MeOH: $H_{2}O $: AcOH=78; 19: 3 solution in Method (II) were suitable, respectively. The value obtained by Method (II) appeared slightly higher than that by Method (I). The effect of some other herbal drugs on the assay of GA quantity in mixed sample was also observed in both above two methods. By Method (I) Cassiae Cortex, Rehmaniae Rhizoma, Paeoniae Radix, and Angelicae Radix gave the subtractive effect on the amount of GA compared with the value from licorice alone. In the case of Method (II) Cassiae Cortex and Rehmaniae Rhizoma appeared to have subtractive effect but Paeoniae Radix and Angelicae Radix scarcely showed any influence. Pachymae Fungus did not affect the GA content at all. It seems that glycyrrhizin in licorice interacts with certain components of other herbal drugs.
Cefetamet pivoxil is a prodrug of cefetamet possessing a broad spectrum of activity against many aerobic gram-positive and -negative organisms. Although many literatures in abroad had introduced its pharmacokinetics about two decays ago, no data have been revealed in Korean subjects. Therefore, this study was aimed to investigate the pharmacokinetics of cefetamet following a single oral administration of cefetamet pivoxil in Korean healthy volunteers. After an overnight fast, a tablet of cefetamet pivoxil (500 mg) was given to eight volunteers, and blood samples were serially taken up to 12 h. Plasma concentrations of cefetamet were determined by HPLC with UV detection. Cefetamet reached the peak concentration ($2.0{\pm}1.3\;{\mu}g/ml$) at $3.0{\pm}0.8$ h, and mono-exponentially decayed at a half-life of $2.6{\pm}0.9$ h. Three volunteers represented very low systemic exposure compared to the others, which provided very large inter-individual variation in Cmax, and AUC. The present results were discussed and reviewed with the previously published data, and a couple of points are suggested for clinical trials of this drug in Korean subject including bioequivalence study.
