Meningococcal Disease, manifesting as meningitis and septicemia, is a life-threatening bacterial infection that results in significant morbidity and mortality, particularly in childhood. Its epidemic potential and limited opportunities for clinical intervention due to its rapid course present unique public health and clinical challenges. Incidence is highest in infants and young children, with a secondary peak of risk in adolescents. Approximately 10% of cases are fatal and survivors can be left with serious and permanent sequelae including amputations, hearing loss and cognitive impairment. Transmission is only from human-to-human, by infected respiratory tract secretions or saliva and therefore crowding poses a tremendously elevated risk for disease development. Military recruits and university students are at high risk due to the high carriage rate in adolescents, their behavior patterns and close contact. Menveo$^{(R)}$ (Novartis Vaccines and Diagnostics), a novel quadrivalent meningococcal conjugate vaccine directed against meningococcal serogroups A, C, W-135 and Y, has been shown to be immunogenic and well tolerated in all age groups and was recently licensed for use in Korea. Recent cases and deaths among military recruits drew public attention to their elevated risk and the Korean government has recommended vaccination of all new military recruits. Many Korean students seek to attend school, university, or language institutes in countries where routine meningococcal vaccination is required - clinicians should be aware of such requirements to ensure that students are vaccinated prior to arrival in the destination country.
Leishmaniasis is a serious parasitic disease caused by Leishmania spp. transmitted through sandfly bites. This disease is a major public health concern worldwide. It can occur in 3 different clinical forms: cutaneous, mucocutaneous, and visceral leishmaniasis (CL, MCL, and VL, respectively), caused by different Leishmania spp. Currently, licensed vaccines are unavailable for the treatment of human leishmaniasis. The treatment and prevention of this disease rely mainly on chemotherapeutics, which are highly toxic and have an increasing resistance problem. The development of a safe, effective, and affordable vaccine for all forms of vector-borne disease is urgently needed to block transmission of the parasite between the host and vector. Immunological mechanisms in the pathogenesis of leishmaniasis are complex. IL-12-driven Th1-type immune response plays a crucial role in host protection. The essential purpose of vaccination is to establish a protective immune response. To date, numerous vaccine studies have been conducted using live/attenuated/killed parasites, fractionated parasites, subunits, recombinant or DNA technology, delivery systems, and chimeric peptides. Most of these studies were limited to animals. In addition, standardization has not been achieved in these studies due to the differences in the virulence dynamics of the Leishmania spp. and the feasibility of the adjuvants. More studies are needed to develop a safe and effective vaccine, which is the most promising approach against Leishmania infection.
Varicella, which is mostly a benign disease, but also can cause considerable health burden in the community, can be prevented by immunization with live attenuated varicella vaccine. Higher uptake of varicella vaccine by universal immunization in North America has apparently been associated with decline in the number of reported cases of varicella, varicella-related hospitalizations, and the number of deaths caused by complications of varicella. On the contrary, there has been some reluctance in endorsing varicella vaccine for universal immunization in most of European countries. Concerns include unanticipated outbreaks of varicella among vaccine recipients, risk of varicella among unvaccinated adults, risk of herpes zoster among vaccinees as well as unvaccinees. Recently developed measles, mumps, rubella, and varicella combination vaccine and herpes zoster vaccine that may be licensed in the upcoming years may be the solution for varicella vaccine to be utilized in a greater scale. In Korea several varicella vaccine products have been utilized since late 1980. The adoption of varicella vaccine for universal immunization since 2005 along with the changing view in varicella prevention strategy mandates more studies for immunogenecity and efficacy of varicella vaccines as well as more surveillance to delineate the changes in epidemiology of varicella in Korea.
In this study, we evaluated the efficacy of a commercial vaccine stress-relieving agent (Anti VS-2 injection) to reduce the adverse effects of foot-and-mouth disease (FMD) vaccination site in 120 pigs on two tested pig farms. The results showed that incidence of pig with abnormal meat, size of abnormal meat and weight of discarded abnormal meat are significantly decreased in Anti VS-2 injection-supplemented FMD vaccinated pig groups (n=60) compared to the non-supplemented FMD vaccinated pig groups (n=60) (P<0.05). And the average economic loss by discard of abnormal meat were estimated 99,382 won or 21,514 won in the non-supplemented FMD vaccinated groups or in the Anti VS-2 injection-supplemented FMD vaccinated groups, respectively, showing that the cost saving effect was 78% (77,869 won). In conclusion, it is expected to reduce the incidence and severity of abnormal meat in the FMD vaccinated pigs and to contribute not only to the reduced economic loss of pig farmers but also to the promotion of food hygiene when the Anti VS-2 injection is mixed with FMD vaccine.
Purpose: Enterovirus 71, a pathogen that causes hand-foot and mouth disease (HFMD) is currently regarded as an increasing neurotropic virus in Asia and can cause severe complications in pediatric patients with blister-like sores or rashes on the hand, feet, and mouth. Notwithstanding the significant burden of the disease, no authorized vaccine is available. Previously identified attenuated and inactivated vaccines are worthless over time owing to changes in the viral genome. Materials and Methods: A novel vaccine construct using B-cell derived T-cell epitopes from the virulent polyprotein found the induction of possible immune response. In order to boost the immune system, a beta-defensin 1 preproprotein adjuvant with EAAAK linker was added at the N-terminal end of the vaccine sequence. Results: The immunogenicity of the designed, refined, and verified prospective three-dimensional-structure of the multi-epitope vaccine was found to be quite high, exhibiting non-allergenic and antigenic properties. The vaccine candidates bound to toll-like receptor 3 in a molecular docking analysis, and the efficacy of the potential vaccine to generate a strong immune response was assessed through in silico immunological simulation. Conclusion: Computational analysis has shown that the proposed multi-epitope vaccine is possibly safe for use in humans and can elicit an immune response.
Relatively little has been studied on the AMA-1 vaccine against Plasmodium vivax and on the plasmid DNA vaccine encoding P. vivax AMA-1 (PvAMA-1). In the present study, a plasmid DNA vaccine encoding AMA-1 of the reemerging Korean P. vivax has been constructed and a preliminary study was done on its cellular immunogenicity to recipient BALB/c mice. The PvAMA-1 gene was cloned and expressed in the plasmid vector UBpcAMA-1, and a protein band of approximately 56.8 kDa was obtained from the transfected COS7 cells. BALB/c mice were immunized intramuscularly or using a gene gun 4 times with the vaccine, and the proportions of splenic T-cell subsets were examined by fluorocytometry at week 2 after the last injection. The spleen cells from intramuscularly injected mice revealed no significant changes in the proportions of CD8$^+$ T-cells and CD4$^+$ T-cells. However, in mice immunized using a gene gun, significantly higher (P<0.05) proportions of CD8$^+$ cells were observed compared to UB vector-injected control mice. The results indicated that cellular immunogenicity of the plasmid DNA vaccine encoding AMA-1 of the reemerging Korean P. vivax was weak when it was injected intramuscularly; however, a promising effect was observed using the gene gun injection technique.
Bordetella pertussis는 유아의 호흡기감염질환인 백일해의 원인균으로 배신도입에 의해 발생률이 크게 낮아졌다. 그러나 최근에 일부 백신접종률이 높은 국가에서 증가된 백일해 발생건수가 보고되고 있으며, 그 추정 원인 중에 하나는 백신주와 유행주 사이의 유전형 흑은 혈청형 변이 이다. 따라서 분리주에 대한 변이 현황을 유전형 혹은 혈청형 분석을 통하여 확인하는 것이 필요하고 국내에서 백일해 발병증가에 대한 가능성을 추정해야 한다. 이에 본 연구에서는 $1999\sim2006$년 사이에 분리된 국내 백일해 분리균주의 변이 양상을 agglutinogen과 fimbriae에 대한 혈청형 조사를 통해 확인하였다. 그 결과 agg 1과 fim 2가 국내 분리주에서 가장 많이 발견되는 혈청형이었고, 두 항원 모두 시대에 따른 혈청형 변이 현상이 확인되었으며, 특히 agglutinogen의 경우는 백신균주(agg 1,2)와는 다른 유형(agg 1)이 증가되는 유형으로 나타났다. 그러나 fimbriae의 경우는 백신균주(fim 2)와 동일한 유형이 증가되어 차이를 나타내었다. 이러한 결과는 백일해 발병증가가 나타나는 국가들에서도 확인되는 현상으로 국내에서의 백일해 발병증가에 대한 보다 정확한 예측을 위해서는 항원결정기 유전자에 대한 변이정도 및 정상인에서의 항체가 분포양상 등에 대한 연구가 필요할 것으로 사료된다.

Newcastle disease vaccine (NDV)-loaded chitosan microspheres (NDV-CM) were prepared. Stimulatory effects of these NDV-CM on antibody response compared to free NDV were examined in vitro and in vivo. In vitro stimulation of macrophages with virus vaccine resulted in higher number of cells compared to saline-treated control. Both NDV and NDV-CM induced secretion of interleukin-1 (IL-1) in dose dependent manner and the secretion of IL-1 by NDV-CM was delayed compared to free NDV. Irrespective of vaccine formulation, NDV subunit antigen was not effective in preventing mortality of the birds after challenge. However, CM loaded with NDV made of whole viron had antibody responses and protection similar to those shown by ND-K, a commercial inactivated oilemulsion vaccine.
Since the first outbreaks of Newcastle disease ( ND) in Korea was reported in 1927, the disease has been a major problem in Korean poultry industry. Several attempts related ND prevention and controls have been tried and significantly reduced the ND outbreaks. However, recently frequencies of ND outbreaks have been increased throughout the country since 1988. Many assumptions such as variances of ND virus pathogenicity, decreased efficacy of ND vaccine, incorrect vaccination program etc, has been issued and discussed. Therefore, to clearly understand this recent situation of ND outbreaks in Korea, we reviewed all available past informations related with ND outbreaks in Korea and compared the virulence of recently isolated ND viruses with that of a reference one. Also, we discussed about some problems related with serological diagnosis. administration methods of vaccine and vaccination program.

Classical swine fever (CSF) is a contagious disease of swine with serious economic losses in pig industry [1]. The disease is caused by CSFV which belongs to the viruses of bovine viral diarrhea (BVDV) and border disease virus (BDV) make up the Pestivirus genus within the family Flaviviridae [2]. Attenuated Korean LOM strains were used in Korea. For these reasons a practical approach for discrimination between vaccine and field strains is needed. Here, we described the deveopment of real-time RT-PCR to discriminate between vaccine strains and Korean field viruses of CSFV. (omitted)
