Upregulation of Kruppel-like Factor 4 Gene expression by Allomyrina dichotoma Hemolymph in the INS-1 Pancreatic β-cells

  • The hemolymph of Korean rhinoceros Allomyrina dichotoma consists of blood and lymph in which various kinds of proteins function physiologically. We have previously demonstrated that A. dichotoma hemolymph has the potential to treatment and prevent diabetes through activating transcription factor 3-gene (ATF3) regulation. In this study, we investigate the expression of Kruppel-like factor 4 (KLF4) in A. dichotoma hemolymph-treated INS-1 pancreatic β-cells. The new findings show that A. dichotoma hemolymph, which upregulates KLF4 gene expression in a dose-dependent and time-dependent manner. In addition, hemolymph combine with mild endoplasmic reticulum (ER) stress, which also differentially regulates KLF4 gene expression. These results may provide insights to KLF4 gene-related disease therapies through KLF4 gene regulation.

Inhibitory Effects of Artemisia capillaris Extract on the Pathogenic Bacteria in Mice (인진쑥 추출물의 병원성 세균에 대한 생체내 증식 억제 효과)

  • Artemisia capillaris THUNB is a perennial herb that belongs to the family Compositae spp and the most common plant among the various herbal folk remedies used in treatment of abdominal pain, hepatitis, chronic liver disease, jaundice and coughing in Korea. This experiment was conducted to investigate the inhibitory effects of orally administrated Artemisia capillaris extracts on the pathogenic bacteria in 200 ICR mice. The experimental groups showed inhibitory effects on the bacteria in $1{\sim}3$ days after inoculation. After 21 days of inoculation, no viable bacterial cells appeared in the feces of both experimental groups while they did appear in the control group. The results of these studies indicate Artemisia capillaris extract exhibited excellent antimicrobial and inhibitory effects on the food poisoning pathogenic bacteria; S. enteritidis, E. coli O157:H7, L. monocytogenes and S. aureus.

  • In Korea, several national cross-sectional surveys monitor the diet, nutritional status, and health status of children. This continual dedicated national surveillance system contributes to the identification of nutritional and health issues, establishment of public health policies, and development of nutrition recommendations. This paper provides recent information about the Korea National Health and Nutrition Examination Survey and the Korean Youth Risk Behavior Web-based Survey and describes key nationwide survey findings published in the last 5 years on infant feeding practices and the dietary intake and nutritional status of Korean infants, children, and adolescents. There have been increasing trends in children, and teenagers who skip breakfast, eat fast food, consume sugary drinks, have vitamin D deficiency, and are obese. This review will inform pediatricians, nutritionists, and other health care practitioners who track children's growth and development. It may also help researchers and policymakers identify diet-related policies and strategies for chronic disease prevention in Korean infants, children, and adolescents.

First detection of ranavirus in a wild population of Dybowski's brown frog (Rana dybowskii) in South Korea

  • Background: Ranavirus is an emerging infectious disease which has been linked to mass mortality events in various amphibian species. In this study, we document the first mass mortality event of an adult population of Dybowski's brown frogs (Rana dybowskii), in 2017, within a mountain valley in South Korea. Results: We confirmed the presence of ranavirus from all collected frogs (n = 22) via PCR and obtained the 500 bp major capsid protein (MCP) sequence from 13 individuals. The identified MCP sequence highly resembled Frog virus 3 (FV3) and was the same haplotype of a previously identified viral sequence collected from Huanren brown frog (R. huanrenensis) tadpoles in South Korea. Human habitat alteration, by recent erosion control works, may be partially responsible for this mass mortality event. Conclusion: We document the first mass mortality event in a wild Korean population of R. dybowskii. We also suggest, to determine if ranavirus infection is a threat to amphibians, government officials and researchers should develop continuous, country-wide, ranavirus monitoring programs of Korean amphibian populations.

Knowledge on Fish in the Imwon-Gyeongje-Ji from a Korean Medical Perspective - A Comparative Study with the Dongui-bogam - (『림원경제지(林園經濟志)』 어류지식의 한의학적 고찰 - 『동의보감(東醫寶鑑)』과의 비교를 중심으로 -)

  • Objectives : This paper summarizes contents on fish in the Injeji and Jeoneo-Ji parts in the Imwon-Gyeongje-Ji and compares them with those in the Dong-uibogam from a Korean Medical perspective. Methods : Most of the knowledge of fish in the Imwon-Gyeongje-Ji is based on preceding material medica studies such as the Dongui-bogam in Joseon, the Bencao-gangmu in China, and the Hwahan Samjaedohoe in Japan. The influential relationships and the process of change among these knowledge information were examined within the context of each literature's development or through identifiable changes in knowledge on each particular kind of fish. Results & Conclusions : In the Imwon-Gyeongje-Ji, the consisting 16 parts are organically connected, sometimes functioning as cross-references. In particular, the medical contents on the efficacy of fish for treating disease is closely linked to contents on fish ecology and environment as written in the Jeoneo-Ji, which deals with the field of living things. On the more detailed knowledge on about 30 kinds of fish, one could grasp the formation process and changes of East Asian traditional knowledge on fishkind among historical study of influential Chinese texts, new stimulation and methodology through Japanese literature on natural history, and dynamic processes of inheriting and selectively accepting traditional knowledge of Joseon.

Lack of Association between the S20G Missense Mutation of Amylin Gene and Essential Hypertension in Korean Population

  • Essential hypertension is a heterogeneously multifactorial disease in which blood pressure is harmfully high without overt cause. Both genetic and environmental factors have been implicated in its etiology. In view of the regulatory role of this peptide in the carbohydrate metabolism and renin-angiotensin system, amylin gene has been proposed to a candidate gene for essential hypertension. Therefore, we scanned the amylin gene for mutations in 133 Korean normotensives and 61 essential hypertensives by single-strand conformational polymorphism, and found a single heterozygous S20G missense mutation. However, no significant difference was observed between normotensives and essential hypertensives in the distribution of allele and genotype frequencies of this mutation at the amylin gene (P>0.05). This finding suggests that S20G missense mutation of the amylin gene are unlikely to contribute to the etiology of essential hypertension in the Korean population.

High ranavirus infection rates at low and extreme temperatures in the tadpoles of Japanese treefrogs (Dryophytes japonicus) that breed in rice paddies in the summer

  • Background: Several species of amphibians in agricultural areas are often infected with ranaviruses; however, the biological or ecological factors that cause this infection are not well understood. In this study, we investigated whether local tadpole density, Gosner developmental stage, and weather conditions affected ranavirus infection in Dryophytes japonicus tadpoles in rice paddies over three months. Results: During the study, eight samplings were undertaken between June 6 and August 21, 2022. No die-off of tadpoles occurred, but 20 of 110 tadpoles (18.8%) were found to be infected with ranavirus. The tadpole density at the sampling site and Gosner stage of the sampled tadpoles were not related to the daily ranavirus infection rate. The mean daily highest temperature during the two weeks prior to the sampling date and the mean daily lowest and highest temperatures during the week prior to the sampling date were negatively related to the daily infection rate. Conclusions: Our results suggest that low and extreme temperatures caused by flooding and draining of paddy fields or climate change in summer could be a significant risk factor for ranavirus infection in summer-breeding frogs in agricultural areas.

Chemopreventive Potential of Salvia miltiorrhiza Fraction Extracts (단삼 분획추출물의 암예방 효과)

  • Six fractions of Salvia miltiorrhiza were tested for their chemopreventive potentials using biochemical markers of carcinogenesis such as quinone reductase (QR), glutathione S-transferase (GST) and glutathione (GSH). Seventy percentage of EtOH extract was potent inducer of QR activity in Hepa1c1c7 murine hepatoma cells. GST activity was increased about 1.4-fold with EtOAc extract at concentration of 50 ${\mu}g/ml$. GSH levels were significantly increased with $H_2O$ extract, 70% EtOH extract and water extract at concentration of 50 ${\mu}g/ml$ (p<0.005). From these results, 70% EtOH extract (250 mg/kg) was intragastrically administered to ICR mice for 14 days. QR, GST and GSH levels were significantly increased with the 70% EtOH treatment. These studies suggest that the 70% EtOH extract of S. miltiorrhiza could be considered as a potential agent for cancer chemoprevention.

  • The effects of climatic factors on organisms lire variable and complex, and it, however, can be interpreted in terms of those on the distribution and those on the population densities. The distribution of an organism may largely be determined by the temperatures, except some temporal organisms which are depended on the air mass movements. Population density of an organism is determined by various climatic factors, such as previous winter temperature, temperature of growing season and rainfall. The start of growing season of the rice plants has been shifted to earlier since last decade in Korea. This may mean that the overall climatic condition during the growing season might be considerably different from those in past years, and such a difference in climatic conditions might have close relation with the recent status of the diseases and insect pests through direct effects on the physiology and population dynamics of the organisms, as well as through on the biotic associations of the pest organisms. The white back planthopper and brown planthopper have become the key insect pests in Korea in recent years. They are migratory and have high reproductive pontentials and more generations than average residential insects. The synchronization of the migrants and physiological condition of the rice plants seems to be the important factors in relation to the recent outbreaks of these insects; the high reproductive rate can be obtained with the growth stage of rice being 30-50 days after transplanting. The modication of the microclimate associated with high plant density and some other introduced new cultural techniques also have some relation with the outbreak. The key diseases of the rice are the blast disease, sheath blight and the bacterial leaf blight. For the rice blast, the seedling blast and leaf blast during the early growing season and the neck blast, have become more serious, the former may be related to hotbed nursery and the later may be related to the high humidity in early August, and synchronization of the heading time which has been shifted to early part from middle or late part of August. In general, for the rice diseases, the development of the new races have been the most serious which are largely resulted from the introduction of the new varieties, but it also seems to be related with the prolonged periods of the favorable condition associated with the shifted growing seasons. In general, the diseases and insect pest problems have become much more variable and complex, and control measures should be based on the thorough knowledge of the ecology of the pest organisms, that is, effects of various environmental factors on the disease cycle; spore release, spore deposition, infection, colonization and sporulation of the disease organisms, and those on the development, reproductive potentials, dispersal, age specific responses of the insects. The well organized real-time pest management systems, such as alfalfa weevil management system developed at the Purdue University in U.S., is the prime importance for the implementation of the pest management principles.

Evaluation of Fungicides, Nozzle Type, and Spray Volume on Control of Typhula Blight on Cool Season Turfgrass (한지형잔디에 설부병 방제에 대한 살균제, 노즐타입 및 살포약량의 평가)

  • Commercial formulation of fungicides was studied in vitro for sensitivity against Typhula species causal agents of Typhula blight. Efficacies of fungicides application, spray volume, nozzle types and fungicides applied time (early fall and late fall) were evaluated for their influence on the chemical control of Typhula blight of turfgrass during the winter season in Wisconsin. All fungicides effectively reduced the mycelial growth of eight isolates of Typhula spp. in vitro on potato dextrose agar (PDA) media. For inhibitory effects on mycelial growth of eight isolates, propiconazole was the most effective at $1.0{\mu}g$ active ingredient (a.i) / ml of PDA. Typhula incarnata two isolates were significantly more sensitive to all fungicides of PDA than six isolates of three varieties of T. ishikariensis. For 2 years in field experiment, unsprayed control has significantly more disease severity than seven fungicides were applied to field plots at two locations. Propiconazole was the most effective for controlling Typhula blight, at two locations in both years. The level of disease control was not dependent on fungicides spray volume or nozzle types at two locations. The disease damage treated with triadimefon applied time (early fall and late fall) was not significantly different at two location for two years.
