구멍갈파래(Ulva pertusa)로부터 Dimethyl-$\beta$-propiothetin 최적추출조건 (Optimal Extracting Conditions Dimethyl-$\beta$-propiothetin from Ulva pertusa)

  • This study was focused on the optimal extracting conditions of dimethyl-$\beta$-propiothetin as bioactive substance from green seaweed. Identification and quantification of dimethyl-$\beta$-propiothetin were measured by headspace gas chromatography after conversion to dimethyl sulfide by treatment with saturated NaOH solution. Dimethyl-$\beta$-propiothetin was extracted through various processes (solvent extraction, ultrasonication, boiling and autoclaving) from Ulva pertusa. The content of dimethyl-$\beta$-propiothetin extracted by autoclaving treatment showed higher than that of various extraction methods. Dimethyl-$\beta$-propiothetin content in extract of Ulva pertusa was 325,800 ng/g after autoclaving 121$^{\circ}C$ for 45 min. Dimethyl-$\beta$-propiothetin in exract of Ulva pertusa was comparative stable under low temperature. The retentions of dimethyl-$\beta$-propiothetin content in extract of Ulva pertusa were 76.6~99.8% by incubation at 10~6$0^{\circ}C$ for 2 hours. Chemical decomposition of dimethyl-$\beta$-propiothetin was observed under laboratory conditions at pH values higher than 9.5.

창자파래(Enteromorpha intestinalis)로부터 Dimethyl-$\beta$-propiothetin 추출 (Extraction of Dimethyl-$\beta$-propiothetin from Enteromorpha intestinalis)

  • 해조류에서 생합성되는 3급 함황화합물인 DMPT를 창자파래로부터 추출하기 위하여 시료의 상태, 용매 의 극성, 초음파 처리, boiling 및 autoclaving 처리에 의한 추출조건을 검토하였다. 용매추출, 초음파 처리후 용매추출, 열수추출 및 가압 열수추출법을 이용하여 창자파래로부터 DMPT를 추출한 결과, 초음파 처리후 가압 열수처리한 경우가 가장 효과적이었고, 다음으로는 초음파 처리후 열수 처리하는 방법이었다. 창자파래로부터 DMPT를 추출 해내는 최적조건으로 분말시료에 물과 acetone (2:1, v/v)의 혼합용매를 15배량 가하여 초음파 처리 후 121$^{\circ}C$에서 60분간 autoclaving하는 방법으로서, 이 때 추출된 DMPT 함량은 시료 g당 311.200ng이었다. 창자파래에서 추출한 DMPT 수용액을 조제하여 온도별로 저장시킬 때 DMPT 분해에 의한 DMS의 발생량은 저장온도에 비례하여 증가되었지만 저온에서는 매우 낮은 분해율을 나타내었다. DMPT 1mg을 저장하였을 때의 잔존율은 6$0^{\circ}C$ 저장의 경우78.5%로 고온에서는 다소 불안정하였으나 $0^{\circ}C$에서는 DMPT의 잔존율이 99.8%로 매우 높게 나타났다. 창자파래에서 추출한 DMPT 수용액을 3$0^{\circ}C$에서 방치하였을 때 pH가 5 이하의 산성영역에서는 DM-PT의 분해가 거의 일어나지 않았으나 pH가 올라감에 따라서 분해가 서서히 일어나기 시작하여 pH 9.5에 이르러서는 DMS의 양이 급속하게 증가하였다.

  • To investigate the effects of growth and body composition of spotted sea bass (Lateolabrix sp.) by the supplemented dimethyl-$\beta$-propiothetin(DMPT). fish was fed the diets each containing 3 mM, 5 mM, 7 mM and 10 mM for eight weeks. The weight gain, feed efficiency and feed intake were significantly increased with dietary DMPT level. Condition factor was reduced by dietary DMPT supplementation. Crude protein content of fish after eight weeks increased with dietary DMPT level, whereas crude lipid content deceased with dietary DMPT level. Amino acid content was higher in DMPT-added group than control group. Amino acid composition was not influenced by dietary DMPT supplementation. Fatty acid content deceased with dietary DMPT level.

  • A eight weeks feeding experiment was conducted to investigate the effects of different dietary dimethyl-$\beta$-propiothetin (DMPT) levels on growth and chemical components in red sea bream (Pagrus major). The content of DMPT extracted from Ulva pertusa by auroclaving was 32.5mg%. Body weight gain, feed efficiency and feed intake were significantly increased with dietary DMPT level. After eight weeks of feeding trial, moisture content decreased with supplemented DMPT level, while crude protein and lipid contents increased with dietary DMPT level. Amino acid contents of muscle was elevated in the DMPT added groups compared to control group. Amino acid composition of muscle in fish fed the diets supplemented DMPT was not different to that of control group. Fatty acid contents of muscle in fish increased with dietary DMPT level.

  • 수산물 중에 다량으로 분포하는 섭이촉진물질로 알려진 DMPT의 추출 이용을 위한 기초 자료를 얻 고자 DMPT를 3 mM, 5 mM, 10 mM로 첨가한 실험용 사료로 8 주간 사육한 조피볼락의 성장효과 및 어체 성분에 미치는 영향을 검토한 결과를 요약하면 다음과 같다. 1. DMPT를 첨가하지 않은 대조구의 체중증가율은 약 1.2 배였으나 10 mM을 첨가한 사료의 경우는 약 1.4배로 성장효과가 크게 증가하였고, 사료효율 및 사료 섭취량도 많아 DMPT가 사료 섭이를 촉진시키는 것으로 나타났다. 2. 일반성분 변화 를 보면 DMPT의 농도가 높을수록 수분 함량은 감소하고 조단백질과 조지방의 함량은 함께 증가하는 경향이었다. 3. 아미노산 함량의 변화에서는 대조구가 3286.3 mg%에 비하여 10 mM 첨가구에서는 3568.3 mg%로 높은 값을 나타내어 DMPT 첨가구가 높데 나타났으며, 구성아미노산 조성에 있어서는 서로 비슷한 경향이었다. 4. 지방산 함량의 변화를 살펴보면, 실험개시전과 대조구에 비하여 DMPT 첨 가구에서 높게 나타났으며, DMPT의 농도가 높을수록 지방산의 함량도 증가하여 지방의 함량 변화와 일치하였다.
