  • The present study was aimed to elucidate the mechanism of stabilization of tubulin by deuterium oxide ($D_2O$). Rate of decrease of tryptophan fluorescence during aging of tubulin at 4$^{\circ}C$ and 37$^{\circ}C$ was significantly lower in $D_2O$ than in $H_2O$. Circular dichroism spectra of tubulin after incubation at 4$^{\circ}C$, suggested that complete stabilization of the secondary structure in D2O during the first 24 hours of incubation. The number of available cysteine measured by DTNB reaction was decreased to a lesser extent in $D_2O$ than in $H_2O$. . During the increase in temperature of tubulin, the rate of decrease of fluorescence at 335 nm and change of CD value at 222 nm was lesser in $D_2O$. Differential Scanning calorimetric experiments showed that the $T_m$ values for tubulin unfolding in $D_2O$ were 58.6$^{\circ}C$ and 62.17$^{\circ}C$, and in $H_2O$. those values were 55.4$^{\circ}C$ and 59.35$^{\circ}C$.

  • The 3-methylglutaconic aciduria (3-MGA-uria) is a heterogeneous group of several inborn errors of metabolism characterized by increased urinary excretion of 3-methylglutaconic acid. In most cases, 3-MGA is only slightly increased and combined with other metabolites. However, repeated and significant excretion of 3-MGA (40->1,000 mmol/mol creatinine) is a hallmark of the disorders of 3-MGA-urias. There have identified five distinct types of disorders: inborn errors of leucine metabolism and four disorders of mitochondrial dysfunction through different mechanism. The range of clinical and biochemical findings in this condition is variable. In the patients with 3-methylglutaconyl-CoA hydratase deficiency, increased 3-hydroxyisovaleric acid is useful in the differential diagnosis. Other forms of 3-MGA-urias are welldefined clinically such as Barth syndrome, Costeff syndrome, TMEM 70 defect, MEGDEL syndrome, and DCMA syndrome. We provide an overview of the expanding clinical spectrum and differential diagnosis of the 3-MGA-urias.

  • Generally, the Box-Culvert in levee is destroyed by various reasons. Especially when Box-Culvert is installed over the pile foundation in soft ground, the failure occurrs for 1) the weakness of compaction in Box-Culvert side by the differential settlement between outer ground and inner soil prism, 2) hydraulic fracturing and disturbance of Box-Culvert side soil by the repeated acting of seepage pressure at flood time. Also the side of Box-Culvert is difficult to compact and the shear resistance is reduced by more than 1/3 for the reduction of friction caused by the difference of material property. In this study, a series of model tests are conducted for the analysis of the development mechanism of outer ground and inner soil prism by the differential settlement using the pile foundation in soft ground, and cavity suppressed technique is suggested by the analysis of base plate enlargement effect.

  • The bimodal flocculation of cohesive sediments in water environments describes the aggregation and breakage process developing a bimodal floc size distribution with dense flocculi and floppy flocs. A two class population balance equation (TCPBE) was tested for simulating the bimodal flocculation by a model-data fitting analysis with two sets of experimental data (low and high turbulent flows) from 1-D flocculation-settling column tests. In contrast to the Single-Class PBE (SCPBE), the TCPBE could simulate interactions between flocculi and flocs and the flocculation mechanism by differential settling in a low turbulent flow. Also, the TCPBE could perform the same quality of simulation as the elaborate Multi-Class PBE (MCPBE), with a small number of floc size classes and differential equations. Thus, the TCPBE was proven to be the simplest model that is capable of simulating the bimodal flocculation of cohesive sediments in water environments and water, wastewater treatment systems.

  • 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD), a prototype of many halogenated aromatic hydrocarbons, is a ubiquitous, persistent environmental contaminant and displays high toxicity in animals and has been implicated in human carcinogenesis. Although the mechanism of carcinogenesis by TCDD is unclear, it is considered to be a non-genotoxic and tumor promoter. In this study, we investigated the tumor promotion effect of TCDD on the two-stage skin chemical carcinogenesis using hairless mouse (SKH1). We induced papillomas after treatment with N-methyl -N'-nitro-N-nitorsoguanidine (MNNG) as a initiator and TCDD as a promoter for 30 weeks. We found that the incidence or multiplicity of papillomas and hyperplastic nodules was maximally induced at MNNG-TCDD group compare to control, MNNG, and TCDD alone. These results suggesting that TCDD can acts as a potent promoter in the hairless mouse skin. In addition, we used cDNA microarray to detect the differential gene expression in normal, tumor surrounding, and tumor regions induced in hairless mouse skin by MNNG plus TCDD protocol. We found that 49 and 42 genes out of 5,592 genes associated with protein synthesis, cell organization, lipid transport and oxidative stress in tumor and surrounding regions were up- or down- regulated two fold or more, respectively. We are currently investigating how these genes play a role in TCDD-mediated chemical carcinogenesis.

  • AMachine vision is a promising tool for the autonomous guidance of farm machinery. Conventional CCD camera for the machine vision needs a desktop PC to install a frame grabber, however, a web camera is ready to use when plugged in the USB port. A web camera with a notebook PC can replace existing camera system. Autonomous steering control system of this research was intended to be used for combine harvester. If the web camera can recognize cut/uncut edge of crop, which will be the reference for steering control, then the position of the machine can be determined in terms of lateral offset and heading angle. In this research, a white line was used as a cut/uncut edge of crop for steering control. Image processing algorithm including capturing image in the web camera was developed to determine the desired travel path. An experimental vehicle was constructed to evaluate the system performance. Since the vehicle adopted differential drive steering mechanism, it is steered by the difference of rotation speed between left and right wheels. According to the position of vehicle, the steering algorithm was developed as well. Evaluation tests showed that the experimental vehicle could travel within an RMS error of 0.8cm along the desired path at the ground speed of $9\sim41cm/s$. Even when the vehicle started with initial offsets or tilted heading angle, it could move quickly to track the desired path after traveling $1.52\sim3.5m$. For turning section, i.e., the curved path with curvature of 3 m, the vehicle completed its turning securely.

  • Basis for differential response of rice and E. crus-galli to cyhalofop-butyl ester (R-butyl 2-(4-(4-cyano-2-fluoro-phenoxy)phenoxy)propionate) was anatomically compared. Cell division in the growing point of E. crus-galli was severely inhibited by the herbicide, leading to malformation and irregular arrangement of cells. The inhibitory symptom of rice by cyhalofop-butyl ester appeared to be very similiar to that of E. crus-galli although its injury was negligible in rice when applied at the rate of 180ppm.

  • The aim of this paper is to investigate the change of pressure differential and smoke propagation characteristics in the elevator lobby with the resident's evacuation scenarios using fire modelling technique. The results showed absolute pressures in the fire room and elevator lobby can significantly increase to cause fire door to the stairway unclosed once it is open. This is due to constant pressure differentials, the increasing reference pressure of fire lobby and pressure leak from elevator lobby to fire lobby. Smoke exhaust mechanism was needed to prevent the continuous pressure rise in the living room. Over 200 Pa was expected upon closing the door during pressurization, which provide difficulties in opening the door for next refugee. Opening both fire door and entrance door may induce smoke flow from fire room to elevator lobby and stairway.

  • Electrochemical behavior of $Ln^{3+},$-ALC complexes($Gd^{3+},$ $Tb^{3+},$ $Dy^{3+},$ $Ho^{3+},$ $Er^{3+},$ $Yb^{3+}$ and $Lu^{3+}$-alizarin complex-one) has been investgated by d.c polarography, differential pulse polarography and cyclic voltammetry. The reduction mechanism of ALC comes to the conclusion that the two electron make one step of reversible processes, and that there is few adsorption in the electrode reaction. The new complex is made from one lanthanide ion and one ALC. This complex is proven to make an adsorptive complex wave, by the experiments of differential pulse polarography and cyclic voltammetry. The reduction potential of complex wave($P_2$)turns up more negatively than ligand wave($P_1$) does. Linear calibration curves of the decreasing P1 and increasing $P_2$ is obtained when the lanthanide concentration varys from $2.5X10^5$M to $1X10^4M$.

  • The electrochemical reduction of O,O-dimethyl-O-(3-methyl-4-nitrophenyl)-phosphorothioate (Fenitrothion) has been studied in acetonitrile solution containing surfactant micelle by direct current (DC)-differential pulse (DP) polarography, cyclic voltammetry (CV) and controlled potential coulometry (CPC). The partially reversible electron transfer-chemical reaction(EC, EC mechanism) of fenitrothion reduction proceeded by four electron transfer to form O,O-dimethyl-O-(3-methyl-4-hydroxyaminophenyl)-phosphorothioate which undergoes single bond of the phosphorus atom and phenoxy group cleaves to give p-amino-m-cresol and dimethyl thiophosphinic acid as major product by two electron transfer-protonation at higher negative potential. The polarograpic reduction waves shown to suppressed due to inhibitory effect of sodium lauryl sulfate micelle solution and split up on selectivity of anionic micelle effect in two step at the first reduction peak.
