재배 조건 및 재배 지역에 따른 콩의 이화학적 특성 및 두부 품질에 미치는 영향 (Physicochemical Characteristics of Soybeans Grown in Different Origins and Cultivation Methods Accompanying Tofu Properties)

  • In this study, the characteristics of soybeans with different cultivation methods depending on their origins and the properties of tofu made from these soybeans were studied. The isoflavone content was higher in the general Korean soybeans compared to the organic American soybeans. There were no significant differences among the Chinese soybean samples, so the cultivar method was considered to have a greater effect on the isoflavone contents rather than the cultivation method. The protein contents were higher in the tofu made from the general Chinese soybeans and the organic Korean soybeans. Furthermore, the tofu made from the organic Chinese soybeans contained the least amount of proteins. Isoflavone contents were not significantly different among the samples. Texture profile analyses consisting of hardness, cohesiveness, adhesiveness, springiness, and gumminess were all shown to be higher in the organic tofu than in general tofu, with the exception of adhesiveness. In sensory evaluation, color and texture were significantly different according to the samples, and the differences were found to be affected more by the origin of soybeans rather than the cultivation methods. According to this study, the physicochemical properties of the soybeans and tofu seemed to be influenced by the cultivation region rather than the methods.

갈색 느티만가닥버섯 추출물의 부위별 항산화 활성 (Antioxidant activities of brown beech mushroom (Hypsizygus marmoreus) pileus and stipe)

  • 본 연구에서는 기능성 식의약품 소재로써 갈색 느티만가닥버섯(Hypsizygus marmoreus)의 이용 가능성을 조사하기 위해 갈색 느티만가닥버섯의 부위별, 추출 용매별 항산화 활성을 조사하였다. 열수 추출물 갓과 대의 총 폴리페놀 함량은 17.15±0.19 mg GAE/g과 7.37±0.16 mg GAE/g이었으며 에탄올 추출물 갓과 대의 총 폴리페놀 함량은 각각 10.23±0.14 mg GAE/g과 3.76.±0.19 mg GAE/g으로 에탄올 추출물에 비해 열수 추출물의 폴리페놀 함량이 높게 나타났고 대에 비해 갓의 폴리페놀 함량이 높게 나타났다. 또한 갈색 느티만가닥버섯 추출물의 DPPH, ABTS, ORAC 지수, 환원력도 10 mg/ml의 농도에서 에탄올 추출물에 비해 열수 추출물에서 높게 나타났고 대에 비해 갓에서 높게 나타났으며 흰색 느티만가닥버섯에 비해 갈색 느티만가닥버섯의 총 폴리페놀 함량, DPPH, ORAC 지수, 환원력이 높은 것으로 나타났다. 추출물의 세포독성은 WST-1 assay (4-[3-(4-iodophenyl)-2-(4-nitrophenyl)-2H-5-tetrazolio]-1,3-benzene disulphonate)를 이용하여 열수 추출물의 처리 농도에 따른 대식세포주 RAW 264.7의 세포 생존율로 확인하였으며 갈색 느티만가닥버섯 열수 추출물 처리구에서 대식세포주 RAW 264.7의 세포 생존율이 증가하였기 때문에 세포독성은 없는 것으로 판단된다. 이상의 결과를 종합하면, 느티만가닥버섯은 품종에 따라 항산화 활성에 차이가 있고, 갈색 느티만가닥버섯은 다른 버섯보다 항산화 활성이 높기 때문에 기능성 식의약품 소재로서 이용 가능성이 있다고 판단된다.

표고 톱밥재배용 무포자 신품종 '대담' 육성 및 특성 (Characteristics of newly bred spore-less cultivar Lentinula edodes 'Daedam' for sawdust cultivation)

  • 표고 톱밥재배용 무포자 품종의 육성을 위해 수집균주 중 우량계통의 단포자 교배를 통해 육성된 '대담'의 주요 특성은 다음과 같다. '대담'의 균사생장 적온은 22~.25℃, 발이 및 생육온도는 16~18℃이었으며, 자실체 형태는 평반구형이며 갓을 형성하지 않는 무포자품종이다. '대담'은 배양기간이 21일, 갈변기간 96일로 대조품종('화담')과 유사하였고, 발이기간은 5일, 생육기간은 9일로 전체 재배기간은 134일로 같았다. '대담'의 개체중은 32.4g으로 대조품종과 유사하였고, 유효개체수는 6.8개로 대조품종 보다 1.3개 낮았으며, 총수량은 222g으로 대조품종의 266g에 비해 낮았다. '대담'의 자실체는 대조품종 대비 갓이 두껍고, 갓 직경은 다소 작았으며, 대의 굵기는 작고 긴 특성을 보였다. '대담'의 배지형태별 생산성 분석결과, 1.2kg 원통형 배지에서 생물학적 효율이 26.7%로 3kg 봉형배지보다 높았으며, '하나참'와 비교 시 재배기간이 40일 짧고 수량도 유사하여, 단기 재배용 품종으로 활용이 가능할 것으로 생각된다.

국내산 감자 품종별 영양 성분 비교 (Nutrient Composition of Domestic Potato Cultivars)

  • The proximates, vitamin C, minerals, and fatty acids of five potato cultivars were evaluated by AOAC methods, the hydrazine method, ICP-AES, and gas chromatography, respectively. The proximate analyses; vitamin C, reducing sugar, and soluble solid contents; and mineral and fatty acid compositions were significantly different among the five cultivars. The Superior cultivar contained a higher carbohydrate content and higher Ca/P ratio and lower levels of crude protein and Na. The Atlantic cultivar contained significantly higher amount of energy, carbohydrate, reducing sugar, vitamin C, SEA, and MUFA, and significantly lower amount of minerals and PUFA. The Shepody cultivar contained significantly higher amount of carbohydrate and MUFA, and significantly lower amount of soluble solid, vitamin C, and SFA. In addition, the P, Fe, Mg, Cu, and Al levels were significantly higher in Shepody, and Zn content was significantly lower. The Jopung cultivar contained significantly higher levels of moisture and Na, and significantly lower levels of soluble content, reducing sugar, carbohydrate, crude protein, and fat. Finally, the Namsuh cultivar contained significantly higher amount of soluble solid, crude protein, K, Mg, and Al, and significantly lower amount of reducing sugar.

Biological and Structural Mechanisms of Disease Development and Resistance in Chili Pepper Infected with the Root-knot Nematode

  • Biological and structural mechanisms of the nematode disease development in chili pepper caused by the root-knot nematode, Meloidogyne incognita, were investigated. Out of 39 chili pepper cultivars/lines tested, six were found resistant, while 33 were susceptible to M. incognita, of which a susceptible cultivar Chilseongcho and three resistant cultivar/lines CM334, 02G132 and 03G53 with different resistance degrees were selected for microscopic studies on the disease development. Gall formation was greatly reduced in the resistant cultivars/lines. Nematode penetration occurred both in the susceptible and resistant chili pepper roots; however, the penetration rates were significantly lowered in the three resistant peppers compared to the susceptible pepper cv. Chilseongcho. In the susceptible pepper, giant cells were extensively formed with no discernible necrosis around the nematode feeding sites. In the highly resistant pepper cultivar CM334, no giant cell was formed, but extensive necrosis formation was observed around the penetrating nematodes. In the other two resistant pepper lines (02G132 and 03G53), both giant cells and prominent necroses were formed, and the necrotic responses appeared to inhibit the further development of giant cells or accelerate their early degeneration. Although the nematode penetration was retarded significantly in the resistant cultivar/lines, all of the above results suggest that the disease resistance of pepper may be related to post-infectional defense mechanisms (nematode growth and development) more than pre-infectional ones (penetration and establishment). Variations in structural modifications in the resistant cultivar/lines may reflect their genetic differences related to the nematode resistance.

저선량의 이온화방사선이 대두품종의 생육과 수량에 미치는 효과 (Effects of Low Dose Ionizing Radiation on the Growth and Yield of Soybean Cultivars)

  • 대두작물의 생육에 대한 방사선 hormesis를 구명하고자 세 품종의 콩종자를 저선량 ${\gamma}$선을 조사하여 포장에서 시험하였다. 생육에 대한 저선량 ${\gamma}$선의 자극효과는 품종에 따라서 다른 반응을 보였는데, 백태인 황금콩과 방사콩보다도 흑태인 S900 품종에서 더욱 효과적이었다. 포장발아율은 S900 품종의 0.5Gy 조사구에서 30%정도, 경장에서는 방사콩과 S900 품종의 4.0G 조사구에서 15%정도 증가하였다. 꼬투리수와 종자 수량에서는 황금콩은 2.0G 조사구에서 35%정도, S900 품종은 4.0Gy 조사구에서 25% 정도 증가하였다.

Molecular discrimination of Panax ginseng cultivar K-1 using pathogenesis-related protein 5 gene

  • Background: The mixed-cultivation of different Panax ginseng cultivars can cause adverse effects on stability of yield and quality. K-1 is a superior cultivar with good root shape and stronger disease resistance. DNA markers mined from functional genes are clearly desirable for K-1, as they may associate with major traits and can be used for marker-assisted selection to maintain the high quality of Korean ginseng. Methods: Five genes encoding pathogenesis-related (PR) proteins of P. ginseng were amplified and compared for polymorphism mining. Primary, secondary, and tertiary structures of PR5 protein were analyzed by ExPASy-ProtParam, PSSpred, and I-TASSER methods, respectively. A coding single nucleotide polymorphism (SNP)-based specific primer was designed for K-1 by introducing a destabilizing mismatch within the 3' end. Allele-specific polymerase chain reaction (PCR) and real-time allele-specific PCR assays were conducted for molecular discrimination of K-1 from other cultivars and landraces. Results: A coding SNP leading to the modification of amino acid residue from aspartic acid to asparagine was exploited in PR5 gene of K-1 cultivar. Bioinformatics analysis showed that the modification of amino acid residue changed the secondary and tertiary structures of the PR5 protein. Primer KSR was designed for specific discrimination of K-1 from other ginseng cultivars and landraces. The developed real-time allele-specific PCR assay enabled easier automation and accurate genotyping of K-1 from a large number of ginseng samples. Conclusion: The SNP marker and the developed real-time allele-specific PCR assay will be useful not only for marker-assisted selection of K-1 cultivar but also for quality control in breeding and seed programs of P. ginseng.

Open-Top chamber 내(內)에서 오존에 노출(露出)시킨 현사시 5개(個) 클론의 생장량(生長量)과 오존에 대(對)한 민감성(敏感性)과의 관계(關係) (Sensitivity of Five Clones of Populus alba × P. glandulosa Cuttings to Ozone Exposure in Open-Top Chambers in Relation to Their Growth Rates)

  • 본 연구는 수목의 오존에 대한 피해정도가 수목 고유의 생장속도와 관련되어 있는지를 검증하기 위하여 실시하였다. 생장이 우수한 현사시(Populus alba ${\times}$ P. glandulosa) 4호 품종 중에서 두 개 클론(72-30, 72-16)과 생장이 비교적 저조한 현사시 2호 품종 중에서 3개 클론(72-28, 72-27, 72-19)을 본 실험에서 비교하였다. 위의 5개 클론의 삽수를 4월초 $2{\ell}$ 화분에 삽목하여 증식시킨 후, 8월 하순에 야외에서 3개의 open top chamber(직경 2.5m, 높이 2.0m) 내에서 30일 동안 70ppb와 130ppb의 오존에 1일 5시간씩 노출하였다. 대조구는 활성탄으로 여과하여 오존의 농도가 30ppb이하로 유지되도록 하였으며, 각 클론별, 처리별로 20개의 화분을 반복으로 사용하였다. 대조구에서 두 품종간의 생장비교는 현사시 4호가 현사시 2호보다 잎 건중량에서 73%, 뿌리 건중량에서 64%, 총건중량에서 38% 더 우수하여 현사시 4호의 생장속도가 2호보다 우수함을 입증하였다. 오존에 의한 가시적 피해는 70ppb 오존처리에서는 나타나지 않았으며, 130ppb 처리에서 현사시 4호에서만 관찰되었으며, 성숙잎에서 전형적인 작은 갈색 반점들로 나타났으며, 성숙잎의 조기낙엽을 가져왔다. 70과 130ppb 오존처리는 5개 모든 클론의 수고생장, 잎과 뿌리의 건중량과 총건중량을 감소시켰는데, 이 중에서 뿌리생장을 가장 크게 감소시켰다. 70ppb 오존에서 뿌리의 감소량은 현사시 4호가 39.7%이고, 현사시 2호는 13.8%이었다. 130ppb 오존에서도 현사시 4호의 뿌리감소량은 47.4%이고, 현사시 2호는 34.9%이었다. 이로 인하여 shoot/root율은 130ppb에서 현사시 4호는 63.4% 증가한 반면, 현사시 2호는 22.1% 증가에 그쳤다. 오존에 의한 가공전도도와 순광합성량의 변화에서도 현사시 4호가 현사시 2호보다 더 많이 감소하였다. 특히 130ppb 오존에서 현사시 4호의 순광합성량은 69.5% 감소한 반면에, 현사시 2호는 31.5% 감소하였다. 따라서 위의 여러 가지 생리적 반응을 근거로 하여 생장이 우수한 현사시 4호가 생장이 상대적으로 적은 현사시 2호보다 오존에 더 민감하다고 판단할 수 있으며, 수목의 오존에 대한 피해정도는 수목 고유의 생장속도가 관련되어 있다고 결론 짓는다.

Comparative Analysis of Defense Responses in Chocolate Spot-Resistant and -Susceptible Faba Bean (Vicia faba) Cultivars Following Infection by the Necrotrophic Fungus Botrytis fabae

  • In this study, resistance responses were investigated during the interaction of Botrytis fabae with two faba bean cultivars expressing different levels of resistance against this pathogen, Nubaria (resistant) and Giza 40 (susceptible). Disease severity was assessed on leaves using a rating scale from 1 to 9. Accumulation levels of reactive oxygen species (ROS), lipid peroxidation and antioxidant enzymes (superoxide dismutase, catalase and ascorbate peroxidase) were measured in leaf tissues at different times of infection. The expression profiles of two pathogenesis-related proteins (PRPs) encoded by the genes PR-1 and ${\beta}$-1,3-glucanase were also investigated using reverse transcription RT-PCR analysis. The accumulation of these defense responses was induced significantly in both cultivars upon infection with B. fabae compared with un-inoculated controls. The resistant cultivar showed weaker necrotic symptom expression, less ROS accumulation, a lower rate of lipid peroxidation and higher activity of the enzymatic ROS scavenging system compared with susceptible cultivar. Interestingly, ROS accumulated rapidly in the resistant leaf tissues and peaked during the early stages of infection, whereas accumulation was stronger and more intense in the susceptible tissues in later stages. Moreover, the response of the resistant cultivar to infection was earlier and stronger, exhibiting high transcript accumulation of the PR genes. These results indicated that the induction of oxidant/antioxidant responses and the accumulation of PRPs are part of the faba bean defense mechanism against the necrotrophic fungus B. fabae with a different intensity and timing of induction, depending on the resistance levels.

Comparison of rice flour properties of different cultivars using wet and dry milling processes

  • We conducted to compare the characteristics of rice flours according to the different milling processes. Five rice varieties (Oryza sativa L.) with different amylose content were prepared by wet and dry milling processes. The moisture contents of wet-milled rice flours (WMR) was mostly three-time higher than those of dry-milled flours (DMR). Water absorption index (WAI), water solubility index (WSI) and swelling power (SP) increased in proportion to temperature ($50-90^{\circ}C$). WAI, WSI, SP of DMR showed higher value than those of WMR. Baeokchal (BOC) which is waxy rice cultivar was significantly high level of WSI. Pasting properties of DMR except BOC cultivar resulted in higher peak viscosity, trough viscosity, final viscosity and Setback. The levels of resistant starch in the four cultivars except Dodamssal (DDS) were under 1% irrespective of Milling processes, whereas the resistant starch contents of DMR and WMR in DDS was 9.18 and 6.27, respectively. Damaged starch content of WMR were less than those of DMR, moreover, negative correlation was observed between amylose content and damaged starch of rice cultivars. These results suggest that the properties of rice flour varied depending on the milling methods and varieties, and it could be a reference for selecting the appropriate processing purposes.
