The Controlling Activity of Several Fungicides against Rice Bakanae Disease Caused by Fusarium fujikuroi in Five Assay Methods (몇 가지 살균제의 벼 키다리병과 병원균에 대한 효과 검정)

  • Recently damage of rice bakanae disease disseminated by infected seeds increased in paddy field in Korea. For controlling rice bakanae disease, the efficacy of 17 fungicides was assessed by 5 kinds of bioassay, spore germination test (SGT), mycelial growth test, detection test on Komada's medium (KDT), pouch test (PT) and greenhouse test (GT). Among ergosterol biosynthesis inhibiting fungicides, prochloraz showed a high controlling activity in all the assay systems while the others showed very low activity except for $500\;{\mu}g/ml$ of hexaconazole in GT and $500\;{\mu}g/ml$ of triflumizole in KDT. Although benomyl and the mixture of benomyl and thiram showed a good activity at 100 and $500\;{\mu}g/ml$ in SGT and PT, respectively, in GT they did a middle activity. Trifloxystrobin and kresoxim-methyl included in strobilurins showed a good activity even at $20\;{\mu}g/ml$ in KDT as well as a middle activity in SGT. Also a high activity not only at $10\;{\mu}g/ml$ in SGT but also at $100\;{\mu}g/ml$ KDT was detected in thiram. The activity of fludioxonil was confirmed in SGT, KDT and PT. Based on these results, it is very important to determine a bioassay system, because the fungicidal activity against rice bakanae disease was fluctuated depending on a assay systems as well as the mechanism of fungicide.

  • Icariside II (ICA II) is used in erectile dysfunction treatment. Adipose tissue-derived stem cells (ADSCs) are efficient at improving erectile function. This study aimed to explore the action mechanism of ADSCs in improving erectile function. ADSCs were isolated from the adipose tissues of rats. Cell proliferation was determined using the Cell Counting Kit-8 (CCK-8) assay. The expressions of mRNA and protein were determined separately through qRT-PCR and western blot. The endogenous expressions of related genes were regulated using recombinant plasmids and cell transfection. A Dual-Luciferase Reporter Assay was performed to determine the interaction between miR-34a and STAT3. Rat models with bilateral cavernous nerve injuries (BCNIs) were used to assess erectile function through the detection of mean arterial pressure (MAP) and intracavernosal pressure (ICP). ICA II promoted ADSCs' proliferation and differentiation to Schwann cells (SCs) through the inhibition of miR-34a. Suppressed miR-34a promoted the differentiation of ADSCs to SCs by upregulating STAT3. ICA II promoted the differentiation of ADSCs to SCs through the miR-34a/STAT3 pathway. The combination of ICA II and ADSCs preserved the erectile function of the BCNI model rats. ADSCs treated with ICA II markedly preserved the erectile function of the BCNI model rats, which was reversed through miR-34a overexpression. ICA II promotes the differentiation of ADSCs to SCs through the miR34a/STAT3 pathway, contributing to erectile function preservation after the occurrence of a cavernous nerve injury.

  • As pervious the SAR(Security Aware Routing)[10] protocol is an secure Ad Hoc network protocol that finds a secure path, it is the security routing protocol that uses the security level of nodes as the routing information. However, the SAR protocol sometimes transfers data through inefficient transmission paths because it always tries to find secure nodes for a safe transmission. Since it is a protocol based on AODV[6], it will cause transmission delay as researching of security routing path. when a node is out of the data transmission range as its battery dying or movement. Although it is possible to connection of nodes because a characteristic of the SAR protocol, the connection is not easy to reconnect when the security level of intermediate node is lower than the level requested by a source node. In this paper, we suggest the MP-SAR based on the SAR to solve the SAR protocol's problem. The MP-SAR seeks multiple secure path for maintenance of data confidentiality using the expanded secure path detection techniques based on the SAR. It can transfer data quickly and reliably by using the shortest efficient path among multiple paths. In the research result, we proved a outstanding performance of MP-SAR than the previous SAR through comparison and analysis.

  • Among Gram-negative pathogens in Korea, the incidence of resistance to third generation cephalosporins is becoming an ever-increasing problem. The production of extended-spectrum ${\beta}$-lactamase (ESBL) is the main mechanism of bacterial resistance to a third-generation cephalosporins and monobactams. Accurate identification of the ESBL genes are necessary for surveillance and epidemiological studies of the mode of transmission in the hospital. This study was conducted to detect the genes encoding ESBL of 46 K. penumoniae isolated from Daejeon, Chungnam and Chungbuk regional university hospitals from February to August in 2012. The phenotypes of the isolated specimens were examined according to the combination disc test (CDT) by the Clinical and Laboratory Standards Institute (CLSI). Forty two ESBL producing K. penumoniae isolates could be detected using ceftazidime (CAZ) discs with and without clavulanate (CLA). By CDT, 42 K. pneumoniae strains were confirmed to be ESBL strains. Genotyping was performed by multiplex PCR with type-specific primers. By PCR analysis, TEM gene in 46 strains, SHV gene in 37 strains and CTX-M genes in 14 strains were identified. Ten isolates did carry genes encoding ESBLs of all types TEM, SHV and CTX-M. The multiplex polymerase chain reaction (PCR) analysis was better to detect and differentiate ESBL producing K. penumoniae strains in clinical isolates.

Change Detection of Damaged Area and Burn Severity due to Heat Damage from Gangwon Large Fire Area in 2019 (2019년 강원도 대형산불지역의 열해 피해로 인한 피해강도 변화 탐색)

  • The purpose of this study is to detect the burned area change by direct burning of tree canopies and post-fire mortality of trees via analyzing satellite imageries from the Korea multi-purpose satellite-2 and -3 (KOMPSAT-2 and -3) for two large-fires over the Goseong-Sokcho and Gangneung-Donghae regions in April 2019. For each case, the burned area was compared between two dates: the day when the fire occurred and 15-18 days after it. As the results, within these two dates, there was no substantial difference in burned area of sites whose severities were marked as "Extreme", but sites with "High" and "Low" severities showed significant differences in burned area between the two dates. These differences were resulted from the lagged post-fire browning of canopies which was detected by images from in-situ observation,satellite, and the unmanned aerial vehicle. The post-fire browning started after 3-4 days and became apparent after 10-15 days. This study offers information about the timing to quantify the burned area by large fire and about the mechanism of post-fire mortality. Also, the findings can support policy makers in planning the restoration of the damaged areas.

  • Eosinophilic gastroenteritis, which shows characteristic eosinophilic inflammation, involves any part of the intestine from esophagus to colon. The immunopathogenesis is expected to be associated with IgE-mediated or non-IgE-mediated reaction, but the precise mechanism is not revealed yet. The clinical manifestation is variably dependent on the extent of eosinophilic infiltration. Usually the symptoms, such as abdominal pain, diarrhea, vomiting, wax and wane for a few months result in failure to thrive, which implicates the importance of early detection. Although the diagnosis is made through clinical and histopathologic evidences, we should suspect the illness in a case of anemia, hypoalbuminemia, and peripheral eosinophilia. Recently, we experienced a case of non-IgE-mediated eosinophilic colitis in a 11-year-old male who complained of diarrhea, right lower abdominal pain, body weight loss, was diagnosed with peculiar histopathologic finding of sigmoid colon specimens obtained by sigmoidoscopy.

Supplementation of the Indoor Location Tracking Techniques Based-on Load-Cells Mechanism (로드셀 기반의 실내 위치추적 보완 기법)

  • Current indoor intrusion detection and location tracking methods have the weakness in seamless operations in tracking the objective because the object must possess a communicating device and the limitation of the single cell size (approximate $100cm{\times}100cm$) exits. Also, the utilization of CCTV technologies show the shortcomings in tracking when the object disappear the area where the CCTV is not installed or illumination is not enough for capturing the scene (e.g. where the context-awarded system is not installed or low illumination presents). Therefore, in this paper we present an improved in-door tracking system based on sensor networks. Such system is built on a simulated scenario and enables us to detect and extend the area of surveillance as well as actively responding the emergency situation. Through simulated studies, we have demonstrated that the proposed system is capable of supplementing the shortcomings of signal cutting, and of estimating the location of the moving object. We expect the study will improve the better analysis of the intruder behavior, the more effective prevention and flexible response to various emergency situations.

  • The mission-critical engagement control computer for air defense has to maintain its operation without any fault for a long mission time. The mission performed by large-scale and complex embedded software is extremely critical in terms of dependability and safety of computer system, and it is very important that engagement control computer has high reliability. The engagement control computer was implemented using four processors. The distributed computer composed of four processors quarantees the dependability and safety, and ASR fault-tolerant technique applied to each processor guarantees the reliability. In this paper, the mechanism and performance of ASR fault-tolerant technique are analysed. And MTBF, reliability, availability, and cost-effectiveness for ASR, DMR and TMR techniques applied to the engagement control computer are analysed. The mission-critical engagement control computer using software-based ASR fault-tolerant technique provides high reliability and fast recovery time at a low cost. The mission reliability of the engagement control computer using ASR technique in 4 processors board is almost same the reliability of the computer using TMR technique in 6 processors board. ASR technique is most suitable to the mission-critical engagement control computer.

Effects of exploration and molecular mechanism of CsV on eNOS and vascular endothelial functions

  • This study aimed to investigate the effects and potential mechanisms of Chikusetsusaponin V (CsV) on endothelial nitric oxide synthase (eNOS) and vascular endothelial cell functions. Different concentrations of CsV were added to animal models, bovine aorta endothelial cells (BAECs) and human umbilical vein endothelial cells (HUVECs) cultured in vitro. qPCR, Western blotting (WB), and B ultrasound were performed to explore the effects of CsV on mouse endothelial cell functions, vascular stiffness and cellular eNOS mRNA, protein expression and NO release. Bioinformatics analysis, network pharmacology, molecular docking and protein mass spectrometry analysis were conducted to jointly predict the upstream transcription factors of eNOS. Furthermore, pulldown and ChIP and dual luciferase assays were employed for subsequent verification. At the presence or absence of CsV stimulation, either overexpression or knockdown of purine rich element binding protein A (PURA) was conducted, and PCR assay was employed to detect PURA and eNOS mRNA expressions, Western blot was used to detect PURA and eNOS protein expressions, cell NO release and serum NO levels. Tube formation experiment was conducted to detect the tube forming capability of HUVECs cells. The animal vasodilation function test detected the vasodilation functions. Ultrasonic detection was performed to determine the mouse aortic arch pulse wave velocity to identify aortic stiffness. CsV stimulus on bovine aortic cells revealed that CsV could upregulate eNOS protein levels in vascular endothelial cells in a concentration and time dependent manner. The expression levels of eNOS mRNA and phosphorylation sites Ser1177, Ser633 and Thr495 increased significantly after CsV stimulation. Meanwhile, CsV could also enhance the tube forming capability of HUVECs cells. Following the mice were gavaged using CsV, the eNOS protein level of mouse aortic endothelial cells was upregulated in a concentration- and time-dependent manner, and serum NO release and vasodilation ability were simultaneously elevated whereas arterial stiffness was alleviated. The pulldown, ChIP and dual luciferase assays demonstrated that PURA could bind to the eNOS promoter and facilitate the transcription of eNOS. Under the conditions of presence or absence of CsV stimulation, overexpression or knockdown of PURA indicated that the effect of CsV on vascular endothelial function and eNOS was weakened following PURA gene silence, whereas overexpression of PURA gene could enhance the effect of CsV upregulating eNOS expression. CsV could promote NO release from endothelial cells by upregulating the expression of PURA/eNOS pathway, improve endothelial cell functions, enhance vasodilation capability, and alleviate vessel stiffness. The present study plays a role in offering a theoretical basis for the development and application of CsV in vascular function improvement, and it also provides a more comprehensive understanding of the pharmacodynamics of CsV.

Epigallocatechin-3-gallate suppresses hemin-aggravated colon carcinogenesis through Nrf2-inhibited mitochondrial reactive oxygen species accumulation

  • Background: Previous studies have presented evidence to support the significant association between red meat intake and colon cancer, suggesting that heme iron plays a key role in colon carcinogenesis. Epigallocatechin-3-gallate (EGCG), the major constituent of green tea, exhibits anti-oxidative and anti-cancer effects. However, the effect of EGCG on red meat-associated colon carcinogenesis is not well understood. Objectives: We aimed to investigate the regulatory effects of hemin and EGCG on colon carcinogenesis and the underlying mechanism of action. Methods: Hemin and EGCG were treated in Caco2 cells to perform the water-soluble tetrazolium salt-1 assay, lactate dehydrogenase release assay, reactive oxygen species (ROS) detection assay, real-time quantitative polymerase chain reaction and western blot. We investigated the regulatory effects of hemin and EGCG on an azoxymethane (AOM) and dextran sodium sulfate (DSS)-induced colon carcinogenesis mouse model. Results: In Caco2 cells, hemin increased cell proliferation and the expression of cell cycle regulatory proteins, and ROS levels. EGCG suppressed hemin-induced cell proliferation and cell cycle regulatory protein expression as well as mitochondrial ROS accumulation. Hemin increased nuclear factor erythroid-2-related factor 2 (Nrf2) expression, but decreased Keap1 expression. EGCG enhanced hemin-induced Nrf2 and antioxidant gene expression. Nrf2 inhibitor reversed EGCG reduced cell proliferation and cell cycle regulatory protein expression. In AOM/DSS mice, hemin treatment induced hyperplastic changes in colon tissues, inhibited by EGCG supplementation. EGCG reduced the hemin-induced numbers of total aberrant crypts and malondialdehyde concentration in the AOM/DSS model. Conclusions: We demonstrated that EGCG reduced hemin-induced proliferation and colon carcinogenesis through Nrf2-inhibited mitochondrial ROS accumulation.
