Desmutagenic Effects of Extracts from Green Tea (녹차 추출물의 항돌연변이원성)

  • Desmutagenic effects of water-soluble and ethanol-soluble extracts of dried green tea toward the mutagenicity of 4-nitroquinoline-N-oxide(4-NQO) and 3-Amino-1,4-dimethyl-5H-pyrido[4,3-b]indole(Trp-P-1) in streptomycin-dependent SD510 strains of Salmonella typhimurium TA98 were investigated. Inhibition effects toward the mutagenicity of 4-NQO and Trp-P-1 of water-soluble and ethanol-soluble extracts from green tea were high as increase of concentration of extracts. Desmutagenic effects toward 4-NQO of water-soluble and ethanol-soluble extracts from green tea harvested in May and August were up to 93% in 1,000 $\mu\textrm{g}$ of extract/plate. Desmutagenic effects toward Trp-P-1 of ethanol-soluble extracts from green tea were 53.3∼92.1%, but the effects of water-soluble extracts decreased as increase of concentration of extracts.

  • Desmutagenic effects of water-soluble and water-insoluble fractions from Aloe vera and Aloe avoresence with different solvents against the mutagenicity of 4-nitro-o-phenylenediamine(NPD) were investigated. Desmutagenic effects of water-insoluble fractions from Aloe vera and Aloe avoresence against NPD-induced mutations in TA97 and TA98 strains were very high compared with water-soluble fractions: whereas water-soluble fractions from two kinds of Aloes showed the inhibition effects of 4∼56% water-insoluble fractions showed the activities of 54∼100% for Aloe vera and 74∼100% for Aloe avoresence. Especially, the addition of 20 $\mu$ι of water-insoluble fractions per plate showed high activities up to 90% in TA97 and TA98 strains.

  • The desmutagenic effects of seaweed and vegetable extracts were investigated on the mutagenicity of Maillard reaction products (MRP) obtained from equimolar amounts of glucose and amino acid (arginine and lysine${\cdot}$HCl) for Salmonella typhimurium TA 100 without S9 mix. The mutagenicities were inhibited by water-soluble extracts of seaweeds(laver, sea-straghorn, sea-mustard and tangle) and vegetables(ginger, garlic, onion, chinese-pepper, green-onion and cabbage). Cabbage, chinese-pepper, green-onion and sea-straghorn exhibited especially high desmutagenic effects. The desmutagenicities of these extracts(cabbage, green-onion and sea-straghorn) except for sea-straghorn were decreased by heat treatment at $100^{\circ}C$ for 10 min. It is assumed that the desmutagenic effect of seaweed and vegetable extract is due to the reducing power and action of enzyme such as peroxidase and catalase.

  • The desmutagenic effects of ${\alpha}$-hydroxycarbonyl compounds, such as glyceraldehyde, glycolaldehyde, dihydroxyacetone, furfural, 5-hydroxymethylfurfural, maltol, acetol and acetoin and ${\alpha}$-dicarbonyls, such as diacetyl, glyoxal, methyl glyoxal and 2, 3-pentanedione were investigated against the mutagenic heterocyclic amines, such as Trp-P-1, Trp-P-2, Glu-P-1, Glu-P-2 and IQ. Most of the carbonyl compounds suppressed the mutagenicity of heterocyclic amines for S. typhimurium TA98, ${\alpha}$-dicarbonyl compounds showing a higher desmutagenic effect than ${\alpha}$-hydroxycarbonyl compounds. Among the ${\alpha}$-hydroxycarbonyl compounds, glyceraldehyde, glycolaldehyde and dihy-droxyacetone showed more effective desmutagenicity, and diacetyl among the ${\alpha}$-dicabonyl compounds had the highest desmutagenic effect. These carbonyl compounds alone also showed mutagenicity to S. typhimurium TA100 without S-9 mix. The reaction of carbonyl compounds with mutagenic heterocyclic amines also eliminated the mutagenicity of the former for S. typhimurium TA100.

Desmutagenic Effect of Water Extract from Areca catechu L. on the Mutagenicity of N-Methyl-N-Nitro-N'-Nitrosoguanidinein in E. coli PQ37 (N-methyl-N-nitro-N'-nitrosoguanidine의 변이원성에 대한 빈랑 물 추출물의 돌연변이 억제효과)

  • The desmutagenic activity of the water extract of Areca catechu L. on the mutagenicity induced by aflatoxin $B_1$ ($AFB_1$), N-methyl-N-nitro-N'-nitrosoguani-dine (MNNG), mitomycin C (MMC) and 4-nitroquinoline 1-oxide (4-NQO) was studied by using the SOS Chromotest with Escherichia coli PQ37. The inhibition rates of water extract of Areca catechu L. at concentration of $100{\mu}g/assay$ were 41.0%, 47%, 46%, and 32% against $AFB_1$, MNNG, MMC and 4-NQO, respectively. The water extract of Areca catechu L. was separated into methanol soluble and methanol insoluble parts. The methanol insoluble part exhibited higher inhibition effect than the methanol soluble part against the mutagenic activities of MNNG. Step-wise fractionation of methanol insoluble part was done to obtain methanol, ethyl acetate and water fractions. Among these fractions, water fraction had the strongest inhibitory effect of 45.0% against mutagenicities of MNNG. The inhibition rates of aqueous fraction of methanol-insoluble from water extracted Areca catechu L. at concentrations of 1.61, 16.13, 161.29 and $322.58{\mu}g/mL$ were 12.0%, 24.0%, 47.5% and 62.0%, respectively. The water fraction showed the inhibitory effects with dose response against the mutagenic activity induced by MNNG.

Desmutagenic Effect of Water Extract from Artemisia capillaris THUNB on the Mutagenicity of Benzo[a]pyrene (Benzo[a]pyrene의 변이원성에대한 인진쑥 물 추출물의 항돌연변이 효과)

  • The antimutagenic activity of the extract of Artemisia capillaris THUNB on the mutagenicity induced by benzo(a)pyrene [B(a)P] in the presense of S9 mixture was studied using bacterial mutagenic assay system. Samples harvested in summer and autumn were extracted using ethanol and hot water. Among these extracts the water extract of summer sample had the strongest inhibitory effect against the mutagenenicity of B(a)P, The water extract of Artemisia capillaris THUNB was separated again into ethanol soluble and insoluble parts. The ethanol insoluble part(El) of water extract exhibited higher inhibition effects than the ethanol soluble part against the mutagenic activity of B(a)P. El showed dose-dependent activity on the mutagenicity of B(a)P in SOS Chromotest and Ames test. The 50% inbibition concentraction $(IC_{50}$ of El were $200{\mu}g/assay$ $600{\mu}g/plate$ and $800{\mu}b/plate$ in E. coil PQ37 S. typhimurium TA100 and TA98 respectively. El were showed desmutagenic effect but had no effect on the DNA repair system for B(a)P-induced mutagenesis. HPLC analysis showed that the formation of aflatoxin M1 by cytochrome P-450 1A1 known as playing an impotant role on B(a) P-induced mutagenicity was highly inhibited by El. Therefore we encluded that B(a)P-induced mutagencity can be reduced possible due to the interference of el with cytochrome P-450 1A1-dependent bioactivation.

  • The desmutagenic activity of the water extract of Cassia tara L on the mutagenicity induced by 4-nitroquinoline 1-oxide (4-NQO) and N-methyl-N-nitro-N'-nitrosoguani-dine (MNNG) was studied using the SOS Chromotest with Escherichia coli PQ37. The inhibition rates of water extract of Cassia tara L. at concentration of $100{\mu}g$/assay were 27.5% and 40% against 4-NQO and MNNG. The water extract of Cassia tara L. was separated into methanol soluble and methanol insoluble parts. The methanol soluble part exhibited higher inhibition effects than the methanol insoluble part against the mutagenic activities of 4-NQO and MNNG. Step-wise fractionation of methanol soluble part was done to obtain methanol, ethyl acetate and water fractions. Among these fractions, water fraction had the strongest inhibitory effects of 23.0 and 19.0% against mutagenicities of MNNG and 4NQO, respectively. The results clearly indicated that the water fraction showed much stronger antimutagenicity against MNNG than 4NQO. The inhibition rates of aqueous fraction of methanol-soluble from water extracted Cassia tara L. at concentrations of 1.0, 10, 100 and $250{\mu}g$/assay were 8.0%, 12.0%, 25.5% and 43.0%, respectively. The water fraction showed the inhibitory effects with dose response against the mutagenic activities induced by MNNG.

Inhibitory Mechanism of Colored Rice Bran Extract Against Mutagenicity Induced by Chemical Mutagen Mitomycin C (유색미 쌀겨 추출물의 화학적 변이원 mitomycin C에 대한 변이원성 억제기작)

  • Inhibitory mechanism of colored rice bran against cellular genotoxicity induced by chemical mutagen was studied using organic solvent extracts from a colored rice cultivar termed as Suwon415, and the mutagen, mitomycin C. Inhibitory effects of 70% ethanol extact and chloroform fraction from rice bran of Suwon415 were higher than those from Chuchung used as control. However, antioxidative activities of each fraction from Suwon415 were slightly lower than those from Chuchung, suggesting the involvement of a different inhibitory mechanism not related to antioxidation pathway. Using E. coli as the indicator cell, inhibitory mechanism of rice bran extract from colored rice against mutagenicity induced by mitomycin C was investigated to reveal the possibility that it acts in a desmutagenic manner. Further investigation to quantify the free mitomycin C in reaction mixture following incubation with rice bran extract demonstrated that rice bran extract might inhibit the cellular genotoxicity of mitomycin C by direct adsorption of the mutagen.

Inhibitory Effect of Methanol Extract of Eleutherococcus senticosus Maxim. on the Direct Mutagen Mutagenicity (직접 돌연변이원에 대한 가시오갈피 추출물의 항돌연변이 효과)

  • Antimutagenic effects og Eleutherococcus senticosus Maxim. on the mutagenicty induced by mutagens, 4-NQO, MNNG, $\textrm{NaN}_{3}$, 2-NF, and 1-NP was studied by the Ames test with Salmonella typhimurium TA98 and TA100. The methanol extract ($500\mu\textrm{g}$/plate) of E. senticosus Maxim. showed inhibitory effect on the mutagenicty induced by 1-NP only among the tested mutagens. In S. typhimurium TA98, the methanol extracts of the root, stem and leaf showed inhibitory effects of 54.9, 29.5, and 32.9% inhibition on 1-NP mutagenicity, respectively. In S typhimurium TA100, the methanol extracts of the root, stem, and leaf showed inhibitory effects of 593, 30.2, and 43.6%, respectively. The methanol extract were further fractionated by a subsequent liquid-liquid partition technique with chloroform, butanol, and water. The chloroform ($300\mu\textrm{g}$/plate) fraction of the root, stem and leaf showed the strong antimutagenic effects on the mutagenicity induced by 1-NP in S typhimurium TA98 and TA100. But none or weak antimutagenicities were observed in the butanol and aqueous fraction. The chloroform fractions of root, stem and leaf showed the antimutagenic effects of 61.6~88.6% in a dose-dependent manner. In the antimutagenic mode test, the inhibition effect of root was mainly bio-antimutagenic, whereas stem and leaf were desmutagenic.

In vitro Antimutagenic Activity of Chitosan and Its Bio-antimutagenic Characteristics (Chitosan의 in vitro 돌연변이 억제효과 및 세포내 작용 특성)

  • The inhibitory effects of chitosan on mutagenicity induced by 3-amino-1-methyl-5H-pyrido [4,3-b] indole (Trp-P-2), sodium azide (SA), 2-nitrofluorene (2-NF), and 4-nitroquinoline oxide (4-NQO) were investigated using Salmonella typhimurium reversion assay and SOS Chromotest. In Salmonella typhimurium reversion assay. Chitosan showed 24-65% of inhibitory effect against the mutagenicity of an indirect-acting mutagen, Trp-P-2. On the other hand, no inhibitory effect was observed against the mutagenicity of direct-acting mutagens (2-NF, SA). In SOS chromotest. chitosan showed 46-49% effects on SOS function induced by 4-NQO. Chitosan inhibited the mutagenicity induced by Trp-P-2 with 9-39% of inhibition rate. It was also evaluated whether inhibitory effect of chitosan is due to its bio-antimutagenic or desmutagenic action. Chitosan at high concentrations showed a bio-antimutagenicity with dose-dependent manner, but it showed a desmutagenicity at low concentrations against the mutation induced by Trp-P-2.
