  • In an epidemiological study of metal workers exposed to metalworking fluids (MWF), the prevalence time of Evolution, seasonal occurrence and clinical type of contact dermatitis were investigated. Compostional analysis of MWF with HPLC, dermatological examination and two consecutive questionnaire surveys were conducted. Study population was divided into two groups ; workers contact to cutting oil and workers contact to rust preventive oil. In the analysis of MWF, aliphatic hydrocarbons, having 12-20 carbons, was most common composition(49.04%) of cutting oil otherwise, major contents (90.99%) of the rust preventives oil were aliphatic hydrocarbons composed of 6-9 carbons. The frequency (point prevalence) of contact dermatitis(CD) was 7(12.7 per 100 subjects) in the dermatological examination of 55 workers. As the result of second survey for contact dermatitis, cumulative prevalence of oil working full-time and recent 1 year prevalence in two groups were 28.0, 16.7 and 15.1, 12.5 per 100 subjects. There were no difference in the prevalence of CD by oil, age, oil contact duration. Summer is the most common evolution season in workers exposed to cutting oil, but not in workers exposed to rust preventive oil. Major clinical type of CD was erythematous papules in both groups. It presents the importance of preventive measures that 51.1% suffer from contact dermatitis had medical care at their own expense, and 47.1% of them felt serious about their contact dermatitis. From the fact that 68.6% think cotton gloves protective apparatus, we emphasize the need for health education.

  • Foeniculum vulgare (FV) has long been used in traditional medicine for the treatment of inflammatory diseases. In addition, it is usually known as an important medicinal and aromatic plant widely used as a carminative, digestive, lactogogue, and diuretic, and for treating respiratory and gastrointestinal disorders. The skin barrier protects against the invasion of pathogens, fends off chemical and physical assaults, and protects against extensive water loss. In this study, the effects of solvent-fractionated FV fruits on strengthening the skin barrier and maintaining moisture, as well as their antifungal activity, were investigated in human keratinocyte (HaCaT) cells. The expression of involucrin, loricrin, filaggrin, hyaluronic acid synthase, human β defensin, and cathelicidin genes and proteins was measured by reverse transcription polymerase chain reaction (RT-PCR) and western blotting. The production of hyaluronic acid was determined by enzyme-linked immunosorbent assay (ELISA). The butanol fraction increased the expression of involucrin and filaggrin. Both the ethyl acetate and the butanol fractions increased hyaluronic acid production by promoting the expression of hyaluronic acid synthase-1. Although the antimicrobial peptides were increased by FV crude extract and its fractions, the samples did not show a significant effect compared to the normal group. These results suggest that the butanol fraction of FV could be very useful in cosmetics for the treatment of dermatological diseases.

  • Raman spectroscopy is known to have strong potential for providing noninvasive dermatological diagnosis of skin cancer. According to the previous work, various well known methods including maximum a posteriori probability (MAP) and multilayer perceptron networks (MLP) showed competitive results. Since even the small errors often leads to a fatal result, we investigated the method that reduces classification error perfectly by screening out some ambiguous patterns. Those ambiguous patterns can be examined by routine biopsy. We incorporated an ambiguous pattern class in MAP, linear classifier using minimum squared error (MSE), MLP and reduced coulomb energy networks (RCE). The experiments involving 216 confocal Raman spectra showed that every methods could perfectly classify BCC by screening out some ambiguous patterns. The best results were obtained with MSE. According to the experimental results, MSE gives perfect classification by screening out 8.8% of test patterns.

  • Melanogenesis is involved in the pigmentation of the hair, eyes, and skin in living organisms. Various signaling pathways stimulated by ${\alpha}-MSH$, SCF/c-Kit, $Wnt/{\beta}-catenin$, nitric oxide and ultraviolet activate melanocyte, leading to melanin production by tyrosinase, tyrosinase-related protein (TRP)-1, and TRP-2 expressed via the microphthalmia-associated transcription factor (MITF). However, the abnormal regulation of melanogenesis causes dermatological issues such as graying hair and vitiligo. Therefore, the activators that promote melanogenesis are crucial for the prevention of graying hair and the treatment of hypopigmentary disorders. Many melanogenesis stimulators have been studied for the development of novel drugs derived from synthesized compounds and natural products. Here, in addition to providing a description of a common signaling pathway in the melanogenesis of graying hair and the vitiligo process for the development of novel anti-hair graying agents, this article reviews natural herbs and the active ingredients that promote melanin synthesis as a pharmaceutical agent for the treatment of vitiligo. In particular, compounds such as Imatinib and Sugen with a stimulating effect on melanogenesis as a side effect of the drugs, are also introduced. Recent advances in research on natural plant extracts such as Polygonum multiflorum, Rhynchosia Nulubilis, Black oryzasativa, and Orysa sartiva, widely known as traditional and medicinal extracts, are also reviewed.

  • Glycosaminoglycans (GAGs) and chitosan have been used as matrix materials to support the dermal part of skin equivalent which is used for both pharmacological and toxicological evaluations of drugs potentially used for dermatological diseases. However, their biological roles of GAGs and chitosan in the skin equivalent are still unknown. In the present study, we evaluated whether GAGs and chitosan directly affect keratinocyte stem cells (KSCs) and their transit-amplifying cells (TA cells). Among supporting matrix materials, chitosan significantly increased the number of ${\alpha}6$ $integrin^{high}/CD71^{high}$ human keratinocyte TA cells by 48.5%. In quantitative real-time RT-PCR analysis, chitosan significantly increased CD71 and CD200 gene transcription whereas not ${\alpha}6$ integrin. In addition, the level of the gene transcription of both keratin 1 (K1) and K10 in the chitosan-treated human keratinocytes was significantly lower than those of control, suggesting that chitosan inhibit keratinocyte differentiation. We also found that N-acetyl-D-glucosamine (NAG) and $\beta$-(1-4)-linked D-glucosamine (D-glc), two components of chitosan, have no effect on the expression of CD71, K1, and K10, suggesting that each monomer component of chitosan is not enough to regulate the number of epidermal keratinocyte lineage. Conclusively, chitosan increases keratinocyte TA cell population which may contribute to the cellular mass expansion of the epidermal part of a skin equivalent system.

  • BACKGROUND/OBJECTIVES: The root of Vitis amurensis Ruprecht, a sort of wild-growing grape, has been used in oriental medicine for treatment of skin ailments; however, its dermatological activity is not sufficiently understood. The aim of this study was to investigate tyrosinase inhibitory and anti-melanogenic activities of V. amurensis Ruprecht root methanol extract (VARM) in B16F10 mouse melanoma cells and to attempt to isolate and identify the active compound issued from VARM. MATERIALS/METHODS: Anti-melanogenic activity of VARM was analyzed in ${\alpha}$-melanocyte stimulating hormone (MSH)-stimulated B16F10 cells through evaluation of antioxidative activity as well as inhibited tyrosinase activity and melanin contents compared with those of kojic acid and arbutin. After anti-melanogenic analysis of VARM, serial fractionation, nuclear magnetic resonance (NMR), and thin layer chromatorgraphy (TLC) were applied for identification of active compounds contained in VARM. RESULTS: VARM significantly inhibited oxidative stress and tyrosinase activity and attenuated ${\alpha}$-MSH-induced melanin production in B16F10 cells. For isolation of active compounds, VARM was fractionated using a series of organic solvents, including dichloromethane ($CH_2Cl_2$), ethyl acetate (EtOAc), and n-butanol (n-BuOH). Among fractions showing anti-melanogenic activity, the CH2Cl2 fraction induced the most potent attenuation of melanogenesis without cytotoxicity and the major compound in the $CH_2Cl_2$ fraction was identified as betulinic acid. Betulinic acid isolated from the $CH_2Cl_2$ fraction of VARM significantly attenuated ${\alpha}$-MSH-induced melanogenesis in a dose dependent manner, which was stronger than that of arbutin used as a positive control. CONCLUSIONS: These results indicate that VARM inhibits oxidative stress, tyrosinase activity, and ${\alpha}$-MSH-induced melanogenesis in B16F10 cells, due primarily to the active compound, betulinic acid, in the $CH_2Cl_2$ fraction.

  • Neospora caninum (Apicomplexa; Sarcocystidae) is a protozoan that causes abortion in cattle, horses, sheep, and dogs as well as neurological and dermatological diseases in dogs. In the central nervous system of dogs infected with N. caninum, cysts were detected that exhibited gliosis and meningitis. Flavonoids are polyphenolic compounds that exhibit antibacterial, antiparasitic, antifungal, and antiviral properties. In this study, we investigated the effects of flavonoids in a well-established in vitro model of N. caninum infection in glial cell cultures. Glial cells were treated individually with 10 different flavonoids, and a subset of cultures was also infected with the NC-1 strain of N. caninum. All of the flavonoids tested induced an increase in the metabolism of glial cells and many of them increased nitrite levels in cultures infected with NC-1 compared to controls and uninfected cultures. Among the flavonoids tested, 3',4'-dihydroxyflavone, 3',4',5,7-tetrahydroxyflavone (luteolin), and 3,3',4',5,6-pentahydroxyflavone (quercetin), also inhibited parasitophorous vacuole formation. Taken together, our findings show that flavonoids modulate glial cell responses, increase NO secretion, and interfere with N. caninum infection and proliferation.

  • Objective Many literature-based researches and clinical studies have been thus far reported of wha-byuong in Oriental medicine but acupunctural approaches to it have been little executed, which motivated this study to be conducted through acupunctural treatment. For that purpose, this study carried out incessant treatment on wha-byuong-having outpatients of this hospital who were suffering from gunhwa of wha in the saam acupuncture in order to inquire into the features of wha-byuong represented recently, and thereupon this got some intellect and knowledge out of clinical cases, which led this study to report them. Methodology Two wha-byuong outpatients were taken as subjects for this study who visited this hospital from January 10, 2002 through May 20, 2002. As for the contents of treatment and the administration of medicine, acupuncture utilized size $30{\times}50mm$ needles and the performance of treatment depended upon yuchim for 20 to 40 minutes at one time by means of simseunggyok (eumgok, sohae bo, taebaek, sinmun sa) on both right and left sides of the outpatients. As the case may be, sobu, baekhoi, and pungji were used at the same time. Bosa of simseunegyok took youngsu bosa. In order to evaluate treatment, symptoms were classified with diagrams into physical symptoms, dorgan symptoms, urinogenital organs symptoms, dermatological symptoms, cardiopulmonary symptoms, musculoskeletal symptoms, somnipathy symptoms, caput and sensual organ symptoms, psychomotor activity symptoms, and other symptoms, and then signs and marks were given to indicate the most objective values according to symptoms of the patients perceived in different sessions both prior and posterior to treatment. Conclusion As a result of having conducted treatment on two outpatients who visited this hospital for their wha-byuong from January 10, 2002 through May 20, 2002, conclusions were obtained, as follows: 1. Treatment depended very largely upon simseunggyok of the saam acupuncture, and as the case may be, baekhoi, pungji and sobu were utilized at the same time. 2. Much more effects were produced only when the patients had got rid of the causes of wha-byuong they had. Furthermore, the perceived seriousness of the symptoms presented had not been improved, regardless of their wha-byuong, 3. The main symptoms complained by the patients such as sangyeolgam, anxieties, impatience, hungmin, difficulty in breathing, cardiopalmus, insomnia, digestive troubles, etc. showed for the most part conspicuous therapeutical effects upon the symptoms of wha-byuong. In particular, a high degree of improvement was represented from melancholia and pyrexia.
