Effect of High Nitrogen Application on Two Components of Dark Respiration in a Rice Cultivar Takanari

  • Plant growth and the two components of respiration, growth and maintenance, were compared between low and high nitrogen applications in hydroponic culture on a high-yielding rice cultivar 'Takanari' (Oryza sativa L.). Grain yield decreased by high nitrogen application, and thus this cultivar has low adaptability to nitrogen. Growth efficiency (GE) and net assimilation rate (NAR) were lower in the high-nitrogen plot. The maintenance coefficient (m) and growth coefficient (g) of dark respiration were 0.0111 $d^{-1}$ and 0.196 in the low-nitrogen plot and 0.0166 $d^{-1}$ and 0.237 in the high-nitrogen plot, respectively. Thus, high nitrogen application increased both g and m. Calculated $R_m$ (maintenance respiration rate) was 70 and 90% of total respiration rate at heading, respectively. The significance of nitrogen adaptability and g was discussed.

Inhibitory Effects of ABA and $Ca^{2+}$ on Dark Respiration in Protoplasts Isolated from the Basal Intercalary Meristematic Tissues of Oat Leaves (귀리잎의 기저부 절간분열조직에서 분이한 원형질체의 암호흡 활성에 미치는 ABA와 $Ca^{2+}$의 억제효과)

  • The present study was made of the effects of abscisic acid(ABA) and calcium ions on dark respiration in protoplasts isolated from the basal intercalary meristematic tissues of oat (Avena sativa L.) seedlings. The influences of calcium channel blockers diitiazem(DTZ), verapamil(VPM), and $LaCl_2$ and the calmodulin antagonist trifluoperazine(TFP) on protoplast respiration activities were also investigated in order to evaluate the possible involvement of calcium channels and calmodulin during the dark respiration. The ABA only caused an 21% inhibition of protoplast respiration at $10^{-6}\;M$, but the extent of inhibition was very low by calcium treatments in the absence of ABA. In the presence of $10^{-6}\;M$ ABA, however, this inhibition of respiration increased by the increment of calcium ions concentrations. Treatments of DTZ and VPM were all found to restore the calcium-dependent inhibition of protoplast respiration by ABA and it was the same in thc $LaCl_2$ treatment except at $10^{-4}\;M$. At concentration from $10^{-6}\;M\;to\;10^{-4}\;M$, TFP also restored an inhibition of respiration. These results support the possibility that ABA increases plasmalemma permeability to calcium ions which might then bind to calmodulin to regulate oat protoplast dark respiration.ration.

Characteristics of Photosynthesis and Respiration Rates in Strobili of Pinus koraiensis S. et Z. (잣나무 구과(毬果)의 광합성(光合成)과 호흡(呼吸)의 특성(特性)에 관(關)한 연구(硏究))

  • The dark respiration, photosynthesis($CO_2$ refixation), $CO_2$ balance and chlorophyll content of 1st-year conelets and 2nd-year cones of Korean pine(Pinus koraiensis S.et.Z.) were investigated after pollination up to the end of maturation. The results obtained are as follows : 1. The growth of 1st-year conelet was 3.6cm in length. 2.4cm in diameter and 3.058 in dry weighs during the first year. The growth of 2nd-year mature cone was 13.5cm in length, 9.3cm in diameter and 141.0g in dry weight in the late of 2nd-year. 2. The refixation of carbon dioxide released from a cone by the dark respiration was less than 50 percent at light saturation through the growing period. The refixation of carbon dioxide released by dark respiration for one year was 7.3 percent in 1st-year conelets and 8.7 percent in 2nd-year cones. 3. The dark respiration rate of cones by increasing temperature was rapidly increased with increasing temperature up to $25^{\circ}C$. The dark respiration rate of cones was much higher in non-growing season than that in growing season at the same temperature. 4. The rates of dark respiration and $CO_2$ refixation, based on the dry weight, were much higher in 1st-year conelet than that in 2nd-year cone. 5. The $CO_2$ balance for a cone was negative from pollination to the end of maturation. The net dark respiration loss for a cone was 7.23g $CO_2$/year in 1st-year conelet and 164.8g $CO_2$/year in 2nd-year cone. 6. The respiratory loss efficiency for a cone(=$CH_2O$ weight calculated by net dark respiration/dry weight of cone) for one year was 1.61 in 1st-year conelet and 0.81 in 2nd-year cone for one year. 7. The total chlorophyll content of surface scale of the cone was lower than 2mg/g dw through the growing period, and chl. a/b ratio was 2 to 3.

  • The influence of respiration on photosythetic electron transport were investigated in the Wid type and psaB mutants from Syneehocystis sp. PCC6803. The amount of glucose uptake in the wild type was proportional to the glucose concentration added in wild type and less than that of psaB mutants in the dark. It was suggested that psaB mutants more depend on the glucose than the wild type. It was investigated how the activities of isocitrate dehydrogenase(IDH) and glucose-6-phos-phate dehydrogenase(G6PDH) were changed. The activities of IDH were very low. While, the ac-tivities of G6PDH were much higher than that of IDH. These results agree to the reports that ex-ogenous glucose was dismilated aerobically via Oxidative Pentose Phosphate Pathway in heterotrophic cyanobacteria. PsaB mutants showed high G6PDH activity in the presence of glucose as well as in the dark and high respiratory activities especially in the dark. It was also investigated how photosynthetic electron transport activities were changed. PsaB mutants showed higher photosynthetic electron tranasport activities than wild type in the presence of glucose as well as in the dark. In the results, it was proposed that photosynthetic electron transport between PS I and PS U was complemented by respiratory electron transport through the NADPH generated by Dxidative Pentose Phophate Pathway in psaB mutant from Synechocystis sp. Seasonal Growth and Root Respiration of North American Ginseng

  • American ginseng plants (Panax quinquefolium L.) of various ages were harvested every two weeks during each of three growing seasons and dry matter yield of components and root respiration determined. Shoot dry weight was about 0.5 g, 2.5 g and 4 g for 2, 3 and 4-year-old plants, respectively and fruit dry weight was as much as 50% of this in 3- and 4-year-.old plants. Root dry weight decrease by 30~50% as shoots emerged and at the end of the season was about 2 g, 3.5 g and 5 g in 2, 3, 4 and 5-year-old plants, respectively. Shoot and root dry weight were linearly related with an approximate 1:2 ratio. Root respiration rate at 2$0^{\circ}C$ in the dark was about 5 $\mu\textrm{g}$ CO2 g-1 DW(dry weight) min-1 in the early season, then doubled within 50 days as shoots emerged, and thereafter declined over the season to 2~5 $\mu\textrm{g}$ CO2 g-1 DW min-1. The Q10 for dark respiration over the interval from 10 to 2$0^{\circ}C$ was 1.58. Root respiration rate and shoot growth rate was positively linearly related in all ages of plants.   • Seedling vigor of introgressive hybrids between birdsfoot refoil 'MO-20"(Lotus corniculatus) used as the maternal parent and tetraploid(2n=4x=24)accession PI302921 narrowleaf trefoil(Lotus corniculatus L.), and parents were compared in terms of dry matter accumulation , net photosynthetic rate, dark respiration rate, and growth analysis. The hybrids showed better seedling vigor than PI302921 as they had lower dark respiration rate and higher relative growth rate (RGR) than PI302921. Hybrids grew faster than MO-20, but there was not significant difference between them. Net photosynthesis per unit area increased during three weeks after emergence without difference among the entries, then decreased sharply with leaf age and shading marking much differences among entries Dark respiration rate was very high just after emergence, after which it decreased rapidly until it reached a stable level at 1 week through 3 weeks of age, and then dropped again. In general, dark respiration, photosynthesis, and seedling growth of the hybrid was similar to those if MO-20. Although there are some reports about interspecific hybrids of Lotus species, it still remains as a new area for genetic improvement of seedling vigor of L. corniculatus.atus.

  • The physiological aspects of tobacco seeds from seeding to germination under light and dark condition, and different temperature were studied to obtain basic information on the production of good seedlings. There were distinct three phases in moisture uptake and respiration process during germination that classifiable into 'Imbibition', 'Lag', and 'Growth' phase under light condition but such growth phase were not observed in the dark germination. Great changes of sugar and fatty acid content of tobacco seeds were observed during germination in light condition but such changes were slight in dark germinated seeds and fatty acid content of seeds during germination were decreased as corresponding to increasing in respiration.

  • Photosynthesis and respiration of leaf and root of field grown Panax ginseng were investigated according to aerial part sensecence. No apparent photosynthesis activity was detected in senescenced leaf(less than 0.7mg total chlorophyll/g FW) and leaf dark respiration was consistent relation with senescence. Leaf respiratory Q$_{10}$ consistently increased with senscence. Root respiration and Q$_{10}$ tended to decrease with aerial part senescence only in the range of optimum temperature of leaf growth. Apparent photosynthesis or respiration of leaf was negatively or positively correlated, respectively with the increase of air temperature. Root respiration with temperature was well accordance with Arrhenius plot which was not consistent with aerial part senescence. Accelerated senescence may be recommendable for better root yield unless any reserve in stem or leaves contributes to root through translocation.

Effects of Cd, Hg and Pb on the Respiration of the Germinating Seeds (발아종자의 호흡작용에 미치는 Cd, Hg 및 Pb의 영향)

  • The present study was carried out to investigate the effects of cadmium(Cd), mercury(Hg) and lead(Pb) on the respiration of the germinating seeds. Two kinds of seeds namely, mungbean (Paseolus radiatus L.) and buckwheat (Fagopyrum esculentum M ench) were used for the plant materials. The concentrations of Cd, Hg, and Pb applied for the treatment were 0.1, 1, 10, 100 and 1,000ppm. The germination test was carried out in the dark at 27$^{\circ}C$. Regardless of the kinds of heavy metals, the respiration of the germinating seeds was greatly increased for 24hrs absorbing the heavy metals, and subsequently the respiration was increased average 0.69mg/g.hr.CO2 for mungbean and 0.7mg/g.hr.CO2 for buckwheat. In the lower concentrations of cadmium and lead than 10ppm the germinating rates, seedling concentraitons. The germinating rates, the seedling growth and the respiration were not increased in ay concentrations of mercury, but were decreased with increase of the mercury concenration. The inhibition of seeds respiration by mercury was 3 times higher than by cadmium, and 5 times higher than that by lead. The toxicity of three heavy metals on the respiration was showed from the highest to the lowest in the order of Hg, Cd, and Pb.

Effects of Sulfur Dioxide on Catalase Activity in Plants (식물체의 catalase 활성에 미치는 아황산가스의 영향)

  • The experiments were conducted to examine the effects of sulfur dioxide on catalase activity in germinating seeds under dark conditions and in green plants under light conditions. Three kinds of plants, bean (Glycine max M.), cabbage (Brassica oleracea L. Chungbang No.1) and rice (Oryza sativa L. Suwon No.213-1) were used in this study. Both thegerminating seeds and the leaves were treated with various concentrations of sulfur dioxide for both one green plants with two and twelve hours. In the results of the measurement of catalase activity it was found that plant injuries by sulfur dioxide for the period of photosynthesis were higher than those for the period of respiration. In the control, apparent catalase activity under the light condition of photosynthesis showed a considerable decrease in comparison with catalase activity under the dark condition of respiration. This tendency also appeared markedly in the gas treatment. In the gas treatment for twelve hours, the decrease of catalase activity was higher than that of one hour in both photosynthesis and respiraton. It was thought that sulfur dioxide was an inhibitor of catalase activity in higher concentration of the gas.
