Mechanisms of Selective Antimicrobial Activity of Gaegurin 4

  • Gaegurin 4(GGN 4), an antimicrobial peptide isolated from a Korean frog, is five times more potent against Gram-positive than Gram-negative bacteria, but has little hemolytic activity. To understand the mechanism of such cell selectivity, we examined GGN4-induced $K^+$ efflux from target cells, and membrane conductances in planar lipid bilayers. The $K^+$ efflux from Gram-positive M. luteus(2.5 ${\mu}g/ml$) was faster and larger than that from Gram-negative E. coli(75 ${\mu}g/ml$), while that from RBC was negligible even at higher concentration(100 ${\mu}g/ml$). GGN4 induced larger conductances in the planar bilayers which were formed with lipids extracted from Gram-positive B. subtilis than in those from E. coli(p<0.01), however, the effects of GGN4 were not selective in the bilayers formed with lipids from E. coli and red blood cells. Addition of an acidic phospholipid, phosphatidylserine to planar bilayers increased the GGN4-induced membrane conductance(p<0.05), but addition of phosphatidylcholine or cholesterol reduced it(p<0.05). Transmission electron microscopy revealed that GGN4 induced pore-like damages in M. luteus and dis-layering damages on the outer wall of E. coli. Taken together, the present results indicate that the selectivity of GGN4 toward Gram-positive over Gram-negative bacteria is due to negative surface charges, and interaction of GGN4 with outer walls. The selectivity toward bacteria over RBC is due to the presence of phosphatidylcholine and cholesterol, and the trans-bilayer lipid asymmetry in RBC. The results suggest that design of selective antimicrobial peptides should be based on the composition and topology of membrane lipids in the target cells.

Effects of Cervus elaphus Acua-Acupuncture's solution on damage of Rat's Kidney induced by Ischemia and Reflow (녹용(鹿茸) 약침액(藥針液)이 허혈(虛血)후 재관류(再貫流)에 의한 신장(腎臟) 조직(組織) 손상(損傷)에 미치는 영향(影響))

  • Cervus elaphus (CE), being known to reinforce Kidney, have tested to study the effects concerning damages of renal tissue induced by oxygen free radicals. I had observed the effects of CE extract on damages of rat's kidney following ischemia and reflow. Before ischemia was caused, CE extract was applied $0.2m{\ell}$ per 250g through femoral vein in ischemia and reflow group and normal sailine was applied in normal group, Ischemia was caused by renal artery's clamp for 60 min and reflowed by clamp remove after 30 min. It was increased on the content of lipid peroxidation, activies and type conversion ratio of xanthine oxidase following ischemia and reflow. by clamp remove after 30 min. It was increased on the content of lipid peroxidation, activies and type conversion ratio of xanthine oxidase following ischemia and reflow. However, they were decreased when CE extract was pre-applied. Glutathione level was decreased in ischemia and reflow group, and increased in CE extract's pre-applied group. However, it could not seen special changes on aldehyde oxidase activities, either. In conclusion, CE extract recovers the damage of kidney due to ischemia and reflow by decreasing the lipid peroxidation.

Effects of Yikgeebohyul-tang Aqueous Extracts on the Rat Hypothyroidism Induced by Propylthiouracil (益氣補血湯 물 추출물이 PTU로 유발된 Rat의 갑상선기능저하증에 미치는 영향)

  • Objectives The object of this study was to evaluate the effect of Yikgeebohyul-tang aqueous extracts (YKBHT) on the propylthiouracil (PTU)-induced rat hypothyroidism. Methods The rats were divided into 6 groups : intact vehicle control, PTU control, LT4, YKBHT 500, 250 and 125 mg/kg treated groups. Hypothyroidism was induced by daily subcutaneous treatment of PTU 10 mg/kg for 28 days. YKBHT aqueous extracts were administered once a day as an oral dose of 500, 250 and 125 mg/kg for 42 days. The changes were observed : weight of body, thyroid gland, liver, testis, epididymis and prostate, serum thyroid hormone levels, serum male sex hormone levels, serum lipid profiles, liver and testis antioxidant system. These results were compared with LT4 0.5 mg/kg intraperitoneally treated rats. Results These PTU induced hypothyroidism and related hepatic and male reproductive organ damages were favorably and dose-dependently inhibited by treatment of YKBHT 500, 250 and 125 mg/kg, and YKBHT also effectively regulated the PTU-induced abnormal antioxidant defense factor changes in the both liver and testis. Although, LT4 also inhibited PTU-induced hypothyroidism and relative liver damages. But it deteriorated the hypothyroidism related testis, epididymis and prostate damages through testicular oxidative damages. Conclusions : The result of this study suggests that YKBHT has favorable effects on the hypothyroidism and related liver and reproductive organ damages with augmentation of antioxidant defense factor in the testis and liver. YKBHT 500, 250 and 125 mg/kg dose-dependently inhibited PTU-induced hypothyroidism and related liver and male reproductive organ damages in rats.

Changes of the lipid and protein components according to the time lapsed after combined treatment of caffeine, iron and vitamin E in MDBK cells (배양세포주(MDBK cell)내에 caffeine, 철분 및 vitamin E 혼합처리시 배양시간 경과에 따른 지질과 단백질 구성성분의 변화)

  • This study was conducted to identify the effects of caffeine or combinations of caffeine and iron or vitamin E on the lipid and protein components in the MDBK(Mardin-Darby Bovine Kidney) cells. For the In vitro test, MDBK cells in ${\alpha}$-MEM(Minimum Essential Medium) were divided into 4 treatment groups according to drug types and dosages as follows; the control(group A), group B was treated with 0.3mM caffeine, group C was treated with 0.3mM caffeine and 0.3mM ferric chloride, group D was treated with 0.3mM caffeine and 0.3mM vitamin E. Those groups were further divided into 5 subgroups according to the time lapsed(control, 4hrs, 8hrs, 24hrs and 48hrs lapsed group). The concentrations of the carbonyl group and malondialdehyde(MDA) and the patterns of the SDS-PAGE(Sodium Dodecyl Sulfate-Polyacrylamide Gel Electrophoresis) and fatty acid compositions were analyzed to determine the oxidative damages and metabolic changes on the lipid and protein components in the MDBK cells. The results obtained from this study were summarized as follows; 1. The concentrations of carbonyl group and malondialdehyde in MDBK cells of group C were significantly higher(p<0.01) in comparison to the control, and increased according to the time lapsed. But the results of groups B and D were little different in comparison to the group C. 2. As the analytical results of fatty acid compositions in MDBK cells, the proportions of palmitoleic acid and linoleic acid in groups B, C and D were lower in comparison to the control, while the proportion of arachidonic acid in groups B, C and D were significantly higher(p<0.01) in comparison to the control. 3. In order to determine the oxidative damages to the protein in MDBK cells, the patterns of the SDS-PAGE were examined and the patterns of SDS-PAGE in groups C and D were significantly different between 43kd and 200kd of molecular weight.

Hepatoprotective Effects of Poly Herbal Formulation (Hepa-1000) on t-BHP Induced Toxicity in Human Hepatoma Cells (간기능 개선용 복합 식물 추출물(Hepa-1000)의 tert-butyl hydroperoxide(t-BHP)로 유도한 간세포 독성에 대한 보호 효과)

  • In the present study, the potential hepatoprotective effects of poly herbal formulation, Hepa-1000, against oxidative damages induced by t-BHP were evaluated in HepG2 cells in order to relate in vitro antioxidant activity with cytoprotective effects. The t-BHP induced considerable cell damage in HepG2 cells was shown by significant glutamic oxaloacetic transaminase (GOT) and lactate dehydrogenase (LDH) leakage, and increased lipid peroxidation. Hepa-1000-treated cells showed an increased resistance to oxidative challenge, as revealed by higher survival capacity than the one of control cells against t-BHP induced oxidative stress and hepatotoxicity. In addition, the Hepa-1000 had hepatoprotective effects lowering the activity of GOT and LDH, simultaneously. That is, it could inhibit the cell membrane damages resulting in the increased activities of GOT and LDH in the cell culture media. Furthermore, the Hepa-1000 could reduce t-BHP enhanced lipid peroxidation, which was evaluated by measuring the production of malonedialdehyde. Based on the data described above, it could be suggested that the Hepa-1000 has significant hepatoprotective effects and plays a protective role against lipid peroxidation by free radicals.

Repressive effects of red bean, Phaseolus angularis, extracts on obesity of mouse induced with high-fat diet via downregulation of adipocyte differentiation and modulating lipid metabolism

  • Obesity is generally caused by quantitative changes in adipocyte differentiation and fat metabolism. Only a few studies have been determined the effect of red beans extract on obesity and plasma cholesterol concentration. We have been studied the functional activities of red-bean extracts including anti-oxidative effect against DNA and cell damages. Histological study including micro CT analysis showed that the accumulation of fat in hepatocytes and intestines was significantly decreased in red bean extract treated group. In addition, plasma cholesterol and triglyceride levels were decreased in blood samples. In addition, it was confirmed that the red bean extract inhibited the expression of $PPAR{\gamma}$, Fabp4 and RETN genes, which regulate total adipocyte differentiation and lipid metabolism. Red bean extract inhibits the expressions of transcription factors associated with adipocyte differentiation in a dose-dependent manner, thereby inhibiting fat accumulation and decreasing blood lipid levels in obese mice induced by high fat diet.

Lipid Peroxidation and Its Nutritional Significance (지방질의 과산화와 영양)

  • A general overview of the lipid peroxidation and its nutritional significance are presented ,with emphasis on the reaction mechaisms, peroxidized products, further interaction and nutritional/biological deterioration in a series of oxidative process. Overall mechanism with various factors and elements for initiation , propagation and termination of free radical reaction is reviewed and the primary /secondary products of peroxidized lipids are defined. Since these products are potentially reactive substances that can cause deterioration of proteins /amino acids and vitamins (carotene, tocopherols and ascorbic acid etc), mechanism and actual damages of their deterioration in some foods and biological models are outlined. Especially , chemical changes caused by interaction of peroxidized products (related hydroperoxides, radicals and malonaldehye etc) and protein are emphasized here. And also, the detailed mechanisms on radical scavenging of the these vitamins which are the most prominent natural antioxidants are presented . Additionally , the possible roles of peroxidicaed lipids and their secondary products in the process of aging an carcinogenesis are briefly discussed . However, it is important to not that more detailed and integrated studies on the reaction kinetics, energetics of peroxidation, their decomposed products , biochemical interaction potential damaging/aging / carcinogenic effects, protection from their oxidative spoilage and novel antioxidants in food and heterogeneous biological systems will be essential in order to assessing the implication of lipid peroxidation to human nutrition and health.

Radioprotective Effect of Ginseng Components on Antioxidant Enzymes, Glutathione and Lipid Peroxidation of Liver in ${\gamma}$-Irradiated Mice (홍삼 분획물이 감마선을 비사한 생쥐 간에서 항산화물질과 지질과산화에 미치는 방사선 보호효과)

  • In the present study, to determine whether the antioxidative components of Korean red ginseng protect against radiation damage and the possible relationship among the radioprotective effects and antioxidant actions, the effects of total saponin (200 mg/kg, ip) and lipophilic fraction (200 mg/kg, oral) preferment of mice on the survival ratio, major antioxidant enzymes (SOD, catalase and glutathione peroxidase) activities, glutathione levels and lipid peroxidation in the liver were exiled for 2 weeks after whole ${\gamma}$-body ${\gamma}$-irradiation (6.5 Gy). The 30-day survival ratio increased from 10% to 57% and 40% for mice treated with total saponin and lipophilic fraction, respectively. On day 14 after ${\gamma}$-irradiation, the ginseng total saponin pretreatment produced a slight increase of antioxidant enzymes activities and significantly Increased reduced glutathione (GSH) contents (p<0.05) in the liver compared with non-treated group. Pretreatment with ginseng total saponin significantly deceased GSSG/total GSH ratio (p<0.05) without change of GSSG in the liver and inhibited the radiation-induced incense in the hepatic malondialdehyde levels. (p<0.05) In these results, GSH plays an important role in the liver in several detoxifications and the reduction of lipid peroxides. Thus, it appears that total saponin of red ginseng exerts its radioprotective effect by accelerating the production of endogenous antioxidants, such as glutathione from radiation induced damages and thereby oxygen free radicals.

Protective Effects of BJS-mix001, in Indomethacin induced Gastric Damages in Rats (BJS-Mix001이 Indomethacin 유발 랫트 위점막 손상에 미치는 영향)

  • The object of this study was to observe the gatro protective effects of BJS-mix001, a mixed herbal formula consisted of 4 herbal drugs Pinelliae Rhizoma : Coptidis Rhizoma : Massa Medicata Fermentata : Ostreae Testa = 1 : 1 : 1 : 1 (g/g) mixtures, which were main component of oriental medicine for treating various digestive diseases, in indomethacin induced gastric damages in rats. Three different dosages of BJS-mix001 (200, 100 and 50 mg/kg) were once orally administered 30 min before indomethacin treatment. Six hrs after indomethacin treatment, changes on the gross lesion scores, fundic histopathology, MPO activity and anti oxidant activities were observed. The results were compared with omeprazole, antioxidant and proton pump inhibitor 10 mg/kg and DA-9601, a standardized extract of the herb Artemisia asiatica 100 mg/kg treated group, respectively. As results of all three different dosages of BJS-mix001 in the indomethacin induced gastric damaged rats, significant decreased gastric damages were detected as compared with the indomethacin treated control rats. BJS-mix001 also strengthened the antioxidative defense systems - decreased the level of lipid peroxidation and catalase activity but increased the level of superoxide dismutase and glutathione contents. BJS-mix001 showed similar gastro protective effects as compared with equal dosage of DA-9601, and BJS-mix001 50 mg/kg showed slighter effects as compared with omeprazole 10 mg/kg, in the present study. The results obtained in this study suggest that BJS-mix001 showed favorable effects in the indomethacin induced gastric damages mediated by strengthening of the antioxidative defense systems.

The Effects of Baekho-tang Extracts on Regulating Th2 Differentiation through Improving Skin Fat Barrier Damage (백호탕 추출물의 지방장벽 손상 개선을 통한 상피 내 Th2 분화 조절 효과)

  • Objectives The purpose of this study is to confirm the regulate effect of T helper (Th) 2 differentiation that Baekho-tang extract may produce to improves skin lipid barrier damages. Methods Four-weeks-old NC/Nga mice were divided into four groups: control group (Ctrl, n=10), lipid barrier eliminated group (LBE, n=10), Dexamethasone treatment after lipid barrier elimination group (DxT, n=10), and Baekho-tang extract treatment group after lipid barrier elimination group (BHTT, n=10). Baeko-tang extract was administered for 3 days after removal of the skin fat barrier in BHTT group. Then, we identified changes in external symptoms of the skin, factors affecting skin barrier such as potential of hydrogen (pH), filaggrin (FLG), transepidermal water loss (TEWL) and Th2 differentiation factors like Interleukin (IL)-4, Kallikrein Related Peptidase 7 (KLK7) and protease activated receptor 2 (PAR-2) through our immunohistochemistry. Results After lipid barrier elimination, the reduction of morphological skin inflammations was less in BHTT group than in LBE group and DxT group. Also, pH and TEWL were significantly decreased with BHTT group. However, FLG was significantly increased in BHTT group compared to LBE, DxT, and Ctrl group. All kinds of Th2 differentiation factors (IL-4, KLK7 and PAR-2) were also decreased in BHTT compared to the LBE and DxT. Conclusions As a result of this study, BHT administration decreased pH, TEWL, and increased FLG, thus participating in recovering damaged skin barrier. Since Th2 differentiation factors were decreased as well, BHT's regulatory effect in sequential immune reactions may be a possible explanation of how it enhances recovery of the damaged lipid barrier.
