Polymorphisms in Epigenetic and Meat Quality Related Genes in Fourteen Cattle Breeds and Association with Beef Quality and Carcass Traits

  • Improvement for carcass traits related to beef quality is the key concern in beef production. Recent reports found that epigenetics mediates the interaction of individuals with environment and nutrition. The present study was designed to analyze the genetic effect of single nucleotide polymorphisms (SNPs) in seven epigenetic-related genes (DNMT1, DNMT3a, DNMT3b, DNMT3L, Ago1, Ago2, and HDAC5) and two meat quality candidate genes (CAPN1 and PRKAG3) on fourteen carcass traits related to beef quality in a Snow Dragon beef population, and also to identify SNPs in a total of fourteen cattle populations. Sixteen SNPs were identified and genotyped in 383 individuals sampled from the 14 cattle breeds, which included 147 samples from the Snow Dragon beef population. Data analysis showed significant association of 8 SNPs within 4 genes related to carcass and/or meat quality traits in the beef populations. SNP1 (13154420A>G) in exon 17 of DNMT1 was significantly associated with rib-eye width and lean meat color score (p<0.05). A novel SNP (SNP4, 76198537A>G) of DNMT3a was significantly associated with six beef quality traits. Those individuals with the wild-type genotype AA of DNMT3a showed an increase in carcass weight, chilled carcass weight, flank thicknesses, chuck short rib thickness, chuck short rib score and in chuck flap weight in contrast to the GG genotype. Five out of six SNPs in DNMT3b gene were significantly associated with three beef quality traits. SNP15 (45219258C>T) in CAPN1 was significantly associated with chuck short rib thickness and lean meat color score (p<0.05). The significant effect of SNP15 on lean meat color score individually and in combination with each of other 14 SNPs qualify this SNP to be used as potential marker for improving the trait. In addition, the frequencies of most wild-type alleles were higher than those of the mutant alleles in the native and foreign cattle breeds. Seven SNPs were identified in the epigenetic-related genes. The SNP15 in CAPN1 could be used as a powerful genetic marker in selection programs for beef quality improvement in the Snow Dragon Beef population.

Physicochemical Properties of Colostrum by Milking Time of Gyeonggi Province (경기지역의 착유회수에 따른 초유의 이화학적 특성)

  • Colostrum samples were collected from 36 dairy farms in Gyeonggi-do and one dairy farm in the National Institute of Animal Science (NIAS) for testing. Colostrum samples were analyzed for phisycochemicals (specific gravity, pH, titratable acidity), general components (fat, protein, lactose, total solid, solid non-fat (SNF)), fatty acids, amino acids, minerals, microflora, somatic cells, and Ig (Immunoglobulin). The first colostrum revealed the following data: fat contents were $6.16{\pm}2.39%$, proteins were $14.78{\pm}4.30%$, lactose $2.57{\pm}0.77%$, total solid $24.28{\pm}4.36%$, and SNF $18.12{\pm}4.08%$, whereas the 2nd (or $12^{th}$) colostrum revealed $5.56{\pm}1.76%$ fat, $3.46{\pm}0.41%$ proteins, $4.19{\pm}0.43%$ lactose, $13.90{\pm}1.76%$ total solid, and $8.34{\pm}0.81%$ SNF. Also, the first colostrum revealed the contents of major amino acids as 0.89% aspartic acid, 0.71% threonine, 0.86% serine, 1.75% glutamic acid, 0.64% valine, 0.95% leucine, 0.83% lysine, and 0.95% proline, and those in the 10th colostrum were 0.25% aspartic acid, 0.15% threonine, 0.19% serine, 0.59% glutamic acid, 0.19% valine, 0.35% leucine, 0.31% lysine, and 0.34% proine. Major amino acid contents rapidly decreased as milking times increased. In the first colostrum, the following mineral contents were observed: there were 2,168 ppm in Ca, 1,959 ppm in P, 914 ppm in K, 761 ppm in Na, 287 ppm in Mg, 1.7 ppm in Fe, 14.3 ppm in Zn, and 1.0 ppm in Cu; while in the 10th colostrum, the following ppm contents were 1,389 in Ca, 1,323 in P, 838 in K, 427 in Na, 131 in Mg, 1.0 in Fe, 4.7 in Zn, and 1.3 in Cu. The mineral contents in a colostrum rapidly decreased as milking times increased.

A Retrospective Study on the Risk Factors and the Effect of Higher Somatic Cell Count in Milk on Reproductive Performance in Dairy Cows (젖소에서 비유초기 체세포 증가 위험 요인 및 번식효율에 미치는 영향 분석 연구)

  • This retrospective study evaluated the effect of somatic cell count (SCC) in milk during early lactation on reproductive performance in dairy cows. Data were collected on 774 cows from six dairy farms, including cow parity, dates of previous calving, artificial insemination, pregnancy diagnosis, incidence of postpartum endometritis, reproductive performance (the intervals from calving to first insemination and conception), milk production and SCC. Data on 774 lactations were grouped based on the average first 3 months postpartum linear somatic cell score (SCS) as T1 (< 3.0, n = 521), T2 (3.0 ${\leq}$ and < 4.0, n = 113), and T3 (${\geq}$ 4.0, n = 140) groups. The odds ratio (OR) for the probability of endometritis increased 1.6 (p < 0.05) and 3.2 times (p < 0.0001) in the T2 and T3 groups, respectively, compared with that in the T1 group. The hazard of first insemination by 150 days in milk (DIM) was lower in the T3 group (hazard ratio [HR]: 0.76, p < 0.01) than in the T1 group. First insemination conception rate did not differ among the 3 groups (28.7-34.2%, p > 0.05). The hazard of pregnancy by 365 DIM in the T3 group was lower (HR: 0.75, p < 0.05 respectively) than in the T1 and T2 groups. The SCS during 4 to 7 months postpartum differed (p < 0.0001) among the 3 groups. Farm and cow parity were important risk factors for higher SCS (${\geq}$ 4.0). Multiparous cows were more likely to have a higher SCS (OR: 2.26, p = 0.0005) compared with primiparous cows. In conclusion, higher SCS (${\geq}$ 4.0) during early lactation was associated with decreased reproductive performance of dairy cows.

  • This paper describes how location influences on the development of dairy farming. It compares Chuncheon, which in the mountains, Daejeon, which is a transportation center and on flat ground, and Incheon, which is the gate to Seoul and an industrial city. The results analyzed are summarized as follows : 1. Incheon, due to her vast market, influenced strongly on the development of management. But shortage of roughage for feeds is the critical problem there. 2. Owing to the side job of raising chicks, which involves 92% of dairy farmers there, Chuncheon does not make smooth utilization of vast grass pasture. 3. In Daejeon's case, running orchards concurrently is the problem. 4. There exists no gap in wages between the region. The labor supply is most affluent in Incheon and there is competition with other forms of agriculture in the mountainous area. 5. The full-time employee tends to be skillful in accordance with his career. Family labor has been skillful, but the full-time employee is very fluid and shows varieties of skill. 6. Because of the obscurity of the distribution of labor with other jobs in Chuncheon and Daejeon, many unspecialized men serve. 7. The prices of milk are different in each region. The income of dairy farmers is strongly influenced by the low purchasing price of milk plants, and this is one of the important factors hindering the development of dairy farm ing in Chuncheon.

Comparison of Forage Yield and Quality of Forage Legume (콩과목초의 사초 수량 및 품질 비교)

  • Forage legumes are being used as an important source of protein for dairy farm as well as nitrogen fertilizer. This experiment was conducted to evaluate forage production and quality of five legumes at Cheonan Yonam College Experimental Livestock Farm for 1 year(2001-2002). Five legumes were crimson clover (Trifolium incarnatum L.) 'Tibbee', hairy vetch(Vicia villosa Roth) 'Common', red clover(Trifolium pratense L.) 'Kenland', persian clover(Trifolium resupinatum L.) 'Leeton', alfalfa(Medicago sativa L.) 'Vernal'. All legumes were shown cold hardiness of 8 rating or higher except crimson clover(7 rating). Flowering stage was observed 23th of April for crimson clover, 13th of May for hairy vetch, 3rd of May for red clover, and 12th of May for alfalfa. But persian clover didn't at harvest. Dry rnatter(DM) of crimson clover(17.6%) was the highest among legumes. In DM and total digestible nutrients(TDN) yields, crimson clover and hairy vetch were higher than those of other legumes. The crude protein of hairy vetch(20.5%) was the highest among legumes. In neutral detergent fiber(NDF) and acid detergent fiber(ADF), persian clover was the lowest(35.0% and 25.0%). Calculations of relative feed value(RFV) based on NDF and ADF of legumes were over 125(Prime degree) except hairy vetch. RFV and TDN of persian clover were the highest among legumes. According to the results of this study, crimson clover and hairy vetch are excellent in forage yield. Persian clover is superior in forage quality among legumes.

Comparison of Genome-wide Association Study (GWAS) Algorithms for Detecting Genetic Variants Associated with Growth Traits in Olive Flounder Paralichthys olivaceus (넙치(Paralichthys olivaceus)의 성장형질 연관 유전자 변이 탐색을 위한 전장유전체연관분석(GWAS) 알고리즘 비교 분석 연구)

  • Genome wide association studies (GWAS) identify genetic loci associated with quantitative traits in genomic selection. Although several studies have compared performance of various algorithms, no study compares them in olive flounder Paralichthys olivaceus. This study compared the GWAS results of four mixed linear model (MLM) algorithms and one Fixed and random model Circulating Probability Unification (FarmCPU) algorithm in olive flounder. Considering gender and genetic association matrices as fixed and random effects, the MLM had stable performance without inflation for λGC (genomic inflation factor) of -log10P. The FarmCPU algorithm had some appropriate λGC of -log10P, and an upward tail was identified in quantile-quantile plots. Therefore, the models were suitable for detecting genetic variants associated with olive flounder growth traits. Moreover, significant genotypes appeared several times at chromosome 22, around which quantitative trait loci are expected to exist. Finally, in both models, some of the most genetic variants were found in genes related to growth traits, confirming their reliability. These results will be helpful when applied to the genomic selection of olive flounder growth traits in the future.

An Investigation on Milking Disturbances of Mammary System (젖소 필유계(泌乳係)의 착유장애(搾乳障碍)에 관한 조사(調査))

  • An investigation on various causes of milking disturbances resulting from injuries and abnormalities of mammary system were made in 2,179 Holstein cows. To perform this investigation, 69 dairy farms of the suburban area of Seoul city, Gyeonggi-do and Chungnam provinces were andomly selected and subjected. Diagnosis was made by means of inspection and palpation of teat, insertion of teat canula, checking milk machines, anamneses and farm records. This investigation was, actively done from March 1977 to February 1979, The results obtained were summerized as follows; 1. It was found that 446 cows, accounting for 20.48% of 2,179 cows inspected, had supernumerary teats. Among them 53.59% had one, 43.72% had two, 2.47% had three, and 0.22% had four supernumerary teats, respectively. 2. Dry off quarters were found in 158 quarters which turned out to be 1.8% of 8,716 quarters inspected. Among dry off quarters, 62.02% seemed to be caused by mastitis, 30.37% by acquired teat obstructions, and 7.59% by congenital blind teats and glands respectively. 3. Teat sphincter stenosis was found in 154 teats of 50 cows, which represents 1.76% of 8,716 teats and 2.29% of 2,179 cows inspected, respectively. Among 154 teats with teat sphincter stenosis, 138 teats (85.7%) of 33 cows were found to be congenital and revealed highest incidence. 4. Loose sphincter was found in 78 teats of 36 cows, which figure 0.89% of 8,7l6 teats and 1.69% of 2,179 cows inspected, respectively. Among 78 teats with loose sphincter, 52 teats (66.66%) of 13 cows were found to be congenital and revealed highest incidence. 5. Injured teat tip caused by over milking of milk machine, was found in 229 teats of 156 cows, which figure 2.63% of 8,716 teats and 7.15% of 2,179 cows observed, respectively. 6. Other miscellaneous injuries and congenital abnormalities of teats and udders were diagnosed as follows: The teat laceration was found in 34 teats (0.39% of 8,716 teats), fissure of teat skin in 24 teats (0.28% of 8,716 teats), stricture of teat cistern in 21 teats (0.24% of 8,716 teats), teat fistula in 12 teats (0.14% of 8,716 teats), papillomas on testes in 8 teats (0.09% of 8,716 teats). Knothole orifice in 7 teats (0.08% of 8,716 teats), subcutaneous abscess of udder in 5 quarters (0.05% of 8,716 teats), membraneous obstruction of teat cistern in 4 teats (0.05% of 8,716 teats), and congenital short teat in 8 teats (0.09% of 8,716 teats), respectively.

Environmental factors influencing acetone and Environmental factors influencing acetone and β-hydroxybutyrate acid contents in raw milk of Holstein dairy cattle (홀스타인 젖소의 원유내 acetone과 β-hydroxybutyrate acid 함량에 영향을 미치는 환경요인)

  • Using 378,086 lactation records on dairy cattle, environmental factors influencing acetone and ${\beta}$-hydroxybutyrate acid contents in raw milk which are used as ketosis diagnosis indicator traits were analyzed in this experiment. Significance testing was conducted on farm, lactation stage, parity, milking time and month of age by traits. The results of this experiment indicated that there was a highly significant (p < 0.01) difference in all factors and lactation stage was the most significant factor. Linear regression coefficients of month of age on daily milk yields and acetone and ${\beta}$-hydroxybutyrate acid contents were all positive, while their quadratic linear regression coefficients were negative. Least square means for milk yield at second lactation stage (36~65 days) was 19.06kg which was higher than that of late lactation stage by 6.51kg. Least square means for acetone and ${\beta}$-hydroxybutyrate acid contents at the first lactation stage (5~35 days) were highest (0.1929mM/L and 0.0742mM/L, respectively), and there was a trend that they decreased as the milking progressed, but increased slightly at the late stage of milking. However, least square means for acetone and ${\beta}$-hydroxybutyrate acid contents at the first parity were 0.1414mM/L and 0.0522mM/L, respectively, which were higher than the average milk yield after the second parity. Least square means for acetone and ${\beta}$-hydroxybutyrate acid contents of PM milk yield (0.1372mM/L and 0.0534mM/L, respectively) were higher than those of AM milk yield collectively.

Effects of Feeding TMR Feed with Dietary Citrus Byproducts on Chemical Properties of Raw Milk from Holstein Cow's Raw Milk (원유의 이화학적 특성에 미치는 감귤 부산물 TMR 사료 급여의 영향)

  • In this study, the effects of feeding providing Holstein cows with with TMR feed including amended with undried citrus byproducts on the properties of their raw milk were investigated. Two samples were used for the experiment: T0 (raw milk produced by dairy cows fed with TMR feed not including citrus byproducts) and T1 (raw milk produced by cows fed with TMR feed including citrus byproducts). The All experiments were conducted with Holstein cows at a dairy farm in the on Jeju island, and were repeated three times, in each, after of which raw milk obtained from 7 cows of each samplein each group was analyzed[ED highlight - please ensure this is correct]. The daily milk yield and somatic cell numbers of T0 and T1 were 24.16 kg and 25.97 kg, and 660,000 thousands and 445,000 thousands, respectively, which means that feeding citrus byproducts to cows increases daily milk yield and reduces somatic cell numbers. There was were no significant differences between T0 and T1 in terms of the raw milk's total plate count, specific gravity, titration acidity, and or the amount of milk protein, lactose, nonfat solids, free amino acids and volatile compounds, while raw milk in T1 showed significantly lower levels of milk fat in raw milk. The antioxidant activity of raw milk was improved by citrus byproducts TMR feed containing citrus byproducts.

Effects of Miling System on Gouda Cheese Characteristics made from Farmstead Milk-processing Plant (착유방식이 목장형 유가공으로 제조된 고다치즈의 품질 특성에 미치는 영향)

  • This investigation was carried out to study effects of different milking systems on the characteristics of Gouda cheese produced at farmstead milk-processing plants. In the first study, raw milk was collected from automatic milking system and conventional milking system farms, and Gouda cheeses were prepared under the same conditions. After 6 months of aging, the chemical composition was analyzed and free fatty acid (FFA) analysis and sensory evaluation were carried out on the products. In the second study, Gouda cheese samples were collected from seven farmstead milk-processing plants and the chemical composition and texture profile analysis were evaluated. No significant differences were found in the chemical composition of Gouda cheese according to different milking systems or different farms except crude ash. FFA analysis showed significant differences among farms, but sensory evaluation showed no significant differences. No significant differences were observed in FFA content and sensory assessment of different milking systems. Texture profile analysis revealed that there were significant differences in each cheese made at different farm plants, but there were no differences when different milking systems were used.
