Rooting and Acclimatization of Shoots Harvested from Bioreactor Culture in Rehmania glutinosa (생체반응기에서 수확한 지황 신초의 발근과 순화)

  • This experiment was carried out to know the effect of media and agar concentrations, aeration and growth regulators on rooting and acclimatization of the shoots harvested from bioreactor culture in Rehmannia glutinosa. Half MS media with 1.2% agar improved rooting and acclimatization of shoots. Shoots were effectively acclimatized and rooted well in case of aeration by using membrane filtered vessels. Shoots acclimatized in vessel with membrane Inter were healthier and had higher ex vitro survival rate than those without membrane Inter on plug tray. Addition of paclobutrazol 0.3-0.4 mg/L, to acclimatization media enhanced shoots growth and root development.

A Study of Cookery of Daily Meal (Soora Sang) in Wonheng Ulmyo Jungri Euigwae (1795) (원행을묘정리의궤(園幸乙卯整理儀軌) 중(中) 조리면(調理面)에서 본 수자상고(水刺床考))

  • To analyze daily meal of royal meal, studied Soora Sang were on record Wonheng Ulmyo Jungri Euigwae(1795). Historic book 'Jungri Euigwae' described the king's visit to his father's royal tomb 'Hyun Neung Won', during the domain of Cheung Jo, the 22nd King of Choson Dynasty. The results obtained from this study are as follows. A dining table was round table with feet as black lacquer. Vessels were brazen vessel, silverware and earthenware. Kinds of dishes indicated as the number of vessels (sets). Food was arranged in two kinds of table, the first one called the main table, the second the side table. The number of sets to be arranged on the table were different according to the royal status of the eater. 15 sets for king's mother, 7 sets for the king and his elder sister, the latter sets being arranged on a single table. Soy and bean paste were ruled out from kind of sets. Kinds of dishes served with a meal generally were cooked rice (飯), soup (羹), heavy soup (助致), jerked meat (佐飯), meat fish and others broiled with seasoning (灸伊), salted fish shrimp and etc, cooked potherbs and potherbs (菜), pickled vegetables (沈菜, 淡沈菜) soy and bean paste (醬). The meat and fish and vegetable in the composition of a menu were well arranged.

  • With increase of people's leisure time for marine facilities it is expected to increase the demand for marine tourism. It is necessary for lighthouse to meet the demand for marine tourism. Now, the lighthouse is not only functioning as a traditional aids to navigation for vessel, but also it becomes the new facility and space for all civilians who might visit and experience the marine culture. This research suggests how to use the facilities of the lighthouse as marine tourism resources by reviewing the cases of domestic lighthouses.

Development of open-top microfluidic chip for visualization of interactions between tumoroids and angiogenic sprouting (튜머로이드-혈관신생 상호작용의 가시화를 위한 개방형 구조 미세유체 칩 개발)

  • Cancer cells secrete angiogenic factors, and nearby vasculatures make new blood vessels essential for cancer development and metastasis in response to these soluble factors. Many efforts have been made to elucidate cancer-endothelial cell interactions in vitro. However, not much is known due to the lack of a suitable co-culture platform. Here, we introduce a 3D printing-based microfluidic system that mimics the in vivo-like cancer-endothelial cell interactions. The tumoroids and endothelial cells are co-cultured, physically separated by porous fibrin gel, allowing communication between two cell types through soluble factors. Using this microfluidic system, we were able to visualize new vessel formation induced by tumoroids of different origins, including liver, breast, and ovary. We confirmed that the ovarian tumoroids most induced angiogenesis while the other two cancer types suppressed it. Utilization of the proposed co-culture platform will help the researchers unveil the underlying mechanisms of the dynamic interplay between tumor and angiogenesis.

Antibacterial Effect of Electrolyzed Water on Oral Bacteria

  • This study investigated the antibacterial effect of electrolyzed water on oral bacteria both in vitro and in vivo. Tap water was electrolyzed in a water vessel using platinum cell technology. The electrolyzed tap water (called Puri-water) was put in contact with five major periodontopathogens or toothbrushes contaminated with these bacteria for 30 sec. In addition, Puri-water was used as a mouthwash for 30 sec in 16 subjects and the antibacterial effect on salivary bacteria was evaluated. Puri-water significantly reduced the growth of all periodontopathogens in culture and on toothbrushes, and that of aerobic and anaerobic bacteria in saliva, when compared to the effect of tap water. It also significantly reduced mutans streptococci growing on mitis salivarius-bacitracin agar. Our results demonstrate that the electrolyzed tap water is effective as a mouthwash and for toothbrush disinfection.

  • A coronary graft fabricated from PLGA poly (lactic-co-glycolic acid) and chitosan electros puns deposited on poly caprolactone (PCL) electro spun tube. Mechanical properties of tube were evaluated through extruder machine depending on thickness of vessel wall. Biocompatible properties were evaluated by SEM morphology, amount of cell counting and MTT assay method for depending on culture days (1, 3, 5 days). MTT assay, counting cell and SEM morphology showed that cells were fast growth and immigration after 5 days. Biodegradability was monitored through loss weigh method for incubator days.

A Feasibility Study on the Application of Safety Assessment to the Fishing Vessels

  • The current paper deals with the safety of fishing vessels, which needs more speculations than other ship types. The characteristics of accidents of fishing vessels are rather different compared with other commercial vessels. In the context, the methodology of the formal safety assessment, one of the important issues in the maritime world to enhance the safety of marine vehicles, are reviewed and consulted along with other methodologies to see its applicability for fishing vessels. It is anticipated that further effective measures for better safety of fishing vessels can be identified including due considerations of the aspect of human elements, i.e. cultural environment.

A Comparison of Microtuberization Efficiency between Normal and Adenosine Deaminase Transgenic Potato Plantlets Cultured In Vitro (Adenosine Deaminase 형질전환식물체와 정상식물체간의 인공씨감자 형성비교)

  • A Study was conducted to investigate comparison of in vitro tuberization between normal and transgenic potato plantlets harboring adenosine deaminase gene in potato cultivar of Desiree. In time course study of in vitro tuberization, the rate of tuberization in four lines were increased till 6 weeks. but maintained stil after 7 weeks. Microtuber initiation of transgenic lines, 43 and 39 were faster than other lines, but no difference was observed after 5 weeks compared with normal plantlets. In all transgenic lines, the majoirty of microtubers produced were small(less than 100 mg) and medium(100-200mg) size rather than large size(more than 200 mg). Among 4 lines , line 9 produced the highest number of microtubers per each culture vessel. The results of this experiment suggest that there is no significant difference in microtuber production efficiency between normal and transgenic potatoes.

  • The Chinese Bronze Epigraphy is based on the study of the Ancient Bronze, which is excavated from the ground and it is also related to Archeology and Historical Science as well. From the Han dynasty, to the Song dynasty and to the Qing dynasty. Chinese Graphonomy had a splendid development. Including ShuoWenJieZi in the Chinese Graphonomy and the study of Bronze got a shining improvement. ShuoWenJieZi as a key for deciphering ancient Chinese characters. With constant reference to the ShuoWenJieZi, Qing scholars pioneered the earliest large scale interpretations of bronze inscriptions. Several Qing Dynasty ShuoWen scholars such as a WangYun and DuanYuCai have made research in the Bronze Epigraphy. Through this research, we can figure out whether there is any relevance between the traditional study of Epigraphy and the Qing dynasty's.

Optimal Culture Conditions for the Shoot Growth of Freesia 'Shiny Gold' during Bioreactor Culture

  • For rapid production of freesia 'Shiny Gold' shoots by using a bioreactor, several culture conditions were investigated. Young shoots (< 1 cm) obtained from freesia corm section in vitro were used as plant materials for this experiment. As a basic experimental environment, 20 young shoots were inoculated into a 5 L balloon type bubble reactor which contained 1 L 1/2 strength MS medium supplemented with 30 g sucrose (3%), and the aeration was 0.1 vvm (vessel volumes per minute). The bioreactors were placed in a growth room with $23^{\circ}C$ temperature, 60% relative humidity and $60{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ light condition (16 h/8 h, day/night). The concentrations of MS media were set with 1/4, 1/2, 1 strength, medium volume 10, 20, 40%, sucrose concentration 3, 6, 9%, and aeration 0.1, 0.2, 0.4 vvm. After 4 weeks of cultivation, the growth indexes including the fresh and dry weight, and plant height were evaluated. At the same time, the consumption, pH, and EC of medium were estimated 4 weeks after incubating. The best results were achieved when 40 young shoots were incubated in a bioreactor in which 1 L of 1/2 strength MS medium supplemented with 6% sucrose was used for the rapid production of freesia shoots. The shoots were 17 cm in plant height and 1.0 g in fresh weight only 4 weeks after incubation which could be a good plant material suitable for corm enlargement in vitro. No correlation was observed between the growth of freesia shoots and the consumption, pH or EC of medium.
