Herbicidin A is a potent herbicide against dicotyledonous plants as well as an antibiotic against phytopathogens. In this study, fermentation parameters for herbicidin A production in submerged culture of Streptomyces scopuliridis M40 were investigated. The herbicidin A concentration varied with the C/N ratio. High C/N ratios (>4) resulted in a herbicidin A production of more than 900 mg/l, whereas maximally 600 mg/l was obtained at ratios between 1 and 3.5. In 5-L batch fermentation, there was a positive correlation between the oxygen uptake rate (OUR) and herbicidin A production. Once the OUR increased, the substrate consumption rate increased, leading to an increase in volumetric productivity. Mechanical shear force affected the hyphal morphology and OUR. When the medium value of hyphal size ranged from 150 to $180{\mu}m$, high volumetric production of herbicidin A was obtained with OUR values >137mg $O_2/l{\cdot}h$. The highest herbicidin A concentration of 956.6 mg/l was obtained at 500 rpm, and coincided with the highest relative abundance of hyphae of $100-200{\mu}m$ length and the highest OUR during cultivation. Based on a constant impeller tip speed, which affects hyphal morphology, herbicidin A production was successfully scaled up from a 5-L jar to a 500-L pilot vessel.
87 microbes were isolated from dyeing wastewater collected at Dongducheon and Banweol industrial complex. Five microbes showed excellent ability of color removal and were identified as Shewanella putrefaciens, Aeromonas salmonicida(3 different strains), and Pseudomonas vesicularis. Five identified strains had optimal pH and optimal temperature as 7.0 and 30$^{\circ}C$ for cultivation, and showed morphological characteristics of Gram negative, oxidase negative, rod shape, and non-motility, but their biochemical characteristics were distinguishable. Each single strain of five microbes were tested in the 500 mL flask to treat dyeing wastewater, and achieved about 35% color removal efficiency in average. When two strains were selected and applied to the treatment at same time, color removal efficiency was increased up to 65%. While three or more associations of each strain did not show the improvement of color removal. Inhibition effects by $Mn^{2+}\;and\;Fe^{3+}$ on the dye degradation were tested and resulted in no effect under 70 ppm concentration.
The sprT gene encoding Streptomyces griseus trypsin (SGT) was introduced into Streptomyces lividans TK24 and Streptomyces lividans 1326 to study which strain would be better to overexpress the extracellular proteinase. Various media with different compositions were also used to maximize the productivity of SGT in heterologous hosts. The SGT productivity was best when the transformants of S. lividans TK24 and 1326 were cultivated in R2YE medium, and their relative trypsin activity of the culture broth measured with an artificial chromogenic substrate, N-${\alpha}$-benzoyl-DL-arginine-${\rho}$-nitroanilide, were 382 units/ml and 221 units/ml, respectively. They produced high levels of SGT in GYE medium but relatively lower than those in R2YE medium, and negligible amount of SGT was produced in Ferm, RASF, LIVID, and NDSK media. Considering non-SGT associated activity in Pronase powder, it was estimated that the transformant of S. lividans TK24 can produce SGT in R2YE 3.5 times more than the amount by S. griseus 10137 from which the sprT gene had been originated. The growth of S. lividans reached the maximum level of cell mass at 5 d of culture, but SGT production started in the stationary phase of cell growth and kept increasing until the ninth day of culture in R2YE medium, but in GYE media the productivity reached at the maximum level at 7 d of cultivation.
Irrigation control plays an important role in improving productivity of paprika which is very sensitive to moisture condition. Among environmental factors, light intensity and distribution are not easily controlled and showed a big difference depending on season and region. For adequate irrigation control, therefore, transpiration and irrigation amounts considering light environmental data should be estimated. In current study, modified transpiration model was used for more precise estimation of transpiration. Seasonal transpiration and irrigation amounts at different regions were compared by using light environmental data provided from Korea Meteorological Administration. The transpiration amount in summer was rather smaller than those in spring and autumn seasons in Korea due to large deviations in light intensity as well as rainy period in summer. Irrigation system capacities at various regions could be recommended by using the transpiration amount in the spring having the longest photoperiod in the year. These results will be useful to the design of irrigation system and optimization of input energy in greenhouse.
The purpose of this study was to optimize the solid state cultivation of Monascus ruber on sterile rice. A single-level-multiple-factor and a single-factor-multiple-level experimental design were employed to determine the optimal medium constituents and to optimize carbon and nitrogen source concentrations for lovastatin production. Simultaneous quantitative analyses of the ${\beta}$-hydroxyacid form and ${\beta}$-hydroxylactone for of lovastatin were performed by the high performance liquid chromatography (HPLC) method with a UV photodiode-array (PDA) detector. The total lovastatin yield ($4{\sim}6\;mg/g$, average of five repeats) was achieved by adding soybean powder, glycerol, sodium nitrate, and acetic acid at optimized levels after 14 days of fermentation. The maximal yield of lovastatin under the optimal composition of the medium increased by almost 2 times the yield observed prior to optimization. The experimental results also indicated that the ${\beta}$-hydroxylactone form of lovastatin (LFL) and the ${\beta}$-hydroxyacid form of lovastatin (AFL) simultaneously existed in solid state cultures of Monascus ruber. while the latter was the dominant form in the middle-late stage of continued fermentation. These results indicate that optimized culture conditions can be used for industrial production of lovastatin to obtain high yields.
Mass production of biocontrol agent is an essential step for its commercial use. Media composition and culture conditions for production of Bacillus amyloliquefaciens SKU-78, a potential biocontrol agent against bacterial wilts, were optimized by a flask culture. Low cost media combining nitrogen and carbon sources were tested. Maximum cell growth (> $2{\times}10^9$ CFU/ml) was obtained in a medium of 5% soy flour combined with 3% corn starch after 24 h cultivation. The optimum initial pH, temperature and shaking speed was 5.5, $30^{\circ}C$ and 150-250 rpm, respectively. Fermentation of SKU-78 was scaled up in 30 L fermenter and the profiles of cell density, pH, dissolved oxygen and spore formation were recorded. After 8 h lag phase, exponential growth occurred and reached at maximum viable cell number ($1.2{\times}10^{11}$ CFU/ml) after 20 h. The SKU-78 strain grown in a low cost medium exhibited the high suppression of bacterial wilts. The results indicate that SKU-78 strain can be produced in a low cost medium and provide a basis for scaling up to industrial level.
The transformant Bacillus subtilis ED213 carrying the pSO100 which cloned the cdd gene encoding cytidine deaminase (cytidine /2'-deoxycytidine aminohydrolase, EC 3.5.4.5, CDase) originated from wild type B. subtilis was cultivated in Spizizen minimal medium (SMM). To overcome poor expression of the cdd gene in SMM medium, the medium compositions and growth conditions were optimized. The optimized medium compositions and growth conditions were cytidine concentration of 80 mg/l, glycerol of 25 g/l, and $(NH_4)_2SO_4$ of 10 g/l, along with $37^{\circ}C$ and pH 7.0. The intracellular CDase production was increased 3 times from 1,000 unit/ml to 3,200 unit/ml, and extracellular CDase also increased from nearly undetectable amounts to 1,500 unit/ml. The cytidine concentration was signified as the most critical compositional factor for overproduction of CDase by increasing the cell density mainly in culture broth. The plasmids were more stable in cells that were grown in original SMM medium with stability of 90% compared to those grown in optimized SMM medium with stability of 80% after 48 hours cultivation. The most active amplification of plasmid was occurred in the logarithmic phase, which showed a value around four times higher than the initial copy number. In the exponential phase, the CDase production was closely related to the plasmid copy number along with the cell density. However, it was not accorded with cell density at the stationary phase.
Miscanthus purpurascens Juice containing potassium (37,952 mg/L), nitrogen (14,000 mg/L), phosphorus (6,800 mg/L), magnesium (5,969 mg/L), calcium (5,910 mg/L), etc., was investigated to develop a novel meidum far the mass cultivation of useful microorganisms. For this research, we first isolated an antagonistic bacterium G-74 from soil, which showed strong growth inhibition against two phytopathogenic fungi, Rhizoctonia solani and Botrytis cinerea, and identified as Bacillus lentimorbus G-74 based on the morphological characteristics and MIDI analysis. Culture conditions for G-74 strain in the M. purpurascens juice medium were optimized. Dilution rate of the medium, temperature and initial pH for the optimum growth of G-74 strain were 30% (V/V), $35^{\circ}C$ and 5.0, respectively. It was found that additions of 2.0% (W/V) corn starch as a carbon source and 1.0% (W/V) yeast extract as a nitrogen source in this medium increased B. lentimorbus G-74 growth to 66% more efficient than Luria Bertani medium.
Studies were made to optimize the meida composition and cultural conditions for the mycelial growth of Agrocybe cylindracea. Media composition for optimal growth was found to be starch 20.0g/l, bacto-soytone 4.0g/l. The media supplemented with $KH_2PO_4\;0.46g/l,\;K_2HPO_4\;1.0g/l,\;MgSO_47H_2O\;0.5g/l$ supported mycelial growth better than the media without mineral salts. Optimum temperature and pH for the growth was $28^{\circ}C$, and 6.0 respectively. Temperature range for the mycelial growth appeared to be $10-35^{\circ}C$ and the mycelium evidently lost the vitality at $40^{\circ}C$.
The effect of phosphate on the production of avermectin B1a was studied. Response surface methodology was applied to optimize the concentration of organic nitrogen sources. The portion of B1b in total avermectins was decreased from 5.8% to 3.0% by the addition of 1.5 g/ι inorganic phosphate to the production medium. Among organic nitrogen sources, soybean meal was the most effective on avermectin biosynthesis. Results showed that B1a productivity was increased by 44.8% in a laboratory scale fermenter cultivation of Streptomyces avermitilis YA99-40 through fed-batch process. A maximal B1a productivity was obtained by repeated 30 and 20 g/ι of glucose feeding at 136 and 206 hour, respectively. The B1a productivity was increased by 86.3% and the proportion of B1a in the total avermectins was improved from 38% to 45% with respect to the control process. These results would be very useful for enhancing productivity of B1a in an up-scaled processes.
