Five day old cotyledon explants of Cucumber (Cucumis sativus L) cv Poinsett 76 were cocultivated with two Agrobacterium strains (EHA105 and LBA 4404) each carrying GUS as the reporter gene and npt-II as the selection marker gene in the T-DNA region of the vector. Transformed shoots were selected at 150 mg/L kanamycin. A two day cocultivation coupled with $20\;{\mu}M$ acetosyringone increased the frequency (8.2 and 15.4 shoots) of GUS expression in the shoots of transformed plant. Among the two Agrobacterium strains, EHA 105 performed better than LBA 4404 in bringing two-fold increase in transformation efficiency (14%) than LBA 4404 (7.4%). PCR analysis was done to confirm the integration of T-DNA into cucumber genome.
This study was performed to select the significant wavelengths for predicting the chlorophyll content of grafted cucumber seedlings using hyperspectral images. The visible and near-infrared (VNIR) images and the short-wave infrared images of cucumber cotyledon samples were measured by two hyperspectral cameras. A correlation coefficient spectrum (CCS), a stepwise multiple linear regression (SMLR), and partial least squares (PLS) regression were used to determine significant wavelengths. Some wavelengths at 501, 505, 510, 543, 548, 619, 718, 723, and 727 nm were selected by CCS, SMLR, and PLS as significant wavelengths for estimating chlorophyll content. The results from the calibration models built by SMLR and PLS showed fair relationship between measured and predicted chlorophyll concentration. It was concluded that the hyperspectral imaging technique in the VNIR region is suggested effective for estimating the chlorophyll content of grafted cucumber leaves, non-destructively.
Mobilization of storage lipids is critical for the germination of oil seeds, as they supply carbon and energy until photosynthesis commences in cotyledons. In this study, we determined the levels of plant carnitine and associated changes in these levels from seed germination to cotyledon senescence. We also examined changes in the content of unsaturated fatty acids throughout seedling development. Carnitine levels peaked on day 3 at 14.5 nM in cotyledons and decreased sharply to 7.2 nM on day 4. On development day 3 carnitine levels were maintained at around 3 nM until day 7. The unsaturated fatty acid content dropped by half at the same time as carnitine peaked (day-3), and storage lipids were almost depleted by day 5. Thereafter, carnitine was hardly detected until the second stage of cotyledon senescence, at which stage the carnitine content was 6.8 nM, similar to that on day 4 at the time of fatty acid depletion in the cotyledons. Unsaturated fatty acids levels remained constant in green cotyledons but slightly increased in the senescing cotyledons. The latter can be explained by intracellular breakdown of membrane lipids. This is the first such discovery in developing cotyledons and may offer clues regarding other roles of the acetyl unit transport system in plants. The expression of BOU was closely associated with carnitine metabolism during seed germination and cotyledon development. The results provide support for the possibility of carbon re-routing during the glyoxylate cycle in the supply of energy for early germination and development.

Cucumber fruit mottle mosaic tobamovirus (CFMMV) causes severe mosaic symptoms with yellow mottling on leaves and fruits, and occasionally severe wilting of cucumber plants. No genetic source of resistance against this virus has been identified. The genes coding for the coat protein or the putative 54-kDa replicase were cloned into binary vectors under control of the SVBV promoter. Agrobacterium-mediated transformation was peformed on cotyledon explants of a parthenocarpic cucumber cultivar with superior competence for transformation. R1 seedlings were evaluated for resistance to CFMMV infection by lack of symptom expression, back inoculation on an alternative host and ELISA. From a total of 14 replicase-containing R1 lines, 8 exhibited immunity, while only 3 resistant lines were found among a total of 9 CP-containing lines. Line 144 homozygous for the 54-kDa replicase was selected for further resistance analysis. Line 144 was immune to CFMMV infection by mechanical and graft inoculation, or by root infection following planting in CFMMV-contaminated soil. Additionally, line 144 showed delay of symptom appearance following infection by other cucurbit-infecting tobamoviruses. Infection of line 144 plants with various potyviruses and cucumber mosaic cucumovirus did not break the resistance to CFMMV. The mechanism of resistance of line 144 appears to be RNA-mediated, however the means is apparently different from the gene silencing phenomenon. Homozygote line 144 cucumber as rootstock demonstrated for the first time protection of a non-transformed scion from soil inoculation with a soil borne pathogen, CFMMV.
When cucumber (Cucumis sativus L.) cotyledon discs were floated on $\delta$-aminolevulinic acid, oxyfluorfen, or rose bengal solution under light condition following 20 h dark incubation, rapid electrolyte leakage from the tissues occurred. The electrolyte leakage from the tissues was dependent on the compounds treated, their concentrations, and the duration of light exposure to the tissues. Dark incubation before exposure to continuous white light enhanced electrolyte leakage from the tissues treated with the compounds and reduced lag period for the activity of the compounds. Electrolyte leakage from the treated tissues was greatly influenced by the light intensity to which they were exposed. Higher light intensities stimulated electrolyte leakage and reduced lag period. Porphyrin biosynthesis inhibitors, gabaculine and 4,6-dioxoheptanoic acid, completely inhibited electrolyte leakage from the oxyfluorfen-treated tissues. Protection against the activity of $\delta$-aminolevulinic acid from electrolyte leakage was complete with 4,6-dioxoheptanoic acid, but not with gabaculine. However, gabaculine and 4,6-dioxoheptanoic acid gave no such protection against rose bengal activity. In summary, our results indicate that $\delta$--aminolevulinic acid, oxyfluorfen, and rose bengal exert their effects by causing electrolyte leakage from the treated tissues in a similar manner, except that oxyfluorfen has an apparent lag period for its action on electrolyte leakage increase. All above compounds require preincubation of treated tissues in darkness and subsequent light exposure with a high intensity for their maximal activities. Our results also support that in the presence of light, $\delta$-aminolevulinic acid and oxyfluorfen cause cellular damage through the indirect generation of singlet oxygen from accumulated tetrapyrroles of porphyrin pathway, whereas rose bengal causes cellular damage through the direct generation of singlet oxygen.
Cucumber (Cucumis sativus L.) plants were regenerated through organogenesis and somatic embryogenesis in cotyledon and hypocotyl cultures. The shoots were efficiently formed on the basal region of cotyledons cultured on MS medium containing 1.0㎎/L zeatin and 0.1㎎/L IAA in all cultivars used. Embryogenic calli were formed on hypocotyl segments cultured on MS medium containing 1.0㎎/L 2,4-D in cv. group 'Nakhab' and maintained by consecutive subculture on the same medium every 2-3 weeks without loss of embryogenic ability. Upon transfer to MS basal medium, high frequency somatic embryogenesis was achieved easily from embryogenic callus. Regenerated plantlets through organogenesis and somatic embryogenesis were transplanted to pots and gradually acclimatized to greenhouse condition where they subsequently produced fruits.

Root colonization of biocontrol agents via seed treatment was investigated and a compatible combination, Gliocladium virens G872B and Pseudomonas putida Pf3, in colonizing cucumber rhizosphere was confirmed through the study. Much higher number of fungal and bacterial propagules were detected when two isolates were inoculated together. The presence of Pf3 in root system was greatly helpful to G872B to colonize at root tip. The mechanism of this phenomenon is partially elucidated through the results of in vitro experiments and the observations of scanning electron and fluorescence microscope. Addition of Pf3 cells resulted earlier germination of G872B conidia and increased mycelial growth. And the more number of germinated conidia on seed coat, the more vigorous hypal streching and sporulation on the root surface were observed in coinoculated treatment. The propagules of G872B on the cucumber root when they were challenged against the pathogenic Fusarium oxysporum, were even higher than that of G872B treated alone, and the magnitude of such a difference was getting grater toward the root ip and the population of F. oxysporum on the root was reduced by seed inoculation of G872B. The rhizosphere competence was obviously reflected to disease suppression and plant growth promotion that induced by the given isolate. Green house experiments revealed that the combined treatment provided long-term disease suppression with greater rate and the larger amount of fruit yield than single treatments. Through this study the low temperature growing Pseudomonas fluorescens M45 and MC07 were evaluated to apply them to the winter crops in field or plastic film house. In vitro tests reveal that M45 and MC07 inhibited the mycelial growth of Pythium ultimum, Rhizoctona solani and Phytophthora capsici and enhanced growth of cucumber cotyledon in MS agar. This effect was more pronounced when the bacteria were incubated at 14$^{\circ}C$ than at 27$^{\circ}C$. And disease suppression and plant growth promotion in green house were also superior at low temperature condition. Seed treatment of M45 or soil treatment of MC07 brought successful control of damping-off and enhanced seedling growth of cucumber. The combined treatment of two isolates was more effective than any single treatment.
The germination of cucumber seeds begins with the degradation of reserved oil to fatty acids within the lipid body, which are then further metabolized to acyl-CoA. The acyl-CoA moves from the lipid body to the glyoxysome following β-oxidation for the production of acetyl-CoA. As an initial carbon source supplier, acetyl-CoA is an essential molecule in the glyoxylate cycle within the glyoxysome, which produces the metabolic intermediates of citrate and malate, among others. The glyoxylate cycle is a necessary metabolic pathway for oil seed plant germination because it produces the metabolic intermediates for the tricarboxylic acid (TCA) cycle and for gluconeogenesis, such as the oxaloacetate, which moves to the cytosol for the initiation of gluconeogenesis by phophoenolpyruvate carboxykinase (PEPCK). Following reserved oil mobilization, the production and transport of various metabolic intermediates are involved in the coordinated operation and activation of multiple metabolic pathways to supply directly usable carbohydrate in the form of glucose. Furthermore, corresponding gene expression regulation compatibly transforms the microbody to glyoxysome, which contains the organelle-specific malate synthase (MS) and isocitrate lyase (ICL) enzymes during oil seed germination. Together with glyoxylate cycle, carnitine, which mediates the supplementary route of the acetyl-CoA transport mechanism via the mitochondrial BOU (A BOUT DE SOUFFLE) system, possibly plays a secondary role in lipid metabolism for enhanced plant development.
This study was conducted to establish an efficient screening method of membrane-disrupting herbicides, based on the result that membrane leakage leading to evolution of carbonyl compounds and aldehydes could be colorimetrically assessed by formation of stable chromophores with MBTH and ferric chloride. Assay conditions including plant material, light intensity, irradiation time, concentrations of reagents for colorization, and reaction time were optimized, and activities of different types of known herbicides were investigated with cucumber cotyledon discs. That result indicated that this assay was sensitive to membrane-disrupting herbicides, such as protoporphyrinogen oxidase inhibitors (acifluorfen, oxyfluorfen, oxadiazon) and an electron interceptor from photosystem I(paraquat). With seven compounds newly synthesized, highly positive correlation was observed between herbicidal activities measured by greenhouse bioassay and MBTH method. Because only a single disc(4 mm diameter) from cucumber cotyledon in 60 ${\mu}L$ of incubation solution was required to assay, a large number of herbicides could be tested simultaneously using 96-well microtitre plates. But the assay efficiency is not likely to significantly increase because of the boiling step during the reaction.
This study was conducted to characterize herbicidal activity of the new type diphenylether compounds inducing bleaching(whitening) in relation to their chemical structures and to find out the herbicidal action mechanism. Bleaching was highly appeared in the compounds which have short chain alkyl or allyl group of cabamoyl type in meta position of A ring and nitro group in para position of B ring, and KC6361 showed the highest efficacy among these derivatives. Rice was tolerant to KC6361 at 2.0kg/ha. The applications of this compound at 0.25~1.0kg/ha provided acceptable control of the various weeds including large crabgrass, barnyardgrass, pigweed and so on. In the dark condition, KC6361 decreased carotenoid contents but increased chlorophyll in etiolated cucumber cotyledon. By the increase of light intensity, the carotenoid biosynthesis was more inhibited than the chlorophyll biosynthesis. The incorporation of $C^{14}$-acetate into lipid was slightly inhibited. Both KC6361 and norflurazon stimulated growth of dwarf rice(cv. Daneunbanju). These results suggest that both KC6361 and norflurazon may have the same action sites. To confirm this hypothesis, the changes of carotenoid intermediates in cucumber cotyledon and barnyardgrass leaf treated with KC6361 were investigated. Phytoene and phytofluene were increased but ${\beta}$-carotene was decreased, indicating that KC6361 inhibited phytoene and/or phytofluene dehydrogenase like norflurazon.
