  • Large bowel cancer is one of the major causes of increasing world-wide cancer mortality. Our group has found a variety of naturally-occurring and synthetic compounds which have chemopreventive potentials against the occurrence of large bowel cancers using animal models. Aberrant crypt foci (ACF) which develop in rodents soon after the carcinogen exposure have been used as a biomarker for screening effective agents for cancer chemoprevention.(omitted)

  • The endocrine cells in the midgut epithelium of the Japanese cockroach, Periplaneta japonica were observed by the light and electron microscopy. The midgut epithelium of the last instar larva and adult cockroach consisted of principal columnar cells, regenerative cells, and secretory granular cells. Midgut endocrine cells were positioned basally as a cone-shaped single cell in the epithelium or underneath the regenerative crypt cells. When midgut epithelium grows and the cell composing it transforms, between the endocrine cells and regenerative cells were made desmosome type junction and large vesicular shaped stretches of loose contact. The endocrine cells were characterized by a clear cytoplasm with abundant Golgi complex and numerous secretory granules. The secretory granules in the cell were spherical and electron dense with their diameter of $200{\sim}400nm$. The secretory granules have been observed as discharged by exocytosis on the basal and lateral side of the cell.

  • Acute hemorrhagic enteritis was diagnosed in a seven-month-old male Shitzu dog dying of blood stained diarrhea and vomiting. Clinical findings were anorexia, dullness and sudden death after massive bloody diarrhea. At necropsy, main lesion was the hemorrhage in small intestine, mainly duodenum and jejunum. Microscopically, Gram positive long bacilli were massively detected on the mucose epithelial cells and necrotic debris of small intestine. Coagulative necrosis of epithelial cells and thrombosis of small intestine were also identified. However, there was no lesion of crypt epithelium. Mineral infiltration in both gastric mucosa and renal tubules was detected and proliferation of fibrous tissue was also shown in corticomedullary regions. In bacterial examination, C perfringens was isolated in anaerobic culture and it was confirmed to type A by multiplex PCR. Therefore, the dog was diagnosed as C perfringens type A associated enteritis with uremia.

Fistula-in-Ano in Children less than 2 Years of Age (2세 이하 소아 치루에 대한 임상적 분석)

  • The clinical characteristics of fistula-in-ano in infants are different from those of older children, and its treatment remains controversial. We suggest that fistula-in-ano in infants has a congenital etiology. To verify this hypothesis and to settle the controversies regarding fistula-in ano in infants, a retrospective analysis of 29 patients less than 2 years of age with anal fistulae treated between 1994 and 2009 at Samsung Changwon Hospital were reviewed retrospectively. Twenty two patients were male and mean age at diagnosis was $7.2{\pm}5.2$ months. Eleven out of 22 cases had previous surgical drainage for perianal abscess. 18 patients had fistulotomy (81.8 %) and four had fistulectomy (18.2 %). Cryptotomies with fistulectomy were performed in 10 patients (45.5 %) who had involved crypt. There was one recurrence. These results suggest that fistula-in-ano in young children less than two years of age is different from those in older children or adults. Fistulotomy is suggested to be the recommended treatment of choice. A future study involving non-operative management would be required to explore all treatment options.

  • Glucagon-like peptide 2 receptor, a GPCR, binds with the glucagon-like peptide, GLP-2 and regulates various metabolic functions in the gastrointestinal tract. It plays an important role in the nutrient homeostasis related to nutrient assimilation by regulating mucosal epithelium. GLP-2 receptor affects the cellular response to external injury, by controlling the intestinal crypt cell proliferation. As they are therapeutically attractive towards diseases related with the gastrointestinal tract, it becomes essential to analyse their structural features to study the pathophysiology of the diseases. As the three dimensional structure of the protein is not available, in this study, we have performed the homology modelling of the receptor based on single- and multiple template modeling. The models were subjected to model validation and a reliable model based on the validation statistics was identified. The predicted model could be useful in studying the structural features of GLP-2 receptor and their role in various diseases related to them.

  • Glucagon like pepide-2, one of the GLPs, is involved in various metabolic functions in the gastrointestinal tract. It plays a major role in the regulation of mucosal epithelium and the intestinal crypt cell proliferation. Because of their therapeutic importance towards the diseases in the gastrointestinal tract, it becomes necessary to study their interaction with its receptor, GLP-2R. In this study, we have developed protein-protein docking complexes of GLP-2 - GLP-2 receptor. Homology models of GLP-2 are developed, and a reliable model out of the predicted models was selected after model validation. The model was bound with the receptor, to study the important interactions of the complex. This study could be useful in developing novel and potent drugs for the diseases related with GLP-2.

An Experimental Study on Radioprotective Effect of DDC, MEA, and WR-2721 (DDC, MEA, WR-2721의 방사선(放射線) 방호효과(防護效果)에 관(關)한 연구(硏究))

  • At present, the treatments of the radiation-induced diseases are only performing by the palliative treatment technique. Moreover, radiation protective agents are a little toxic for human being and this seriously limits their applicability with various complications in clinical uses. Accordingly, as a part of the aim of gain of the basic data for protective roles of some radioprotectors, the present investigation was carried out to evaluate the comparative radioprotective effects by the administration of DDC, MEA, WR-2721. Results are shown for statistically significant analysis and correlation with each group as follows; 1. The proper doses of the radioprotectors were DDC; 1,550 mg/kg, MEA; 450 mg/kg, WR-2721; 780 mg/kg of the mouse body weight. 2. DMF(Dose modification factor) of LD 50/10 and LD 50/30 for whole body irradiation was DDC; 1.2, MEA; 1.4, WR-2721; 1.9 and DDC; 1.7, MEA; 1.8, WR-2721; 2.5 respectively. 3. DMF for radiation reaction of jejunal crypt was DDC: 1.07, MEA: 1.21 and WR-2721: 1.76 and that of jejunal crypt cell was DDC: 1.04, MEA: 1.08 and WR-2721: 1.38 respectively. 4. Conclusively, WR-2721 was the most effective drung among the three radioprotectors and this result must be a supportive data for further study for clinical application.

Effects of Feeding and Processing Methods of Diets on Performance, Morphological Changes in the Small Intestine and Nutrient Digestibility in Growing-Finishing Pigs

  • These experiments were conducted to investigate the effects of different feeding and processing methods of diets on performance, morphological changes in the small intestine and nutrient digestibility of growing-finishing pigs. One-hundred fifty growing pigs ($Yorkshire{\times}Landrace{\times}Duroc$; initial body weight of $23.33{\pm}0.75kg$) and one-hundred twenty finishing pigs ($Yorkshire{\times}Landrace{\times}Duroc$; initial body weight of $59.22{\pm}0.56kg$) were used in Exp. 1 and Exp. 2, respectively. Pigs were grouped on the basis of body weight and gender, and randomly allotted into 6 different treatments with 5 replications in each treatment in a $2{\times}3$ factorial arrangement. Treatments were 1) dry feeding with a mash diet (DM), 2) dry feeding with a pelleted diet (DP), 3) dry feeding with an expanded crumble diet (DEC), 4) dry/wet feeding with a mash diet (WM), 5) dry/wet feeding with a pelleted diet (WP), and 6) dry/wet feeding with an expanded crumble diet (WEC). In Exp. 1 (growing phase), there was no significant difference in average daily gain (ADG) and average daily feed intake (ADFI) among treatments during the entire experimental period, but feed conversion ratio (FCR) was significantly (p<0.05) improved in pigs fed pelleted diets regardless of feeding method. FCR was best in pigs fed a DP diet and worst in pigs fed a WM diet. Pigs fed a pelleted diet showed a 6.2% or 4.0% improvement in FCR compared with those fed a mash diet or an expanded crumble diet. Water disappearance was not significantly affected by dry/wet feeding or feed processing. Significant differences in villus height were not found among treatments, but villus height tended to be improved by dry/wet feeding. Dry/wet feeding or feed processing did not affect crypt depth. Digestibilities of calcium and phosphorus were significantly (p<0.05) improved in pigs fed an expanded crumble diet compared with pigs fed mash diets. Especially, pigs fed a WEC diet digested 8.1% more P than those fed a DM diet. Feed cost per kg weight gain (FCG) tended to be increased by dry/wet feeding rather than dry feeding. In Exp. 2 (finishing phase), ADG and ADFI were not significantly different among treatments, but a significant difference in FCR was found among feed processing forms. The best FCR was obtained in pigs fed a pelleted diet. Pigs fed a DP diet showed a 11.3% improvement compared with those fed a DEC diet. Water disappearance was significantly (p=0.0408) decreased by feeding the mash diet. However, water disappearance was not affected by dry/wet feeding during the finishing period. The villus height and crypt depth were not significantly different among treatments. However, crypt depth tended to be decreased by dry/wet feeding at the mid part of the small intestine. Fat digestibility was improved by dry feeding rather than dry/wet feeding, and was improved by 4.8% by feeding pellet diets compared with expanded crumble diets. Except for carcass grade, carcass characteristics were not significantly (p<0.05) different among treatments. Carcass grade was the best in pigs fed a WP diet. Feed cost per kg weight gain (FCG) was significantly decreased in pigs fed a pelleted diet compared with those fed an expanded crumble diet, and tended to be decreased by dry/wet feeding. In conclusion, these studies suggest that feeding the pelleted diet to growing-finishing pigs can be beneficial in terms of FCR and production cost. Dry/wet feeding can be helpful for the maintenance of villus height, but may not be reflected in improved growth performance or reduction of production costs.

Immunohistochemical study on distribution of proliferating cells and tumor formations in intestinal tracts of 1,2-dimethylhydrazine-treated rats (1,2-dimethylhydrazine을 투여한 rat 장관의 증식세포 분포와 종양발생에 대한 면역조직화학적 연구)

  • The present investigation was focussed mainly on the development of the tumors and proliferating cells on the intestinal tracts of 1, 2-dimethyl-hydrazine(DMH)-treated young or adult rats. 26 rats(Wistar, 14 young rats weighting approximately 130~180gm and 12 adult rats weighting approximately 500~550gm) were given subcutaneously once weekly with 20mg of DMH/kg body weight(BW)/week for 8~22 weeks. Individual body weight were recorded weekly at the same day and time. The rats were killed at 8, 13, 15. 17, 19, 21 and 22 weeks. The intestinal tracts were opened longitudinally and carefully examined for tumors. The localization, number, and size of tumors were noted. Tumor-bearing areas were dissected out and fixed on neutral buffered 10% formalin and normal-looking mucosa from 8~22 weeks rats were also taken for fixation. Paraffin sections were stained by H-E for histopathological examination or with immunohistochemical stain for bromodeoxyuridine(Brdur) positive cells. 1. The growth proportion of body weight appeared to be decreased in the DMH-treated young rats than in control young rats and body weight of DMH-treated adult rats appeared to be 13.4% or less lower than weighted on 0 week. 2. Macroscopically, the developed tumors in the intestinal tracts were not observed as early as the 13 weeks after DMH treatment. The number of developed tumors per rat was found to be 14.3, 18.8, 22.3 in 15, 17 and 22 weeks. The numbers of tumors in intestinal regions per rat were 2.1, 4.3, 5.4, 2.5 in duodenum, jejunum, ilium and colon on 15 weeks, 2.3, 6.4, 7.8, 2.3, on 17 weeks, and 2.7, 9.3, 9.0, 1.3 on 22 weeks, respectively and the ileum and jejunum were higher in appearance rate of tumors and tumor types are dome shapes and diameter of largest tumor were 6.3mm. 3. Histopathologically, intestinal mucosa were thickened by the irregular distorted and distended crypts following hyperplasia. The tumors developed on the mucosa and submucosa and were recognized to be adenocarcinoma. 4. Immunohistochemically, the labeling index(LI) was calculated as the ratio of the number of Brdur-labeled cells to the total number of column cells of the crypts with longitudinal axis. LI of Brdur positive cells per crypt were 5.6%, 8.0% on small intestine of control and 22 week group, respectively and 3.7%, 12.7% on large intestine of control and 22 week group, respectively and were appeared to be increase in 22 week group than in control group and to be more number of proliferating cells in 22 week group than in control group. 5. LI of Brdur positive cells in 1, 2, 3, 4, 5 segments of crypt column were 11.7%, 10.7%, 3.8%, 0.6%, 0% in small intestine of control group and 23.5%, 11.8%, 2.3%, 2.4%, 0.8% in small intestine of 22 week group, and 5.4%, 7.4%, 3.8%, 1.0%, 0.4% in large intestine of control group and 29.5%, 20.3%, 5.9%, 6.3%, 1.3% in large intestine of 22 week group respectively. So results indicate that the number of proliferating cells by DMH treatment increase and were concentrated on the 1, 2 segments of crypt columns.

Effect of Bacillus amyloliquefaciens-based Direct-fed Microbial on Performance, Nutrient Utilization, Intestinal Morphology and Cecal Microflora in Broiler Chickens

  • The present study was conducted to evaluate the effect of the dietary supplementation of Bacillus amyloliquefaciens-based direct-fed microbial (DFM) on growth performance, nutrient utilization, intestinal morphology and cecal microflora in broiler chickens. A total of two hundred and eighty eight 1-d-old Arbor Acres male broilers were randomly allocated to one of four experimental treatments in a completely randomized design. Each treatment was fed to eight replicate cages, with nine birds per cage. Dietary treatments were composed of an antibiotic-free basal diet (control), and the basal diet supplemented with either 15 mg/kg of virginiamycin as antibiotic growth promoter (AGP), 30 mg/kg of Bacillus amyloliquefaciens-based DFM (DFM 30) or 60 mg/kg of Bacillus amyloliquefaciens-based DFM (DFM 60). Experimental diets were fed in two phases: starter (d 1 to 21) and finisher (d 22 to 42). Growth performance, nutrient utilization, morphological parameters of the small intestine and cecal microbial populations were measured at the end of the starter (d 21) and finisher (d 42) phases. During the starter phase, DFM and virginiamycin supplementation improved the feed conversion ratio (FCR; p<0.01) compared with the control group. For the finisher phase and the overall experiment (d 1 to 42) broilers fed diets with the DFM had better body weight gain (BWG) and FCR than that of control (p<0.05). Supplementation of virginiamycin and DFM significantly increased the total tract apparent digestibility of crude protein (CP), dry matter (DM) and gross energy during both starter and finisher phases (p<0.05) compared with the control group. On d 21, villus height, crypt depth and villus height to crypt depth ratio of duodenum, jejunum, and ileum were significantly increased for the birds fed with the DFM diets as compared with the control group (p<0.05). The DFM 30, DFM 60, and AGP groups decreased the Escherichia coli population in cecum at d 21 and d 42 compared with control group (p<0.01). In addition, the population of Lactobacillus was increased in DFM 30 and DFM 60 groups as compared with control and AGP groups (p<0.01). It can be concluded that Bacillus amyloliquefaciens-based DFM could be an alternative to the use of AGPs in broilers diets based on plant protein.
