A study on the effect of Ni-rich phase on the stress-rupture properties of Ni microalloyed W (W 활성소결체의 Ni-rich 상의 양이 응력 파단 성질에 미치는 영향에 관한 연구)

  • The effect of Ni-rich phase on the stress-rupture properties of Ni mlcroalloyed W were studied using direct load creep tester at 100$0^{\circ}C$, 110$0^{\circ}C$, and 120$0^{\circ}C$ in $H_2.$ The stress rupture strength of 100hrs. of W-0.4wt% Ni was 43% higher at 100$0^{\circ}C$ and 35% higher at 110 $0^{\circ}C$than that of W-0.2wt% Ni due to the larger initial grain size, the higher relative density and the higher grain growth during test. That of W-0.8wt% Ni was 90% higher at 100$0^{\circ}C$ and 60% higher at 110$0^{\circ}C$ than that of W-0.2wt% Ni. The activation energy of W-0.4wt% Ni for creep was 81.3 Kcal/mole. It was considered that creep deformation was controlled by the diffusion of W in the Ni rich phase between the grains and the deformation of grains. All of the specimens showed intergranular fracture by grain boundary cavitation and growth of cavity throughout the entire spcimen cross-section.

The effects of face velocity and path length on the uptake rates of volatile organic compounds measured by diffusive samplers (확산포집기로 공기중 ppb 농도수준의 휘발성유기물질 포집시 확산길이와 기류변화가 시료포집속도에 미치는 영향)

  • Passive samplers have been used for personal, indoor, and outdoor air monitoring of VOCs at ppb concentrations in community and office environments. The path length of modified passive sampler was shortened, so it was intended to increase an uptake rate. The performance of the modified 3M 3500 organic vapor monitor(OVM) as a tool for assessing exposures to toxic air pollutants in nonoccupational community environments was evaluated using combined controlled test atmospheres of six selected target volatile organic compounds(VOCs): benzene, methyl tert-butyl ether(MTBE), chloroform, 1,4-dichlorobenzene, tetrachloroethylene, and toluene. The experiments were conducted by exposing the dosimeters to concentrations of $50{\sim}100{\mu}g/m^3$ on six face velocity(0.00, 0.02, 0.06, 0.12, 0.20, 0.30 m/sec) for 24 hours. If the uptake rate was increased, that means that we could use the passive sampler more effectively. The uptake rates were increased linearly according to reduce the path length. Although the diffusion path length was shortened, the change of uptake rate was within ${\pm}25%$ of theoretical value, indicating that the modified passive sampler(TM) can be effectively used over the range of concentrations and environmental conditions tested with a 24-h sampling period if the face velocities were over 0.12 m/s for 6 components of VOCs. But when the face velocities were less than 0.12 m/s, uptake rates were reduced more than expected values. So, the passive sampler with the shortened path length should be used at indoor or outdoor environment where the face velocity should be over about 0.10 m/s. If the path length was shortened more, the uptake rate was more effected by starvation.

Immunochemical Studies for the Characterization of Purified $(Na^+,\;K^+)-ATPase$ and Its Subunits with a Special Reference of Their Effect on Monovalent Cation Transport in Reconstituted $(Na^+,\;K^+)-ATPase$ Vesicles

  • A highly purified $(Na^+,\;K^+)-ATPase$ from the rectal gland of Squalus acanthias and from the electric organ of Electrophorus electricus has been used to raise antibodies in rabbits. The 97,000 dalton catalytic subunit and glycoprotein derived from the rectal gland of spiny shark were also used as antigens. The two $(Na^+,\;K^+)-ATPase$ holoenzymes and the two shark subunits were antigenic. In Ouchterlony double diffusion experiments, these antibodies formed precipitation bands with their antigens. Antibodies prepared against the two subunits of shark holoenzyme also formed precipitation bands with their antigens and shark holoenzyme, but not with eel holoenzyme. These observations are in good agreement with inhibitory effect of these antibodies on the catalytic activity of $(Na^+,\;K^+)-ATPase$ both from the shark and the eel, since there is very little cross-reaction between the shark anticatalytic subunit antibodies and the eel holoenzyme. The maximum antibodies titer of the anticatalytic subunit antibodies is found to be 6 weeks after the initial single exposure to this antigen. Multiple injections of the antigen increased the antibody titer. However, the time required to produce the maximum antibody titer was approximately the same. These antibodies also inhibit catalytic activity of $(Na^+,\;K^+)-ATPase$ vesicles reconstituted by a slow dialysis of cholate after solubilization of the enzyme in a presonicated mixture of cholate and phospholipid. In these reconstituted $(Na^+,\;K^+)-ATPase$ vesicles, effects of these antibodies on the fluxes of $Na^+$, $Rb^+$, and $K^+$ were investigated. Control or preimmune serum had no effect on the influx of $^{22}Na^+$ or the efflux of $^{86}Rb^+$. Immunized sera against the shark $(Na^+,\;K^+)-ATPase$ holoenzyme, its glycoprotein or catalytic subunit did inhibit the influx of $^{22}Na^+$ and the efflux of $^{86}Rb^+$. It was also demonstrated that these antibodies inhibit the coupled counter-transport of $Na^+$ and $K^+$ as studied by means of dual labeling experiments. However, this inhibitory effect of the antibodies on transport of ions in the $(Na^+,\;K^+)-ATPase$ vesicles is manifested only on the portion of energy and temperature dependent alkali metal fluxes, not on the portion of ATP and ouabain insensitive ion movement. Simultaneous determination of effects of the antibodies on ion fluxes and vesicular catalytic activity indicates that an inhibition of active ion transport in reconstituted $(Na^+,\;K^+)-ATPase$ vesicles appears to be due to the inhibitory action of the antibodies on the enzymatic activity of $(Na^+,\;K^+)-ATPase$ molecules incorporated in the vesicles. These findings that the inhibitory effects of the antibodies specific to $(Na^+,\;K^+)-ATPase$ or to its subunits on ATP and temperature sensitive monovalent cation transport in parallel with the inhibitory effect of vesicular catalytic activity by these antibodies provide direct evidence that $(Na^+,\;K^+)-ATPase$ is the molecular machinery of active cation transport in this reconstituted $(Na^+,\;K^+)-ATPase$ vesicular system.

Comparative molecular field analyses (CoMFA) on the antifungal activity against phytophthora blight fungus of 3-phenylisoxazole and 3-phenyl-2,5-dihydroisoxazol-5-one derivatives (고추 역병균에 대한 3-phenylisoxazole과 3-phenyl-2,5-dihydroisoxazol-5-one 유도체들의 살균 활성에 관한 비교 분자장 분석 (CoMFA))

  • 3D-QSAR between fungicidal activitives ($pI_{50}$) against metalaxyl-sensitive (SPC: 95CC7105) or metalaxyl-resisitant (RPC: 95CC7303) isolate of phytophthora blight fungus (Phytophthora capsici), and a set of 3-phenylisoxazole (A) and 3-phenyl-2,5-dihydroisoxazole (B) derivatives as substrates were conducted using comparative molecular field analyses (CoMFA). The antifungal activities of (A) were generally higher than those of (B). And it is assumed that the most stable conformation of the active substrate was approximately planar from conformational search. The CoMFA models proved a good predictive ability and suggested that the electronic field of substrates were higher than hydropohobic field and steric field requirements for recognition forces of the receptor site. And the factors were strongly correlated (cross-validated $q^2>0.570$ & conventional $r^2>0.968$) with the fungicidal activitives. According to the CoMFA analyses, the selectivity factors for RPC suggested that the sterically bulky groups (C14 & C15) and electron withdrawing groups (C15 & C16) have to be introduced to the ortho, meta and para-position on the benzoyl moiety of substrates.

Electrical Properties in $Pt/SrTiO_3/Pb_x(Zr_{0.52}, Ti_{0.48})O_3/SrTiO_3/Si$ Structure and the Role of $SrTiO_3$ Film as a Buffer Layer ($Pt/SrTiO_3/Pb_x(Zr_{0.52}, Ti_{0.48})O_3/SrTiO_3/Si$ 구조의 전기적 특성 분석 및 $SrTiO_3$박막의 완충층 역할에 관한 연구)

  • $Pt/SrTiO_3/Pb_x(Zr_{0.52}, Ti_{0.48})O_3/SrTiO_3/Si$ structure was prepared by rf-magnetron sputtering method for use in nondestructive read out ferroelectric RAM(NDRO-FEAM). PBx(Zr_{0.52}Ti_{0.48})O_3}$(PZT) and $SrTiO_3$(STO) films were deposited respectively at the temperatures of $300^{\circ}C and 500^{\circ}C$on p-Si(100) substrate. The role of the STO film as a buffer layer between the PZT film and the Si substrate was studied using X-ray diffraction (XRD), Auger electron spectroscopy (ASE), and scanning electron microscope(SEM). Structural analysis on the interfaces was carried out using a cross sectional transmission electron microscope(TEM). For PZT/Si structure, mostly Pb deficient pyrochlore phase was formed due to the serious diffusion of Pb into the Si substrate. On the other hand, for STO/PZT/STO/Si structure, the PZT film had perovskite phase and larger grain size with a little Pb interdiffusion. the interfaces of the PZT and the STO film, of the STO film and the interface layer and $SiO_2$, and of the $SiO_2$ and the Si substate had a good flatness. Across sectional TEM image showed the existence of an amorphous layer and $SiO_2$ with 7nm thickness between the STO film and the Si substrate. The electrical properties of MIFIS structure was characterized by C-V and I-V measurements. By 1MHz C-V characteristics Pt/STO(25nm)/PZT(160nm)/STO(25nm)/Si structure, memory window was about 1.2 V for and applied voltage of 5 V. Memory window increased by increasing the applied voltage and maximum voltage of memory window was 2 V for V applied. Memory window decreased by decreasing PZT film thickness to 110nm. Typical leakage current was abour $10{-8}$ A/cm for an applied voltage of 5 V.

The High-speed train model for reducing the micro-pressure wave in railway tunnel (고속철도 터널에서의 미기압파 저감을 위한 모형실험)

  • The purpose of the study was to verify deduction of each coefficient necessary to analysis on micro-pressure waves and reliability of the analysis result. The tunnel running train model testing device used in the test was manufactured by scale of 1:60 and the study used a train model with ten cars long according to specifications of KTX model. The study applied tunnels with cross sections of $107.9m^3\;and\;95.1m^3$ and applied tunnel extensions with 1km, 0.75km and 0.5km. Also, the study tested train speed by changing it into 275, 300, 325 and 350km/h. The test device was a hydraulic launch system composed of a train model, a hydraulic launcher, a tunnel model and a brake. The study measured speed of a model trainby a speed sensor installed in the point of each 1.2m from the front of tunnel entrance and a pithead of tunnel exit and measured pressure change of internal tunnel continuously by installing pressure sensors in the entrance part of tunnel, in the middle part of tunnel and in the exit part of tunnel. As the result of the measurement, it was known that pressure slope of pressure wave happened in the entrance part of tunnel was increased by a nonlinear effect while spreading the tunnel or its pressure slope was reduced by diffusion. Also, the study compared and analyzed micro-pressure waves happened in the exit part of the tunnel by installing each kind of hoods in the entrance part of the tunnel to prevent reduction of micro-pressure waves.

The Effect of a Au Based Bonding Agent Coating on Non-Precious Metals-Ceramic Bond Strength (비귀금속 합금에 적용한 Au Based Bonding Agent가 금속-도재 결합에 미치는 영향)

  • The purpose of this study investigated the effect of Au coating on adhesion between porcelain matrix and metal substructure interface. Titanium, Ni-Cr alloy and Co-Cr alloy are well known as proper metal for the dental restorations. The success of a porcelain fused to metal (PFM) restoration depends upon the quality of the porcelain-metal bond. However, adhesion between dental alloys and porcelain is related to diffusion of oxygen during ceramic firing. The excessive oxidized layers make hard adhesion between dental alloy and ceramic. Ni-Cr and Co-Cr specimens were divided into test and a control group and Titanium specimens were divided into three test groups and a control group. Each group had 20 specimens. The adhesion characteristics of porcelain and metal with Au coating layer and without Au coating layer were observed with scanning electron microscopy(SEM). The adhesion was evaluated by a biaxial flexure test and volume fraction of adherent porcelain was determined by SEM/EDS analysis. Result of this study suggest that Au coating layer is effective barrier to diffuse oxide layer completely protect non-precious alloys from oxidation during the porcelain firing. The SEM photomicrographs of cross-section specimens showed a smooth interface between Au coating layer and metals and porcelain which suggested proper chemical bonding, and no gap, porosity were observed. The mode of failure was mainly adhesive for Ti tested specimens, but mixed failures with adhesive and cohesive were observed in Ni-Cr and Co-Cr specimens. The adhesion between non-precious metals and porcelain would not be improved by Au coating agent. However, It is suggested that the continuous study is required further investigation and development.

Elastic Behavior of Zeolite Mesolite under Hydrostatic Pressure (제올라이트 메소라이트의 수압 하 탄성특성)

  • Powder diffraction patterns of the zeolite mesolite ($Na_{5.33}Ca_{5.33}Al_{16}Si_{24}O_{80}{\cdot}21.33H_2O$), with a natrolite framework topology were measured as a function of pressure up to 5.0 GPa using a diamond-anvil cell and a $200{\mu}m$-focused monochromatic synchrotron X-ray. Under the hydrostatic conditions mediated by pore-penetrating alcohol and water mixture, the elastic behavior of mesolite is characterized by continuous volume expansion between ca. 0.5 and 1.5 GPa, which results from expansion in the ab-plane and contraction along the c-axis. Subsequent to this anomalous behavior, changes in the powder diffraction patterns suggest possible reentrant order-disorder transition. The ordered layers of sodium- and calcium-containing channels in a 1:2 ratio along the b-axis attribute to the $3b_{natrolite}$ cell below 1.5 GPa. When the volume expansion is completed above 1.5 GPa, such characteristic ordering reflections disappear and the $b_{natrolite}$ cell persists with marginal volume contraction up to ca. 2.5 GPa. Further increase in pressure leads to progressive volume contraction and appears to generate another set of superlattice reflections in the $3c_{natrolite}$ cell. This suggests that mesolite in the pressure-induced hydration state experiences order-disorder-order transition involving the motions of sodium and calcium cations either through cross-channel diffusion or within the respective channels.

Bioequivalence of Two Clarithromycin Tablets (클래리스로마애신 정제의 생물학적 동등성 평가)

  • The bioequivalence of two clarithromvcin products was evaluated with 16 normal male volunteers (age 23-28 yr, body weight 57.5-75.517g) following single oral dose. Test product was ReYon Clarithromycin tablets (ReYon Pharm. Corp., Korea) and reference product was Klarici $d_{R}$ tablets (Abbott Korea). Both products contain 250 mg of clarithromucin. One tablet of the test or the reference product was administered to the volunteers, respectively, by randomized two period cross-over study (2$\times$2 Latin square method). The determination of clarithromycin was accomplished using a modified agar well diffusion bioassay. As a result of the assay validation, the quantification of clarithromycin in human serum by this technique was possible down to 0.03$\mu$g/ml using 100$\mu$l of serum. The coefficient of variation (C.V.) was less than 10%. Average drug concentrations at each sampling time and pharmacokinetic parameters calculated were not significantly different between two products P>0.05); the area under the curve to last sampling time (24 hr) (AU $Co_{24hr}$ (8.10$\pm$ 1.26 vs 8.22$\pm$ 1.627g . hr/ml), AUC from time zero to infinite (AU $Co_{\infty}$) (8.61 $\pm$ 1.28 vs 8.84$\pm$ 1.71 $\mu$g . hr/ml), maximum plasma concentration ( $C_{msx}$) (0.87$\pm$0.22 vs 0.88$\pm$0.19 $\mu$g/ml) and time to maximum plasma concentration ( $T_{max}$) (2.69 $\pm$0.48 vs 2.56$\pm$ 0.51 hr). The differences of mean AU $Co_{24h}$, $C_{msx}$ and $T_{msx}$ between the two products (1.44, 1.39, and 4.65%, respectively) were less than 20%. The power (1-$\beta$) and treatment difference ($\Delta$) for AU $Co_{24hr}$, and $C_{max}$ were more than 0.8 and less than 0.2, respectivly. Although the power for $T_{max}$ was under 0.8, $T_{max}$. of the two products was not significantly different each other (p>0.05). These results suggest that the bioavailability of ReYon Clarithromycin tablets is not significantly different from that of Klarici $d_{R}$ tablets. Therefore, two products are bioequivalent based on the current results. results.sults.sults.s.s.s.s.s.s.s.

A study on enclosure in Daesoeng-dong Site, Kimhae (김해(金海) 대성동(大成洞) 환호유적(環濠遺蹟) 연구(硏究))

  • There is no definite theory, which archeologists have believed to be built either for the protection or ceremonial purpose, of enclosure historical site exhibited from Bronze Age to Samhan era in Korea. The main conditions of choosing right enclosure were natural levee-backswamp-plateau, which all found in village on a hill. For this reasons Kimhae Daesoeng-dong enclosure discovered in 2001 is not an exception, and therefore this enclosure has been believed to be a beginning point of the farming society. In Korea there have been 2 types of enclosure. One is that enclosure entirely surrounds whole historic site itself and the other is that enclosure partially does. Although Kimhae Daesoeng-dong historic site has been discovered only part of it, it's not believed to have the first type-which surrounds the whole enclosure historic site In this research, 3 developing steps of enclosure historic site were defined. Enclosure surrounds only a part of the historic site is 1st step. Enclosure surrounds all around the site is 2nd step. The site discovered with big round clay pottery and developed into multiple enclosure is 3rd step. Since some researches have not been accomplished, the hypothesis above was suggested to have a better understanding of the development of enclosure historic site. The purpose of enclosure has been separately explained for the protection or for the ceremony. It is difficult to assert that excavation sites could be applied to only one of two purposes. But it is possible that both purposes of enclosure such as protection and as division from ceremonial area could be applied together. Kimhae Daesoeng-dong enclosure was shown for the purpose of division from ceremonial area. But we can not ignore that since Kimhae Daesoeng-dong enclosure exposed to the Sea and dwelling area was not shown from village hill, it would be probably for the protection. However enclosure of Gujibong village area in Daesoeng-dong had used for ceremonial purpose since the Bronze Age, even this enclosure would be possible to play an important role of being a center of legend of Gaya foundation. Many unsolved questions are still lying ahead. The enclosure was used for a short time, even if dwelling area was found in and out of enclosure, there were no differences between them, and although duplicate and triplicate enclosure were found, we could not sure which were before and after. Also we could not confirm the type of enclosure with location condition which was the flat land or the top of hill on floodplain. We should not divide the types of enclosure historic site due to differences from cross section of stratum. And I expect that we will have much data such as distribution of enclosure historic site in Yeongnam area where is concerned with origin and diffusion of Japan and China.
