Physical Mapping of rDNAs Using McFISH in Anemarrhena asphodeloides Bunge (지모에서 McFISH를 이용한 rDNAs의 물리지도 작성)

  • Anemarrhena asphodeloides, a medicinal plant, has chromosome number of 2n=2x=22. To characterize the somatic metaphase chromosomes, physical mapping of 45S and 5S rDNAs using McFISH (multi-color fluorescence in situ hybridization) was applied. Two pairs of 45S rDNA loci were detected on the terminal regions of the short arm of chromosomes 1 and 3. A pair of 5S rDNA signal was observed on the short arm of chromosome 3. 5S rDNA site seemed to be the same locus as one of the 45S rDNA site. McFISH was very useful tool for the localization and identification of rDNAs on the metaphase chromosomes in A. asphodeloides.

Use of Remotely-Sensed Data in Cotton Growth Model

  • Remote sensing data can be integrated into crop models, making simulation improved. A crop model that uses remote sensing data was evaluated for its capability, which was performed through comparing three different methods of canopy measurement for cotton(Gossypium hirsutum L.). The measurement methods used were leaf area index(LAI), hand-held remotely sensed perpendicular vegetation index(PVI), and satellite remotely sensed PVI. Simulated values of cotton growth and lint yield showed reasonable agreement with the corresponding measurements when canopy measurements of LAI and hand-held remotely sensed PVI were used for model calibration. Meanwhile, simulated lint yields involving the satellite remotely sensed PVI were in rough agreement with the measured lint yields. We believe this matter could be improved by using remote sensing data obtained from finer resolution sensors. The model not only has simple input requirements but also is easy to use. It promises to expand its applicability to other regions for crop production, and to be applicable to regional crop growth monitoring and yield mapping projects.

QTL Mapping of Resistance to Gray Leaf Spot in Ryegrass: Consistency of QTL between Two Mapping Populations

  • Gray leaf spot (GLS) is a serious fungal disease caused by Pyricularia oryzae Cavara, recently reported on the important turf and forage species, perennial ryegrass (Lolium perenneL.). This fungus also causes rice blast, which is usually controlled by host resistance, but durability of resistance is a problem. Few instances of GLS resistance have been reported in perennial ryegrass. However, two major QTL for GLS resistance have been detected on linkage groups 3 and 6 in an Italian x perennial ryegrass mapping population. To confirm that those QTL are still detectable in the next generation and can function in a different genetic background, a resistant segregant from this population has been crossed with an unrelated susceptible perennial clone, to form a new mapping population segregating for GLS resistance. QTL analysis has been performed in the new population, using two different ryegrass field isolates and RAPD, RFLP, and SSR marker-based linkage maps for each parent. Results indicate the previously identified QTL on linkage group 3 is still significant in the new population, with LOD and percent of phenotypic variance explained ranging from 2.0 to 3.5 and 5% to 10%, respectively. Also two QTL were detected in the susceptible parent, with similar LOD and phenotypic variance explained. Although the linkage group 6 QTL was not detected, the major QTL on linkage group 3 appears to beconfirmed. These results will add to our understanding of the genetic architecture of GLS resistance in ryegrass, which will facilitate its use in perennial ryegrass breeding programs.

Comparative Cytogenetic Characteristics and Physical Mapping of the 17S and 5S Ribosomal DNAs between Atractylodes japonica Koidz. and Atractylodes macrocephala Koidz.

  • This study was carried out to compare chromosomal characteristics between Atractylodes japonica and A macrocephala. Cytogenetic analysis was conducted based on karyotype analysis and physical mapping using fluorescence in situ hybridization. As a result of karyotype analysis by feulgen staining, somatic chromosome numbers of A. japonica and A. macrocephala were 2n=24. The length. of the mitotic metaphase chromosomes of A. japonica ranged from $0.70\;to\;1.60{\mu}m$ with a total length. of $12.11{\mu}m$ and the homologous chromosome complement comprised six metacentrics, five submetacentrics and one subtelocentrics. On the other hand, the length of the mitotic metaphase chromosomes of A. macrocephala ranged from $0.90\;to\;2.35{\mu}m$ with a total length of $16.58{\mu}m$ and the homologous chromosome complement comprised seven metacentrics and five submetacentrics. The total length of A. japonica chromosomes was shorter than that of A. macrocephala, but A. japonica had one subtelocentrics (chromosomes 4) different from A. macrocepha1a. chromosomes. The F1SH technique using 17S and 5S rDNA was applied to metaphase chromosomes. The signals for 17S rDNA were detected on the telomeric regions of chromosomes 4 and 5 in both A japonica and A. macrocephala. The 5S rDNA signal was found in the short arm of chromosome 1.

  • Quantitative trait loci, a genetic concept for explaining the inheritance of non-Mendelian traits in 1940s, have been realized as particular fragments of chromosome even unique genes in most crops in 21st century. However, only very a small portion of QTL has been screened out by geneticists comparing to a great number of genes underneath the quantitative traits. These identified QTL even have been seldom used into breeding program because crop breeders may not find the QTL in their breeding populations in their field station. Several key points will be proposed to meet the challenges of QTL analysis today: a fine mapping population and the related reference genetic map, QTL evaluation in multiple environments, recognizing real QTL with small genetic effect, map integration.

Improvement in Seasonal Prediction of Precipitation and Drought over the United States Based on Regional Climate Model Using Empirical Quantile Mapping (경험적 분위사상법을 이용한 지역기후모형 기반 미국 강수 및 가뭄의 계절 예측 성능 개선)

  • The United States has been known as the world's major producer of crops such as wheat, corn, and soybeans. Therefore, using meteorological long-term forecast data to project reliable crop yields in the United States is important for planning domestic food policies. The current study is part of an effort to improve the seasonal predictability of regional-scale precipitation across the United States for estimating crop production in the country. For the purpose, a dynamic downscaling method using Weather Research and Forecasting (WRF) model is utilized. The WRF simulation covers the crop-growing period (March to October) during 2000-2020. The initial and lateral boundary conditions of WRF are derived from the Pusan National University Coupled General Circulation Model (PNU CGCM), a participant model of Asia-Pacific Economic Cooperation Climate Center (APCC) Long-Term Multi-Model Ensemble Prediction System. For bias correction of downscaled daily precipitation, empirical quantile mapping (EQM) is applied. The downscaled data set without and with correction are called WRF_UC and WRF_C, respectively. In terms of mean precipitation, the EQM effectively reduces the wet biases over most of the United States and improves the spatial correlation coefficient with observation. The daily precipitation of WRF_C shows the better performance in terms of frequency and extreme precipitation intensity compared to WRF_UC. In addition, WRF_C shows a more reasonable performance in predicting drought frequency according to intensity than WRF_UC.

Karyotype Analysis and Physical Mapping of rDNAs in Bupleurum longeradiatum (개시호 (Bupleurum longeradiatum)의 핵형분석과 rDNAs의 Physical Mapping)

  • Karyotype analysis and chromosomal localization of 5S and 45S rDNAs using multi-color fluorescence in situ hybridization (McFISH) technique were carried out in Bupleurum longeradiatum. Somatic metaphase chromosome number was 2n=12. Karyotype was composed of three pairs of metacentrics (No.3, 4 and 6) and three pairs of submetacentrics (No. 1, 2 and 5). The length of somatic prometaphase chromosomes ranges from 2.55 to $5.05{\mu}m$ with total length of $18.15\;{\mu}m$. In FISH experiment, one pair of 5S rDNA signals was detected on the pericentromeric region of chromosome 4 and one pair of 45S rDNA signals was detected on the telomeric region of chromosome 2.

A Study on the Geometrical Features of Soil Doundaries and Mapping Units for Consolidation Works of Arable Land (농경지(農耕地)의 기반조성(基盤造成)을 위한 작도단위(作圖單位) 및 배계(培界)의 기하학적(幾何學的) 형태(形態)에 관한 조사(調査))

  • This investigation was conducted to get basic informations on land consolidation works and soil management for arable land. The characteristics of geometrical features of mapping units in the detailed soil maps of Korea were measured from 70 soil series (565 soil phases) by using a picture analysis system. The results obtained were as follows: 1. The mean values of the mapping units in extent, periphery, diameter of long axis and short axis, roundness and number of acute angles were 22.0ha, 2.267m, 911m, 0.104, and 3.5, respectively. It was suggested that "Simplicity Index of Mapping Unit", $=\frac{Extent\;in\;ara}{Length\;of\;boundary\;periphery\;in\;meter}{\times}\frac{1}{number\;of\;acute\;angle}$ be valuable to judge the complexity degrees of mapping units. 2. The size of mapping unit among physiographyic position was shown in order of Alluvial plains > Fluviomarine plains > Terraces > Hills > Fans > Mt. foot slopes > Valleys, and the simplicity index of mapping units also showed similar order of the extent. 3. The size and the simplicity index of the soils developed on plains with silty textured imperfectly drained were higher than the soils developed on sloped land with loamy textured. As the slopes getting steeper or relatively better in soil drainage, the size and the simplicity index became smaller. 4. The relationship between the simplicity index and the size of the farm unit divides by the land consolidation works was positively correlated. And it was concluded that the parcelling of the farm unit divides by the planning of land consolidation should be based on the soil boundaries of the detailed soil maps for mechanized or collaborated farming.
