Effects of Dietary Creatine on Tissue Metabolite Contents and Uptakes in Rat Liver (랫드 간장에서 Creatine 섭식에 의한 metabolites의 변화)

  • 흰쥐 간정맥 및 간문맥에 PE10 튜브를 마취하에 이식시킨 후 5일째부터 고형사료와 10% creatine을 함유한 고형사료를 5일 동안 섭식 시켜 급속 냉동 시켰다. 냉동시킨 흰쥐 간장, 뇌 및 근육에서 creatine을 포함한 간장 metabolites을 조사하여 다음과 같은 결과를 얻었다. 또한 간장세포내의 water compartment의 변화를 동시에 실시 하였다. Creatine을 섭식시킨 실험군은 정상 대조군과 비교하여 섭식에 의한 간세포내의 water compartment 에 벼노하를 주지 않았고 간장 세포내의 creatine농도에 있어서는 약 43배 이상 세포내 증가를 유도하였으며, 이는 세포외 및 세포내의 creatine의 gradient에 의해 영향을 받고 있음을 알 수가 있었고, 또한 동시에 뇌, 근육 등에서는 섭식에 의해 유의성 있는 농도 변화는 보이지 않았다. 이는 세포 외액속에 함유된 creatine 이 여러 수송기전에 의하여 간세포내로 흡착이 이루어진 것을 알 수 있었으나, 간세포 분리 시험법에로 Creatine Kinase 효소 활성도를 측정한 결과 non-parenchymal hepatocytes에서만 거의 100% 효소 활성도가 있음을 증명 하였다.

MR Spectoscopic Patterns Early and Late Cerebral Ischemic Infarct: Correlation with Clinical Findings (초기 및 지연기 허혈성 뇌경색의 양자 자기공명분광양상 : 임상소견과의 비교)

  • Purpose : To evaluate the proton MR spectroscopy (MRS) findings of early and late ischemic infarcts and to compare these MRS findings with clinical symptoms. Materials and Methods : We obtained MRs spectra of 28 consecutive patients with early ischemic infarct (15 me, 13 women) between 2-10 (mean 6.2) days after stroke onset. Follow-up MRS was carried out between 20-32 (mean 25) days in 12 patients. The MRs spectra were acquired at 1.5T MR unit using single voxel technique with PRESS sequence, TR of 2000ms, TE of 288 (144)ms, and voxel size of 2cm x 2cm x 2cm in the three areas; an infarct lesion, the brain parenchyma adjacent to the lesion, and contralateral normal brain parenchyma. The NAA/creatine, choline/creatine, and lactate/creatine ratios were calculated in each spectrum. The spectra of MRS were compared with clinical symptoms. Results : In early infarct, decreased NAA/creatine ratio (n=22) and increased lactate/creatine ratio (n=25) were found in the infarct lesion. Choline/creastine ratio was within normal range (n=25). On follow-up MRS in late stage, NAA/creatine ratio in the infarct lesion decreased further (n=5), did not change (n=6), or increased (n=1). Lactate/creatine ratio became less elevated (n=10), or did not changed (n=2). Choline/creatine ratio had a trend for increase. The decreased NAA/creatine and increased lactate/creatine ratios were correlated well with the severity of symptoms, respectively. Conclusion : Decreased NAA/creatine and increased lactate/creatine ratios were common MRS findings characteristic in early ischemic infarct and correlated well with clinical severity. On follow-up MRS in late stage, NAA/creatine ratio decreased further or did not change, and lactate/creatine ratio became less elevated.

The effect of short-term creatine intake on blood lactic acid and muscle fatigue measured by accelerometer-based tremor response to acute resistance exercise

  • [Purpose] The purpose of this study was to investigate the effects of short-term creatine intake on muscle fatigue induced by resistance exercise in healthy adolescent men, i.e., lactic acid concentration and wrist and head tremor measured by an accelerometer. [Methods] Twelve healthy adolescent men who had no experience with creatine intake were included. The subjects were randomly assigned to the creatine group and the placebo group, followed by 5 days of creatine and placebo intake, and 5 times of 5 sets of leg press, leg extension, bench press, and arm curl exercises at 70% repetition maximum (RM). The lactic acid concentration before and after exercising, rate of perceived exertion (RPE), and accelerometer-based wrist tremor and head tremor during exercise were measured. Subsequently, after 7 days to allow for creatine washout, the same exercise treatment and measurement were performed in each group after switching drug and placebo between the groups. [Results] The level of lactic acid before and after the acute resistance exercise trial was significantly lower in the creatine group than in the placebo group (P <0.05). The mean RPE during the resistance exercise was significantly lower in the creatine group than in the placebo group (P <0.05). There was no difference between the two groups in the mean wrist tremor during resistance exercise, but the mean head tremor values were significantly lower in the creatine group than in the placebo group in the arm curl, the last event of the exercise trials (P <0.05). [Conclusion] Short-term creatine intake reduces the blood fatigue factor increased by resistance exercise, and is thought to suppress fatigue, especially in the latter half of resistance exercise. Therefore, these findings indicate that short-term creatine intake can have an improved effect on anaerobic exercise performance.

  • N-Methylamidinoglycine, an isomer of creatine which was postulated to be formed enzymatically in vitro, has been synthesized by coupling glycine with N, S-dimethylthiopseudouronium iodide in a yield of approxirnately 60%. On heating in acidic solution, it was converted to a cyclized form (isocreatinine) in analogy with the conversion of creatine to creatinine(anhydrous form). Structures were confirmed by an elemental analysis and proton NMR spectroscopy. Further studies on their characteristics were compared with those of creatine and creatinine in regard to isoelectric points(pI), retardation coefficients(Rf) on thin layer chromatography, and elution profiles on amino acid analyzer. In order to facilitate the comparison, $^{14}C$-labeled creatine, creatinine, isocreatine and isocreatinine were also synthesized.

Effects of Combined Exercise and Creatine Intake to Muscular Function and Pain in Back Pain Patients (복합운동과 크레아틴 복용이 요통환자의 근기능과 통증에 미치는 영향)

  • The purpose of this study was to examine the effects of combined exercise and creatine consumption to muscular function and pain in 24 middle-aged men in back pain patients. The patients were categorized into three groups; 8 patients underwent combined exercise alone, 8 patients underwent creatine consumption as well as combined exercise and the control group was 8 patients without these options. The follow-up period was 10 weeks doing aerobic exercise, resistance exercise or creatine consumption. The group that underwent creatine intake as well as combined exercise simultaneously showed significant increase on the 0°, 12°, 24°, 36°, 48°, 60° and 72° of back muscular power in the muscular function changes compared to control group, furthermore, it showed statistically significant improvement on the 24°, 36°, 48° and 60° in the combined exercise group compared to control group. However, between the group that underwent creatine consumption as well as combined exercise and the group that underwent combined exercise, the back muscular power was high in the all angles, but it was not statistically significant. The group that performed both creatine consumption and combined exercise showed pain relief relatively, comparing with either combined exercise or control group and the pain was also alleviated in the combined exercise group.

  • Two short-term bioassays were employed to asses the mutagenic and clastogenic activities in browning reaction of pentose-creatine, pentose-glycine and pentose-creatine-glycine browning reaction model system. Methylene chloride extract of rhamnose-creatine-glycine browning reaction exhibited the strongest mutagenicity toward Salmonella typhimurium TA98 with S-9. Methylene chloride extract of pentose-creatine and pentose-glycine browning reaction solutions was also tested for mutagenicity, with positive responses. Methylene chloride extract of pentose-creatine-glycine browning reaction solutions induced significant increase in chromosome aberrations in the treated Chinese hamster ovary(CHO) cells. Each of pentose-creatine and pentose-glycine browning reaction solutions induced a relatively low frequency of chromosome aberrations in the treated cells.

  • Cerebral creatine deficiency syndrome (CCDS) is a disorder where a defect is present in transport of creatine in the brain. Creatine plays an essential role in the energy metabolism of the brain. This is a genetic disorder, autosomal recessive or X linked, characterized by intellectual disability, speech and language delay, epilepsy, hypotonia, etc. Until recently very few number of cases have been reported. Here we report a case of 1.5-year-old boy who had epilepsy (epileptic spasm and generalized tonic clonic seizure), intellectual disability, microcephaly, hypotonia and speech delay. His magnetic resonance imaging of brain showed cortical atrophy and electroencephalography showed burst suppression pattern. The diagnosis was confirmed by clinical exome sequencing which showed novel mutation of SLC6A8+ in exon 9, suggestive of X linked recessive CCDS. The patient was then treated with glycine, L-arginine and creatine monohydrate with multiple antiepileptic drugs.

  • The purpose of this study is to investigate how effects creatine dosage has on the improvement of rowing athletes' performance ability. Rowing athletes were administered with creatine, through which to examine the change of athletic performance ability, blood fatigue substances, and muscular activity. The subjects (participants) of this Study consisted of 12 male rowing athletes at P University, with at least 5 years of rowing experiences, which divided into two groups - creatine dosing group of 6 persons and control group of 6 persons - for random sampling measurement. Enzymatic-colorimetric method using lacrate oxidase and 4-aminoantipyrine was performed for blood lactate level analysis, and wireless EMG system (QEMG-4: Lxtha Korea) for muscular activity analysis, with 4 channels set for data analysis. As body parts to be measured, two muscular parts - latissimus dorsi and lumbar spinel - were chosen. Then, on the 5th day from the date of administering them with creatine (that is, 4 days after dosing them with creatine), rowing movement with the highest level of activity was calculated as peak value, which was measured twice. The test data used for this Study were SPSS/PC 18.0, pre-movement and post-movement two-way ANOVA for repeated measurement for comparative analysis of each muscle, with significant level at .05. As a result, the change of blood lactate level was significantly higher in creatine dosing group than in non-dosing group (p<.05). As for the change of muscular activity, both latissimus dorsi and lumbar spinel showed a significantly higher change of muscle in creatine dosing group than in non-dosing group (p<.05 and p<.05, respectively).

  • The main purpose of the present study was to find the effect of short term creatine loading on repeated bouts of kicking, plasma creatine phosphokinase and anaerobic power in male taekwondo player from high school. Eighteen male were randomly assigned to two groups. They were tested before and after 6 days of placebo($4{\times}5$ glucose d-1, N=9) or Creatine monohydrate loading(consume 0.3g kg-1, N=9). Only creatine loading group were significantly increased of repeated bouts of kicking, plasma creatine phosphokinase and anaerobic power These results suggest that short creatine loading was effective diet protocol in taekwondo player from male high school.

  • The planar-type amperometric creatinine biosensor employing an additional enzyme layer containing creatine kinase and adenosine triphosphate was developed to eliminate severe interference from creatine. In the additional enzyme layer, an interfering substance, creatine is converted to noninterfering product, phosphocreatine. Furthermore, the carbon electrode electroplated with Pt black(Pt-B) was employed to fabricate creatinine biosensors with improved sensor performance(e.g., sensitivity, reliability, and reproducibility). The creatinine levels in an unknown sample were determined within less than 5% errors using creatinine microsensors equipped in a flow-cell cartridge.
