The rate of utilization of the oak cork (bark of Quercus variabilis Blume) ranges only 40-50%. Therefore, this study was carried out to investigate the chemical properties of the oak cork for increasing the utility. 1) The contents of alkali extractives, organic solvent extractives and ash in the oak cork are similar to those of other barks, but hot and cold water extractives and lignin contents are lower than those of the wood. The carbohydrate (cellulose and hemicellulose) content of the oak cork is similar to that of other barks. The suberin contents in the first and the second bark of the oak cork are 34.8 and 32.2% respectively, in the dry weight. 2) Inorganic component contents of the first bark are similar to those of the second. The pH of the first and the second bark are 3.9 and 4.2%. The caloric values of the first and second bark are 6,263 Kcal/kg, and 5,828 Kcal/kg, respectively, and these caloric values are higher than those of other barks. The sclerencymatous cell content of the first bark which is related to the quality of the oak cork is lower than that of second bark, the contents of the sclerenchymatous cell and lignin show the positive correlation. 3) In the dimension of the cross sectioned cork cell, the first bark is bigger than that of the second. The shape of the cork cell is globular shape in the early bark and discoid shape in the late bark. The cross and the radial section are the same shape, but the tangential section shows difference from the other section.
This study examines the differences in structures, physical and mechanical properties between domestic(Quercus variabilis Blume) and foreign(Quercus suber L.) corks. The results obtained are as follows: 1. The cork tissue consists of cork cells, lenticels, sclereids and dark-brown zone. There was a significant difference in ratio of cork cells between foreign cork(93 %) and domestic cork(87 %). The ring width and width of late cork of the foreign cork were wider than those of domestic cork. But the percentage of late cork of domestic cork was richer than that of foreign cork. The size of cork cell of foreign cork was larger by about two times than that of domestic cork. 2. The density was slightly higher in domestic cork(0.22 g/$cm^3$) than in foreign cork(0.19 g/$cm^3$). During first 24 hours, the amount of water absorption of the foreign cork was greater than that of domestic cork. However, after 24 hours, the tendency was reversed. The level of EMC was higher in domestic cork than in foreign cork. Total shrinkage in the radial and tangential directions was larger in domestic cork than tn foreign cork. In the longitudinal direction, the tendency was reversed. There was no difference in total swelling in three woody directions between the two corks. 3. The modulus of elasticity in compression in the logitudinal and tangential direction was higher in domestic cork than in foreign cork, but the tendency was reversed in the radial direction. Both corks showed 95% of recovery rate after 24 hours when they were compressed by 0.5. There were no differences in tension strength and Brinell,s hardness bet ween domestic and foreign corks.
Various culture conditions for cell growth and berberine production in cork tree (Phellodendron amurense Rupr.) were investigated. Callus was induced from cambium tissue of cork tree, and cultured on LS liquid medium supplemented with 0.5 mg/1 2,4-D, 0.1mg/1 BA, and 3% sucrose. Several factors enhancing berberine production and cell growth in cork tree cell cultures were found. Some of them enhanced both cell growth and berberine production, but others resulted in a decoupling of cell growth and berberine production with significant in the specific levels. High level of nitrate (80mM), high level of phosphate (8.98mM), and sucrose (7%), 1.0mg/l IAA were effective in berberine production, whereas low level of nitrate (40mM), and phosphate (2.25mM), and high level of sucrose (7%) in the medium were effective in cell growth. Two stage culture(first stage for cell growth, and second stage for berberine production) increased berberine production almost twice (5.06mg/g dry weight) as much as single stage cultures in berberine production.
In this study, we investigated on antioxidative activity, antibacterial activity and tyrosinase inhibitory activity in 75% ethanol extracts of bark and cork layer from Prunus sargentii. The total polyphenol content of cork and bark extract was found to be 217.4 mg/g and 184.3 mg/g. And flavonoid content was found to be 31.9 mg/g and 58.6 mg/g respectively. In DPPH radical scavenging ability, cork extract was exhibited stronger scavenging ability than bark extract. Moreover, cork extract was showed higher inhibitory activity than bark extract in tyrosinase inhibitory activity. In antibacterial activity, bark extract was showed higher growth inhibition effect than cork extract against tested bacterial strains. In measurement of cytotoxicity by MTT assay, bark and cork extract showed fine cell viabilities(95.7~120.9%) against RAW 264.7 cell.
Filter media are one of themost important choices when bilfiltration is applied. Filter media provide adsorbing microbes with available sites for residence and the residential cells grow to degrade or decompose target waste gases. Up to date, filter media were only qualitatively analyzed. As a quantitative approach to filter material for biofiltration, a simple model based on reaction kinetics and mass action law (Ottengraf's Model in 1983) was presented. Cork, zeolite, and granulated activated carbon were tested in terms of effective surface area, cell covered fraction of adsorbing sites, surface roughness, and pore size distribution. The cell covered fraction, surface roughness and hydrophilicity was found to be closely related to the efficiency of gas degradation in biofiltration. The cork was the best candidate for cell residence and growth in this work.
Abortive infection systems (Abi) are phage resistance systems that can be prophage-encoded. Here, two genes encoding an Abi system were identified on a prophage sequence contained by the chromosome of the Levilactobacillus brevis strain UCCLBBS124. This Abi system is similar to the two-component AbiL system encoded by Lactococcus lactis biovar. diacetylactis LD10-1. The UCCLBBS124 prophage-derived Abi system (designated here as AbiL124) was shown to exhibit specific activity against phages infecting L. brevis and L. lactis strains. Expression of the AbiL124 system was shown to cause reduction in the efficiency of plaquing and cell lysis delay for phages of both species.
본 연구는 '만풍배'의 생육단계 및 괘대 유무에 따른 과실의 해부학적 차이를 알아보고, 수확기 과실의 품질 및 착색특성 관찰을 통하여 '만풍배'의 생리적 특징을 이해하기 위하여 수행되었다. 만개기의 과실구조는 바깥쪽으로부터 외표피층, 아표피층, 유조직층, 내표피층으로 구성되어 있으며 아표피층을 이루는 세포 층수는 만개기부터 만개 후 77일까지 9-13층까지 최대로 증가하여 '만풍배'의 세포 분열 기간은 만개 후 77일까지로 조사되었다. 이 기간 동안 아표피층에서 유관속조직까지의 두께는 $73.1{\mu}m$부터 $195{\mu}m$까지 증가하였다. 석세포는 만개 후 7일부터 관찰되어 만개 후 49일에는 석세포군이 형성되어 있었다. 코르크층은 만개 후 49-77일 사이에 형성되었다. 성숙기인 만개 후 161일 '만풍배' 과실의 과피는 바깥쪽부터 코르크층은 4.5층, 아표피층은 7개의 세포층으로 구성되어 있었고 아표피층에는 탄닌이 분포해 있었다. '만풍배'의 괘대 유무에 따른 과실비대 및 조직발달을 비교한 결과, 괘대 초기에는 코르크층이 유대재배구에서 더 빨리 생겨나는 것으로 관찰되었으나 성숙기에는 3층 정도 무대재배구에서 더 두꺼워지는 것을 관찰할 수 있었다. 석세포군의 크기는 두 실험구 모두 불균일하게 분포되어 있었으며 과중은 무대재배구에서 더 높았으나 수확기의 당도는 괘대 유무에 따른 차이를 보이지 않았다. '만풍배'의 수확기 과중 등급은 301g부터 900g 이상까지 분포하였고, 그 중 551-800g의 과실이 총 수확량의 71.6%로 가장 많았으며 이 범위의 평균 과중은 677.2g이었다. 과실품질은 가용성 고형물 함량 $10.1-12.1^{\circ}Brix$, 산도 0.10-1.24%, 당산비 9.76-14.31로 분포하였으며 과중과 가용성 고형물 함량은 매우 높은 정의 상관관계를 나타내어 과실 크기가 클수록 가용성 고형물 함량도 높았다. 반면에 경도는 과중 및 가용성 고형물 함량과 높은 부의 상관관계를 나타내어 과실크기가 클수록 낮은 경향을 나타냈다. 과피의 황갈색 부분과 녹색 부분을 구분하여 코르크층 수와 함께 엽록소 함량을 조사한 결과, 코르크층은 황갈색 부분은 3.8층, 녹색부분은 3.5층으로 거의 차이를 보이지 않았으나 엽록소 함량은 녹색부분이 많은 것으로 조사되었다.
In this study, we investigated the antibacterial activity, antioxidative activity and whitening effect of 75% ethanol extracts from different parts of Prunus sargentii. The total phenolic compound content of the branch extract was 277.92 mg/g as the highest level. In the measurement of DPPH radical scavenging ability, $SC_{50}$ values of the cork layer and branch extract were 26.79 ${\mu}g/m{\ell}$ and 30.13 ${\mu}g/m{\ell}$. In nitric oxide (NO) scavenging ability, $SC_{50}$ values of the branch and leaf extract were 49.19 ${\mu}g/m{\ell}$ and 55.55 ${\mu}g/m{\ell}$. All extracts exhibited higher NO scavenging ability than ascorbic acid used as positive control. On the other hand, in antibacterial activity against Staphylococcus epidermidis and Staphylococcus aureus by disc diffusion assay, the pure bark extract showed the highest activity. Moreover, tyrosinase inhibitory activity of cork layer, pure bark and branch extracts showed higher activity than arbutin used as positive control. In the cytotoxicity measurement by MTT assay, leaf extract was exhibited Raw 264.7 cell viabilities of 44.68~61.83% as cytotoxic result in tested concentration. In conclusion, the branch extract of Prunus sargentii will be a functional materials without damage compared to other parts such as pure bark or cork layer in the plant.

Seventeen compounds (1 - 17), ${\beta}$-sitosterone (1), lupenone (2), arborinone (3), ${\beta}$-sitosterol (4), lupeol (5), epi-lupeol (6), taraxerol (7), betulinic acid (8), 24R-methyllophenol (9), germanicol (10), hexatriacontane (11), nonacosan-1-ol (12), benzoic acid (13), tetradecyl(E)-ferulate (14), di(2-ethylhexyl) phthalate (15), trilinolein (16) and monopalmitin (17), were isolated from the methylene chloride-soluble fraction of the cork of Euonymus alatus Sieb. The structures of these compounds were elucidated on the basis of spectroscopic evidence. Compounds 6, 11, 13 and 14 were isolated for the first time from this plant. Compound 4 showed moderate cytotoxic activity with an $IC_{50}$ value of 6.22 ${\mu}M$ in HL-60 cell line. Compound 9 exhibited moderate cytotoxic activity with $IC_{50}$ values of 63.31, 15.45, 15.14 and 21.72 ${\mu}M$ in four kinds of human cancer cell lines, Jurkat T, HeLa, HL-60 and MCF-7, respectively. Compound 17 showed moderate cytotoxic activity with an $IC_{50}$ value of 70.71 ${\mu}M$ in Jurkat T cell line. In addition, compounds 2, 3, 14 and 16 exhibited weak antioxidant activity with $IC_{50}$ values of 151.76, 170.79, 137.46 and 139.37 ${\mu}M$, respectively.
