Coordination chemistry of mitochondrial copper metalloenzymes: exploring implications for copper dyshomeostasis in cell death

  • Mitochondria, fundamental cellular organelles that govern energy metabolism, hold a pivotal role in cellular vitality. While consuming dioxygen to produce adenosine triphosphate (ATP), the electron transfer process within mitochondria can engender the formation of reactive oxygen species that exert dual roles in endothelial homeostatic signaling and oxidative stress. In the context of the intricate electron transfer process, several metal ions that include copper, iron, zinc, and manganese serve as crucial cofactors in mitochondrial metalloenzymes to mediate the synthesis of ATP and antioxidant defense. In this mini review, we provide a comprehensive understanding of the coordination chemistry of mitochondrial cuproenzymes. In detail, cytochrome c oxidase (CcO) reduces dioxygen to water coupled with proton pumping to generate an electrochemical gradient, while superoxide dismutase 1 (SOD1) functions in detoxifying superoxide into hydrogen peroxide. With an emphasis on the catalytic reactions of the copper metalloenzymes and insights into their ligand environment, we also outline the metalation process of these enzymes throughout the copper trafficking system. The impairment of copper homeostasis can trigger mitochondrial dysfunction, and potentially lead to the development of copper-related disorders. We describe the current knowledge regarding copper-mediated toxicity mechanisms, thereby shedding light on prospective therapeutic strategies for pathologies intertwined with copper dyshomeostasis.

Effects of Seed Soaking of Kinetin on Alleviating Copper Toxicity during Germination in Rice (Kinetin 침종처리가 벼 발아중 구리 독성 경감에 미치는 영향)

  • The study was carried out to determine the proper concentration of plant growth regulator, kinetin on alleviating copper toxicity for two rice cultivars of seed germination. The results were as followings : Soaking treatment of kinetin 10$^{-3}$ M increased the germination rate of both cultivars, Ilpumbyeo and Hyangmibyeo 1 by 92% and 88% as compared with copper treatment (60ppm). But the soaking treatment effect of plant growth regulator, kinetin was not recognized at the kinetin 10$^{-4}$ M and 10$^{-5}$ M. Chlorophyll content of both rices was higher than that of Hyangmibyeo 1. Copper content of Ilpumbyeo was higher in leaf than in seed part. At the 3 days after treatment of copper 60ppm, both cultivars of treatment of kinetin 1O-3M showed the somewhat thin bands at the 35 and 40kDa compared with others. A new protein band pattern was only appeared to kinetin 1O-3M at approximately 54.4kDa(M. W) at the 7 days after treatment of copper 60ppm in llpumbyeo cultivar, SOD activity of copper 60ppm treatment increased in 3DAT, but there were no significant differences in 5 and 7DAT of two cultivars. Free proline contents of copper 60ppm treatment in llpumbyeo were remarkably increased about 4.996$\mu$M. In particular, free proline content of kinetin l0$^{-3}$ M in Ilpumbyeo was 5.008$\mu$M in 3DAT. In case of Hwangmibyeo 1, free proline content of copper 60ppm was 5.825$\mu$M compared with an untreated control showing 2.34l$\mu$M. The effects of kinetin treatment were recognized to promote the root growth and germination rate under copper toxicity(60ppm) condition in both cultivars.

Role of Proline Accumulation in Response to Toxic Copper in Microcystis aeruginosa

  • The blue green alga, Microcystis aeruginosa, was found to accumulate proline under the stressful concentration of cupric ions. The changes of proline level in Microcystis aeruginosa in response to copper(Cu) have been monitored and the function of the accumulated proline was studied with respect to its effect on Cu uptake. Exposure of Microcystis aeruginosa elevated concentrations of Cu led to accumulation of fee proline depending on the concentrations of the metal in the external medium. The greater the toxicity or accumulation of the metal, the higher the amount of proline in algal cells were found. When proline was exogenously supplied prior to Cu treatment, the absorption of Cu was markedly reduced. When exogenous proline was supplied after Cu treatment, it resulted in a remarkable desorption of the adsorbed Cu immediately after the addition of proline. Pretreatment of Microcystis aeruginosa with proline counteracted with metal-induced lipid peroxidation. The results of the present study showed a protective elect of proline on metal toxicity through inhibition of lipid peroxidation and suggested that the accumulation of proline may be related to the tolerance mechanism for dealing with Cu stress.

Reaction Route to the Crystallization of Copper Oxides

  • Copper is an important component from coin metal to electronic wire, integrated circuit, and to lithium battery. Copper oxides, mainly including $Cu_2O$ and CuO, are important semiconductors for the wide applications in solar cell, catalysis, lithium-ion battery, and sensor. Due to their low cost, low toxicity, and easy synthesis, copper oxides have received much research interest in recent year. Herein, we review the crystallization of copper oxides by designing various chemical reaction routes, for example, the synthesis of $Cu_2O$ by reduction route, the oxidation of copper to $Cu_2O$ or CuO, the chemical transformation of $Cu_2O$ to CuO, the chemical precipitation of CuO. In the designed reaction system, ligands, pH, inorganic ions, temperature were used to control both chemical reactions and the crystallization processes, which finally determined the phases, morphologies and sizes of copper oxides. Furthermore, copper oxides with different structures as electrode materials for lithium-ion batteries were also reviewed. This review presents a simple route to study the reaction-crystallization-performance relationship of Cu-based materials, which can be extended to other inorganic oxides.

Potential of Marine Ciliate Mesodinium rubrum as a Standard Test Species for Marine Ecotoxicological Study (해양생태독성 평가용 표준시험생물로서 섬모충류 Mesodinium rubrum에 대한 연구)

  • The mixotrophic marine ciliate Mesodinium rubrum possesses a highly modified algal endosymbiont as a nutrition source for the species. Accordingly, we assumed that the species can reflect the ecotoxicity on marine producer (as phytoplankton) and consumer (as zooplankton) both. A series of experiments were conducted to identify the potential of the species as a standard test species for marine ecotoxicological study. The comparison of species sensitivity on reference toxic materials was made using potassium dichromate for phytoplankton and copper chloride for zooplankton. The ciliate revealed the highest sensitivity on both reference materials among the seven test species including phytoplankton, benthic copepod and rotifer species. The toxicity end point of the species was 72hr-$EC_{50}$=1.52 mg/L (as potassium dichromate) estimated by population growth inhibition (PGI), which is more sensitive than the most sensitive phytoplankton Skeletonema costatum (72hr-$EC_{50}$=3.05 mg/L). As comparison to rotifer, it also revealed higher sensitivity on copper chloride; 72hr-$EC_{50}$=0.38 mg/L for ciliate and 48hr-$EC_{50}$=0.48 mg/L for rotifer. Also, the elutriate toxicity test of various ocean disposal wastes were conducted to identify the potential of ciliate toxicity test application using industrial waste sludges. The toxicity of leather processing waste sludge was highest on the ciliate, followed by dyeing waste sludge and dye production waste sludge as an increasing order of toxicity. 72h-$EC_{50}$ of ciliate PGI test was 1.83% and that of S. costatum 3.84% for leather waste sludge which showed highest toxicity. The toxicity test results also revealed that the highest sensitivity was observed on ciliate species on ocean disposed sludge wastes. Also, ciliate toxicity test well discriminated the degree of toxicity between sludge sources; 72h-$EC_{50}$ values were 1.83% for leather processing waste sludge, 16.75% for dye production waste sludge and 27.75% for textile production waste sludge. Even the laboratory culture methods of the species were not generally established yet, the species has high potential as the standard test species for marine toxicity test in terms of the dual reflection of phyto- and zooplankton toxicity from single test, sensitivity and test replicability.

Comparative Toxicity of some Pesticides to the Predatory Mites, Neoseiuius fallacis Carman (Acari: Phytoseiidae) (팔라시스이리응애에 대한 농약의 독성)

  • The toxicities of 37 commercial pesticides for citrus pests were evaluated to H, fallacis. Eight fungicides (difenoconazole, imibenconazole, azoxystrobin, dithianon, fluazinam, procymidon, streptomycin, tribasic copper sulfate), three insecticides (dichlovos, imidacloprid, thiamethoxam) and four acaricides (milbemectin, tetradifon, dicofol, spirodiclofen) had lower effect to the hatchability of eggs N. fallacis. Six fungicides (propineb, difenoconazole, imibenconazole, azoxystrobin, dithianon, procymidon) and three acaricides (bifenazate, tetradifon, spirodiclofen) showed lower contact toxicity to adult N. fallacis. The secondary toxicity of 26 pesticides to N. fallacis adult were evaluated. Two fungicides (fluazinam, streptomycin) and 3 acaricides (machine oil, cyhexatin, halfenprox) showed low toxicity when the prey (eggs of spider mite) was treated with pesticides.

A Study on the Fundamental Characteristics of a Copper Slag Mixed with Granite Soil (동슬래그 혼합토의 기본 성질에 관한 연구)

  • This paper presents fundamental characteristics of a copper slag when used geotechnical materials. For this study, it was conducted laboratory tests such as compaction, large direct shear, hydraulic conductivity, leaching, TDR, frost heave test and so on. The results of laboratory tests shown gradually increase in draining capacity and shearing resistance more slag mixing. The unfrozen water in temperature changes and frost heave amounts in condition of -17 $^{\circ}C$ appeared to decrease. Also, toxicity tests based on the domestic solid waste regulations were satisfied with nonhazardous. By this research results, a copper slag mixed with granite soil may been used as granular base and embankment materials, fill etc.

  • Toxicity of ten commercial disinfectants(hydrogen peroxide, sodium hypochlorite, chlorine dioxide, povidon iodine, formaldehyde, glutaraldehyde, quaternary ammonium compounds(QAC), didecyl dimethyl ammonium chloride(DDAC), ortho-dichlorobenzene, and copper sulfate) was measured by chinook salmon embryo-214 cell line and three fish species: flounder Paralichthys olivaceus, black rockfish Sebastes pachycephalus, and black sea bream Acanthopagrus schlegelii. The $LC_{50}$ levels of 24 hours acute toxicity with the ten disinfectants were tested in three species of fish. Effectiveness of ten chemical disinfectants were varied by each chemical as well as by species. Hydrogen peroxide showed the higest activity at 201, 269, and 139 ppm in the flounder, the black rockfish, and black sea bream, respectively. DDAC showed the lowest activity at 2.1, 1.0, and 1.5 ppm in the flounder, the black rockfish, and black sea bream, respectively. The highest variation was observed in copper sulfate by both the chemicals and the species.

Effects of Copper (II) Treatment in Soil on Tetracycline Toxicity to Growth of Lettuce (Lactuca sativa L.) (토양에서 상추의 생장에 대한 Tetracycline의 독성에 미치는 구리 (II)의 효과)

  • Tetracycline (TC) groups, widely used veterinary antibiotics, can enter into environment through animal manure application. TC forms a ligand complex with multivalent metal cations via chelation that can affect sorption and mobility of TC in soil. So far, however, it has been confirmed through the reaction of the soil outside in the aqueous solution and the evaluation of the performance in the soil cultivation process is insufficient. The purpose of this study was to examine effects of copper on TC toxicity to lettuce growth. In this research, $750mg\;kg^{-1}$ of TC and 2.5, 7.5, $17.5mg\;kg^{-1}$ of Cu are treated in soil and lettuce was cultivated in the treated soil. Growth difference of lettuce by treatment was observed. As a result, $750mg\;kg^{-1}$ of TC treated soil showed toxic effect to lettuce and the effect is alleviated by copper treatment.
