Development of the Optimal Composting Condition for the High Quality of Pig manure compost (고품질의 돈분 퇴비를 위한 합리적인 퇴비화 조건 개발)

  • This study was conducted to induce the optimal composting conditions of pig manure mixed with sawdust and dried paper-mill sludge in the composting for production of high quality compost. Pig manure contains high water content and How C/N ratio because of comparatively high nitrogen content than sawdust and dried paper-mill sludge. Therefore the addition of dried paper-mill sludge and sawdust to the raw materials helps controlling the C/N and the water content of compost pile. The composting system used in the experiment was agitated static bed system. The physical properties of the mixed raw materials was not good at the working conditions in the early stage of composting. The temperature of compost heap reaches at $60^{\circ}C$within 5 day after starting composting in P-2 treatment mixed with pig manure and sawdust(56.6 : 43.4). Then the water content of P-2 was 58%. The pH in all treatments were slowly decreased as the composting was proceeded. Although the changes of T-C and T-N were not extended because of the short composting experiment period. Reduction rates of T-C in treatments were 5-12% without special difference. By considering the efficiency of composting in each of five treatments with pig manure the optimal water contents was about 57% level. Mixing a sawdust as a bulking agent was more positive than dried paper-mill sludge from a viewpoint of compost quality.

Insecticidal Activities of Polymers and Surfactants Against Sweet Potato Whitefly, Bemisia tabaci (Hemiptera: Aleyrodidae) (담배가루이에 대한 폴리머와 전착제의 살충효과)

  • This study was performed to evaluate the polymers and surfactants as the potential control agents of sweet potato whitefly Bemisia tabaci, which is causing problems in ornamental garden and greenhouse. Polymers have an insecticidal activity to knock down and to be lethal to small winged insects by its viscosity. Among five polymers tested at 0.2% concentration, polinol P-24 showed the highest insecticidal activity as 59.4% against B. tabaci adult in cylindrical chamber, and followed by polinol P-20 (insecticidal activity, 57.1%). When treated at 0.1 % or 0.3% concentrations, Polinol P-24 also showed the highest insecticidal activity with 43.3% and 54.5%, respectively. Among eight surfactants tested, insecticidal activity was the highest in 0.0005% NP10 treatment (70.0%), and followed by 0.001% NP7 (67.4%). The synergistic effect between polinol P-24 and eight surfactants was evaluated. After bioassays, the 0.2% polinol P-24 plus 0.005% NP10 was selected as a candidate control agent for controlling of B. tabaci adults. Polinol P-24/NP10 was showed the highest control efficacy against B. tabaci adults applied three times at three day-intervals in square rearing cage. In the greenhouse, the mixture treatment showed good control value over 70% seven days after treatment.

Factors Associated with Poor Glycemic Control among Patients with Type 2 Diabetes Mellitus: The Fifth Korea National Health and Nutrition Examination Survey (2010-2012) (제2형 당뇨병 환자의 혈당 비조절 관련 요인분석: 국민건강영양조사(2010-2012) 자료이용)

  • Background: Glycemic control is an effective way to reduce the cardiovascular complications of diabetes, but more than half of the adults with diabetes in Korea are improperly controlling their glycemic levels. The purpose of this study is to identify the factors associated with poor glycemic control in type 2 diabetes patients. Methods: This study analyzed 1,261 subjects ${\geq}30years$ old diagnosed with type 2 diabetes who participated in the fifth Korean National Health and Nutrition Examination Survey (2010-2012). Poor glycemic control rates were defined as hemoglobin A1c (HbA1c) level ${\geq}7%$. To shed light on the causes of poor glycemic control, socio-demographics, diabetes severity, health status, and health behavior factors were adjusted and logistic regression was done. Results: Of the total 1,261 patients, 53.0% of patients with type 2 diabetes had HbA1c ${\geq}7%$. After running a logistic regression model, the odds ratio of poor glycemic control was higher in high school graduates than elementary school graduates; in people living in Chungcheong and Jeolla/Jeju than those living in Seoul; in the group with diabetes for over 5 years had diabetes less than 5 years; in a group with insulin and oral hypoglycemic agent treatment than non-treatment; in a group with hypertriglyceridemia than without hypertriglyceridemia; and in the group with slept less than 6 hours slept 7-8 hours. Conclusion: We need a comprehensive public health policy to reduce the poor glycemic control rates in type 2 diabetes patients. We should recognize the education levels, duration of diabetes, diabetes treatment, hypertriglyceridemia, and sleep duration were associated with poor glycemic control.

Chemical Pesticides and Plant Essential Oils for Disease Control of Tomato Bacterial Wilt

  • Efficacy of different control methods was evaluated for disease management of tomato bacterial wilt caused by $Ralstonia$ $solanacearum$. All six chemical pesticides applied to the bacterial suspension showed $in$ $vitro$ bactericidal activities against $R.$ $solanacearum$. Minimal inhibitory concentrations (MICs) of copper hydroxide (CH), copper hydroxide-oxadixyl mixture (CH+O), and copper oxychloride-dithianon mixture (CO+D) were all 200 ${\mu}g/ml$; MIC of copper oxychloride-kasugamycin (CO+K) mixture was 100 ${\mu}g/ml$; MICs of both streptomycin- validamycin (S+V) and oxine copper-polyoxine B mixture (OC+PB) were 10 ${\mu}g/ml$. Among these chemical pesticides, treatment of the detached tomato leaves with the 5 pesticides (1 mg/ml), except for OC+PB delayed early wilting symptom development caused by the bacterial inoculation ($10^6$ and $10^7$ cfu/ml). Four pesticides, CH, CH+O, CO+K and S+V, showed disease protection in pot analyses. Six plant essential oils, such as cinnamon oil, citral, clove oil, eugenol, geraniol and limonene, differentially showed their antibacterial activities $in$ $vitro$ against $R.$ $solanacearum$ demonstrated by paper disc assay. Among those, cinnamon oil and clove oil exert the most effective activity for protection from the wilt disease caused by the bacterial infection ($10^6$ cfu/ml). Treatment with cinnamon oil and clove oil also suppressed bacterial disease by a higher inoculum concentration ($10^7$ cfu/ml). Clove oil could be used for prevention of bacterial wilt disease of tomato plants without any phytotoxicity. Thus, we suggest that copper compounds, antibiotics and essential oils have potency as a controlling agent of tomato bacterial wilt.

The Characterizations of Tape Casting for Low Temperature Sintered Microwave Ceramics Composite (저온소성 마이크로파 유전체 세라믹스 복합체의 Tape Casting특성)

  • Sintering behavior of $BaO-Nd_{2}O_3-TiO_2$ with a Pb-based glassceramics frit were investigated in order to understand an effect of glassceramics as a low temperature sintering agent on dielectric ceramics. A green sheet form was fabricated through tape casting method with the glassceramic fut added $BaO-Nd_{2}O_3-TiO_2$. The dispersion properties, rheological properties and final density of dielectric composit slurry as a function of amount and composition of organic additives was examined. The dispersants' addition was effective in controlling dispersion of the ceramics in solution. The addition of excessive dispersant showed adverse effect on dispersion. The prepared slurries, containing ceramic : powders, glass-ceramics and various kinds of organic viechles, exhibited typical shear thinning behavior. The best properties of tape casting appeared powder to solvent ratio 65 : 35 and amount of the binder 6 wt$\%$ and plasticizer 3 wt$\%$. The viscosity of the slurry was 677 cps and green/sintered density in the tape was $3.3 g/cm^3,\;5.56 g/cm^3$ respectively.

  • A series of coffee middlemen has come to be regarded as an exploitative force in the lives of small and poor coffee farmers, which is called 'coyote', controlling production, paying unfair prices for labor and goods, and participating in fraudulent practices to maximize their own profits. However, the reality of gains captured by coffee middlemen in the value chain might be exaggerated and even unfair. Therefore, the main aim of this paper is to reconsider the implication of coffee middlemen for the development of coffee value chain in Vietnam. It also attempts to identify their characteristics by investigating relations among coffee farmers, middlemen and processing/exporting firms. In terms of middlemen's margin in the coffee sub-sector, their margin is quite small when compared to other actors higher in the value chain. Rather, coffee middlemen in Vietnam have played a critical role as an intermediary of change in local economies. More specifically, coffee middlemen in Dak Lak has played a significant role as a market facilitator by stimulating the access of farmers to markets by providing buyers for farmers thanks to changes in institutional environment. Also, they have played a critical role as an agent of sustainable coffee production by encouraging sustainable coffee production in accordance with the demand of processing and exporting firms.

Inhibitory Effects of Allium sacculiferum Max. Methanol Extracts on ROS Production and Lipid Accumulation during Differentiation of 3T3-L1 Cells (참산부추(Allium sacculiferum Max.) 메탄올 추출물의 지방세포 내 ROS 생성 및 지질 축적 억제 효능)

  • Allium sacculiferum Max. (ASM) is a perennial plant of the Liliaceae family and grows over the entire regions of Korea. Obesity is a serious health problem worldwide and has currently become a prevalent chronic disease. Adipocytes produced by preadipocyte differentiation during adipogenesis and adipocytes combined with abnormal accumulation cause obesity. Recently, intracellular reactive oxygen species (ROS) were shown to accelerate lipid accumulation in 3T3-L1 cells. In this study, we investigated the effects of ASM methanol extracts on ROS production and lipid accumulation in 3T3-L1 adipocytes. Our results indicate that the 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity of ASM methanol extracts increased in a dose-dependent manner. ASM methanol extracts suppressed ROS production and lipid accumulation during adipogenesis. In addition, ASM methanol extracts inhibited the mRNA expression of both pro-oxidant enzymes such as glucose-6-phosphate dehydrogenase as well as the transcription factors, including sterol regulatory element-binding proteins 1c, peroxisome proliferator-activated receptor ${\gamma}$, and CCAAT/enhancer-binding protein ${\alpha}$. Our results suggest that ASM methanol extracts inhibit ROS production and lipid accumulation by controlling ROS regulatory genes and adipogenic transcription factors. Thus, ASM has potent natural antioxidant, anti-adipogenic properties and have potential in the development of a potent anti-obesity agent.

Production of Soluble Recombinant Human Granulocyte Colony Stimulating Factor in E. coli by Control of Growth Rate. (대장균에서 증식속도 조절에 의한 수용성 재조합 인간 과립구 콜로니 촉진인자의 생산)

  • Human granulocyte colony-stimulating factor (hG-CSF) is a hematopoiesis agent that principally affects the differentiation of neutrophils in the bone marrow. At present, recombinant hG-CSF is used successfully in the treatment of chemotheraphy-induced neutropenia and its indication has been expanded to bone marrow transplantation and aplastic anemia. In this study, we have constructed rhG-CSF secretion plasmid pYRC1 in which OmpA signal sequence/hG-CSF gene was expressed under the control of the T7 promoter. rhG-CSF produced in E. coli BL21 (pYRC1) grown at $37{\circ}C$ was found in aggregates. However, 15% of the periplasmic protein was soluble rhG-CSF when the E. coli BL21 (pYRC1) was cultured at $25^{\circ}C$ for 7 h in the modified MBL medium containing 10 g/$\ell$ glucose with 10 $\mu$M IPTG induction. The production of soluble rhG-CSF in E. coli BL21 (pYRC1) using fed batch culture was also studied. In the fed batch culture system, the final yield of rhG-CSF produced from E. coli BL21 (pYRC1) was increased from 4.4 mg/$\ell$to 24 mg/$\ell$by controlling the specific growth rate from $0.43 h^{-1}$ to $0.14 h^{-1}$, and optimizing the time of induction.

Determination and preconcentration of Cu(II) using microcrystalline p-Dichlorobenzene loaded with salicylaldoxime (Salicylaldoxime이 내포된 p-Dichlorobenzene 미세결정을 이용한 Cu(II)의 예비농축 및 정량)

  • A technique for the determination of trace Cu(II) in various real samples by FAAS after the column preconcentration onto p-dichlorobenzene-SA adsorbent, which is microcrystalline p-dichlorobenzene loaded with salicylaldoxime (SA) has been developed. Several experimental conditions such as pH of the sample solution, the amount of chelating agent salicylaldoxime, the amount of adsorbent p-dichlorobenzene-SA, and flow rate of sample solution were optimized. The interfering effects of various concomitant ions were investigated. $CN^-$ interfered more seriously than any other ions. However, the interference by $1\;{\mu}g\;mL^{-1}\;CN^-$ could be overcome completely by controlling the concentration of Ni(II) to $20\;{\mu}g\;mL^{-1}$. The linear range, correlation coefficient ($R^2$) and detection limit obtained by this technique were $3.0\sim100\;ng\;mL^{-1}$, 0.9901, and $3.1\;ng\;mL^{-1}$, respectively. For validating this technique, the aqueous samples (wastewater, reservoir water and stream water) and the food samples (orange juice, fresh egg and skim milk) were used. Recovery yields of 93~104% were obtained. These measured mean values were not differents from ICP-MS data at 95% confidence level. The good results were obtained from the experiments using the rice flour certified reference material (CRM) sample. Based on the experimental results, it was found that this technique could be applied to the preconcentration and determination of Cu(II) for various real samples.

TAp73 and ΔNp73 Have Opposing Roles in 5-aza-2'-Deoxycytidine-Induced Apoptosis in Breast Cancer Cells

  • The p73 gene contains an extrinsic P1 promoter and an intrinsic P2 promoter, controlling the transcription of the pro-apoptotic TAp73 isoform and the anti-apoptotic ${\Delta}Np73$ isoform, respectively. The DNA methylation status of both promoters act equally in the epigenetic transcriptional regulation of their relevant isoforms. The aim of this study was to analyze the different effects of these p73 isoforms in 5-aza-2'-deoxycytidine (5-aza-dC)-induced apoptosis in breast cancer cells. We investigated the effects of the DNA demethylation agent, 5-aza-dC, on the T-47D breast cancer cell line, and evaluated the methylation status of the p73 promoters and expression of TAp73 and ${\Delta}Np73$. Furthermore, we assessed the expression of p53 and p73 isoforms in 5-aza-dC-treated T-47D cells and p53 knockout cells. 5-aza-dC induced significant anti-tumor effects in T-47D cells, including inhibition of cell viability, G1 phase arrest and apoptosis. This was associated with p73 promoter demethylation and a concomitant increase in TAp73 mRNA and protein expression. In contrast, the methylation status of promoter P2 was not associated with ${\Delta}Np73$ mRNA or protein levels. Furthermore, demethylation of P2 failed to inhibit the expression of ${\Delta}Np73$ with 5-aza-dC in the p53 knockdown cell model. Our study suggests that demethylation of the P1 and P2 promoters has opposite effects on the expression of p73 isoforms, namely up-regulation of TAp73 and down-regulation of ${\Delta}Np73$. We also demonstrate that p53 likely contributes to 5-aza-dC-induced ${\Delta}Np73$ transcriptional inactivation in breast cancer cells.
