Bioremediation of Diesel-Contaminated Soil by Bacterial Cells Transported by Electrokinetics

  • The electrokinetic technology was applied in bioremediation for the purpose of supplying a Pseudomonas strain capable of degrading diesel to contaminated soil bed, and their biodegradation of diesel was carried out after a desired cell distribution was obtained. Electrokinetic injection of the strain was made possible because the cells acted as negatively charged particles at neutral pH, and thus the cells were transported with a precise directionality through the soil mostly by the mechanism of electrophoresis and in part by electroosmosis. A severe pH change in the soil bed was formed due to the penetration of electrolysis products, which was harmful to the cell viability and cell transport. To achieve a desirable cell transport and distribution, the control of pH in soil bed by a recirculating buffer solution in electrode chambers was essential during the appliation of an electric field. The judicious selections of electrolyte concentration and conductivity were also important for achieving an efficient electrokinetic cell transport since a higher electrolyte concentration favored the maintenance of pH stability in soil bed, but lowered electrophoretic mobility on the other hand. With electrolyte solution of pH 7 phosphate buffer, a 0.05 M concentration showed a better cell transport buffer, a 0.05 M concentration showed a better cell transport than 0.02 M and 0.08 M. The cell under pH 8 were obtained, compared to the cells under pH 7 or pH 9 in a given time period Up to $60\%$ of diesel was degraded in 8 days by the Pseudomonas cell, which were distributed electrokinetically under the conditions of pH 8 ($1,800{\mu}S/cm$, a mixture of phosphate and ammonia buffers) and 40 mA in a soil bed of 15 cm length.

Determination of Cadmium and Zinc Contamination Source in Arable Soil in the Vicinity of a Zinc Smelting Factory

  • Agricultural area in the vicinity of the ${\triangle}{\triangle}$ smelting factory in Kyeongbuk province, the third largest zinc smelting factory in the world, was contaminated by high concentration of heavy metals. However, the heavy metals source was not yet directly traced and thus, resulted to a conflict between the factory and residents within its vicinity. In order to determine the level of heavy metal contamination in the arable lands located at the north eastern part of the factory, soils were sampled systematically. To find out the major reason for the occurrence of this problem, waters and aerosols were sampled with constant intervals to the upward and downward direction from the factory and were analyzed to find out the heavy metal concentrations. Cadmium (Cd) and zinc (Zn) of the heavy metals were highly accumulated more than the Korean warning criteria (Cd 1.5, Zn 300 mg $kg^{-1}$) with mean values 1.7 and 407 mg $kg^{-1}$, respectively, at the surface soils (0-20 cm), and heavy metal concentration significantly decreased with increasing soil depth In addition, the concentration of both metals slightly decreased with increasing distance from the factory to the surface soils. Cadmium and Zn were detected in the upward stream water with low concentration and concentrations increased significantly in the downstream after passing across the factory. Aerosol samples also showed traces of Cd and Zn which could be attributed to the contamination of the water system and the surface soils. Conclusively, Cd and Zn emitted from the ${\triangle}{\triangle}$ smelting factory moved with the aerosol in the atmosphere and thus, contaminated the agricultural areas and the water system within it vicinity.

  • Contaminated liver fluke egg in the environment has led to the high prevalence of human opisthorchiasis associated with cholangiocarcinoma in Southeast Asia. To find the effective lessening methods of Opisthorchis viverrini eggs in the contaminated environment, we investigated the temperature conditions for killing of these trematode eggs in vitro. Numerous O. viverrini eggs were obtained in the proximal part of uteri of adult worms from experimental hamsters. Mature eggs with miracidium were allocated by experimental groups (2 control: positive and negative and 4 treatment: 50, 60, 70, and $80^{\circ}C$) with 0.85% saline, and treated by the experimental plan. Eggs in each experimental groups were observed under the confocal microscope after stain with Propidium Iodide (PI) to evaluate the effect of temperatures. Eggs in 70 and $80^{\circ}C$ groups were all killed after over 10 min heated. Majority of eggs in $60^{\circ}C$ (10, 15, and 30 min heated), 70 and $80^{\circ}C$ (5 min heated) groups were inactivated. However in $50^{\circ}C$ group, below half of eggs were to be killed in all time lapse (10, 15 and 30 min). In order to prevent O. viverrini infection and cholangiocarcinoma, direct treatment of sewage by heating at 70 or $80^{\circ}C$ at least 10 min is essential. Therefore, treatment of O. viverrini eggs at a high temperature is a potential method for controlling egg contamination in sewage.

  • Recently, deep learning technology that processes images has been widely used in fire detection, autonomous driving, and defective product detection. In particular, in order to determine whether a product is contaminated or not, it can be identified through the contaminants passed from the existing sensor data, but technologies for recognizing cracks in products or contaminants themselves as images are being actively studied in various fields. In this paper, a system for classifying uncontaminated normal cups and contaminated cups through images was designed and implemented. The image was analyzed using an open image and a photographed image, and the image was analyzed by extracting the upper part of the cup image using Google Objectron for 3D object recognition. Through this study, it is thought that it will be used in various ways for research that can extract the contamination level of products required in the hygiene field based on images.

  • In this paper, we implemented a real-time system improving the performance of the paper currency discrimination by integrating a weighted region of interest matching algorithm with a weighted shape feature matching algorithm of the blocked image. The system classifies the paper currency by comparing a query image with compared images based on the database that contain images of paper currency. Especially, the system has good efficiency at the contaminated, rotated, and translated paper currency. The system hardware consists of three parts as follows : the paper currency image acquired by CIS(contact image sensor) is applied to the pre-processing part with A/D converter and PLD. Finally the pre-processed image data are classified by the main image processing part with a high-speed DSP based on the proposed algorithm.

Change of Heavy Metals and Sediment Facies in Surface Sediments of the Shihwa Lake (시화호 표층퇴적물의 중금속 및 퇴적상 변화)

  • In order to determine the changes of sediment facies and metal levels in surface sediments after the construction of Shiwha Lake, surface sediments were sampled at 8 sites located on the main channel monthly from June, 1995 to August, 1996 and analysed for 12 metals (Al, Fe, Mn, V, Cr, Co, Ni, Cu, Zn, Cd, As and Pb) by ICP/AES and ICP/MS. Two groups of sampling sites(the inner lake with 3 sites and the outer lake with 5 sites) are subdivided by the surface morphology ; the inner lake is a shallow channel area with a gentle slope, while the outer lake is relatively deep and wide channel with a steep slope which has many small distributaries. After the construction of dam, fine terrestrial materials were deposited near the outer lake, which resulted in the change of major sediment facies from sandy silt to mud. With the deposition of fine sediments in the outer lake, anoxic water column induced the formation of sulfide compounds with Cu, Cd, Zn and part of Pb. Metal (Cr, Ni, Cu, Zn and Cd) contents in sediments increased up to twice within 2 years after the construction of dam. This is due to the direct input of industrial and municipal wastes into the lake and the accumulation of metals within the lake. In addition, frequent resuspension of contaminated sediments in a shallow part of the lake may make metal-enriched materials transport near the outer lake with fine terrestrial materials. As the enrichment of Cu, Zn, Cd and part of Pb in the Shiwha Lake may be related to the formation of unstable sulfide compounds by sulfate reduction in anoxic water or sediment column, the effect of mixing with open coastal seawater is discussed.

Identification of a Proper Phytoavailable Arsenic Extraction Method Associated with Arsenic Concentration in Edible Part of three Crops in Soils Near Abandoned Mining Areas

  • This study aimed to investigate correlations between concentrations of extractable Arsenic (As) with varying chemical solutions (0.1 M $Ca(NO_3)_2$, 0.1 M $(NH_4)2HPO_4$, 0.5 M EDTA, Mehlich 3, and 0.5 M $NaHCO_3$) and those of As in crops, and then to seek the most suitable soil extraction method for predicting the potential of As uptake in crops cultivated in soils contaminated with As. For a mesocosm experiment, pepper (Capsicum annuum L.), soybean (Glycine max L.), and rice (Oryza sativa L.) were cultivated for three months in pots containing soils taken from the arable areas near abandoned mines in Korea. Following the cultivation, soil pH and DOC significantly increased by treatments of lime and lime plus compost, respectively, while insignificant influences in changing total and all extractable As concentrations were found in all soils. Arsenic concentration in edible part of all crops considerably depended on the extractable As concentration in the soils, particularly with Mehlich 3. All extractable As concentrations in the soils of C. annuum and G. max were significantly correlated with As concentration in their edible parts. For O. sativa, the extractable concentrations of Mehlich 3 ($R^2$: 0.18 at p: 0.006) and EDTA ($R^2$: 0.11 at p: 0.036) showed only marked relationships with As concentration in the edible part. These results may indicate that the Mehlich 3 and EDTA are soil extractants to determine phytoavailable As in soil that provide better prediction for As transfer from soil to crop.

Microbial Community Structures Related to Arsenic Concentrations in Groundwater Occurring in Haman Area, South Korea (함안지역 지하수의 비소(As) 함량과 미생물 군집 특성과의 연관성 검토)

  • This study evaluated the characteristics of arsenic production in groundwater through microbial community analysis of groundwater contaminated with high arsenic in Haman area. Groundwater in Haman area is contaminated with arsenic in the range of 0-757.2 ㎍/L, which represents the highest arsenic contamination concentration reported in Korea as natural groundwater pollution source. Of the total 200 samples, 29 samples (14.5%) showed higher arsenic concentration than that of 10 ㎍/L, which is the standard for drinking water quality, and 8 samples (4%) found in wells with 80-100 m depth were above 50 ㎍/L. In addition, seven wells with arsenic concentration more than 100 ㎍/L located in the northern part of Haman. As a result of microbial community analysis for high arsenic-contaminated groundwater, the microbial community compositions were significantly different between each sample, and Proteobacteria was the most dominant phyla with an average of 61.5%. At the genus level, the Gallinonella genus was predominant with about 12.8% proportion, followed by the Acinetobacter and Methermicoccus genus with about 7.8 and 7.3%, respectively. It is expected that high arsenic groundwater in the study area was caused by a complex reaction of geochemical characteristics and biogeochemical processes. Therefore, it is expected that the constructed information on geochemical characteristics and microbial communities through this study could be used to identify the origin of high arsenic groundwater and the development of its controlling technology.

Derivation of On-site Major Exposure Factor using NDD Analysis when Landfilling NORM Waste (NORM 폐기물 매립 시 NDD 분석을 활용한 부지 내 주요 피폭인자 도출)

  • As part of the social response to the radon bed incident in 2018, the Nuclear Safety and Security Commission took measures to collect and dispose of all radon beds. The Waste Management Act provides landfill disposal as one of the disposal methods for natural radioactive product waste, which is one of the NORM wastes. When NORM wastes are landfilled, workers and the public at the landfill site are exposed to radiation through various pathways, such as leaching of radionuclides through soil and groundwater, and multiple exposure factors are involved simultaneously. In order to improve the reliability of radiological impact assessment, the values of main exposure factors should be selected more accurately. Therefore, before developing the main exposure factors for site characteristics, it is necessary to prioritize main exposure factors reflecting domestic characteristics of NORM waste landfills. Therefore, in this study, the main exposure factors for NORM waste landfill were derived using NDD analysis. To derive the main exposure factors, the analysis tool was first selected as RESRAD-ONSITE computer code, and the exposure scenarios were mainly selected as a resident farmer and suburban resident scenario, recreation scenario, and industrial worker scenario. Then, the priority 1 and 2 factors were selected for sensitivity analysis, and a Korean standard model was established to reflect Korean characteristics. Finally, the sensitivity analysis was conducted through NDD, and the main exposure factors were derived based on this. In the resident farmer scenario, the contaminated zone distribution coefficients of 226Ra, 210Pb, 232Th, 228Ra, 234U, and 238U, as well as precipitation and evapotranspiration factors, were derived as the main exposure factors. In the suburban resident scenario, the contaminated zone distribution coefficients of 226Ra, 210Pb, 232Th, 228Ra, 234U, and 238U, as well as precipitation and evapotranspiration coefficients, were derived as the main exposure factors. In the recreation scenario, the contaminated zone distribution coefficient of 232Th was derived as the main exposure factor. For the industrial worker scenario, the erosion rate was derived as the main exposure factor. The main exposure factors for each scenario were analyzed to be different depending on the scenario characteristics. The results of this study can be utilized as a basis for radiological environmental impact assessment of NORM waste landfill in Korea.

Studies on the Effects of Atmospheric Pollution in Sericulture -Injuries of Sulphur Dioxide and Cadmium on Parent Silkworms Rearing- (대기공해가 양잠에 미치는 영향에 관한 연구 -원잠종 사육에 있어서 아황산 가스 및 카드미움의 해를 중심으로-)

  • Those studies were examined rearing two varieties on Japanese descent and two varieties of Chinese descent by feeding polluted mulberry leaves, non-polluted ones and water-cleaned ones respectively to find effects of air-pollution on the economic characters of silkworms and analysis of contents of Sulphur and Cadmium in the mulberry leaves and silkworms as followings; 1) Japanese descent of polluted part was delayed about 2.5 days than non-polluted part, Chinese descent was delayed about 4 days or inequal and water-cleaned part was medium in the silkworm larval duration. 2) Results of maximum weight of 5th instal, cocoon layer weight and cocoon weight were decreased in due order non-polluted, water-cleaned and polluted in the factors of mulberry 3) Pupal ratio of Japanese descent was not shown statistical significance, but Chinese descent was revealed it obviously in the factors of mulberry leaves. 4) In the resistance of polluted mulberry leaves, Chinese descent was feeble obviously than Japanese and there were some difference even through among the same varieties. 5) The content of S and Cd of polluted area mulberry leaves was increased about 30% respectively than non-polluted area. 6) The fed part of non-polluted mulberry leaves was S 0.41% and Cd 0.013 ppm water-cleaned part was S 0.47% and Cd 0.024 ppm and polluted part was S 0.52% and Cd 0.042 ppm in the contents of S and Cd of silkworm larvae. 7) The contents of S and Cd didn't make visible injury in mulberry leaves but made it seriously in silkworm larvae. 8) The injury of dust on mulberry leaves was more serious than that of quality of mulberry leaves by air-pollution in the economic characters of silkworm. 9) As above results, Chinese descent should avoid contaminated area and it rearing by water-cleaned mulberry leaves can get noticable results in inevitable case on selection of parents silkworm rearingzone.
