Effect of Bopaewon-tang on Allergic Reaction (보폐원탕(補肺元湯)이 알러지반응에 미치는 영향)

  • The purpose of this research was to investigate the effects of Bopaewon-tang (BT) on allergic reaction. In the present study, we examined the effect of BT on type 1 and type tV allergic reaction. BT (500 mg/kg) did not affect the systemic anaphylaxis induced by compound 48180 and the passive cutaneous anaphylaxis induced by anti-dinitrophenyl (DNP)-IgE and DNP-human serum albumin in vivo. Also, BT did not affect the release of histamine from peritoneal mast cells in rats. In addition, BT did not affect the permeability of evans blue into peritoneal cavity, but inhibited the writhing syndrome induced by acetic acid. BT inhibited the delayed type hypersensitivity induced by SRBC and the contact dermatitis induced by dinitrofluorobenzene. These results indicate that BT may be useful for the prevention and treatment of type IV allergy related disease.

Effects of Crataegus Pinnatifida (CP) on Allergic Contact Dermatitis (ACD) Induced by DNCB in Mice (산사(山楂)가 DNCB로 유발된 생쥐의 알레르기성 접촉피부염에 미치는 영향)

  • Allergic contact dermatitis (ACD) is a type IV delayed hypersensitivity reaction that results from exposures and subsequent sensitization to an environmental chemical. Crataegus Pinnatifida (CP) is commonly used to improve spleen function, remove retention of food, and promote blood circulation. This study is designed to investigate the effects of CP on ACD induced by 2,4-dinitrochlorobenzene (DNCB) in mice. In this experiment, the effects of CP on changes in body weights, ear and dorsum skin thicknesses, ear weights, clinical aspects on the dorsum skin, histopathological changes, spleen weights, cytokines were investigated. In addition, the effects on the proliferation rates of splenocytes were also investigated in vivo and vitro study. In results, CP spread (CPS) group and CP spread and administered (CPS+Adm) group showed decrease in spleen weights. In CPS+Adm group, dorsum skin thicknesses were decreased significantly compared to control group. CP treatment diminished erythema, desquamation and keratosis which were induced by repeated painting of DNCB. In histopathological observation, spongiosis and edema were diminished in CPS and CPS+Adm group. CP led to decrease in the proliferation rates of splenocytes in vivo and vitro. In conclusion, these data suggest that CP can decrease symptoms of ACD, so CP is useful to treat patient with ACD.

1 YEAR FOLLOW-UP STUDY OF DIRECT AND INDIRECT COMPOSITE RESTORATIONS (직접법과 간접법으로 수복한 복합레진의 1년간의 임상적인 평가)

  • Background : The purpose of the present study was to evaluate the direct and indirect composite restorations which had been placed for 1 year Methods : The composite restorations which had been placed between 1999. Mar and 1999, Dec was evaluated after 1 year For direct restorations. Spectrum (Dentsply, USA) and Z100 (3M, USA) were used in the anterior teeth and Surefil (Dentsply, USA) were used. For class V restorations of anterior and posterior teeth. Spectrum was used. For indirect restorations, Targis/Vectris system (Vivadent/Ivoclar, Liechtenstein) was used 2 examiners evaluated marginal quality, proximal contact. discoloration, presence of 2$^{nd}$ caries, loss of filling and hypersensitivity of restorations. The restorations was clinically evaluated by modified methods based on USPHS. Results : 60 teeth were evaluated. 59 were clinically acceptable and 1 restoration which was placed in class v cavity in the posterior tooth was fallen out. In most cases, the restorations were clinically accept-able. For restorations which had been directly placed in the class II cavities, loose proximal contact was indicated as the main complaints. Conclusions : Most of Anterior and posterior restorations which bad been directly or indirectly placed for 1 year were clinically acceptable. For posterior teeth, loose proximal contact was indicated as the main problem in the directly placed Class II restorations. Long term clinical study is needed.

The Effects of Gyejijakyakjimo-Tang on the Allergic Contact Dermatitis induced by DNCB (계지작약지모탕(桂枝芍藥知母湯)이 DNCB로 유발된 알레르기성 접촉피부염에 미치는 영향)

  • Objectives : The purpose of this study is to investigate the effects of Gyejijakyakjimo-Tang on the Allergic Contact Dermatitis caused by 2,4-dinitro-chlorobezene(DNCB). Methods : Twenty eight mice were divided into four groups ; normal, control, experimental group A and B. Control and experimental groups were induced allergic contact dermatitis by DNCB. Experimental group A was orally administered the Gyejijakyakjimo-Tang and experimental group B was orally administered the prednisolone. In this study, ear thickness measurement, auricle microphotograph observation, MPO(Myeloperoxidase) activity measurement, Reverse transcription-polymerase chain reaction(RT-PCR) analysis of the mRNA level of TNF-$\alpha$, IL-$1{\beta}$ were performed on these four groups. In addition, the effect of Gyejijakyakjimo-Tang on cell viability and the effect of Gyejijakyakjimo-Tang on the compound 48/80-induced histamine release from HMC and RPMC were measured. Results: 1. Both experimental group A and B had decreased ear thickness compared with control group In contact hypersensitivity assay. 2. In experimental group A, inflammatory edema was similarly observed comparing to control group. Nevertheless, inflammatory edema was obviously reduced in experimental group B. In both experimental group A and B, pathological lesion of dermatitis were alleviated. In addition, the numbers of infiltrated inflammatory cells were decreased compared with control group. 3. Compared to the normal group, there was a noticeable increase in MPO activity in control group. However, in experimental group A and B, it showed remarkable inhibition of the increase in MPO activity comparing with control group. 4. The level of expression of TNF-$\alpha$, IL-$1{\beta}$ in experimental group A and B were meaningfully lower than those in control group. 5. In MTT assay, the concentrations of Gyejijakyakjimo-Tang that were used on the test had no cytotoxicity. 6. Gyejijakyakjimo-Tang dose-dependently inhibited the compound 48/80-induced histamine release from both HMC and RPMC. Conclusions : According to above experiments, Gyejijakyakjimo-Tang was effective on allergic contact dermatitis.

Lonicerae Caulis(LC) can clear away heat and relieve toxin, disperse wind and heat, dredge the channel. The present study was designed to investigate the effects of LC on allergic contact dermatitis(ACD) induced by 2,4-dinitrochlorobenzene(DNCB) in mice. Methods : In this experiment, the effects of LC on changes in body weights, ear and dorsum skin thicknesses, ear weights, clinical aspects on the dorsum skin, histopathological changes, spleen/body ratio, cytokines were investigated. In addition, the effects on proliferations of splenocytes were also investigated in vitro and vivo study. Results : LC spread(SPR) group and LC spread and administered(SPR+ADM) group showed diminished ear thicknesses. In SPR+ADM group, ear weights were lowered significantly compared to contact dermatitis control(CTL) group. LC treatment diminished erythema, desquamation and keratosis which were induced by repeated painting of DNCB. In histopathological observation, spongiosis and edema were diminished in SPR and SPR+ADM group. In cytokines, SPR+ADM group were increased in IL-10, and SPR and SPR+ADM group were decreased in TNF-${\alpha}$ compared with control group. Conclusions : These data suggest that LC can decrease symptoms of ACD, then LC is useful to treat patient with ACD.

  • Objective To study the effectiveness of Yuldahansotang against Allergic Contact Dermatitis, the change of cutaneous shape, histochemistry, immunohistochemistry and distribution of apoptotic cells was researched. materials and methods 4-month-old rats were divided into three groups of 10. One is a contrastive group which has applied Acetone olive oil only. Another is ACD group which has intentionally activated Allergic Contact Dermatitis by DNCB. And the other is YST group which has given medication of Yangkyuksanhawtang extract. Each group of mice were observed after 24, 48 and 72 hours. results 1. With the result of Contact hypersensitivity assay, YST group shows appreciably less ear swelling than ACD group. 2. Comparing YST and ACD groups to each other regarding general change of skin, YST group shows less hyperplasia of epidermis, less migration of inflammatory cells and less damage of epidermis than ACD group. 3. Regarding the change of collagen fiber, ACD group has appeared to be low in number of collagen fiber while YST shows similarity with the contrastive group. 4. In dermis YST group has showed lower number of mastocyte than ACD group and is granulated type. 5. In dermis YST group has showed less MAC-1,IL-1, $IL-2R-{\alpha}$, ICAM-1 and VCAM-1 than ACD group. 6. The distribution of apoptotic cells has appeared littler in YST group than in ACD 7. Among signal molecule of apoptosis Bcl-2 has distributed more in YST group than ACD group and Bax and Fas has distributed less in YST group than ACD group.

Inhibition of Contact Dermatitis in Animal Models and Suppression of Proinflammatory Gene Expression by Topically Applied Flavonoid, Wogonin

  • Wogonin (5,7-dihydroxy-8-methoxyflavone) is a down-regulator of cyclooxygenase-2 and inducible nitric oxide synthase expression, contributing to anti-inflammatory activity in vivo. For further characterization of modulatory activity on ploinflammatory gene expression in vivo, the effect of wogonin was examined in this experiment using animal models of skin inflammation. By topical application, wogonin inhibited an edematic response as well as ploinflammatory gene expression against contact dermatitis In mice. Wogonin inhibited ear edema ($19.4-22.6\%$) at doses of $50-200\;{\mu}g$/ear and down-regulated interleukin-$1{\beta}$ induction ($23.1\%$) at $200{\mu}g$/ear in phenol-induced simple irritation. Wogonin ($2{\times}50-2{\times}200{\mu}g$/ear) also inhibited edematic response ($51.2-43.9\%$) and down-regulated ploinflammatory gene expression of cyclooxygenase-2, interleukin-$1{\beta}$, interferon-$\gamma$, intercellular adhesion molecule-1 and inducible nitric oxide synthase with some different sensitivity against picryl chloride-induced delayed hypersensitivity reaction. All these results clearly demonstrate that wogonin is a down-regulator of ploinflammatory gene expression in animal models of skin inflammation. Therefore, wogonin may have potential for a new anti-inflammatory agent against skin inflammation.

Behavioral Response and Immune Alterations by Electric Footshock in Mice (생쥐에서 전기자극 스트레스에 의한 행동반응과 면역 기능 변화)

  • The present experiment was designed to investigate the effects of behavioral, response to immune function in response to electric footshock in mice. Mice were subjected to electric footshock for 3 days(two sessions a day, 11 times of shock for about 31 minutes a session). The humoral immune response was measured using mice immunized with rat RBC. The cell-mediated immune responses were evaluated by contact hypersensitivity to 2, 4-dinitrofluorobenzene(DNFB) and by phytohemagglutin(PHA)-stimulated splenocytes proliferation assay. In stressed group, electric footshock suppressed significantly anti-rat RBC antibody production(p<0.05), but enhanced significantly $T_{48}$ relative to $T_{24}$ in contact hypersenstivry (P<.01) and T-cell proliferation response(P<.05) by PHA stimulation elative to control group. T-cell proliferation response by PHA stimulation was significantly correlated to the movement than the sensitivity and coping behavior in the mouse, in response to the electric footshock. These data supper the importance of behavioral response in stress-induced changes of immune functions.

The more industrialization advanced, the more materials that could induce the allergic contact dermatitis have been increased. Therefore in oriental medicine, various studies have been performed. The objective of this study is to investigate the effects of Naetakchunkeum-san on the Allergic Contact Dermatitis induced by 2,4-dinitro-chlorobezene(DNCB). Meterial and Methods : Twenty eight mice were divided into four groups ; normal, control, experimental group A and B. Control and experimental group were induced allergic contact dermatitis by DNCB. Experimental group A was orally administered the Naetakchunkeum-san and experimental group B was orally administered the prednisolone. In this study, ear thickness measurement, observation auricle microphotograph, Myeloperoxidase(MPO) activity measurement, Reverse transcription-polymerase chain reaction(RT-PCR) analysis of the mRNA level of $TNF-\alpha$, $IL-1\beta$, $INF-\gamma$ were performed on these four groups. In addition, the effect of Naetakchunkeum-san on cell viability and the effect of Naetakchunkeum-san on the compound 48/80-induced histamine release from HMC and RPMC were measured, Results : 1. In contact hypersensitivity assay, experimental group A and B showed decreased ear thickness compared with control group, 2. In addition, the numbers of infiltrated cells were reduced, and cleft was not shown compared with control group, In experimental group B, similar results were shown. 3. There was a significant increase in MPO activity in control group compared with normal group, Experimental group A and B significantly inhibited the increase in MPO activity compared with control group. 4, The level of expression of $TNF-\alpha$, $IL-1\beta$, $INF-\gamma$ in experimental group A and B were significantly lower than those in control group. As the internal control, cyclophilin mRNA was also reverse-transcribed and amplified. 5, In MTT assay, there were no statistically significant differences in 100 ${\mu}g/ml$, 200 ${\mu}g/ml$, 500 ${\mu}g/ml$, 1000 ${\mu}g/ml$ Naetakchunkeum-san treated group from 0 ${\mu}g/ml$ Naetakchunkeum-san treated group as determined by the Tukey test. 6. Naetakchunkeum-san dose-dependently inhibited the compound 48/80-induced histamine release from both HMC and RPMC. Conclusions : According to above experiments, Naetakchunkeum-san may be applied to allergic contact dermatitis.

Topical Irradiation of UVA to The Eye Induces Immunosuppression in The Mice via Nitric-Oxise Dependent Neuronal Pathways

  • It has been well documented that dermal irradiation by ultraviolet A (UVA) locally decreases the number of Langerhans cells and suppresses contact hypersensitivity of the skin. We found that topical irradiation of UVA to the eye systemically decreased the number of Langerhans cells (LC) in the dorsalskin and lymph nodes and elicited lymphocyte apoptosis in the latter tissues but not in the thymus. Optic nerve resection, but not ciliary ganglionectomy, eliminated the UVA-induced decrease in dermal Langerhans cells by a mechanism that was partially inhibited by hypophysectomy. The immunosuppressive effect of UVA was not observed in knockout mice lacking inducible-type of nitric oxide synthase (iNOS). These results suggested that topical irradiation of UVA to the eye induced immunosuppression via NO-dependet neuronal pathways.
