Expression of E. coli Phosphofructokinase Gene in an Autotrophic Bacterium Acidithiobacillus thiooxidans

  • A plasmid pSDK-l containing the Escherichia coli phosphofructokinase-l gene (pfkA) was constructed, and transferred into extremely acidophilic Acidithiobacillus thiooxidans Tt-7 by conjugation with the aid of plasmid RP4 at a frequency of $10^{-5}$ per recipient. This plasmid was stable in A. thiooxidans. The pfkA gene from E. coli could be expressed in this obligately autotrophic bacterium, but the enzyme activity (21.6 U/g protein) was lower than that in E. coli (K12: 85.9 Dig protein; DF1010 carrying plasmid pSDK-l: 96.6 U/g protein). In the presence of glucose, the Tt-7 transconjugants consumed glucose, leading to a better growth yield.

  • Eosin-doped boric acid glass saturable absorber has a relatively low saturation intensity (700 mW/$\textrm{cm}^2$) and low power optical phase conjugation is achived by the degenerate four-wave mixing (DFWM) experiments. Polarization properties of optical phase conjugation by DFWM have been demonstrated in this materials using a cw argon ion laser at wavelength 488 nm. The dependence of the phase conjugated reflectivity on the intensity and wavelength of the pump beam is examined. The magnitude of the energy exchange by the nondegenerate two-wave mixing (NDTWM) is maximized by frequency difference between the two beams of $\varphi\simeq$ 1100 Hz.

  • The spectinomycin resistant strain of slow growing R. japonicum R-168 was selected to be participated in conjugation with E. coli WA803/pGS9. Tn5 was introduced from suicide vector pGS9 into R. japonicum R-168 $spr^{r}$ chromosome at the frequency of $1.0\times 10^{-5}-5.0\times 10^{-7}$ and the transconjugante were selected on the yeast extract-mannitol plate containing kanamycin ($50{\mu}$g/ml) and spectinomycin ($100{\mu}$g/ml) after 8-9 days incubation. All transconjugants we tested were found to contain Tn 5 DNA on their genome, which was confirmed by Southern hybridization experiments. R. japonicum RNa75, which had been selected through plant test, was found to be defective in symbiotic nitrogen fixing ability and the production of leghemoglobin in soybean nodules formed by the inoculation of this mutant. In addition, this mutant strain hardly developed nitrogenase activity asymbiotically in contrast with the wild type strain, indicating that some nitrogen fixing gene might be blocked in this strain and the production of leghemoglobin could be decreased by the interference in nitrogen fixing genes.

  • The induced linewidth, frequency shift and absorption spectrum for a molecular dipole in the vicinity of a phase -conjugate mirror have been investgated within a classical phenomenological model, with particularreference to the technique of optical phase conjugation by a surface. While the shifts and the widths show similar characteristics as those obtained recently by Bochove who considered the problem within the context of four-wave mixing, the results obtained in the present model can be defined uniquely with the possibility of an infinite lifetime for the excited admolecule . Furthermore, the absorption lineshape obtained here some interesting features which depend on both the magnitude and the phase of the complex reflectivity of the mirror.

  • The carbonyl stretching vibration, νC=O of 2-cyclohexene-1-one, is in Fermi resonance with a combination tone. The amount of Fermi resonance interaction between these two modes is dependent upon the amount of solute/solvent interaction due to hyd rogen bonding between the carbonyl oxygen and the solvent proton. The corrected νC=O frequency of 2-cyclohexene-1-one occurs at a lower frequency than the observed νC=O mode of cyclohexanone, possibly caused by expanded conjugation effects. The carbonyl stretching modes of cyclic ketones were also affected by interaction with the ROH/CCl4 mixed solvent system.

  • To investigate the epidemiological trait of intestinal diseases of animals caused by thermophilic Campyllobacter spp., isolation of etiological agent was carried out and the profiles of plasmids and the transfer of resistance plasmid in the isolated Campylobacter spp. were examined. The results were as follows. 1. A total of 110 isolates of C jejuni and C coli were subjected to the test for the presence of plasmid DNA. Of the isolates examined, 60% of the isolates were noted to harbor plasmid DNA. Plasmid occurrencer ate from pigs, chickens and cattle were 76.2%, 61.7% and 37.7%, respectively. The plasmids of a large molecular weight, ranging from 36 Md to 86Md, were identified with the strains of tetracycline resistant. 2. Transfer frequency of tetracycline resistant plasmids was higher in the case of the filter mating method than in the broth mating method by the factor of 10~1,000. 3. Tetracycline resistant plasmids of C jejuni were transferrable to C jejuni and C coli by conjugation. In a low frequency, the transfer of tetracycline plasmid was also possible to Vibrio parahemolyticus. However, it was impossible to transfer to Streptococcus fecalis, E coli and Vibrio cholerae. 4. Tetracycline resistant plasmids of C jejuni were impossible to transfer to Campylobacter spp. and related bacteria by transformation.

Curing and segregation of pSL100 and recombination of its segregants (Plasmid pSL100의 curing, segregation 및 segregants 들의 재조합에 관한 연구)

  • A study was undertaken to examine the effect of curing agents on the stability, curing and segregation of R plasmid pSL100. And also the stability, transfer frequency, and recombination of its segregants obtained from curing agent treatment were studied. Ethidium bromide, acridine orange, and mitomycin-C were used as curing agent. The results obtained were as follows ; 1. The curing agent ethidium bromide, acridine orange, and mitomycin-C were not effective for curing the multiple antibiotic resistant determinant of pSL100 in Salmonella typhimurium and Escherichia coli. However, they induced plasmid segregation with high frequency in S.typhimuruim LT-2strains. TcApSmCm, TcSmCmKm, TcApCm, TcAp, TcKm, Tc segregants were obtained. 2. The resistant markers of the segregents were transferred to S.typhimurium LT-2 strains with high frequencies whereas they were transferred to E.coli K-12 only with low frequencies. 3. The transconjugants obtained from conjugation between two different S.typhimurium segregants were similar to the phenotype of the original R factor pSL100 and the resistant markers were transferred to the S.typhimurium LT-2 or E.coli strain with equal frequencies, indicating that they are recombinants. 4. The transconjugants obtained from conjugation between pSL100 segrgants and pKM101, or pBR322 possessed the resistant markers of the two parental plasmids and they were transferred to both S.typhimurium and E.coli K-12 strains with the same frequencies and maintained stably, suggesting that they are also recombinants. 5. The recombinant pSL100 could be also obtained in rec A-strains of E.coli, suggesting that the gene function of rec A is required for the recombination of pSL100 segregants in E.coli.

Distribution and Drug Resistance of Salmonella Causing Enteric Fever in Taegu Area of Korea (대구지방에서의 장열성 Salmonella의 분포와 항균제내성)

  • Salmonella strains isolated from blood in the Dong-san hospital, Taegu during the period from 1971 to 1986 were studied for species distribution, drug resistance, and R plasmids. The number of strains was 2,527 and all of them were classified into S. typhi and S. paratyphi A. Approximately 300 strains were isolated in the period from 1974 to 1976 and 1978, 268 in 1982, and 204 in 1983, but the numbers isolated in the 1980's have a tendency to decrease as compared with those of the 1970's. S. typhi occupied 85% or more of strains isolated until 1976, but the isolation frequency decreased yearly with some variation, and S. paratyphi A increased gradually from 1974. Only 4 strains of S. paratyphi A were resistant to some drugs, and the resistance was not transferred to E. coli by conjugation. S. typhi resistant to drugs were 15 in 1971 through 1973, 24 in 1974, and 13 in 1975, but afterwards only few resistant strains were isolated. These strains were resistant to two or more drugs; chloramphenicol(Cm), tetracycline(Tc), streptomycin(Sm), sulfisomidine(Su), ampicillin(Ap), and kanamycin(Km) and no strain resistant to other drugs tested was found. Strains resistant to 3 or less drugs didn't transfer the resistance to E. coli by conjugation. There were 15 strains resistant to four or more drugs, and were isolated in years from 1972 to 1976. These strains transferred the resistance to E. coli, and the resistance was considered to be mediated by R plasmids. Transfer frequency was higher at $25^{\circ}C$ than at $37^{\circ}C$ and patterns of transferred resistance were Cm, Tc, Sm, Su; Ap, Km; Cm. R plasmids having markers of Cm, Tc, Sm and Su were classified into Inc H1.

Application of a New Conjugation Method to Fish Pathogenic Bacteria Containing R Plasmid for the Analysis of Drug-Resistant Status in Aquaculture (새로운 conjugation 방법을 응용한 R plasmid 함유 어병세균의 분리와 양식장 내성균의 현황 분석)

  • To develop a new method of conjugation and to determine the distribution of R plasimds, we isolated multi-drug resistant strains from fish pathogenic bacteria in the farms of south and east seacoasts of Korea. Out of the 134 isolates examined, 10 showed resistance to chloramphenicol, tetracycline, streptomycin, ampicillin, colistin, nalidixic acid, oxolinic acid and kanamycin. One out of 10 multi-drug resistance bacteria, Vibfio damsela JE1 (V. damsela JE1), contained transferable R plasmid of chlorarnphenicol- tetracycline resistance genes and other nucleic acids encoding ampicillin and kanamycin resistance. The presence of the R plasmid was confirmed by conjugation using the chromocult medium (CC) as a selective and differential medium for transconiugants with identification based on the growth or colors of the colonies. The frequency of R plasmid transfer with filter mating method was come out much higher than that of broth mating method and appeared to be dependent upon the mating time and temperature. The optimum conditions for filter mating method were found to be 30$^{\circ}C$ and 24hrs as mating temperature and period, respectively, Moreover, donor cells with R plasmid, both isolate and standard bacteria, were shown to have an ability to transfer the plasmid against Escherichia coli K-12 HB101 (E. coli HB101) and Edwardsiella tarda (E. tarda) RE14 at fairly high frequencies, finally, we isolated 3 isolates of Sphingomonas sp., carrying R plasmid from 12 multi-drug resistant bacteria in normal microflora of the flounder (Paralichthys olivaceus) group used for the isolation of V emsela JE1 four months before. The same size and gene transfer chayateristics of R plasimds with those of V damsela JE1 confirmed that normal microflora have the reservoir activity for R plasmid in natural aquatic environment.

The Mental Representation of English Verb in Korean-English Moderate Bilingual (한국어-영어 이중언어화자의 외국어 동사 표상)

  • This research was investigate to understand how moderate bilingual subjects represent the lexicon in second language. Although most researches have focused only on high proficient bilinguals, we analysed how moderate bilinguals who have learned English mostly in school represent the prototype of verb and its inflected form of verb. Results of lexical decision task showed that moderate bilingual subjects used different mental representation depending on whether the verb have regular or irregular conjugation. With regular verbs, the identification of an inflected form was affected by both the frequency of its prototype and that of inflected form, but with irregular verbs, it is affected only by the frequency of inflected form.
