In March of 2003, the gray mold occurred severely on pea (Pisum sativum) grown in the green houses and open fields in Changseon-myon, Namhae-gun, Gyeongnam Province, Korea. The symptoms started with water-soaking lesions in the leaves, stems and pods. The typical symptoms of the leaves, stems and pods became withered, discolored and blighted, and then eventually died. The mycelia, conidia and conidiophore of the pathogen appeared on leaves, stems and pods. The conidia were gray, 1-celled, mostly ellipsoid or ovoid in the shape and were $8{\sim}17{\times}5{\sim}13\;{\mu}m$ in size. Conidiophores were $14{\sim}30\;{\mu}m$ in size. The sclerotia were formed abundantly on potato dextrose agar. The optimum temperature for sclerotial formation was $20^{\circ}C$. Pathogenicity of the causal organism was proven according to Koch's postulate. The causal organism was identified as Botrytis cinerea based on mycological characteristics. This is the first report on gray mold of Pisum sativum caused by B. cinerea in Korea.
Fusarium verticillioides is an important pathogen of maize, being responsible for ear rots, stalk rots, and seedling blight worldwide. During the past decade, F. verticillioides has caused several severe epidemics of maize seedling blight in many areas of China, which lead to significant losses. In order to understand the molecular mechanisms regulating fungal development and pathogenicity in this pathogen, we isolated and characterized the gene fpk1 (GenBank Accession No. EF405959) encoding a homolog of the cAMP-dependent protein kinase catalytic subunit, which included a 1,854-bp DNA sequence from ATG to TAA, with a 1,680-bp coding region, and three introns (lengths: 66 bp, 54 bp, and 54 bp), and the predicated protein precursor had 559 aa. The mutant ${\Delta}fpk1$, which was disrupted of the fpkl gene, showed reduced vegetative growth, fewer and shorter aerial mycelia, strongly impaired conidiation, and reduced spore germination rate. After germinating, the fresh hypha was stubby and lacking of branch. When inoculated in susceptible maize varieties, the infection of the mutant ${\Delta}fpk1$ was delayed and the infection efficiency was reduced compared with that of the wild-type strain. AU this indicated that gene fpk1 participated in hyphal growth, conidiophore production, spore germination, and virulence in F. verticillioides.
The resistance of 10 varieties of cucumber (Cucumis sativus L.) to powdery mildew, caused by Sphaerotheca fusca, was evaluated by a leaf disk assay. Leaf disks (10 mm in diameter) were removed from fully expanded leaves and then placed in petri dishes containing 0.16% water agar amended with benzimidazole. Leaf disks were inoculated by dropping a 10 $\mu$l of conidia suspension. Conidiophore formation of powdery mildew was the greatest at $25^{\circ}C$. The response of the host to powdery mildew, based on the inoculation onto disks of the first leaf, highly correlated with results obtained from harvesting stage of cucumber plants in greenhouse test (r = 0.99$^{**}$). It is indicating that a leaf disk assay may precisely predict the response of cucumber plant to S. fusca.a.
Strain NR $15-1^T$ isolated from traditional Korean Nuruk is described as a new species and named as Aspergillus coreanus NR $15-1^T$ sp. novo Strain NR $15-1^T$ grew rapidly to form yellow-green colonies whose surfaces were velvety on Czapek solution agar. Conidial heads were yellow to light and elliptical, whereas the conidiophore was colorless and typically long. In addition, vesicles were from flask-shaped to globose, and sterigmata are uniseriate. Conidia were spherical and deep yellow-green, and their surfaces were lightly roughened. The G+C content of strain NR $15-1^T$ was 51 mol% and strain NR $15-1^T$contained a dihydrogenated ubiquinone with Q9 (94.9%) as a major quinone. The nucleotide sequences of strain NR $15-1^T$ in the two Internal Transcribed Spacers (ITS 1 and 2) and 5.8S rDNA showed highest similarity when compared with that of A. tubingensis and A. phoenicis NRRL $365^T$. However, based on morphological and chemotaxonomic characteristics, this strain was different from A. tubingensis and A. phoenicis NRRL $365^T$. On the basis of the data presented, it is proposed that strain NR $15-1^T$ should be placed in the genus Aspergillus as a new species, Aspergillus coreanus sp. novo Therefore, the type strain of the new species is strain NR $15-1^T$(=KCTC 18075P^T,=KCCM 80006^T$.
The powdery mildew of broad bean (Vicia faba) extensively occurred in farmers' fields in Changseon-myon, Namhae-gun, Gyeongsangnam-do, Korea in 2001. Both sides of the leaves and stems were covered with white fungal spores and mycelia, and then the leaves and stems colored brown and dark brown and eventually died. Conidia and conidiophores were formed on the lesion. Conidia were cylindric, 29~45$\times$13~23 um (av, 36~18 um) in size and born singly on the conidiophore. Fibrosin bodies were not observed. Conidiophores were straight with 3-4 cylindric cells and 51~100$\times$6~10 um (av.70$\times$8 um) in size. Foot cell was 28~53$\times$7~10 um (av,39$\times$8 um) in size. Appressorium was a lobed type. Cleistothecia were not formed. This is the first report on powdery mildew of broad bean caused by Oidium sp. in Korea.
A severe pink mold rot on matured asian pear (Pyrus serotina Rehder) fruit occurred in the organic farmers' orchard in Jinju, Korea in October, 2012. Decay of pear fruit appeared as a softened water-soaked symptom that was easily punctured by pressure. Later pink mycelium appeared on the surface of pear fruit and produced a mass of powdery pink conidia spores. Optimum temperature for mycelial growth of T. roseum was $25^{\circ}C$. Conidia showed hyaline, smooth, 2-celled, thick-walled with truncate bases, ellipsoidal to pyriform, and characteristically held together zig-zag chains and $10{\sim}22(34){\times}6{\sim}10(12){\mu}m$ in size. Conidiophore was erect, colorless, unbranched type, and 4-5 ${\mu}m$ width. On the basis of mycological characteristics, pathogenicity test, and molecular identification with the ITS region, the causal fungus was identified as Trichothecium roseum (Pers.) Link ex Gray.
Cylindrocarpon spp. were isolated from the soil where strawberry was grown in Suweon by soil plate method: colonies reaching 10 mm diam. in seven days at about $20^{\circ}C$; sporodochia with cream to beige to conidial slime commonly produced; conidiophore repeatedly branched and bearing subulate phialides; macroconidia cylindrical in the center part, straight or slightly curved and mostly $1{\sim}3\;septate,\;22{\sim}45\;{\times}\;5.0{\sim}6.0\;{\mu}m$; chlamydospore abundantly produced, intercalary or terminal on mycelium, singly or in chains and smooth or warted. The hypha and spore were easily fused each other on water agar. This fungus was pathogenic strawberry as a result of inoculation test. The symptom showed dwarf and yellowing at top and rotted roots under the ground. The fungus was identified as Cylindrocarpon destructans Scholten from the shape of conidiophores and conidia, mycelial growth and pathogenicity test.
The powdery mildew of Phlox (Phlox paniculata) extensively occurred in the experimental farm of Cheongam-myon, Hadong-gun, Gyeongnam province in 2002. Both sides of the leaves and stems were covered with the white fungal spores and mycelia, and then the leaves became brown to dark brown in color, eventually died. The conidiophores on the lesion were observed. Conidia were cylindric in shape, colorless, $25.3{\sim}49.4{\times}12.3{\sim}17.2{\mu}m$ in size and borne singly on conidiophore. Fibrosin bodies were not observed. Conidiophores were straight with 2-4 cylindric cells, and $65.7{\sim}124.8{\mu}m$ in size. Appressoria were lobed. Cleistothecia were not formed. This is the first report on powdery mildew of Phlox caused by Oidium sp. in Korea.
Twenty-two strains of nematophagous fungi were isolated from 100 soil samples. Nematophagous fungi were classified into three categories; 3-dimensional adhesive nets (A group), 2-dimensional adhesive nets (B group) and constricting ring (C group). Nine strains were selected and identified on the basis of morphological characteristics (hypha, conidiophore, form and size of conidia, number of conidia, node of conidophore, number and location of septa, size and color of chlamydospore) and ITS (internal transcribed spacer) region of rDNA sequences. As the results, the isolated were identified as belonging to the species of Monacrosporium thaumasium (Kan-2, Kan-4, Kan-11), Arthrobotrys oligospora (Kan-9, Kan-13, Kan-20, Kan-21), A. musiformis (Kan-12), and A. dactyloides (Kan-22).
Aspergillus niger van Tieghem was cultured by the method of synchronous and submerged culture. Throughout the culture, sporulation was occured. Highly phosphorylated nucleotides in sporulating mycelia were detected to assure whether the eucaryotic Aspergillus niger produce these substances or not during the differentiation. Phosphorylated nucleotides were extracted from the conidiophore, bearing mycelia and spore forming body, these nucleotides were identified by TLC with P.E.I. cellulose. Guanosine tetraphosphate was found in both phialide forming mycelia and spore forming body. The contents of free amino acids were assayed and its level was found to increase at early stage of sporulation. The effects of 8-azaguanine examined, it was found to prevent spore formation and to made abnormal structure. The effects of inosinic monophosphate and guanosine monophosphate on spore formation were examined, spore formation was enhanced by these nucleotides.
