In vitro anti-skin-aging effects of dried pomegranate concentrated powder

  • Purpose : In this study, we intended to observe the anti-wrinkle and moisturizing effects of dried pomegranate juice concentration powder (PCP) using in vitro test. Materials and methods : Antioxidant effects of PCP were determined by free radical scavenging capacity (DPPH assay) and the cytotoxicity of PCP was examined in human keratinocyte (HaCaT) and human primary dermal fibroblast-neonatal (HDF) cells. To investigate the moisturizing effect of PCP, hyaluronan synthesis was examined in HaCaT cells. Activity of procollagen production were assessed in HDF cells and elastase inhibition properties of PCP were evaluated in cell free condition, to determine their anti-wrinkle effects. Metalloproteinase 1 (MMP-1) activity was also assessed following UVB irradiation, in the current in vitro experiment. Results : No PCP treatment related significant cytotoxic effects were demonstrated against to the both HDF and HaCaT cells. PCP showed favorable free radical scavenging activities in dose-dependent manner. In PCP-treated HaCaT cells, hyaluronan synthesis was non-significantly but markedly increased, and pro-collagen productions were significantly increased in HDF cells, at all three different concentrations (0.25, 0.75 and 1 mg/ml), and elastase inhibitory activities were observed by PCP treatment. A significant decrease in UVB-induced MMP-1 activity was also observed in 1 mg/ml PCP-treated HDF cells as compared to those of UVB-exposed cells. Conclusions : Taken together, these results suggest that PCP has favorable antioxidant, anti-wrinkle and moisturizing effects without meaningful cytotoxicity on HDF and HaCaT cell lines.

Detection of Metastases of Primary Hepatocellular Carcinoma with $^{99m}Tc-HIDA$ Scintigraphy ($^{99m}Tc-HIDA$를 이용(利用)한 원발성간세포암전이(原發性肝細胞癌轉移) 병소(病巢)의 진단(診斷))

  • $^{99m}Tc-Sulfur$ Colloid is concentrated in Kupffer cells of the liver, whereas the new biliary agents such as $^{99m}Tc-HIDA$ are processed by hepatic parenchymal cells. The distant metastatic lesiors in skull and lung of the primary hepatocellular carcinoma in 38-year old Korean male were detected with $^{99m}Tc-HIDA$ scintigraphy. The chest PA, skull bone X-ray and radionuclide scintigraphic studies are illustrated. This observation suggests that $^{99m}Tc-HIDA$ scintigraphy is useful for detection of distant metastases of primary hepatocellular carcinoma.

Improved Purification Process for Cholera Toxin and its Application to the Quantification of Residual Toxin in Cholera Vaccines

  • A simplified method for the purification of cholera toxin was developed. The 569B strain of Vibrio cholerae, a recognized hyper-producer of cholera toxin, was propagated in a bioreactor under conditions that promote the production of the toxin. The toxin was separated from the bacterial cells using 0.2-${\mu}m$ crossflow microfiltration, the clarified toxin was passed through the membrane into the permeate, and the bacterial cells were retained in the retentate. The 0.2-${\mu}m$ permeate was then concentrated 3-fold and diafiltered against 10 mM phosphate buffer, pH 7.6, using 30-kDa crossflow ultrafiltration. The concentrated toxin was loaded onto a cation exchange column, the toxin was bound to the column, and most of the impurities were passed unimpeded through the column. The toxin was eluted with a salt gradient of phosphate buffer, pH 7.0, containing 1.0 M NaCl. The peak containing the toxin was assayed for cholera toxin and protein and the purity was determined to be 92%. The toxin peak had a low endotoxin level of $3.1\;EU/{\mu}g$ of toxin. The purified toxin was used to prepare antiserum against whole toxin, which was used in a $G_{M1}$ ganglioside-binding ELISA to determine residual levels of toxin in an oral inactivated whole-cell cholera vaccine. The $G_{M1}$ ganglioside-binding ELISA was shown to be very sensitive and capable of detecting as little as 1 ng/ml of cholera toxin.

Heavy Metal Accumulation in Cell of Heavy Metal-Tolerant Bacteria by Some Physical and Chemical Treatments (물리화학적 전처리에 의한 중금속 내성세균의 균체내 중금속 축적 변화)

  • Heavy metal-tolerant microorganisms, such as Pseudomonas putida, P. aeruginosa, P. chlororaphis and P. stutzeri which possessed the ability to accumulate cadmium, lead, zinc and copper, respectively, were isolated from industrial wastewaters and mine wasewaters polluted with various heavy metals. Metal binding sites in the cells were investigated by extracting the components of the cells through pretreatments with hot water, acid, alkli, chloroform-methanol or chloroform-methanol/concentrated alkali. The heavy metal accumulation was drastically decreased by pretreatment with alkali or chloroform-methanol/concentrated alkali, but the heavy metal accumulation was not changed by pretreatment with chloroform-methanol. The amount of heavy metal accumulation was remarkably decreased by decreasing crude protein remaining in the cell. These results suggested that proteins of cell components played an important role on the heavy metal accumulation.

Effects of Cuscutae Semen Water Extract on Apoptosis of MCF-7 Human Breast Cancer Cells (토사자 추출물이 MCF-7 유방암 세포의 세포자멸사에 미치는 영향)

  • Objectives: This study aimed to evaluate the effects of Cuscutae Semen water extract (CS) on MCF-7 human breast cancer cells. Methods: To clarify the results, we cultivated MCF-7 cells in cell culture plates. And then we extracted each of $100{\mu}g/ml$, $300{\mu}g/ml$, $600{\mu}g/ml$ CS, gave it to MCF-7 cell. After these process we performed MTT assay to elucidate the ability of apoptosis. The result of mRNA was analyzed by RT-PCR. Results: Each of concentrated extracts CS decreased the survival rate of MCF-7 cells. CS decreased Bcl-2 which is known as a blocking cell apoptosis. Bax, caspase-3, P21 and RIP-1 that accelerate apoptogenic activity factors increased by CS. CS did not change the condition of caspase-8, caspase-9, P53 factors on MCF-7 cells. Furthermore caspase-8, caspase-9, P53 factors on MCF-7 cells does not make it more active but turn it on. Conclusions: According to the above results, we could suggest that CS can occur the apoptosis on MCF-7 cells.

  • Different kinds of forces can be applied to the biological tissue. The analysis of the applied force is highly important to explain the mechanism of cellular response. In this study, the applied force to the collagen gel was analyzed by the finite elements analysis. The model received two different kinds of static force (compression and tension). The force range was 50g to 400g. In results, von Mises stress was concentrated in the peripheral region in the compression model. It was concentrated in the central area in the tension model. However, the compressive force was high in the peripheral area of the compression model and the tensional force was also high in the same area of the tension model. In conclusion, the applied force could be different to the region and it should be considered in the experiment to analyze the effects of the mechanical force on the cells.

The Effects of Acorus gramineus on Changes of Nasal Tissue in Allergic Rhinitis Model (석창포가 알레르기성 비염 유발 동물 모델에 미치는 영향)

  • Background & Objectives : Rhinitis is an inflammation of nasal mucosa. The major symtoms are watery rhinorrhea, sneezing, itchy nose, and nasal obstruction. Allergic rhinitis is an immune reaction by allergen. So, we aimed to determine therapeutic effects of Acorus gramineus by observing changes in IL-4, $IFN-{\gamma}$ and the nasal mucosal tissue. Materials and Methods : Fifteen BALC/c mice were divided into three groups : m group(which ate low concentrated herbal medicine ), M group(which ate high concentrated herbal medicine) and control group. Control and experimental group were induced allergic rhinitis by Ovalbumin as the method of Levin and Vaz. Experimental group was orally administered the Acorus gramineus extract for 28days. We observed changes in IL-4, $IFN-{\gamma}$ and trans aminase(AST, ALT) in blood and nasal mucosa and submucosa. Results : There were no significant changes statistically in IL-4 and $IFN-{\gamma}$ in blood(p<0.05). And there were no hepatotoxicity with Acorus gramineus extract. Histologically, almost no inflammatory response in treatment group(m,M) against that there were inflammatory response(increased goblet cells, dilated vessels, edema of bowman's glands and injured olfactory hairs) in control group. Conclusion : According to above results, it is supposed that Acorus gramineus has no immunological effects on allergic rhinitis.

A Study on the Residual Stress Analysis of a-Si Thin Film Solar Cell (a-Si 박막형 태양전지의 잔류응력 해석에 관한 연구)

  • The size and distribution of residual stresses and the effect of the minimum mesh size were investigated by the a-Si thin film solar cell. Attributed to the difference in coefficient of thermal expansion of the a-Si and Ag concentrated residual stresses at the joint interface of dissimilar materials. The ${\sigma}y$ and ${\tau}xy$ didn't appear in the central part, but ${\sigma}x$ existed. However, ${\sigma}x$, ${\sigma}y$ and ${\tau}xy$ appeared in the edge part and concentrated residual stresses at the interface between a-Si and Ag. Minimum mesh size gets smaller, the concentration of ${\sigma}y$ was significantly and existence area was reduced. As a result, the failure of thin film solar cells during the cutting process can be explained by the residual stresses.

Histochemistry of Mucosubstances on the Pedal Sole of Five Abalone Species

  • The present study describes histochemical properties of the mucosubstances in the pedal sole of abalone, in relation to two main additional functions known to date. Only neutral mucosubstances were demonstrated in the lateral epithelium of the pedal sole of the abalone, however, two distinct types of cells, one containing neutral and the other containing acidic mucosubstances, were identified in the ventral epithelium. The epithelial mucocytes distributed throughout both epithelial and subepithelial glands located in the ventral subepithelial region showed a mixture of neutral and acidic mucosubstances. However, acidic mucosubstances were found in larger amounts than neutral ones in epithelial mucocytes, and vice versa in subepithelial glands. Among these acidic mucosubstances, sulphated substances were dominant. Lesser amounts of carboxylated substances also occurred together with low levels of neuramic and sialic acid. Collectively, the notable morphological differences between abalone and other gastropoda were observed in the ventral surface of the foot, which showed a tall columnar epithelium, peripherally concentrated mucocytes, and subepithelial glands. Mucosubstances are presumably produced by these cells and may also play a role in clinging to some substances to support abalones in addition to the main functions of lubrication and protection.

Inhibitory Effect of Extracts from Woody Plants on Tumor Necrosis $Factor-{\alpha}$ Production in Lipopolysaccharide-Stimulated RAW264.7 cells (한국산 자생 수목 유래 수피추출물의 종양괴사인자 억제효과)

  • Naturally occurring substances are important biomedical resources with low toxicity and ethnopharmacology-based efficacy. Four out of 45 extracts (Celastrus orbiculatus, Cercis chinensis, Stephanadra incisa, and Weigela subsessilis) prepared from the bark of Korea Forest plants exhibited more than 50% of inhibition on $TNF-{\alpha}$ production in lipopolysaccharide (LPS)-activated RAW264.7 cells at $100\;{\mu}g/m{\ell}$. In particular, potential inhibitory components of 4 extracts showed more than 50% inhibition seemed to be concentrated in methylene chloride (MC) fraction from C. orbiculatus, in ethyl acetate (EtOAc) fraction from C. chinensis and in hexane (Hx) fraction from S. incisa, whereas inhibitory activities of W. subssilis were broadly seen in non-polar solvent fractions such as Hx, MC and EtOAc. Therefore, our results suggest that extracts from C. orbiculatus, C. chinensis, S. incisa and W. subsessilis may be developed as a therapeutic remedy against $TNF-{\alpha}-mediated$ diseases such as rheumatoid arthritis or further fractionated to isolate active components having $antiTNF-{\alpha}$ inhibitory activity.
