To investigate effects of vanous herbal-acupuncture, which were Daebangpungtang, Oyaksungisan, Dokhwalgisaengtang, and Binsosan on Adjuvant Arthritis in rats, the edema rate, the number of WBC, the quantity of TNF-${\alpa}$, COX -2, and IL-6, and histological test of the muscular tissue were measured in the arthritis part. After elicilating arthritis of Sprague Dawely(SD) rats by injection of Freund's complets adjuvant for 2 weeks, saline was injected for the control group, Daebangpungtang acua-acupuncture was injected for the Daebangpungtang group, Oyaksungisan acua-acupuncture was injected for the Oyaksungisan group, Dokhwalgisaengtang acua-acupuncture was injected for the Dokhwalgisaengtang group, and Binsosan acua-acupuncture was injected for the Binsosan group during 30days. Selected point was ST35 in all groups. 1. The number of WBC was $8.24{\pm}0.51$($10^3$/ml) in the normal group and $27.35{\pm}3.51$($10^3$/ml) in the control group. the Daebangpungtang group and the Oyaksungisan group decreased each $20.42{\pm}2.75$($10^3$/ml) and $19.78{\pm}4.99$($10^3$/ml) in the Exp.ll group. This fact showed that the group Exp.ll was more effective than the control group effectively. (P<0.05) 2. The volume of the paw were checked. The volume of paw was $0.62{\pm}0.11mm in the control group and $0.45{\pm}0.08$mm in the Daebangpungtang group and $0.47{\pm}0.07$mm in the Oyaksungisan group, the swelling of the paw was restricted significantly in the Daebangpungtang and Oyaksungisan group(P<0.05) 3. The bands of the TNF-${\alpa}$, COX -2 in the muscular tissue of the control group wcre certain and thick than other groups(except normal group), and the band of the IL-6 in the muscular tissue of all group(except normal group) were similar forms. 4. In histological finding, because of severe inflammatory reaction, remarkably irregular tissue and large amount of inflammatory cells were found in thc control and Dokhwalgisaengtang group. But the Oyaksungisan and Daebangpul1gtang group showed small amount of inflammatory cells, the refrained inflammatory state and even recovering state. From these results, it is showed Oyaksugisan and Daebangpungtang acua-acupuncturc refrain inflammatory reaction and muscular tissue necrosis in SD rats paw were induced by Freund's complete adjuvant

Objectives : The present study aimed to examine the analgesic effect of electroacupuncture(EA) applied to the brook point of the Liver meridian in the rat model of inflammatory pain and to investigate involvement of endogenouse opioid in the EA-induced analgesia. Methods : Knee arthritis was induced by injection of $125{\mu}l$ of complete Freund's adjuvant into the knee joint cavity unilaterally. To examine the level of pain, weight bearing forces(WBFs) of affected limb was measured. EA treatment was given at the LR2, LI4 or non-acupoint on the contralateral limb with frequency of 2 Hz and intensity of 2 mA under gaseous anesthesia. Results : After induction of arthritis, rats subsequently showed a reduced stepping force of the affected hindlimb due to a painful knee. EA applied to the LR2 point on the contralateral hind limb produced a significant improvement of stepping force of the hind limb lasting for at least 2 h, and this effect was equivalent to that obtained by 5 mg/kg of oral indomethacin. The effect of EA was specific to the acupoint since it could not be mimicked by EA applied to the LI4 point or the arbitrary non-acupoint. The analgesic effect was blocked by pretreatment with naltrexone(10 mg/kg, i.p.). Conclusions : These results suggest that acupuncture applied to LR2, brook point of Liver meridian suppresses inflammatory pain in a rat model of knee arthritis and this effect seems to be mediated by endogenous opioid systems.

To study the antigenicity of BR92021(Vi polysaccharide typhoid vaccine), active systemic ana-phylaxis and passive cutaneous anaphylaxis were tested in guinea pigs. The groups were as follows: group I(low dose, 30 $\mu\textrm{l}$/kg), group II(high dose, 300 $\mu\textrm{l}$/kg), group III(300 $\mu\textrm{l}$/kg plus complete Freund's adjuvant), group IV(positive control, ovalbumin plus complete Freund's adjuvant) and group V(saline-treated control). Male Hartley guinea pigs at 7 weeks of age were sensitized subcutaneously with the test article or saline three times per week for three weeks(j.e., total 9 times). For groups III and IV, animals were sensitized subcutaneously with either the test article or ovalbumin plus complete Freund's adjuvant once per three week for 6 weeks(i.e., total 3 times). Twelve days after the last sensitization, the blood was collected from the sensitized animals for the passive cutaneous anaphylaxis test. In addition, the sensitized animals were subjected to the active systemic anaphylaxis test on fourteen days after the last sensitization by an intravenous challenge with either the test article or ovalbumin. In group I, mild(1/5) or moderate(4/5) symptoms of anaphylactic shock were observed. In group II, no sign(1/5), moderate(3/5) and severe(1/5) symptoms were observed. In group III, four animals of revealed moderate signs and one of 5 showed no signs of anaphylactic shock. In group IV, all 5 animals showed severe signs of shock. In group V, one of 5 revealed moderate and four of 5 showed no signs. The necropsy findings related to the active systemic anaphylaxis were observed in most animals of groups I to V In the passive cutaneous anaphylaxis test, the antiserum was diluted 10- to 5120- fold and was injected intradermally on the clipped back of recipient animals, followed by an intravenous challenge with either the test article or ovalbumin. No animals in groups I, II, III and V showed the positive reaction, whereas all animals in group IV, the positive control, showed the positive reaction at the dilution range of x1280 to x5120. Our results indicate that the test article, BR92021, may have weak antigenic potential in male guinea pigs.

Antigenicity of Recombinant Granulocyte macrophage colony stimulating factor(LBD-005), a newly developed drug for hematopoietic growth, was investigated in mice and guinea pigs. 1. Mice showed production of antibodies against LBD-005 (100mg/kg) with alumin]m hydroxide gel(alum) as an adjuvant, judged by the heterologous anaphylaxis(PCA) test using rats. On the other hand, antibodies against ovalbumin(OVA) inoculated with alum were definitely detected. 2. In the studies with guinea pigs, both the inoculation of LBD-005 only and of LBD-005 with complete Freund's adjuvant(CFA) as an adjuvant did not produce positive reactions in homologous passive cutaneous anaphylaxis (PCA).

본 연구는 랫드에서 Freund's complete adjuvant를 랫드의 후지에 투여하여 인의적으로 유발된 관절염을 진단하기 위한 방법으로 $^{99m}Technetium-MDP (^{99m}Tc_MDP$)을 이용한 골 스캔의 임상적 의의를 찾고자 하였다. 성숙 Sprague-Dawley 암컷 랫드를 대조군과 실험군으로 분리하고, 골의 스캔은 0.05mCi의 $^{99m}Tc_MDP$를 정맥 주사하여 3시간 후에 전신 영상을 얻었다. 골의 스캔과 방사선학적 평가를 비교한 겨과 관절염 유발 7일 후부터 관절염의 병변이 관찰되었지만, 방사선학적 소견에서는 14일 후부터 병변이 관찰되었다. 골의 스캔에서 관절염 유발 7일 후에 유의적으로 골의 이상적 섭취 증각가 관찰되었고(p<0.05), 21일 후에는 전지와 후지의 모든 부위에서 골의 이상적 섭취 증가가 관찰되었지만, 방사선학적 소견에서는 후지에서만 병변 부위가 관찰되었다. 이상의 결과에서 $^{99m}Tc_MDP$을 이용한 골의 스캔은 류마티스 관절염을 임상적으로 진단하는데 방사선학적 진단보다 효과적인 방법임을 알 수가 있었고, 골의 스캔은 류마티스 관절염을 초기에 진단하는데 유효한 진단방법임을 확인하였다.

The methanolic extract of Lysimachiae christinae Herba, which has been used for diuretic, calculi remover and jaundice remedy in oriental countries, was found to possess analgesic and antiinflammatory effect in Freund's complete adjuvant treated rat. From ethyl acetate fraction of the herb a phenyl propanoid isoferulic acid was isolated as an active principle.

'Ssangwha-Tang,' which have known as a tonic and restorative preparation in Korean traditional medicines, was shown to have signification antiinflammatory activity in the edema-induced carrageenin inflammation of rats, and also indicated remarkable preventive effect to chronic arthritis model in rat induced by Freund's complete adjuvant method.

Daidzein and its methyl derivatives were synthesized and their anti-inflammatory activities were examined. Daidzein suppressed the carrageenin-induced edema, but its methyl derivatives showed decrease or abolition of the anti-inflammatory effect. Daidzein did not significantly suppress the complete Freund's adjuvant-induced arthritis, and the mixed phlogistics (histamine+serotonin)-induced edema. Daidzein inhibited the leukocyte emigration and protein exudation when it was administered into the CMC pouch at low doses (5,25mg/pouch). Daidzein significantly suppressed the cotton-pellet granuloma formation.

Objectives : To investigate effects of Achyranthis Radix aqua-acupuncture on adjuvant arthritis in rats. Methods : After eliciting arthritis in Sprague dawley(SD) rats by injection of Freund's complete adjuvant for 2 weeks, normal saline was injected for the Exp. I group and Achyranthis Radix aqua-acupuncture was injected for the Exp. II group during 30days. The selected point was Dokbi (ST35) in both groups. Then the edema rate, the number of WBC, the quantity of total protein, albumin and globuline in the blood serum, and histological test of the muscular tissue were measured in the arthritis stricken areas. Results : I) The volume of the paws were checked. The volume of the paws was $0.84{\pm}0.l4mm$ in the Exp. I group and $0.47{\pm}0.14mm$ in the Exp. IIgroup. The swelling of the paws was restricted significantly in the Exp. II group(P<0.05). 2) The number of WBC was $10.34{\pm}0.14(l03/ml)$ in the normal group and $37.47{\pm}5.46(103/ml)$ in the Exp. I group. It was $21.24{\pm}2.58(103/ml)$ in the Exp. II group. This fact showed that treatment with Achyranthis Radix aqua-acupuncture adminstered to the Exp. II group was more effective than the Exp. I group in the treatment of arthritis(P<0.05). 3) The content of the total protein in the blood serum was $6.14{\pm}43g/dl$ in the normal group, $7.95{\pm}0.94g/dl$ in the Exp. I group, and $6.41{\pm}0.68g/dl$ in the Exp. II group. There was no significance in total protein between the Exp. II group and the Exp. I group from the statistical analysis. 4) The content of albumin in the blood serum was $2.94{\pm}0.l3g/dl$ in the normal group, $2.01{\pm}0.48g/dl$ in the Exp. I group and $3.15{\pm}0.27g/dl$ in the Exp. II group, which revealed a significant increase in the serum albumin from the statistical analysis of the Exp. II group compared with the Exp. I group. 5) The content of the globulin in the blood serum was $3.19{\pm}$0.48g/dl in the normal group, $4.70{\pm}1.26g/dl$ in the Exp. I group and $3.26{\pm}0.57g/dl$ in the Exp. II group. There was no significance in the serum globulin between the Exp. II group and Exp. I group from the statistical analysis. 6) In histological findings, because of severe inflammatory reaction, remarkably irregular tissue and large amount of inflammatory cells were found in the Exp. I group. But the Exp. II group showed a small amount of inflammatory cells, the refrained inflammatory state and even recovering state. Conclusions : These results show that Achyranthis Radix aqua-acupuncture reduces the inflammatory reaction and muscular tissue necrosis in SD rat's paws induced by Freund's complete adjuvant.
