Comparative Studies on Concentration of Decursinol in Plasma after Oral Administration of Angelicae Gigantis Radix Extract and Combined Use of Decursin and Cnidii Rhizoma Extract or Bupleuri Radix Extract in Rats (당귀와 천궁 및 시호의 병용 경구 투여에서 흰쥐 혈중 Decursinol 농도의 비교 검토)

  • This study was performed to elucidate the influence on the plasma decursinol concentration after administration of combined decursin and Cnidii Rhizoma or decurisin and Bupleuri Radix extracts in comparison with single decursin administration. The identification of decursinol isolated from Angelica gigas was carried out by GC/MS and the concentration of decursinol in the plasma after oral administration was determined by HPLC. The value of area under the plasma concentration (AUC) and the maximal concentration (Cmax) of decursinol after administration of methanol and ether Angelicae Gigantis Radix extract was higher than those of single decursin-treated group. It was also found that the AUC and the Cmax of decursinol after combined administration with decursin and Cnidii Rhizoma were higher than those of decursin administration, whereas those of combined with decursin and Bupleuri Radix was lower. Moreover, the studies were performed with decursinol only or with combined decursinol and Cnidii Rhizoma/Bupleuri Radix, both of combined treatment increased the plasma decursinol concentrations compared with decursinol-treated group in rats. On the basis of these results, it is concluded that co-existing substances or co-administration with other drugs may influence the plasma levels of decurisinol after oral administration.

Effect of Ethanol on the Immunotoxicity of Ampicillin in Mice (Ampicillin의 면역독성에 미치는 Ethanol의 영향)

  • Experiments were performed on mice to investigate the influences of ampicillin and ethanol on the immune response. Ampicillin was injected intraperitoneally and ethanol was administered in the drinking water. Mice were sensitized and challenged with sheep red blood cells. Immune responses were evaluated by humoral immunity, cellular immunity, peripheral circulating white blood cell and phagocyte activity. 1. The combined administration of ampicillin and ethanol as compared to ampicillin had not influence on the weight of spleen, but increased the weight of thymus. 2. Humoral immune response was slightly reduced by ampicillin. Especially, the combined administration of ampicillin and ethanol significantly reduced hemclysin production. 3. Cellural immune response was reduced by ampicillin. The combine administration of ampicillin and ethanol significantly reduced cellural immune response. 4. Peripheral circulating white blood cell was reduced by the combined administration of ampicillin and ethanol as compared to ampicillin. 5. The combined administration of ampicillin and ethanol as compared to ampicillin had not influence on the phagocyte activity.

  • Experiments were performed on mice to investigate the influences of doxycycine and ethanol on the immune responses. Doxycycline was injected intraperitoneally and ethanol was administered in the drinking water. Mice were sensitized and challenged with sheep red blood cells. Immune responses were evaluated by humoral immunity, cellular immunity, peripheral circulating white blood cell and phagocyte activity. Pathotoxicological influences were measured by serum protein and albumin. The weight of spleen, thymus and liver were measured. Doxycline and ethanol combined administration decreased the weight of thymus and spleen. Humoral and cellular immune response were reduced by doxycycline administration. Especially ethanol combined administration significantly reduced humoral and cellular immune response. Phagocyte activity was increased by ethanol combined administration and peripheral circulating white blood cell was significantly increased by ethanol adminstration. Ethanol combined administration decreased serum A/G ratio.

  • Cardiopulmonary depression of long-term constant rate infusion (CRI) administration of multiple analgesic drugs is important, especially in critically ill dogs. Therefore, this study was conducted to evaluate the effects of lidocaine, ketamine or combined lidocaine-ketamine combination CRI treatment on vital signs and left ventricular (LV) function in healthy dogs. Six adult Beagle dogs were administered either ketamine (initial loading dose of 0.5 mg/kg followed by $10{\mu}g/kg/min$ CRI), lidocaine (initial loading dose of 2 mg/kg followed by 0.025 mg/kg/min CRI), or combined lidocaine-ketamine intravenously. Arterial blood pressure (BP), heart rate (HR), respiratory rate (RR), body temperature (BT) and echocardiographic LV dimensions were measured before administration of medications, immediately after administration of drugs, and then every 10 min for 2 h. There were no significant changes in HR, RR, BT and BP after the administration of either lidocaine CRI, ketamine CRI, or combined lidocaine and ketamine CRI. There were also no significant changes in LV dimensions and stroke volume. The results revealed that treatment with either lidocaine, ketamine or combined lidocaine-ketamine may not cause cardiopulmonary suppression in healthy dogs.

Effect of DHEA Administration Alone or Exercise combined with DHEA before Steroid Treatment on Rat Hindlimb Muscles (스테로이드 치료 전 DHEA 단독투여와 DHEA 투여와 운동의 동시적용이 스테로이드에 의해 유발되는 쥐 뒷다리근의 위축 예방에 미치는 효과)

  • Purpose: The purpose of this study was to determine the effect of Dehydroepiandrosterone(DHEA) administration alone or exercise combined with DHEA before steroid treatment on rat hindlimb muscles. Methods: Male Sprague-Dawley rats were assigned to one of three groups: a steroid group(S, n=10) that had no treatment for 7 days before steroid treatment; a DHEA-steroid group(DS, n=8) that had 0.34 mmol/kg/day DHEA injection once a day for 7 days before steroid treatment and an exercise+DHEA-steroid group(EDS, n=9) that ran on the treadmill combined with 0.34 mmol/kg/day DHEA injection for 7 days before steroid treatment. At 15 days all rats were anesthetized and soleus, plantaris and gastrocnemius muscles were dissected. Body weight, food intake, muscle weight, myofibillar protein content and cross-sectional area of the dissected muscles were determined. Results: The DS group showed significant increases(p<.05) as compared to the steroid group in body weight, and muscle weight of gastrocnemius muscles. The EDS group showed significant increases(p<.05) as compared to the S group in body weight, muscle weight, myofibrillar protein content, and Type II fiber cross-sectional area of soleus, plantaris and gastrocnemius muscles. Conclusion: Exercise combined with DHEA administration before steroid treatment prevents steroid induced muscle atrophy, with exercise combined with DHEA administration being more effective than DHEA administration alone in preventing muscle atrophy.

Soft Set Theory Oriented Forecast Combination Method for Business Failure Prediction

  • This paper presents a new combined forecasting method that is guided by the soft set theory (CFBSS) to predict business failures with different sample sizes. The proposed method combines both qualitative analysis and quantitative analysis to improve forecasting performance. We considered an expert system (ES), logistic regression (LR), and support vector machine (SVM) as forecasting components whose weights are determined by the receiver operating characteristic (ROC) curve. The proposed procedure was applied to real data sets from Chinese listed firms. For performance comparison, single ES, LR, and SVM methods, the combined forecasting method based on equal weights (CFBEWs), the combined forecasting method based on neural networks (CFBNNs), and the combined forecasting method based on rough sets and the D-S theory (CFBRSDS) were also included in the empirical experiment. CFBSS obtains the highest forecasting accuracy and the second-best forecasting stability. The empirical results demonstrate the superior forecasting performance of our method in terms of accuracy and stability.

  • The extract of Mawhang-tang showed considerable inhibition of 70.7 and 57.2% at oral doses of 200 and 400mg/kg, respectively, on carrageenin edema in rat paw. In case of combined oral administration of the extract at each dose of 100, 200 and 400mg/kg with 100mg/kg of aspirin, the inhibition percentages were 78.2, 87.2 and 72.5%, respectively. The combined oral administration of 200mg/kg of the extract with 200mg/kg of aspirin exhibited 87.6% inhibition of the edema. On the writhing syndrome induced by 0.7% acetic acid solution, the oral administration of 200mg/kg of the extract with 200mg/kg of aspirin showed remarkable inhibition of 90.3%. In the inhibitory effect of the leakage of dye into peritoneal cavity were shown to be 51.9, 56.6 and 58.1% at the combined administration of the extract at the doses of 100, 200 and 400mg/kg with 100mg/kg of aspirin, respectively.

The Effect of Panax Ginseng Extract on the Immunotoxicity of Cimetidine in Mice (마우스에 있어서 Cimetidine의 면역독성에 미치는 인삼유출물의 영향)

  • The immunopotenciating effects of petroleum ether extract, ethanol extract and butanol fraction of panax ginseng on the immunotoxicity of Cimetidine were investigated in ICR mice. Immune responses were evaluated by antibody production, Arthus reaction, delayed type hypersensitivity (DTH), and rosette forming cell (RFC) in mice, sensitized and challenged with sheep red blood cells. To investigate the change of the non-specific immune responses, phagocyte activity and number of leukocytes in peripheral blood were measured also. The results of this study are summarized as followings; 1. Cimetidine treated group as compared with normal group generally decreased HA, 2-MER, RFC, number of circulating leukocytes and phagocyte activity whereas in-creased Arthus reaction and DTH. 2. The panax ginseng petroleum ether extract combined administration group as compared with the control group remarkably increased HA, 2-MER, number of circulating leukocytes and phagocyte activity. 3. The panax ginseng ethanol extract combined administration group as compared with the control group remarkably increased Arthus reaction, DTH, HA, RFC, number of circulating leukocytes and phagocyte activity. 4. The panax ginseng butanol fraction combined administration group as compared with the control group remarkably increased Arthus reaction, HA, 2-MER, RFC, number of circulating leukocytes and phagocyte activity.

Benorylate Interaction with Ethoxybenzamide and Lorazepam (Benorylate와 Ethoxybenzamide 밍 Lorazepam 과의 상호작용)

  • Benorylate and ethoxybenzamide have been used alone or in combination as an analgesic, antipyretic and antiinflammatory agent. We investigated the significance of the differences of analgesic activities between single and concurrent administration of benorylate and ethoxybenzamide and lorazepam in mice and also antipyretic activity between single and concurrent administration of benorylate and ethoxybenzamide in rats. 1). Concurrent administration of each half dose of benorylate and ethoxybenamide showed much inhibiting effect on the acetic acid-induced writhing syndrome of mice than the above drug alone, and the some increased analgesic response by hot plate method. 2). The synergistic and analgesic effect of combined administration of benorylate and lorazepam was found to be significant. 3). Antipyretic effect of half-dose combined administration of benorylate and ethoxybenzamide on the rat pyrexia induced by yeast(s.c.) and T.T.G. (i.v.) was shown to be similar to the effect of each drug.

Synergistic antibacterial effect of disinfectants and microbubble water to Salmonella Typhimurium

  • Salmonella is a pathogenic bacterium that has long been important industrially because it has a wide host range and can be transmitted to humans through direct contact as well as indirect contact such as food contaminated with animal waste. Understanding how to reduce Salmonella contamination in pig farms is important for public health and the livestock industry from an economic perspective. In the swine industry, high concentrations of disinfectants have been applied because it is difficult to effectively control Salmonella in environments contaminated with organic substances. In order to evaluate the synergetic effect of disinfectants, the efficacy of two commercial disinfectants diluted in hard water and microbubble water (MBW) were compared under the laboratory condition. Different concentrations of both disinfectants combined with 1% detergent diluted in the two diluents were evaluated for their antibacterial effect. In the case of monopersulfate-based disinfectant groups, the growth of Salmonella was not observed at 1:200 dilution with both the hard water and MBW combined with 1% detergent. In the case of citric acid-based disinfectant, the bacterial growth was not observed at 1:800 dilution with MBW combined with 1% detergent. Our results show that the use of MBW as a diluent might improve the biological activities of acid-based disinfectant.
