인공 항원제시세포의 표면 분자의 특성 및 상호작용이 T 세포 활성화에 미치는 영향 (The effects of properties and interactions of surface molecules in antigen presenting cells on T cell activation)

  • 인체 적응 면역 반응을 일으키는데 중요한 항원 특이적 T 세포를 활용한 면역 세포 치료에서 T 세포를 체외에서 배양하고 클론 확장시키는 과정은 매우 섬세하고 복잡하여 조절하기가 쉽지 않아 T 세포의 활성화와 클론 확장을 유도하면서도 조절 및 취급이 용이한 인공 항원제시세포 개발의 필요성이 대두되고 있다. 인공 항원제시세포는 인체의 항원제시세포의 세포 표면 분자와 작용을 모방하게 되는데, 기본적인 신호 분자인 MHC-항원 복합체, 공동 자극 분자, 그리고 용해성 면역 조절 분자를 필수적으로 발현하여야 한다. 또한 T 세포가 항원과 접촉할 때, 이들 분자들이 잘 조직화되어 작용하는 것이 효과적인 T 세포 활성화에 중요하다. 본 논문에서는 여러 인공 항원제시세포 제작 방법과 세포 표면 분자들의 결합 방법과 물리적인 특성이 T 세포와의 상호작용에 중요함을 고찰하였으며, 효과적인 T 세포 활성화를 유도하며 면역세포치료에 적용 가능한 인공항원제세세포의 제작 방법을 살펴보았다.

Nanoliposomes of L-lysine-conjugated poly(aspartic acid) Increase the Generation and Function of Bone Marrowderived Dendritic Cells

  • Background: Biodegradable polymers have increasingly been recognized for various biological applications in recent years. Here we examined the immunostimulatory activities of the novel poly(aspartic acid) conjugated with L-lysine (PLA). Methods: PLA was synthesized by conjugating L-lysine to aspartic acid polymer. PLA-nanoliposomes (PLA-NLs) were prepared from PLA using a microfluidizer. The immunostimulatory activities of PLA-NLs were examined in mouse bone marrow-derived dendritic cells (BM-DCs). Results: PLA-NLs increased the number of BM-DCs when added to cultures of GM-CSF-induced DC generation on day 4 after the initiation of cultures. Examination of the phenotypic properties showed that BM-DCs generated in the presence of PLA-NLs are more mature in terms of the expression of MHC class II molecules and major co-stimulatory molecules than BM-DCs generated in the absence of PLA-NLs. In addition, the BM-DCs exhibited enhanced capability to produce cytokines, such as IL-6, IL-12, TNF-${\alpha}$ and IL-$1{\beta}$. Allogeneic mixed lymphocyte reactions also confirmed that the BMDCs were more stimulatory on allogeneic T cells. PLA- NL also induced further growth of immature BM-DCs that were harvested on day 8. Conclusion: These results show that PLA-NLs induce the generation and functional activities of BM-DCs, and suggest that PLA-NLs could be immunostimulating agents that target DCs.

생약복합조성물(HemoHIM)의 수지상세포 활성화 효과 (Effects of a Herbal Composition (HemoHIM) on the Activation of Dendritic Cells)

  • 특이적 면역반응을 유도하는 전문 항원제시세포인 수지상세포의 분화와 항원제시기능에 미치는 HemoHIM의 면역기능 증강 효과에 대해 알아보았다. HemoHIM은 수지상세포의 분화/증식 과정에 첨가하였을 경우에 공동자극 분자인 CD40, CD86 분자의 세포 표면 발현량을 증가시키는 것으로 나타났다. 이 수지상세포는 동종항원 반응 T세포를 활성화시켜 IL-2, $IFN-\gamma$ 분비량과 증식반응을 유의하게 증가시켰다. 그리고 HemoHIM을 처리한 수지상세포는 대조군에 비해 OVA항원을 T세포주에 충분히 전달하여 IL-2와 $IFN-\gamma$의 분비량을 유의하게 증가시켰을 뿐만 아니라 T세포주의 증식반응도 유의하게 증가시켰다. 따라서 HemoHIM은 생체 내의 특이적 면역반응을 유도하는 수지상세포의 기능을 강화하는 면역기능 조절제로서 사용이 가능할 것으로 생각된다.

  • Dendritic cells (DCs) are known to professional antigen presenting cell (APC). Due to the role as an effective activator of cytotoxic T Lymphocytes by expressing MHC, adhesion and co-stimulatory molecules, DCs are now widely recognized to play an important role in the immune responses to tumors.We investigated the effect of cultured DCs in murine melanoma pulmonary metastasis model. To follow the metastasis protocol, syngenic melanoma cells were inoculated intra-venously into the mouse (B16F10 into the C57BL/6)8 days prior to the first DC injection (1$\times$106 DCs/ mouse, i.p.) and the autologous tumor cell lysate pulsed-DCs were injected as a therapeutic module twice in two weeks. 일부 한약재의 수지상세포 활성화 효과 (Effect of Some Herbal Plant Extracts on the Activation of Dendritic Cells)

  • 본 실험에서는 일부 한약재를 이용하여 수지상세포의 활성화에 미치는 영향을 알아보았다. 그 결과, 실험에 사용한 여섯 가지 한약재 중 선별된 천궁과 당귀는 수지상세포의 항원 제시 능력에 영향을 미쳐, T 세포 증식 반응을 증가시켰고, IL-2와 IFN-r의 분비를 증가시키는 것으로 나타났다. 또한 이러한 T 세포의 활성은 수지상세포의 세포표면 단백질인 MHC classII와 CD86, 그리고 CD11c의 발현 증가에 의한 것임을 확인 할 수 있었다. 이상의 실험 결과, 본 실험에서 선별된 천궁과 당귀는 수지상세포를 활성화시키는 효과가 있는 것으로 생각된다.

The Stimulatory Effect of Essential Fatty Acids on Glucose Uptake Involves Both Akt and AMPK Activation in C2C12 Skeletal Muscle Cells

  • Essential fatty acid (EFA) is known to be required for the body to function normally and healthily. However, the effect of EFA on glucose uptake in skeletal muscle has not yet been fully investigated. In this study, we examined the effect of two EFAs, linoleic acid (LA) and ${\alpha}$-linolenic acid (ALA), on glucose uptake of C2C12 skeletal muscle cells and investigated the mechanism underlying the stimulatory effect of polyunsaturated EFAs in comparison with monounsaturated oleic acid (OA). In palmitic acid (PA)-induced insulin resistant cells, the co-treatment of EFAs and OA with PA almost restored the PA-induced decrease in the basal and insulin-stimulated 2-NBDG (fluorescent D-glucose analogue) uptake, respectively. Two EFAs and OA significantly protected PA-induced suppression of insulin signaling, respectively, which was confirmed by the increased levels of Akt phosphorylation and serine/threonine kinases ($PKC{\theta}$ and JNK) dephosphorylation in the western blot analysis. In PA-untreated, control cells, the treatment of $500{\mu}M$ EFA significantly stimulated 2-NBDG uptake, whereas OA did not. Phosphorylation of AMP-activated protein kinase (AMPK) and one of its downstream molecules, acetyl-CoA carboxylase (ACC) was markedly induced by EFA, but not OA. In addition, EFA-stimulated 2-NBDG uptake was significantly inhibited by the pre-treatment of a specific AMPK inhibitor, adenine 9-${\beta}$-D-arabinofuranoside (araA). These data suggest that the restoration of suppressed insulin signaling at PA-induced insulin resistant condition and AMPK activation are involved at least in the stimulatory effect of EFA on glucose uptake in C2C12 skeletal muscle cells.

Cytokine Reporter Mouse System for Screening Novel IL12/23 p40-inducing Compounds

  • Cytokines interleukin (IL) 12 and 23 play critical roles in linking innate and adaptive immune responses. They are members of heterodimeric cytokines, sharing a subunit p40. Although IL12/23 p40 is mainly induced in macrophages and dendritic cells (DCs) after stimulation with microbial Toll-like receptor ligands, methods to monitor the cells that produce IL12/23 p40 in vivo are limited. Recently, the mouse model to track p40-expressing cells with fluorescent reporter, yellow fluorescent protein, has been developed. Macrophages and DCs from these mice faithfully reported p40 induction using the fluorescent marker. Here we took advantage of these reporter mice to screen bio-compounds for p40-inducing activity. After screening hundreds of compounds, we found several extracts inducing IL12/23 p40 gene expression. Treatment of DCs with these extracts induced the expression of MHC class II and co-stimulatory molecules, which implies that these might be useful as adjuvants. Next, the in vivo target immune cells of candidate compounds were examined. The reporter system can be useful to identify cells producing IL12 or IL23 in vivo as well as in vitro. Thus, our cytokine reporter system proved to be a valuable reagent for screening for immunostimulatory molecules and identification of target cells in vivo.

천식에서 기도평활근의 증식과 합성 반응에 대한 최신지견 (Proliferative and Synthetic Responses of Airway Smooth Muscle in Asthma)

  • New evidence is emerging that airway smooth muscle(ASM) may act as an immunomodulatory cell by providing pro-inflammatory cytokines and chemokines, polypeptide growth factors, extracellular matrix proteins, cell adhesion receptors and co-stimulatory molecules. ASM can promote the formation of the interstitial extracellular matrix, and potentially contribute to the alterations within the extracellular matrix in asthma. In addition, extracellular matrix components can alter the proliferative, survival, and cytoskeletal synthetic function of ASM cells through integrin-directed signaling. Increased ASM mass is one of the most important features of the airway wall remodeling process in asthma. Three different mechanisms may contribute to the increased ASM mass : cell proliferation, increased migration and decreased rate of apoptosis. The major signaling pathways of cell proliferation activated by ASM mitogens are those dependent on extracellular signal-regulated kinase and phosphoinositide 3'-kinase. The key signaling mechanisms of cell migration have been identified as the p38 mitogen-activated protein kinase and the p21-activated kinase 1 pathways. ASM cells contain ${\beta}2$-adrenergic receptors and glucocorticoid receptors. They may represent a key target for ${\beta}2$-adrenergic receptor agonist/corticosteroid interactions which have antiproliferative activity against a broad spectrum of mitogens.

Activation of Macrophages by Exopolysaccharide Produced by MK1 Bacterial Strain Isolated from Neungee Mushroom, Sarcodon aspratus

  • Background: The MK1 strain, a novel bacterial isolate from soft-rotten tissue of the Neungee mushroom, produces copious amounts of exopolysaccharide (EPS) in a dextrose minimal medium. This study examined the molecular characteristics and immunomodulatory activity of MK1 EPS. Methods: The EPS in the culture supernatant was purified by cold ethanol precipitation, and characterized by SDS- PAGE/silver staining and Bio-HPLC. The immunomodulatory activities of the EPS were examined using the mouse monocytic cell line, RAW 264.7 cells. Results: The molecular weights of the purified EPS were rather heterogeneous, ranging from 10.6 to 55 kDa. The EPS was composed of glucose, rhamnose, mannose, galactose, and glucosamine at an approximate molar ratio of 1.00 : 0.8 : 0.71 : 0.29 : 0.21. EPS activated the RAW cells to produce cytokines, such as TNF-${\alpha}$ and IL-$1{\beta}$, and nitric oxide (NO). EPS also induced the expression of co-stimulatory molecules, such as B7-1, B7-2 and ICAM-1, and increased the phagocytic activity. The macrophage-activating activity of EPS was not due to endotoxin contamination because the treatment of EPS with polymyin B did not reduce the macrophage-activating activity. Conclusion: The EPS produced from the MK1 strain exerts macrophage-activating activity.

Identification of Immunostimulatory Oligodeoxynucleotide from Escherichia coli Genomic DNA

  • Bacterial DNA containing immunostimulatory CpG motifs can stimulate antigen-presenting cells to express co-stimulatory molecules and to produce various cytokines in vivo and in vitro. In this study, we fragmented macromolecular E.coli genomic DNA with DNase I, and analyzed the ability of the resulting DNA fragments to induce the NF-${\kappa}B$ activation and humoral immune response. Furthermore, using computational analysis and luciferase assay for synthetic ODNs based on the sequence of the immunostimulatory DNA fragments (DF-ODNs), an active component of DF-ODNs sequences was investigated. Experimental results demonstrated that DF-ODN is optimal for the NF-${\kappa}B$-responsive promoter activation in the mouse macrophage cell line and the humoral immune response in vivo. In agreement with the activity of the DF-ODNs processed by DNase I, a synthetic ODN based on the DF-ODN sequences is potent at inducing IL-12 mRNA expression in primary dendritic cells. These results suggest that the discovery and characterization of a highly active natural CpG-ODN may be achieved by the analyses of bacterial DNA fragments generated by a nuclease activity.
