As for Services of the Public Libraries toward the Youth (공공도서관의 청소년들에 대한 봉사에 대하여 - 학교도서관과의 협조적 측면에서)

  • The following results have been driven out from a survey of the actual conditions of the public libraries service in Korea, made through questionnaire in reference with the actual service for youth that are presently done by the foreign countries' libraries, in order to prepare a perfect and objective base in establishing a service activities of the public libraries in this country: (1) It was found that the public libraries' loarning hours during the week days which accords with the schooling hours of the schools are shown to 50 %. On Saturdays during Summer season seven libraries and during Winter season twelve libraries would close their loaning before 17 : 00 hours, and on Holidays and Sundays fifteen libraries are not doing their loarning. (2) The followings are the present status of the service activities for the youth by the public libraries: a. It was found that they do introduction activity (50 %) of newly published books as a service for inducing reading interestings of the youth, and except this it was found that others are on beginning. b. The service activities by the data for the visual and auditory sense are almost negelected. and the rate of doing such activity is shown to 9.74%. c. It has been found that little information service activity is for inducing the youth to the libraries. d. As a cooperative activity with the school's, a passive service activity to have pursuaded to visit the public libraries by each class, are mostly being done (27.27%). Some research meeting for solving all the matters between the school libraries are not held mostly.

Molecular Cloning of a cDNA Encoding a Cathepsin B Homologue from the Mulberry Longicorn Beetle, Apriona germari

  • A cDNA encoding a putative member of cathepsin B of the thiol pretense superfamily was cloned from a cDNA library of the mulberry longicorn beetle, Apriona germari. Sequence analysis of the cDNA encoding the cathepsin B of A. germari (AgCatB) revealed that the 972 bp cDNA has an open reading frame of 324 amino acid residues. The deduced protein sequence of the AgCatB showed high homology with cathepsin B of the insects, Bombyx mori (47.3% amino acid identity), Helicoverpa armigera (46.6%) and Sarcophaga peregrina (45.6%), and the lowest homology with Aedes aegypti (33.2%). The AgCatB contains six disulfate bonds typical for cysteine pretenses. The three amino acid positions Cys-109, His-267, and Asn-287 which are conserved, active sites characteristic for cathepsin B, were also found. Phylogenetic analysis further confirmed that the AgCatB has a close relationship with that of B. mori, H. armigera and S. peregrina.

Gene Identification and Molecular Characterization of Solvent Stable Protease from A Moderately Haloalkaliphilic Bacterium, Geomicrobium sp. EMB2

  • Cloning and characterization of the gene encoding a solvent-tolerant protease from the haloalkaliphilic bacterium Geomicrobium sp. EMB2 are described. Primers designed based on the N-terminal amino acid sequence of the purified EMB2 protease helped in the amplification of a 1,505-bp open reading frame that had a coding potential of a 42.7-kDa polypeptide. The deduced EMB2 protein contained a 35.4-kDa mature protein of 311 residues, with a high proportion of acidic amino acid residues. Phylogenetic analysis placed the EMB2 gene close to a known serine protease from Bacillus clausii KSM-K16. Primary sequence analysis indicated a hydrophobic inclination of the protein; and the 3D structure modeling elucidated a relatively higher percentage of small (glycine, alanine, and valine) and borderline (serine and threonine) hydrophobic residues on its surface. The structure analysis also highlighted enrichment of acidic residues at the cost of basic residues. The study indicated that solvent and salt stabilities in Geomicrobium sp. protease may be accorded to different structural features; that is, the presence of a number of small hydrophobic amino acid residues on the surface and a higher content of acidic amino acid residues, respectively.

cDNA Cloning, Expression and Homology Modeling of a Luciferase from the Firefly Lampyroidea maculata

  • The cDNA of a firefly luciferase from lantern mRNA of Lampyroidea maculata has been cloned, sequenced and functionally expressed. The cDNA has an open reading frame of 1647 bp and codes for a 548-residue-long polypeptide. Noteworthy, sequence comparison as well as homology modeling showed the highest degree of similarity with H. unmunsana and L. mingrelica luciferases, suggesting a close phylogenetic relationship despite the geographical distance separation. The deduced amino acid sequence of the luciferase gene of firefly L. maculata showed 93% identity to H. unmunsana. Superposition of the three-dimensional model of L. maculata luciferase (generated by homology modeling) and three dimensional structure of Photinus pyralis luciferase revealed that the spatial arrangements of Luciferin and ATP-binding residues are very similar. Putative signature of AMP-binding domain among the various firefly species and Lampyroidea maculata was compared and a striking similarity was found. Different motifs and sites have been identified in Lampyroidea maculata by sequence analysis. Expression and purification of luciferase from Lampyroidea maculata was carried out using Ni-NTA Sepharose. Bioluminescence emission spectrum was similar to Photinus pyralis luciferase.

Analysis for nucleotide sequence of the small membrane (sM) protein gene of porcine epidemic diarrhea virus Chinju99 isolated in Korea

  • To provide information for the molecular pathogenesis and antigenic structures of Korean isolates of porcine epidemic diarrhea virus (PEDV), the small membrane (sM) protein gene of Chinju99 strain, which was previously isolated from piglets suffering from severe diarrhea was characterized and further analyzed with other PEDV strains. The sM gene of Chinju99 generated by reverse transcription and polymerase chain reaction had a single open reading frame with 231 bases consisting of 24.2% adenine, 18.6% cytosine, 18.1% guanine and 39.0% thymine nucleotides. Nucleotide sequence of the gene revealed 97.8% homology to those of Belgian strain CV777 and British strain Br1/87, and 97.0% to Chinese strain LZC. The gene encoded a protein with 76 amino acids, and putative amino acid sequence of the gene revealed 98.7% homology to those of CV777 and Br1/87, and 96.1% to LZC. The amino acids of Chinju99 sM gene consisted of mostly hydrophobic residues, and there were one potential N-myristylation site and one potential threonine (T)-linked phosphorylation site recognized. Also, there was a transmembrane region with 46 amino acids, and Chinju99 was more close to CV777 and Br1/87 than to LZC in phylogenetic analysis on the sM amino acid sequences.

  • Rhodopsin, a dim-light receptor, is a model system for the study of G protein-coupled receptors that transduce extracellular signals into cells. To study the molecular mechanisms of visual systems in fish, the rod opsin gene of olive flounder Paralichthys olivaceus was characterized. The full-length P. olivaceus opsin gene was obtained by PCR amplification of genomic DNA, as well as cDNA synthesis. A comparison of clones obtained from both methods indicated that the olive flounder rod opsin gene lacks introns. Sequence analysis of the opsin gene indicated that it contains a 1,056-bp open reading frame encoding 352 amino acids. The deduced amino acid sequence contains features of typical rod opsins, such as sites for Schiff's base formation (K296) and its counterion (E113), disulfide formation (C110 and C187), and palmitoylation (C322 and C323). An opsin sequence alignment showed the highest similarity between P. olivaceus and Solea solea (95.1%), followed by Hippoglossus hippoglossus (94.5%). An opsin phylogenetic tree revealed a close relationship between olive flounder and teleost rod opsins.

Genomic Organization of Penicillium chrysogenum chs4, a Class III Chitin Synthase Gene

  • Class III chitin synthases in filamentous fungi are important for hyphal growth and differentiation of several filamentous fungi. A genomic clone containing the full gene encoding Chs4, a class III chitin synthase in Penicillium chrysogenum, was cloned by PCR screening and colony hybridization from the genomic library. Nucleotide sequence analysis and transcript mapping of chs4 revealed an open reading frame (ORF) that consisted of 5 exons and 4 introns and encoded a putative protein of 915 amino acids. Nucleotide sequence analysis of the 5'flanking region of the ORF revealed a potential TATA box and several binding sites for transcription activators. The putative transcription initiation site at -716 position was identified by primer extension and the expression of the chs4 during the vegetative growth was confirmed by Northern blot analysis. Amino acid sequence analysis of the Chs4 revealed at least 5 transmembrane helices and several sites for past-transnational modifications. Comparison of the amino acid sequence of Chs4 with those of other fungi showed a close relationship between P chrysogenum and genus Aspergillus.

  • This essay exams the issues of aesthetic elements that come up when Korean novels get translated into Chinese language. The short story collection titled "An old well" written by Jeong Heui Oh in both languages are compared and analyzed by focusing on the mode of narrations. There are various narrative modes in "An old well". Each narrative mode properly functions for aesthetic effects and drawing meanings. In short, we can find a way to grasp its leitmotif the writer wants to indicate only when we carefully interpret the narrative modes in the original text. However, the narrative modes in Korean text have been simplified by changing its modes into direct narrative in Chinese-translated text. Thus the aesthetic effects in the original text have been spoiled and the Chinese text fails to deliver its meaning involved in the original narrative mode. Translation of novel invites consideration on both of its form and content on account of the text's uniqueness. Accordingly, a close examination and study of the original text should be completed beforehand.

John of Plano Carpini, Papal Diplomat and Spy along the Silk Road

  • In March 1245, Pope Innocent IV authorized three missions to the Mongols, seeking information about this menace from the East and summoning Eastern Christian support against an anticipated Mongol onslaught. Only one of the missions, led by John of Plano Carpini, reached Mongolia-the first-known Western European party to reach East Asia by a land route. Traveling along the Silk Road's new "Grasslands Route," John and his companion Benedict reached the camp of Güyüg Khan, where they witnessed his installation as the Great Khan. Upon their return to the papal court in 1247, they delivered Güyüg's letter demanding the submission of the pope and all the West's princes. John also presented a detailed report on what he and Benedict had learned. A close reading of it reveals a master intelligence operative at work. In addition to presenting an overview of Mongol history and culture, Friar John's report provides detailed information on the Mongols' grand strategy, their military organization and armaments, and their battle tactics. Turning from intelligence gathering to military operations, he offered practical advice on how to meet and defeat the coming Mongol onslaught, an attack that, providentially for the West, never came. What did occur was a modest but significant migration of Western missionaries and merchants to East Asia in the century following this pioneering journey.

Effects of CSR Motives on Authenticity and Attitude in the Food and Beverage Franchise Sectors (식음료 프랜차이즈 기업의 CSR 활동 동기에 대한 지각이 진정성 및 태도에 미치는 영향)

  • Purpose: Previous studies show that perceived CSR motives have a significant impact on company evaluations. However, consumer responses to CSR motives vary depending on CSR motives. From this perspective, this study investigates the impact of CSR motives on consumers' responses in the context of food and beverage franchise companies using a scenario. Research design, data, and methodology: For achieving the purposes of the study, an example of a domestic food and beverage franchise company actively carrying out CSR activities was presented. Data was collected from 304 respondents aged 20 or older who were aware of CSR activities. The respondents answered the questionnaire after reading the scenario. The data was analyzed with SPSS 28.0 and SmartPLS 4.0 program. Result: Values-driven motive and strategic motive influence authenticity, while stakeholder-driven motive and egoistic motive did not influence authenticity. Values-driven motive influences on attitude, while stakeholder-driven motive, strategic motive and egoistic motive didn't. Lastly, authenticity influences attitude. Conclusions: Companies need to be aware that consumers may infer different motives for their CSR activities, and pay close attention to consumers' perceived motives from the planning stage of CSR activities. In particular, companies should focus on the values-driven motive and the strategic motive when planning CSR activities.
