Antioxidant Activities of Extracts from Medicinal Plants (약용식물 추출물의 항산화 활성)

  • In this study, the optimal conditions for phenolic-compound extraction from medicinal plants were found to be 24 h and about 50% ethanol. The electron-donating scavenging activities (DPPH), ABTS radical-cation decolorization (ABTS), antioxidant protection factor (PF), and thiobarbituric acid reaction substance (TBAR) were measured to determine the antioxidant activities of the extracts of Sanguisorba officinalis Linn., Citrus unshiu Markovich, Melia azedarach L., Asparagus cochinchinensis Merr., Citrus unshiu S., Polygonum aviculare L., and Leonurus sibiricus L. The total phenolic contents of the extracts of medical plants were determined to be 0.45-3.00 mg/g in the water extracts and 0.33-3.15 mg/g in the 50% ethanol extracts. The electron-donating abilities (EDA) of the water and ethanol extracts were both above 85% at the $50{\mu}g/ml$ concentration. The ABTS radical-cation decolorization was above 80% at the $100{\mu}g/ml$ concentration in all the extracts of various medicinal plants. The antioxidant protection factor (PF) of the Melia azedarach L. extracts was found to be $1.65{\pm}0.40$ PF in the water extracts at the $100{\mu}g/ml$ concentration, and was higher than those of the other medicinal-plant extracts. The TBAR inhibition rates of all the medicinal-plant extracts, except Asparagus cochinchinensis Merr., were above 85% at the $100{\mu}g/ml$ concentration. These results confirmed that the various oriental medicinal plants (Sanguisorba officinalis Linn., Citrus unshiu Markovich, Melia azedarach L., Asparagus cochinchinensis Merr., Citrus unshiu S., Polygonum aviculare L., and Leonurus sibiricus L.) that were included in this study are useful functional-food sources.

Anti-Oxidant Effect and Anti-Inflammatory of Fermented Citrus unshiu Peel Extract by using Schizophyllum commune (치마버섯을 이용한 진피 발효 배양물의 항산화 및 항염 효과)

  • Citrus unshiu (C. unshiu) Markovich were dried peel of mandarin orange, of which fresh fruit was one of the famous foods in Korea and Eastern Asia. In the oriental medicine, C. unshiu peel was known to have a diuretic effect and to strengthen spleen function. Recently, natural flavonoids of C. unshiu peel have been investigated. In this study, C. unshiu peel extract containing flavonoid-glycosides was cultured with Schizophyllum commune (S. commune) mycelia producing ${\beta}$-glu- cosidase and its biological activities were investigated. ${\beta}$-glucosidase of S. commune mycelia converted the flavonoid-glycosides (rutin and hesperidin) into aglycones (naringenin and hesperetin). Fermented C. unshiu peel extract compounds were analyzed by HPLC system. The photoprotective potential of fermented C. unshiu peel extract was tested in human dermal fibroblasts (HDFs) exposed to UVA. Fermented C. unshiu peel extract extract also showed notable in vitro anti-inflammatory effect on cellular systems generating cyclooxygenase-2 (COX-2) and 5-lipoxygenase (5-LOX) metabolites. Also, UVB-induced production of interleukin-$1{\alpha}$ in human HaCaT cells was reduced in a dose-dependent manner by treatment with fermented C. unshiu peel extract. These results suggest that fermented C. unshiu peel extract may mitigate the effects of photoaging in skin by reducing UV-induced adverse skin reaction.

Inhibitory Effect of Citrus Unshiu Peel and Cimicifuga Dahurica Extracts on Melanogenesis accroding to the Various Extraction Methods (추출조건에 따른 진피(陳皮) 및 승마(升麻) 추출물의 미백 활성효과)

  • Objectives: Excessive melanin production leads to skin pigmentation, which causes various cosmetic and health problems. Citrus unshiu Markovich peel (CS) and Cimicifuga dahurica (CD) have long been widely used as a oriental medicinal plant because of their pharmacological properties including anti-ulcer, anti-oxidant, and anti-inflammatory properties. Methods: In this study, we investigated the inhibitory effects of CS, CD extracts or CS:CD=1:2 mixture on melanogenesis according to the various extraction methods. CS and CD extracted were prepared by ethanol extraction (EE), ultrasonification extraction (USE), Supercritical extraction (SCE), reflux extraction (RE), respectively. Results: DPPH radical scavenging and tyrosinase inhibitory activity of CD extracts or CS:CD=1:2 mixture were increased in dose-dependent manners. In addition, we evaluated the effect of CS, CD extracts or CS:CD=1:2 mixture on tyrosinase activity and melanogenesis in α-melanocyte stimulating hormone-induced B16-F10 melanoma cells. CS, CD extracts or CS:CD=1:2 mixture significantly inhibited tyrosinase activity and melanogenesis at 10-200 ㎍/mL. Conclusions: Therefore, our study suggests that CS and CD extracts have potential as a safe treatment for excessive pigmentation or as a natural ingredient in cosmetics.

  • Sinomegoura citricola (van der Goot, 1917) is recognized on Citrus unshiu Markovich (Rutaceae), Photinia glabra (Thunb.) Maxim (Rosaceae), and Cinnamomum lourelirii Nees (Lauraceae) from Jeju-do, Korea. Apterous viviparous female, alate viviparous female, and an intermediate morph between the apterous female and alate female are described. This is the first record of the genus Sinomegoura in Korea.

  • Objectives : The present study was undertaken to determine whether the ethanol extact of Citri Pericarpium (CP, Pericarp of Citrus unshiu Markovich, Rutaceae) is effective against atopic dermatitis according to it's storage period. Methods : To evaluate antiallergic effect of CP and OCP (Old Citri Pericarpium) evaluated in vivo their inhibitory effects against passive cutaenous anaphylaxis (PCA) reaction induced by IgE-antigen complex and scratching behaviors by compound 48/80. The Anti-atopic effects were measured by contact dermatitis, prurient animal model and PCA reaction. Contact dermatitis in mice as a model of the Type IV reaction caused by Oxazolone. Results : The results showed that anti-pruritus effects, analgesic effects of CP was depends on its hesperetin contents. And It also showed that keep longer in storage appeared to be higher in hesperetin contents. Both CP and OCP(Old CP) have a dose-dependent analgesic action in acetic acid induced writhing syndrome. OCP Potently inhibited PCA reaction in mice, although OCP weakly inhibited in long term contact dermatitis model in mice. Conclusions : These results suggest that the Proportional to the storage period, Citri Pericarpium possesses analgesic effects and anti-allergic effects.

  • Objectives: This analysis of prescription used for low back pain in the Yomun(腰門) chapter of ${\ll}$Donguibogam(東醫寶鑑); The Precious Mirror of Oriental Medicine${\gg}$ is designed to be helpful to practical use of clinics. Methods : Proscriptions used for low back pain in the Yomun(腰門) chapter of ${\ll}$Donguibogam(東醫寶鑑)${\gg}$ were classified and analyzed according to the frequency of proscriptions and the characteristics of each herbs in proscriptions(efficacy, used frequency, related organs etc.) Results and conclusions : After analysis, we obtained the following results : 1. The causes of low back pain are mainly eohyeol(瘀血), yangheo(陽虛), punghanseub(風寒濕). In care of low back pain, I suppose more efficiency that if Angelica gigas NAKAI(當歸) Cnidium officinale MAKINO(三芎) Prunus persica BATSCH(桃仁) is added when the cause is eohyeol(瘀血), or if Psoralea corylifolia L.(破古紙), Cinnamomum cassia PRESL(肉桂), Foeniculum vulare MILL(茴香), Eucommia ulmoides OLIV.(杜冲), Citrus unshiu MARKOVICH(陳皮) are added when the cause is yangheo(陽虛), or if Phellodendron amurense RUPR.(黃柏), Notopterygium incisum TING(羌活), Atractylodes Japonica KOIDZ.(蒼朮) are added when the cause is punghanseub(風寒濕).

A Review of Domestic Study Trends of Korean Medicine Treatment for Hyperemesis Gravidarum (임신오조의 한약 치료에 대한 국내 증례 보고 고찰)

  • Objectives: The purpose of this review is to anaylze the domestic study trends of Korean medicine treatment for hyperemesis gravidarum. Methods: We investigated the studies on Korean medicine treatment on hyperemesis gravidarum via searching four online databases up to October 2021. After searching studies, we analyzed selected studies. Results: 13 clinical studies were selected and all studies were case reports. The most common Korean Medicine used in hyperemesis gravidarum were Boseng-tang -gamibang, Gyullyeongboseng-tang, Taesanbanseok-san-gami. Rhizome of Atractylodes japonica Koidz., Pericarp of Citrus unshiu Markovich were the most commonly used herb except Root of G. uralensis Fisch.. Although the evaluation index was different for each study, most of the cases showed that hyperemesis gravidarum was improved. Conclusions: Korean medicine treatment is effective for hyperemesis gravidarum. Further large, well-designed studies are needed to establish the foundation of Korean medicine treatment for hyperemesis gravidarum.

Quilitative certificational plan of chenpi (진피(陳皮)의 품질인증(品質認證) 방안(方案))

  • Now many sustitution and false articles is used in korea instead of chenpi. To use chenpi correctly, we will make a quilitative certificational plan of chenpi to investigate all of lieraturea, records and documents. And we could reach conclusions as folloews. 1) Source In china source of Chen-pi is pericarp of citrus reticula Blanco(Family;rutaceae)and in korea source of Chen-pi is pericarp of citrus unshiu Markovich(Family;rutaceae). Though source of both countries are not same, it has no problems because of containing family plants and culture variants. 2) Harvesting and processing After the peel attained full growth, wash cleanly in water. And dry in shade or in low temporature at state of eliminating pericarp. 3) Quality (1) Functional standards Exocarp is soft and clear yellow with numerous oil sports. The less white mesocarp is the better. (2) Physicochemical standards We need to suggest new standards about hesperidin to various conditions by processing methods and storing time. The loss on drying is less than 13.5%. Ash content is less than 4.0%. A standard capacity of hesperidin is more than 4.0%. The content of heavy metal is less than 30 ppm. We can not detect reminding agricultural medicines.

Anti-mycoplasmal and anti-inflammatory effect of Origanum vulgare extract against Mycoplasma hyopneumoniae (Mycoplasma hyopneumoniae에 대한 Origanum vulgare 추출물의 항마이코플라즈마 및 항염증 효과)

  • In the present study, ten herbal extracts, Citrus unshiu Markovich, root and stem of Berberis koreana, Morus alba, Dendrobium moniliforme, Aster gramineus, A. scabar, Alisma canaliculatum, Fallopia japonica and Origanum (O.) vulgare were determined to examine anti-mycoplasmal activity. Among them, O. vulgare extract (OVE) showed strong anti-mycoplasmal activity and was analyzed by gaschromatography/ mass spectrometry (GC/MS). As the results, OVE was consisted of carvacrol (68.78%), o-cymene (9.80%), terpinene (7.61%) and thymol (4.03%) as main ingredients. To investigate inflammatory activity by intact pathogenic Mycoplasma hyopneumoniae (M. hyo) at 30 $\mu$g/ml, we examined induced transcription of proinflammatory cytokines such as cyclooxygenase-2, tumor necrosis factor-a, interleukin (IL)-1, IL-6 and inducible nitric oxide synthase in RAW 264.7 cells. With the above results, we further investigated whether OVE could reduce inflammation induced by M. hyo at minimal inhibitory concentration. The result showed that 32 $\mu$g/ml of OVE inhibited nitric oxide production by 60%. This study also evaluated the combination of OVE with antibacterials against M. hyo for application. Based on these results, it could be concluded that M. hyo induces inflammation in RAW 264.7 cells and OVE protects this inflammation, indicating that OVE may be useful for industrial animals.

Hepatocyte protection and antioxidant effect of Citri Unshius Pericarpium against cadmium-induced oxidative stress (카드뮴으로 유발된 산화 스트레스에 대한 진피의 간세포 보호 및 항산화 효과)

  • Objective : Citri Unshius Pericarpium is the dried peel of mature fruit of Citrus unshiu Markovich and has been used to treat indigestion, vomiting, and removal of phlegm. This study investigated the hepatoprotective and antioxidant effect of CEE (Ethanol extract of Citri Unshius Pericarpium) in cadmium (CdCl2)-treated HepG2 cells. Methods : Component analysis of Citri Unshius Pericarpium was analyzed by UPLC with C18 column. Cell viability was determined by MTT assay. The enzyme activity of superoxide dismutase (SOD) and the level of reactive oxygen species (ROS) and reduced glutathione (GSH) were analyzed using commercially available kits. Results : Cadmium caused severe HepG2 cell death. Cadmium also increased ROS production, consistent with depletion of GSH and inhibition of the SOD enzyme. However, CEE treatment reduced cell death and relieved oxidative stress caused by cadmium toxicity. CEE lowered ROS levels and improved depletion of GSH levels. CEE also enhanced the enzymatic activity of SOD. In component analysis, hesperidin was the most abundant of the five marker compounds (Narigenin, Narigin, Narirutin, Hesperidin and Hesperidin), which assumes that hesperidin partially contributed to the antioxidant activity of CEE. Conclusion : These results suggested that CEE could be a potential substance to solve heavy metal-related health problems. In particular, inhibition of oxidative stress by CEE can be a way to treat liver damage caused by cadmium.
