인간 항균펩타이드인 LL-37 유래의 FK-13의 화장품보존제로 활용에 대한 연구 (A Study on the Use of Human Antibacterial Peptide LL-37-derived FK-13 as a Cosmetic Preservative)

  • 본 연구에서는 인체에 보다 안전한 천연 화장품보존제 발굴을 목표로 하여 인간유래 항균펩타이드 LL-37의 짧은 유사체인 FK-13을 화장품보존제로 사용하고자 연구를 진행하였다. 이를 위해 13개의 아미노산으로 구성된 FK-13 펩타이드를 고상 펩타이드 합성법로 합성하였고 reversed phase-high performance liquid chromatography (RP-HPLC)로 정제 후 liquid chromatography-mass spectrometry (LC-MS)를 이용하여 순도와 분자량을 확인하였다. 합성된 FK-13을 이용하여 미생물에 대한 방부력을 검정하였고 화장품에 적용 가능성을 살펴보았다. FK-13은 3개의 그람-양성균 (Staphylococcus aureus, Bacillus subtilis, Staphylococcus epidermidis)과 3개의 그람-음성균 (Escherichia coli, Salmonella typhimurium, Pseudomonas aeruginosa), 그리고 진균인 Candida glabrata 에서도 높은 항균 활성을 보였으며 넓은 항균활성 스펙트럼을 나타내었다. 이를 바탕으로 화장품보존제로서의 적합성을 확인하였다. 또한 FK-13은 고온에서 분자의 열안정성을 보였으며 기존의 천연 한방화장품 방부제 및 화학보존제들과의 항균활성 비교 실험에서도 보다 월등한 항균활성 효능을 나타내었다. 따라서 FK-13 펩타이드는 인체 친화적이고 효과적인 항균활성을 나타내므로, 기존 화학보존제를 대체할 새로운 천연 화장품보존제로 적용이 가능할 것으로 판단된다.

  • This study was to examine the anti-bacteria effect and the changes of chemical properties of chrome-tanned garment leathers on the preservative treatment. Various test methods, such as investigation of preservative treatment process, resistance test and chemical analysis by cleaning, antbbacterial test by shake flask method are carried out in this study. The results can be obtained as follows: 1. Bacterial reduction percentage of chrome-tanned garment leathers on the preservative treatment was 28.6%. 2. In the antbbacterial effect by dry cleaning, preservative treated leathers has no resistance. 3. Fats content has been removed by dry cleaning using perchloroethylene, so garment leathers properties were altered. 4. PH value was changed by dry cleaning. But once after fats removing, it was changed the little by dry cleaning numbers.

Efficacy of Chemical Preservatives to Control Perilla Rust

  • The rust fungi are biotrophic fungal pathogens that cause serious disease on Perilla leaves in Korea. The effect of some commonly used chemical preservatives (sorbic acid, D-sorbitol, propionic acid and benzoic acid) against Perilla rust was investigated in this study. Results of this investigation showed that all the preservatives were effective against Perilla rust except benzoic acid. There was no growth of rust spores on the Perilla leaves treated with 0.1% of preservative even after 21 days of preservation. However, 0.01% of preservative also showed remarkable reduction of rust spores on the Perilla leaves as compared to the control groups. Thus, the results of this study indicated that the chemical preservatives used might be useful to control the growth of rust fungi on Perilla leaves.

가자, 오배자, 계피 추출물을 이용한 화장품 제형에서의 방부효과 (Antimicrobial Plant Extracts as an Alternative of Chemical Preservative: Preservative Efficacy of Terminalia chebula, Rhus japonica (gallut) and Cinnmomum cassia Extract in the Cosmetic Formular)

  • 본 연구는 천연 식물 추출물을 이용하여 화장품 제형 내에서 천연 방부제로서 효능을 확인하기 위한 목적으로 진행하였다. 선별된 15종의 천연 식물 추출물을 이용하여 병원성 세균(Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli)과 효모균(Candida albicans), 곰팡이균(Aspergillus niger)에 대한 paper disc법(disc diffusion method)과 최소저해농도측정법(MIC)을 이용하여 항균 활성을 측정하였다. 그 결과 가자, 오배자 추출물은 그람 양성인 S. aureus와 그람 음성균인 P. aeruginosa, E. coli 에 대한 항세균 활성이 우수하였고, 계피 추출물은 효모균인 C. albicans와 곰팡이균인 A. niger에 대해 우수한 항진균 활성을 나타냈다. 최종 선별된 3종 외 식물추출물은 효과가 없었다. 항균 활성 효과가 우수한 최적 조합비로 구성된 가자, 오배자, 계피 추출물의 혼합 추출물을 화장품 제형에 첨가하여 제형 내 방부 활성을 측정한 결과 상업적으로 널리 사용되고 있는 파라벤 혼합 방부제와 유사한 정도의 방부 활성을 나타냄을 확인하였다. 결론적으로, 화학 방부제를 대신하여 천연 식물 추출물로만 이루어진 넓은 항균 스펙트럼을 갖춘 방부 시스템의 개발이 가능할 것으로 사료되며 최근 다양한 문제가 제기되고 있는 화학 방부제의 단점을 극복한 천연 방부제를 함유한 화장료의 개발에 응용 가능할 것으로 기대된다.

에멀젼 제형에서 수종의 폴리올이 방부 시스템에 미치는 영향 (Effects of Various Polyols on Antiseptic System in Emulsions)

  • It is inevitable to use chemical germicidal agents like paraben, imidazolidinyl urea and phenoxyethanol to preserve the emulsions which is usually used in cosmetics. Although these chemical preservatives are good enough to reduce the microbiological contamination, they are irritative, allergenic to the skin. Several kinds of polyols are used in cosmetics as moisturizer and solvent. In this study, we evaluate the effects of polyols on anti-microbial activities, safety and resistant index. MIC(minimal inhibitory concentration) of polyols determined against 6 germs including Staphylococcus aureus. The order of MIC was PG $\cong$ DPG $\cong$ 1,3BG > HG > 1,2-PD > 1,2-HD $\cong$ 1,2-OD. The $2{\sim}3\;wt%$ of 1,2-HD(hexanediol) shows good anti-microbial effects in emulsions without allergenic response. Resistant index of 1,2-HD was less than 2 and this value was smaller than that of chemical preservatives. The mechanism of antimicrobilogical effect might be disturb the membrane of germs by investigating using electron microscope. Added to that, using this paradigm, low preservative contents, paraben-free system, and even preservative-free systems can be expected from these results.

Antimicrobial Activity of Propolis Extract and Their Application as a Natural Preservative in Livestock Products: A Meta-Analysis

  • This study aimed to evaluate the effectiveness of propolis extract as a natural preservative for livestock products in term of chemical and microbiological characteristics by meta-analysis. The stages carried out in this study were identification, selection, checking suitability, and the resulting selected articles were used in the meta-analysis. The selection results obtained a total of 22 selected journal articles consisting of 9 articles for analysis of the antimicrobial activity of propolis extract and 13 articles for analysis of the chemical and mirobiological characteristics of livestock products. The articles were obtained from electronic databases, namely Science Direct and Google Scholar. The model used in this study is the random-effect model involving two groups, control and experimental. Heterogeneity and effect size values were carried out in this study using Hedge's obtained through openMEE software. Forest plot tests and data validation on publication bias was obtained using Kendall's test throught JASP 0.14.1 software. The results showed that there is a significant relationship between propolis extract with the results of the antimicrobial activity (p<0.05). In addition, the results of the application of propolis extract on the livestock products for the test microbes and the value of thiobarbituric acid reactive substances (TBARs) showed significant results (p<0.05). Conclusion based on the random-effect model on the effectiveness of antimicrobial activity of propolis extract and their apllication as a natural preservative of the chemical and microbiological characteristics of livestock products is valid by Kendall's test (p>0.05). Propolis in this case effectively used as natural preservatives in livestock products.

신규 화장품용 천연유래 보존제의 물성 측정, 용액 거동 및 보존제 포함 화장품의 제형 안정성 (Basic Properties and Solution Behavior of New Naturally Derived Cosmetic Preservative, and Stability of Cosmetic Formulation)

  • 화장품 보존제는 화장품의 유효 사용 기간에 매우 중요한 성분으로, 최근에는 저독성 자연유래 화장품 보존제에 대한 관심이 급증하고 있다. 본 연구에서는 기존 다이올(diol) 계통 보존제를 대체할 수 있으며 천연유래 성분으로부터 얻어진 신규 보존제(laurimino bispropanediol, LB)의 기초적인 물성을 측정하고 신규 보존제를 포함한 수용액을 제조하여 용액 거동을 고찰하였다. 특히, 신규 보존제의 수용액에서의 micelle 형성 가능성을 pyrene 분자를 이용한 실험법으로 고찰하였다. 이를 통해 신규 보존제는 상대적으로 긴 소수성 탄화수소와 늘어난 하이드록실 그룹(hydroxyl group)으로 인해 micelle 형성이 가능함을 추론할 수 있었다. 보존제의 제형 속에서의 성능을 검증하기 위해 유화력 테스트를 수행하였으며, 유기용매와 유지 성분에 대해 유화력을 지님을 확인할 수 있었다. 추가로, 점도가 상대적으로 낮은 에센스 및 비교적 점도가 높은 로션 제형에 도입하였을 때 제형의 분산 안정성이 확보됨을 파악할 수 있었다. 본 연구는 향후 새로운 저독성 천연유래 화장품 보존제의 개발 및 사용에 중요한 정보를 제공할 것이다.

폴리리신을 함유한 Streptomyces albulus 배양액의 김치미생물 성장억제 효과 (Effects of Crude ε-poly-L-lysine in Streptomyces albulus Broth on Suppression of Microbial Growth in Korean Kimchi)

  • The Streptomyces albulus broth, when the polylysine in the broth, that has powerful growth inhibiting effect far many microbes, is its maximum, had filtered off the cells, to use the broth as preservative for keeping favorable taste of Korean Kimchi. Some microorganisms in their specific growth medium, known to deteriorate the useful nutrient of the Kimchi, has grown with different amounts of the inhibiting broth, to determine the minimum growth inhibition concentration. The ${\varepsilon}$-poly-L-lysine had been identified from the IR spectroscopic analysis of the purified poly lysine of the broth from ion exchange chromatographic separation. The content of the polylysine had been determined by methyl orange decoloration effect. Though the minimum inhibition concentration, evaluated by the naked eye based on the conventional method measuring the turbid feature after 18 hours of culture, has different values each other, the observed effects confirmed that the crude broth could be used as a natural preservative for the Kimchi in extending the fair taste.

  • 천연보존제 나린진 및 키토산과 화학보존제 K 제품의 독성을 토끼 각막의 상피와 내피를 통하여 주사현미경적으로 관찰하였다. 나린진은 자몽씨 추출물로서 이는 항산화를 일으키는 f lavonid의 구성성분중 하나로 식품과 화장품의 보존제로서도 이미 많이 사용되고 있다. 키토산은 갑각류의 외피나 곤충의 표피, 곰팡이 및 효모 등에 널리 분포되어져 있다. 키토산은 인체의 독성이 없으면서도 다양한 미생물의 항균제로 널리 인식되어져 있다. 본 연구는 천연보존제와 화학보존제의 독성을 검증하였다.

Decay Resistance of Fire-Retardant Treated Wood

  • In this study, the Korean pine wood (Pinus densiflora Sieb. et Zucc) and Italian poplar wood (Populus euramericana Guinier) was treated with a mixture of monoammonium phosphate (MAP) and boric acid. Their usability as fire retardant and as decay-resistant construction and interior materials were evaluated by testing of chemicals, corrosion rate and absorption rate, weight loss and chemical contents. An experiment was performed to compare treated pine wood and Italian poplar wood. According to the results, Italian poplar wood had higher specific gravity and retention of chemicals than pine wood, and treated wood showed higher decay-resistance than untreated one. Weight loss was less in treated wood than untreated one because the degree of decay was lower in the former than the latter. Corrosion rate and absorption rate met the KS standard for wood preservative performance. The chemical contents analysis was carried out to determine the degree of decay and it was found that the preservative effect of chemical treatment was lower in Italian poplar wood than in pine wood.
