Studies on Light-Induced Mitochondrial ATPase in Pleurotus ostreatus -Effects of Organic Compounds- (느타리버섯 중의 Light-Induced Mitochondrial ATPase에 관한 연구 -유기물 효과-)

  • Mitochondria in Pleurotus ostreatus were isolated and purified by stepped sucrose density gradient centrifugation, to compare the effects of organic compound on the activities of mitochondrial ATPase in Basidiomycotina with those in mammalian cell. The effects of N, N'-dicycio-hexylcarbodiimide (DCCD), carbonyl cyanide m-chlorophenylhydrazone (CCCP), sodium azide and aurovertin known as compounds to be related to electron transfer system in mitochondria were studied. A activity of mitochondrial ATPase was inhibited by 64%, 57% and 53% in the presence of 0.25 mM DCCD, 0.02 mM sodium azide and 1.5 $({\mu}g/mg\;of\;protein)$ aurovertin B, respectively. It was stimulated by 22% in the presence of 0.15 ${\mu}M$ CCCP.

Refolding of Acid-Unfolded Globin to Hemoglobin

  • Hemoglobin is oxygen carrier protein within erythrocyte in blood. Apoprotein of this, globin, is synthesized in the cytosol but it's cofactor, heme, is synthesized in the mitochondria. It has not been known very well how globin receives the heme from mitochondria and folds to hemoglobin. In this folding process, the initial structure of globin seems to be very important. A small volume of globin at acid pH was added rapidly into the bulk of an egg phosphatidylcholine $60\%$ liposome, containing hemins, at neutral pH according to the Rapid Dilution method. It was observed that an acid-induced unfolding structure of globin is initially needed to receive hemins from the lipid bilayer of liposomes. Also, this conclusion was confirmed with the absorption spectrum of the refolded globin separated by centrifugation.

Biological Function of Lactoferrin in Milk

  • Lactoferrin is an iron-binding glycoprotein and its bacteriostatic and bactericidal effects on Gram-positive and Gram-negative bacteria have been well-known. However, certain kind of lactic acid bacteria are resistant against its antibacterial effects. Moreover, it is reported that lactoferrin promotes the growth of bifidobacteria by in vitro and in vivo experiments. In this experiment, lactoferrin-binding protein was found both in the membrane and cytosolic franctions of Bifidobacterium. Bifidobacterium was grown in anaerobic conditions in MRS broth containing cysteine, gathered by centrifugation and processed by sonication. The lactoferrin-binding proteins on the PVDF-membrane transferred after SDS-PAGE were detected by far-western method using biotinylated lactoferrin and streptavidin-labeled horse radish peroxidase. Observation in growth effects of lactoferrin on Bifidobacterium suggested that there is a relation between the presence of lactoferrin-binding proteins on the cells and their growth.

  • The main objective of modern reproductive technologies in pig reproduction is to increase reproductive efficiency and rates of genetic improvement. They also offer potential for greatly extending the multiplication and transport of genetic materials and the conservation of unique genetic resources in reasonably available forms for possible future use. The development and refinement of these technologies is concentrating on gamete and embryo collection, sorting and preservation, in vitro production of embryos, culturing, manipulation of embryos (splitting, nuclear transfer, production of chimeras, establishment embryo stem cells, and gene transfer) and embryo transfer. Also, the development of these novel technologies is facilitated by modern equipment for ultrasonography, microscopy, cryopreservation, endoscopy, and flow cytometry, microinjectiors, micromanipulators and centrifugation. The real impact on herd productivity will come from combining new reproductive techniques with powerful DNA technologies. The new reproductive techniques will allow a rapid turnover of generations, whereas the DNA technology can provide selection, which does not need phenotypic information when the selection decisions are made.

Killing Activity and Molecular Properties of Bacteriophage Sigma FA1 of Bacillus circulans (Bacteriophage Sigma FA1의 치사활성과 구조특성)

  • In the previous paper (10). a new temperate phage, Sigma FA1 had been isolated from B. circulans. Sigma FA1 had an icosahedral head with a diameter of about 70 nm, and a tail about 15 nm long, and beared a circularly permuted, linear duplex DNA. Signla FA1 killed sensitive cells by a single-hit process. Phage DNA injected into the cell immediately after infection was degraded slowly. Our results indicate that the killing action of Sigma FA1 is different from the phenomenon of abortive infection and suggest that the killing might be caused by a proteinaceous component of Sigma FA1.

  • Lysosomal enzyme latency was demonstrated for hydrolases from porcine leukocyte by suspending sediment sfrom differential centrifugation in 0.125 to 0.250 M sucrose. Specific activities pH optima and activation energies were determined for hydrolases distributed in various sedimentation fractions and for enzymes solubilized by n-butyl alcohol extraction. Specific activities of the hydrolases revealed the heterogeneity of the Iysosomal fractions relative to enzyme content. pH optima identified the enzyme as acid hydrolases with optima for cathepsin D and aryl sulfatase also at pH 6.8. Activation energies of some hydrolases were low revealing that these enzymes could function efficiently during low temperature aging of meat.

  • The present paper describes a colorimetric method of determining the free and total unsaturated fatty acids in animal tissues. The procedure is based in three steps on the following principles : First step is that the tissue homogenates are extracted in chilled acetone in order to eliminate the interfering substance, such as phospholipids, sulfatides and proteins. At next step, after centrifugation acetone layer is decanted and evaporated to dryness. Then the extract is shaken with heptane to solve in the solvent. That the characteristic nature of copper salts of unsaturated fatty acids are freely soluble in heptane and those of saturated acids are not is the bases of separating one from another. Thus unsaturated fatty acids can be isolated in heptane as their salts from saturated acids and other lipid mixture by shaking with copper reagent. Finally the yellowish brown color developed by adding color reagent (0.2% sodium diethyldithiocarbamate in n-butanol solution) which reacts with the copper salts of the acids and produces the color is measured colorimetrically.

  • Throughout the studies the following experimental results were obtained and so are summarized here. 1) What caused tussah silkworms terrible disease broken ous all over the Korea in 1965, was examined to be “Inclusion body of virus” through microscope. 2) The examined inclusion bodies are easily stained as purple by seller's staining. 3) The proper speed of centrifugation for the purification of inclusion bodies is 1,000 r. p. m for 10 minutes. 4) It is possible, cleanly resulted, to attempt the “Rapid Agglatination Test & Complement Fixation test” with autigen of inclusion bodies. 5) The Anti-polyhedra rabbit serum from antigen of the dilution of 2${\times}$10$\^$6//l$m\ell$ polyhedra made the Ropid Agglutination test possible even with dilution of 1/8 infected pupa blood(antigen).

Quality Characteristic of Drink and Tea-Bag Processed with Safflower Seed Powder (홍화씨분말을 이용한 음료 및 티백차의 품질특성)

  • Drink and tea-bag were processed with safflower seed powder. Drinks were processed with 90$^{\circ}$C hot water extraction. Yield, soluble solid, pH and centrifugation residue of drinks were ranged in 79.2~89.3%, 0.6~0.99%, 5.98~6.40 and 1.00~1.18, respectively. Sensory score of overall acceptance in drinks were highest at that of enzyme treated drink. Tea-bag was processed with roasted safflower seed powder, alone(1.2g) and the mixtured tea-bags were consisted of persimmon leaves, pine needle and angelica gigas powder in same amount, respectively. Teas were prepared with extraction at 80$^{\circ}$C water for 2 min. Soluble solid and sensory evaluation score of teas were the highest when the tea was processed with safflower seed alone.

Ag Nanowires Prepared by a Modified Polyol Method with 1,4-Benzoquinone Additives

  • This paper describes a selective synthetic method of fabricating Ag nanowires by using a modified polyol process. To synthesize the Ag nanowire, an ethylene glycolic solution of silver nitrate and an ethylene glycolic solution of polyvinylpyrrolidone solution containing a small amount of organic oxidant, 1,4-benzoquinone, were slowly added to a hot ethylene glycol medium at $160^{\circ}C$ for 8 min using a syringe pump. The reaction mixtures were heated for an additional 45 min and cooled to room temperature. Finally, the silver nanomaterials were isolated from the mixture by centrifugation. The crystal structure of the nanomaterials was investigated by powder X-ray diffraction analyses, and their morphology was investigated by scanning electron microscopy. A small amount of organic oxidant, 1,4-benzoquinone, played a significant role in controlling the morphology during crystal growth. Consequently, Ag nanowires rather than Ag nanoparticles were selectively obtained.
