Differentiation and Characterization of Cystic Fibrosis Transmembrane Conductance Regulator Knockout Human Pluripotent Stem Cells into Salivary Gland Epithelial Progenitors

  • The differentiation of pluripotent stem cells has been used to study disease mechanisms and development. We previously described a method for differentiating human pluripotent stem cells (hPSCs) into salivary gland epithelial progenitors (SGEPs). Here, cystic fibrosis transmembrane conductance regulator (CFTR) knockout hPSCs were differentiated into SGEPs derived from CFTR knockout hESCs (CF-SGEPs) using the same protocol to investigate whether the hPSC-derived SGEPs can model the characteristics of CF. CF-a disease that affects salivary gland (SG) function-is caused by mutations of the CFTR gene. Firstly, we successfully generated CFTR knockout hPSCs with reduced CFTR protein expression using the CRISPR-Cas9 system. After 16 days of differentiation, the protein expression of CFTR decreased in SGEPs derived from CFTR knockout hESCs (CF-SGEPs). RNA-Seq revealed that multiple genes modulating SG development and function were down-regulated, and positive regulators of inflammation were up-regulated in CF-SGEPs, correlating with the salivary phenotype of CF patients. These results demonstrated that CFTR suppression disrupted the differentiation of hPSC-derived SGEPs, which modeled the SG development of CF patients. In summary, this study not only proved that the hPSC-derived SGEPs could serve as manipulable and readily accessible cell models for the study of SG developmental diseases but also opened up new avenues for the study of the CF mechanism.

  • The main objective of this paper is to obtain a formula containing eleven interesting results for the reducibility of Kamp$\acute{e}$ de F$\acute{e}$riet function. The results are derived with the help of two general results for the series $_2F_1(2)$ very recently presented by Kim et al. Well known Kummer's second theorem and its contiguous results proved earlier by Rathie and Nagar, and Kim et al. follow special cases of our main findings.

  • We study spaces of continuous-function-valued functions that have the property that composition with evaluation functionals induce $weak^*$ to norm continuous maps to ${\ell}^p$ space ($p{\in}(1,\;{\infty})$). Versions of $H{\ddot{o}}lder^{\prime}s$ inequality and Riesz representation theorem are proved to hold on these spaces. We prove a version of Dixmier's theorem for spaces of function-valued matrix operators on these spaces, and an analogue of the trace formula for operators on Hilbert spaces. When the function space is taken to be the complex field, the spaces are just the ${\ell}^p$ spaces and the well-known classical theorems follow from our results.

A Study of the Hierarchy of the Central Place and the Shopping Area in the Kyeong-ju Si Area (경주시지역 중심지계층과 생활권에 관한 연구)

  • The purpose of this study is to analyze the classification of the hierarchy in terms of centrality and the demarcation of shopping areas in the context of goods-purchasing activity in the Kyeong-ju si area (The old Kyeong-ju city and Wol-song Kyn). The basic data for this study include mail questionnaire materials and sources by the on-the-sport survey. The main results of this study are as follows. (1) When forty-two centers within the wide Kyeong-ju area are classified into hierarchies in terms of the functional index and the number of central functions, it is confirmed that there are five hierarchies or classes. Hierachy A is the City of Kyeong-ju. Hierarchy B is the Up of An-gang. Hierarchy Cis made up of three Up of Kam-p'o, Oe-dong and Kon-ch' on. Hierachy D consist of seven Myons and two Ris. Hierarchy E is composed of one Myon and twenty-seven Ris. (2) Hierarchy A has five proper functions of the center. Hierarchy B holds eleven of them. Hierarchy C takes forty-one. Hierarchy D cherish twenty-seven. And hierarchy E has three. Therefor Hierachies C and D have the most functions of all. The threshold populations of the lowest central function is fifty-eight of miscellaneous stores in the central place of Hierarchy E (3) The purchase distance of the central functions of a center becomes farther in the order of barbers' and beauty salons, dental clinics, TV and refrigerator shops, and furniture stores. (4) When the shopping areas are classified according to goods purchase activity, they can be divided into four: the Myon, small Up, big Up, and City areas. Each shopping area can be classified into two categories: the independent areas and compoung areas. The Kyeong-ju city area is the largest city shopping areas of Po-hang and Ull-san are very small.

Moving Path Tracing of Image Central Position with Autocorrelation Function

  • For an complete image composition to be stitched on several mosaic images, tracing displacement of direction and distance between successive images are important parameters. The input image is modeled by using a general second order two-dimensional Taylor-series and then converting it to a $3{\times}3$ correlation block and storing the data. A moving factor and coordinate is calculated by comparing the continuous correlation blocks. The experimentation result has a success rate of 85% for moving path tracing as continuous images are moved to 10% of image central position.

  • 최근 널리 보급되고 있는 IPTV의 주요 서비스인 주문형 비디오 서비스의 시청 순위는 Zipf의 법칙을 따른다고 알려져 있으며 이를 기반으로 계층형 네트워크 스토리지 구조로 서비스를 제공하여 네트워크 비용과 스토리지 비용을 절감하고 있다. 그러나, 최근 연구에 의하면 미디어 시청 순위는 stretched exponential 함수에 더 근사함이 알려졌다. 본 연구에서는 국내에서 현재 상용 서비스 중인 대규모 주문형 비디오 서비스의 시청 순위-회수 데이터를 분석하여, 시청 순위로부터 stretched exponential 함수의 모수를 추정하여 시청 순위의 분포함수를 파악하고자 한다. 특히, 콘텐츠 파일 크기, 콘텐츠 파일 전달방식, 콘텐츠 장르 등의 요인과 시청 순위 분포함수의 모수 사이에 상관관계가 있는지 분석하고자 한다.

On the Motion of the Structure Varying Multibody Systems with Two-Dimensional Dry Friction

  • In the present paper the dynamics of the structure varying multibody systems caused by stick-slip motion with two-dimensional dry friction are analyzed. The methods to determine friction force both in stick and slip states are described. The direct method of considering the wagon bogie system as a structure varying system was used to consider two dimensional friction at the wheelset-side frame connection. The concept of friction direction angle used to determine the friction force components of two-dimensional dry friction both in the stick and slip motion states was used. A speed depended friction coefficient was used and described approximately by hyperbolic secant function. All switch conditions were derived and friction forces both for stick and slip states. Some simulation results are provided.

  • We consider functions that map the open unit disc conformally onto the complement of an unbounded convex set with opening angle ${\pi}{\alpha}$, ${\alpha}{\in}(1,2]$, at infinity. We derive the exact interval for the variability of the real Taylor coefficients of these functions and we prove that the corresponding complex Taylor coefficients of such functions are contained in certain discs lying in the right half plane. In addition, we also determine generalized central functions for the aforesaid class of functions.

  • This study aimed at analysing on consumer's behaviour to central functions in center villages of Hub-myun. Consumers in the center villages were classified as follows; residents in the center village and in the hinterland area, visitors. Through the interview works on the customers in 8-pilot project sites and differential analysis works of answered results, consumers' attitude in the center villages were grouped into two types; self-sufficient one and higher center-dependent one.

  • This paper presents the reverse design of a cam mechanism using NURBS(Nonuniform Rational B-spline curve). Cam is very difficult to make the accurate shape on the design and the manufacture. Because the cam shape is commonly made in order to move in special functions. The reverse design can be used to check accuracy between the designed data and the manufactured data of the cam shape and also reproduce the cam without the design data. The reverse design procedures consist of motion analysis and curve fitting. The motion analysis is used the central difference method and the relative velocity method to find the displacement and velocity. The curve fitting is used NURBS to develope the whole curve. The central difference method is derived in the 3 dimensional space.
