Investigation of Bordered Pit Ultrastructure in Tracheid of Korean Red Pine (Pinus densiflora) by Confocal Reflection Microscopy (공초점반사현미경을 이용한 소나무 유연벽공의 초미세구조 연구)

  • Confocal reflection microscopy (CRM) was utilized to create 3-dimensional images of bordered pits and cell wall in the tracheid of Korean red pine (Pinus densiflora). Ultrastructures of torus, margo, and pit border were clearly observable in the CRM micrograph. Micrograph of cross-field pit revealed the connecting and supporting structure between tracheid and ray parenchyma cell. The CRM micrographs enabled to investigate detailed structures of tracheid cell wall such as S1, S2, S3 layers, transition layers between these layers, and microfibril (MF) orientation in S3 and S2 layers as well as complicated distribution of MF orientation around bordered pits. Not only concentric MF orientation of border thickening in the pit border was observed, but also changes in MF orientation from the cell wall to the border. From the experimental results, the CRM was thought to be a versatile microtechnique to investigate detailed structures of cell wall and bordered pit in the tracheid and cross-field pit between tracheid and ray parenchyma cell.

  • The discussion about the structural reformation of Korean rail industry has been consistently conducted from the evaluation on Korean National Railroad's management in 1998, the foundation of KORAIL in 2005 to the approval of Suseo KTX corporation establishment in 2013 and the controversy over its privatization. This research explores the changes of government policies by applying to ACF in the assumption that those changes have been implemented due to the interaction of various external variables and main agents in policy which had existed for a long-term period that the rail industry's structural reformation has been practiced for more than 10 years. The unions failed to reach an agreement for the rail industry's structural reformation, the policies regarding the reformation have been led by superior political unions. And this research concludes that a mediator who can compromise unions' different policy preferences and means and bring up a compromised decision in conflicted situation plays a pivotal role to successfully practice these policies.

Comparison of Helminth Infection among the Native Populations of the Arctic and Subarctic Areas in Western Siberia Throughout History: Parasitological Researches on Contemporary and the Archaeological Resources

  • The aim of this parasitological study is examining contemporary (the late 20th century) specimens of the arctic or subarctic areas in Western Siberia and comparing them with the information acquired from archaeological samples from the same area. In the contemporary specimens, we observed the parasite eggs of 3 different species: Opisthochis felineus, Ascaris lumbricoides, and Enterobius vermicularis. Meanwhile, in archaeoparasitological results of Vesakoyakha, Kikki-Akki, and Nyamboyto I burial grounds, the eggs of Diphyllobothrium and Taenia spp. were found while no nematode (soil-transmitted) eggs were observed in the same samples. In this study, we concluded helminth infection pattern among the arctic and subarctic peoples of Western Siberia throughout history as follows: the raw fish-eating tradition did not undergo radical change in the area at least since the 18th century; and A. lumbricoides or E. vermicularis did not infect the inhabitants of this area before 20th century. With respect to the Western Siberia, we caught glimpse of the parasite infection pattern prevalent therein via investigations on contemporary and archaeoparasitological specimens.

Detection of CTX-M Type ESBL Producing Salmonella in Retail Meat in Korea

  • This study was performed to evaluate antimicrobial resistance of food-borne pathogens isolated from retail meat in Korea. A total of 157 samples of beef, pork, and chicken were collected and analyzed for E. coli, Salmonella, Campylobacter. Resistances to tetracycline were declined in accord with reduced usage of tetracycline in live stock production. E. coli stains from chicken meat had higher multi-drug resistance ratio than strains from other meat. One extended spectrum beta lactamase (ESBL) producing E. coli and two ESBL producing Salmonella were identified in this study. ESBL producing Salmonella strains were confirmed to carry CTX-M-1 type genes.

FISH Karyotype and GISH Meiotic Pairing Analyses of a Stable Intergeneric Hybrid xBrassicoraphanus Line BB#5

  • xBrassicoraphanus line BB#5, a new synthetic intergeneric hybrid between Brassica rapa L. ssp. pekinensis and Raphanus sativus L. var. rafiphera induced by N-methyl-N-nitroso-urethane mutagenesis in microspore culture, shows high seed fertility and morphological uniformity. Dual-color fluorescence in situ hybridization (FISH) using 5S and 45S rDNA probes and genomic in situ hybridization (GISH) using B. rapa genomic DNA probe were carried out to analyze the chromosome composition and the meiosis pairing pattern compared to its parental lines. The somatic chromosome complement is 2n = 38, which consists of 17 metacentric and two submetacentric chromosomes with lengths of 2.18 to $5.01{\mu}m$. FISH karyotype analysis showed five and eight pairs of 5S and 45S rDNA loci. GISH meiosis pairing analysis showed that 19 complete bivalents were most frequent and accounted for 42% of the 100 pollen mother cells examined. Based on chromosome number, size, morphology, rDNA distribution, and meiosis pairing pattern, both parental genomes of B. rapa and R. sativus appear to exist in xBrassicoraphanus line BB#5, demonstrating its genome integrity. Such stable chromosome constitutions and meiotic pairing patterns in somatic and meiotic cells are very rare in natural and synthetic intergeneric hybrids. Chromosomal studies and genetic and phenotypic changes in allopolyploids a re discussed.   • Recycled fibers usually give slow drainage in the paper forming zone on papermachine, which limit the application of more refining to the fibers for improving paper strength and formation. To use recycled fibers, especially, OCC, more effectively, developing very efficient handling technique of short fibers and fines is inevitable. We tried to make hard flocs of fractionated short fibers and fines, which were the main cause of slow drainage, by adding excessive amount of retention aid on them. This technique was proved to increase drainage with no difference in strength properties, compared to the conventional technique of adding the same amount of polymers to the whole furnish in the lab test. The bonding capability of short fibers and fines in Korean OCC were very poor to be considered as ´fillers´in paper products. Various chemical treatment on the short fibers and fines of the Korean OCC did not improve their bonding and optical properties. One of the reasons of no improvement in their properties was thought to be their high amount of ashes (over 30% in the fractionated samples).

Fracture Behavior of Polycarbonate/Polyestercarbonate Blends (폴리카보네이트/폴리에스터카보네이트 블렌드의 파괴 거동)

  • Fracture behaviors of polycarbonate (PC)/polyestrercarbonate (PEC) blends and their miscibility have been examined to find out the mechanism of ductilie-brittle transition of fracture behavior which would be a main governing factor on the thickness sensitivity of impact strength of PC. $T_g$ measurement showed that PEC with a carbonate content higher than 30 mol% was miscible with PC. In the notched Izod impact test of PC, ductile-brittle transition occurred in the range of 4 to 5 mm thickness. The impact strength of miscible PC/PEC5 blends ductile-fractured in the thin specimens decreased with increasing PEC5 content, which was in accordance with the decrease of elongation at break in tensile test. In the brittle fracture of the thick specimens, the impact strength was well correlated with the plastic zone size in the vicinity of the notch tip.

Quantification and comparison of functional phytochemicals in steamed and freeze-dried mature silkworm powders and freeze-dried mulberry leaves

  • Various health promoting effects of steamed and freeze-dried mature silkworm powder (SMSP) have been reported. However, it is not still clear which substances in SMSP are responsible for those health promoting effects yet. In this study, we examined and compared the quantities of phytochemicals in SMSP and freeze-dried mulberry leave powder (FMLP). To investigate the optimal solvent for extracting phytochemicals from SMSP and FMLP, we used four different solvents. Among them, 80% ethanol extracts from SMSP and FMLP contained the highest amount of five flavonoids examined. In addition, FMLP had high contents of flavonoids compared with those of SMSP. The amounts of rutin, isoquercetin, astragalin, quercetin, and kaempferol in FMLP were $5.078{\pm}0.187mg/g$, $5.162{\pm}0.083mg/g$, $2.989{\pm}0.061mg/g$, $3.317{\pm}0.236mg/g$, and $2.243{\pm}0.237mg/g$, respectively, while the amounts of rutin, isoquercetin, astragalin, quercetin, and kaempferol in SMSP were $0.171{\pm}0.024mg/g$, $0.252{\pm}0.032mg/g$, $0.374{\pm}0.031mg/g$, $0.645{\pm}0.063mg/g$, and $0.0512{\pm}0.047mg/g$, respectively. Taken together, SMSP could be a source for providing various and readily absorbable flavonoids.

A Study on the Implementation of Official Family-related Service History Archives in Korea (공적 가족사업역사 아카이브 구축을 위한 기초 연구)

  • This study aims to provide basic resources for the construction of the archives on the official family service history in Korea. The study collected records of family policies and official family service from 1948 to 2019 and thereafter attempted to classify the official family service. In addition, experts were consulted to diagnose compliance with research contents and directions, investigate priorities of the family policies, and provide opinions on the elements of each process of archiving. The results of this study were as follows. First, there were five areas of concern to be considered in the process of establishing an archive of official family services: concept, scope, collection, arrangement, preservation, and utilization. Second, the findings suggest which processes are critical to developing a sustainable and systematic archive system for the official family service history. The construction of the archives will provide a platform for understanding the contents of integrated family policies and inform the direction of future family policies in Korea. In addition, these archives will be an important factor in building a solid Korean national identity.

Evaluation of the Suitability of Establishing Biological Exposure Indices of Styrene

  • Background: This study was designed to provide logical backgrounds for the revision of biological exposure indices (BEIs) for styrene exposure in Korea. In order to investigate the correlation between airborne styrene and biological exposure indices, we measured urinary mandelic acid (MA) and phenylglyoxylic acid (PGA) in workers exposed to styrene occupationally, as well as airborne styrene at workplaces. Methods: Surveys were conducted for 56 subjects. The concentrations of airborne styrene and urinary metabolites of styrene were measured in 36 workers who were occupationally exposed to styrene, and in 20 controls. Air samples were collected using personal air samplers and analyzed by gas chromatography. Urine samples were collected at the end of the shift and analyzed by high performance liquid chromatography. Results: The geometric mean concentration of airborne styrene was 9.6 ppm. The concentrations of urinary MA, PGA, and MA+PGA in the exposure group were 267.7, 143.3, and 416.8 mg/g creatinine, respectively. The correlation coefficients for correlation between airborne styrene and MA, PGA, and MA+PGA were 0.714, 0.604, and 0.769, respectively. The sum of urinary MA and PGA corresponding to an exposure of 20 ppm styrene was 603 mg/g creatinine. Conclusion: The correlation of the sum of urinary MA and PGA with airborne styrene was better than the correlation of each individual urinary determinant. It is considered appropriate to amend the concentration of urinary MA+PGA to 600 mg/g creatinine as a BEI, which corresponds to an airborne styrene concentration of 20 ppm in Korea.
