Exploring the DNA methylome of Korean patients with colorectal cancer consolidates the clinical implications of cancer-associated methylation markers

  • Aberrant DNA methylation plays a critical role in the development and progression of colorectal cancer (CRC), which has high incidence and mortality rates in Korea. Various CRC-associated methylation markers for cancer diagnosis and prognosis have been developed; however, they have not been validated for Korean patients owing to the lack of comprehensive clinical and methylome data. Here, we obtained reliable methylation profiles for 228 tumor, 103 adjacent normal, and two unmatched normal colon tissues from Korean patients with CRC using an Illumina Infinium EPIC array; the data were corrected for biological and experiment biases. A comparative methylome analysis confirmed the previous findings that hypermethylated positions in the tumor were highly enriched in CpG island and promoter, 5' untranslated, and first exon regions. However, hypomethylated positions were enriched in the open-sea regions considerably distant from CpG islands. After applying a CpG island methylator phenotype (CIMP) to the methylome data of tumor samples to stratify the CRC patients, we consolidated the previously established clinicopathological findings that the tumors with high CIMP signatures were significantly enriched in the right colon. The results showed a higher prevalence of microsatellite instability status and MLH1 methylation in tumors with high CMP signatures than in those with low or non-CIMP signatures. Therefore, our methylome analysis and dataset provide insights into applying CRC-associated methylation markers for Korean patients regarding cancer diagnosis and prognosis.

Emission Characteristics of 0.7' Monochrome MOSFET-Controlled Field Emission Display in a High Vacuum Chamber

  • MCFEDs (MOSFET-Contoolled Field Emission Displays) were fabricated to evaluate the validity of MCFEA for display application. The electrical properties of FEAs (Field Emitter Arrays), HVMOSFETs (High-Voltage MOSFETs), and MCFEAs (MOSFET-Controlled Field Emitter Arrays) were measured. The extraction gate voltage of the FEAs to obtain the anode current of 10 nA/tip was around 71 V. The breakdown voltages of the HVMOSFETs were above 81 V for all the samples. The I-V characteristics of the MCFEAs showed that the emission currents of the FEAs were well controlled depending on the control gate voltages of the HVMOSFETs. To avoid the harmful effects during the packaging process, the performance of the MCFEDs was evaluated in a high vacuum chamber. The emission images of the MCFEDs were controlled through very-through operation. From the comparison with a conventional FED, it was proven that the poor uniformity of FED could be improved through the integration with HVMOSFET.

  • Catechol 2, 3-dioxygenase (C230) catalyses the oxidative ring cleavage of catechol to 2-hydroxymuconic semialdehyde. This is one of the key reactions in the metabolism of the widespresd pollutant aniline. We have cloned a gene encoding C230 from cells of the aniline degrading bacteria, Pseudomonas acidovorance KCTC2494 strain and expressed in E. coli, A 11.3-kilobase Sau3A partial digested DNA fragment from KCTC2494 was cloned into phagemid vector pBluescript and designated as pLP201. The C230 gene was mapped to a 2.8-kb region, and the derection of transcription was determined. The cloned C230 gene contains its own promoter which can be recognized and employed by E. coli transcriptional apparatus. C230 activities of subclones were identified by enzyme assay and activity staining. The T7 RNA promoter/polymerase system and maxicell analysis showed that a polypeptide with Mw of 35 kDa is the C230 gene product.

  • The adhesion of activated and normal platelets to fibrinogen requires the receptor binding site of GPIIb/IIIa. These recognition sites exists in the A ${\alpha}$ chain(RGDS at 572-575 and RGDF at 95-98) and the carboxy-terminal of ${\gamma}$ chain (HHLGGAKQAGDV at 400-411) of fibrinogen. In this study, we developed RGDF-immobilized surface to detect the unctional state of platelet. RGDF-immobilized surface was prepared on the glass using photolithographic technology. Platelet adhesion to RGDF-immobilized surface was observed by staining platelets with mepacrine using a fluorescence microscope using mepacrine. Using the RGDF peptide of fragment E, we observed that the platelets pretreated with PGE1 interacted incompletely with RGDF-immobilized surface, whereas ADP activated platelets interacted with the surface extensively. These results show that the distinct selectivity of RGDF-immobilized micro-patterned surface can be used to detect the unctional state of platelets.

  • In order to realize the high luminance and luminous efficacy MFFLs for LCD backlights, we optimized the phosphor profile to enlarge the surface area. The MFFL with the new phosphor profile shows a very wide luminance range from 2,600 to 17,000 nit with the corresponding luminous efficacy from 66 to 32.5 lm/W. The results were obtained with the color coordinate of the phosphor to be around (0.25, 0.23). And the single cell is expanded into a multi-structured configuration to realize a 32 inch-sized lamp by a simple repetition of the single cells, and a new driving scheme is proposed for an adaptive local dimming and scanning drive using dual auxiliary electrodes and bipolar drive scheme. Especially the ultra high luminance and luminous efficacy results suggest the duty time of illumination of the backlight unit can be significantly reduced.

  • The galaxy cluster is an important object for investigating the large scale structure and evolution of galaxies. Recent wide and deep near-IR surveys provide an opportunity to search for galaxy clusters in the high redshift universe. We have identified candidate clusters of 0.8< z <1.2 from the $25deg^2$ SA22 field using an optical-near-IR dataset from merged UKIDSS DXS, IMS and CFHTLS catalogs. Using these candidates, we investigate the star forming activity of member galaxies. Consequently, at z ~1, the star forming activity of cluster galaxies is not distinguishable from those of field galaxies, which is different from members in local clusters. This means the environmental effect becomes more important for $M_{\ast}>10^{10}M_{\odot}$ galaxies at z <1.

  • A bird's-eye view on preparation, structure and properties of polymeric complexes in the field of Inorganic-Organic-Hybrids is presented in the view point of solid state and materials chemistry. These materials are useful precursors for preparing nanoparticles and fine grain oxides. Some of them are electroactive and are used as protonic or lithium electrolytes, electrochromic materials or membranes for sensors and actuators. New results on bio-hybrids, a class of material not far from polymeric complexes, are also described.

  • Twenty-four cinereous vultures that had been taken to a wildlife center due to starvation and exhaustion were studied to evaluate approaches for determining sex. Coelioscopy was performed to identify sexes of two vultures, whereas, DNA testing was performed to identify the sexes of the 24 vultures. Testes and ovaries could be unambiguously identified with an endoscope and DNA analyses could identify sex sex in most, but not all of the specimens. Although the coelioscopy examination can unambiguously confirm sex, the approach is invasive and requires anesthesia. Thus, coelioscopic examination should only be performed when sex cannot be determined through DNA analysis.

  • The broadband noise can be dominant or important for total characteristics for marine propeller noise representing the minimum base of self-noise. Accurate prediction of such noise is crucial for survivability of underwater military vessels. While the FW-H Formulation 1B can be used to predict broadband trailing edge noise, the method required experiment measurements of surface pressure correlations, showing its limitations in generality. Therefore, in this study, the methods are developed to utilize wall pressure spectrum models to overcome those limitations. Chase model is adopted to represent surface pressure along with the developed formulations to reproduce pressure statistics. Newly developed method is validated with the experiments of airfoils at different velocities. Thereafter, with its feasibility and generality, the procedure incorporating computational fluid dynamics is established and performed for a propeller behind submarine hull. The results are compared with the experiments conducted at Large Cavitation Tunnel, thus showing its usability and robustness.

Pediatric blunt pancreatic trauma at a single center in Korea: a retrospective review from 2007 to 2022

  • Purpose: Blunt pancreatic trauma in pediatric patients is relatively rare, yet it is associated with high risks of morbidity and mortality This study aimed to review pediatric patients with blunt pancreatic trauma treated at a single center and provide treatment guidelines. Methods: This study included patients under the age of 18 years who visited our center's pediatric emergency department and were diagnosed with pancreatic injury due to abdominal trauma via radiological examination between January 2007 and December 2022. Patients' medical records were retrospectively reviewed and analyzed. Results: Among 107 patients with abdominal trauma, 14 had pancreatic injury, with a median age of 8.2 years (interquartile range, 3.1-12.3 years). Eight patients were male and six were female. The most common mechanism of injury was falls from a height and bicycle handlebars (four cases each). Six patients had associated injuries. Two patients had American Association for the Surgery of Trauma grade I or II, eight had grade III, and four had grade IV or V injuries. Eight patients underwent surgical resection, and four were discharged with only an intervention for duct injuries. Conclusions: Patients with blunt pancreatic trauma at our center have been successfully treated with surgical modalities, and more recently through nonsurgical approaches involving active endoscopic and radiologic interventions.
