Mapping Cellular Coordinates through Advances in Spatial Transcriptomics Technology

  • Complex cell-to-cell communication underlies the basic processes essential for homeostasis in the given tissue architecture. Obtaining quantitative gene-expression of cells in their native context has significantly advanced through single-cell RNA sequencing technologies along with mechanical and enzymatic tissue manipulation. This approach, however, is largely reliant on the physical dissociation of individual cells from the tissue, thus, resulting in a library with unaccounted positional information. To overcome this, positional information can be obtained by integrating imaging and positional barcoding. Collectively, spatial transcriptomics strategies provide tissue architecture-dependent as well as position-dependent cellular functions. This review discusses the current technologies for spatial transcriptomics ranging from the methods combining mechanical dissociation and single-cell RNA sequencing to computational spatial re-mapping.

Synthesis of Diacetoxy Acetal Derivatives of Santonin and their Enhancing Effects on HL-60 Leukemia Cell Differentiation

  • Several diacetoxy acetal analogues have been synthesized from santonin and assessed for their ability of inducing or enhancing the differentiation of human HL-60 leukemia cells. The compounds themselves had little effect on HL-60 cell differentiation. However, three analogues, 2a, 3a, and 5b, synergistically enhanced 1,25-dihydroxyvitamin $D_3[1,25-(OH)_2D_3]-induced$ HL-60 cell differentiation when combined with 5 nM of dihydroxyvitamin $D_3[1,25(OH)_2O_3]$, a well-known differentiation inducer. Especially, the compound 5b profoundly enhanced the $1,25-(OH)2O_3]-induced$ HL-60 cell differentiation.

The Effect of Shape of Core Cell on Shock Absorption Characteristics of Biomimetically Inspired Honeycomb Structures

  • The effect of the core cell shape on shock absorption characteristics of biomimetically inspired honeycomb structures has been numerically investigated. The finite element models of honeycomb test specimen composed of five core cells of identical mass have been constructed, and numerical simulations have been run on PAMCRASH. The dimensions of the sides of core cells as well as the angle between the sides have been shown to influence the shock absorption characteristics of the honeycomb structure. The specimen with regular hexagonal core cell shape is found to show the best shock absorbing capacity, and specimen with rectangle-like core cell are found to provide good shock absorbing characteristics.

Wnt signaling in cartilage development and degeneration

  • The Wnt signaling network, which is composed of Wnt ligands, receptors, antagonists, and intracellular signaling molecules, has emerged as a powerful regulator of cell fate, proliferation, and function in multicellular organisms. Over the past two decades, the critical role of Wnt signaling in embryonic cartilage and bone development has been well established, and much has been learnt regarding the role of Wnt signaling in chondrogenesis and cartilage development. However, relatively little is known about the role of Wnt signaling in adult articular cartilage and degenerative cartilage tissue. This review will briefly summarize recent advances in Wnt regulation of chondrogenesis and hypertrophic maturation of chondrocytes, and review data concerning the role of Wnt signaling in the maintenance and degeneration of articular chondrocytes and cartilage.

BIR Containing Proteins (BIRPs): More Than Just Cell Death Inhibitors

  • BIRPs (BIR containing Proteins) which contain one to three BIR domains constitute a highly conserved family from yeast to human. BIR domains mediate the interaction of BIRPs with various other proteins. Some of the members acquire a Ring domain which acts as an E3 ubiquitin ligase. The first member of BIRPs identified in the baculovirus was found as an inhibitor of apoptosis and most of the family members in the other species have been recognized to have the same function which bind to and inhibit caspases, thereby suppresses apoptotic cell death. But an increasing number of evidences indicate that BIRPs are involved in various cellular events such as cell division, control of cell cycle, signal transduction, cell migration, innate immunity as well as regulation of apoptosis. In this review, we summarize the structural and functional features of the BIRPs, especially focus on the various functions of BIRPs unrelated to regulation of apoptosis by the recent findings.

CD137-CD137 Ligand Interactions in Inflammation

  • The main stream of CD137 studies has been directed to the function of CD137 in $CD8^+$ T-cell immunity, including its anti-tumor activity, and paradoxically the immunosuppressive activity of CD137, which proves to be of a great therapeutic potential for animal models of a variety of autoimmune and inflammatory diseases. Recent studies, however, add complexes to the biology of CD137. Accumulating is evidence supporting that there exists a bidirectional signal transduction pathway for the CD137 receptor and its ligand (CD137L). CD137/CD137L interactions are involved in the network of hematopoietic and nonhematopoietic cells in addition to the well characterized antigen-presenting cell-T cell interactions. Signaling through CD137L plays a critical role in the differentiation of myeloid cells and their cellular activities, suggesting that CD137L signals trigger and sustain inflammation. The overall consequence might be that the amplified inflammation by CD137L enhances the T-cell activity together with CD137 signals by upregulating costimulatory molecules, MHC molecules, cell adhesion molecules, cytokines, and chemokines. Solving this outstanding issue is urgent and will have an important clinical implication.

석유탄화수소를 이용한 단세포단백질의 생산에 관한 연구 3 (Production of Single-Cell Protein on Petroleum Hydrocarbon)

  • The growth characteristics of Candida tropicalis KIST 351 on gas oil substrate under different culture conditions were investigated and the preliminary animal feeding experiments using this yeast as a partial substitute of fish meal was also conducted. The yeast assimilates effectively n-paraffins in gas oil ranging from $C_{16}$ to $C_{16}$ with its maximum cell growth at $33^{\circ}C$ and pH 5.5 with aeration of 3 vvn and agitation of 900 rpm. The optimal concentrations of nitrogen sources, $HK_2PO_4$ and $Na_2HPO$ were 4, 2 and 0.5g/1, respectively. Ferrous sulfate, manganese sulfate and zinc sulfate showed positive effect to cell growth with the optimal range of 5-10 ppm. In the feeding experiment with 3 and 5% incorporation of the gas oil grown yeast, neither adverse effects on growth of chicks nor toxic effect were observed. Protein content of the dried cell was 58.8% and its amino acid composition compared well with other single-cell protein products and FAO reference protein.

SMD 타입 태양전지 어레이를 이용한 white GaN LED용 전원 공급 장치 (Power Supply for White GaN LED by Using SMD Type Solar Cell Array)

  • Using six SMD(surface mount device) type AlGaAs/GaAs single junction solar cells connected in series, a power source was fabricated for a white GaN LED. The electrical properties of the power source was measured and analyzed under one sun (100mW/$cm^2$) and various indoor light (300 - 900 lux) conditions. Under 600 lux indoor light condition, output power was 17.06 ${\mu}W$ and it was 30.75 ${\mu}W$ under 900 lux indoor light condition. Using the fabricated solar cell power supply, we have turned on the white GaN LED. It was worked well under 15 ${\mu}W$(at 480 lux) power supplied from solar cell array. This kind of solar cell power supply can be used as a power source for ubiquitous sensor network (USN).

  • Autophagy is an evolutionarily well-conserved cellular homeostasis program that responds to various cellular stresses and degrades unnecessary or harmful intracellular materials in lysosomes. Accumulating evidence has shown that autophagy dysfunction often results in various human pathophysiological conditions, including metabolic disorders, cancers, and neurodegenerative diseases. The discovery of an autophagy machinery protein network has revealed underlying molecular mechanisms of autophagy, and advances in the understanding of its regulatory mechanism have provided novel therapeutic targets for treating human diseases. Recently, reports have emerged on the involvement of autophagy in oral squamous cell carcinoma (OSCC). Although the role of autophagy in cancer therapy is controversial, the beneficial use of the induction of autophagic cell death in OSCC has drawn significant attention. In this review, the types of autophagy, mechanism of autophagosome biogenesis, and modulating molecules and therapeutic candidates affecting the induction of autophagic cell death in OSCC are briefly described.

Investigation of gene expression in primary embryonic cell line (FGBC8) from olive flounder Paralichthys olivaceus

  • In this paper, we have reported the expression of immune-related gene in a new embryonic cell line (FGBC8) which was established from olive flounder (Paralichthys olivaceus) embryos. To explore the cell biotechnological applicability, the FGBC8 cells were incubated with the several mitogens such as lipopolysaccharide (LPS), polyinosinic-polycytidylic acid (poly I:C), flagellin, and interferon (IFN)-γ. After incubation, the expression of immune-related gene was observed in FGBC8 cells through the quantitative real-time PCR. Our results indicate that FGBC8 cells will serve as a valuable research tool for investigating host-pathogen interactions as well as cell biotechnological applications.
