우분에 퇴비화 첨가제 수준이 퇴비화 과정 중 이화학적 성상변화와 줄무늬 지렁이 생존에 미치는 영향 (Effects of Mixing Ratios of Cow Manure and Composting Bulking Agent on Physico-Chemical Characteristics and Earthworm (Eisenia foetida) Survival)

  • 본 연구는 가축분의 퇴비화에 일반적으로 첨가되는 왕겨를 수준별(0(CRH0구), 10(CRH10구), 20(CRH20구), 30(CRH30구) 및 40%(CRH40구))로 우분과 혼합한 다음 부숙기간별 이화학적 변화를 측정하고 이를 바탕으로 생존율을 조사하여 가축분의 효율적인 vermicomposting의 기초자료로 이용하고자 실시하였다. 그 결과를 요약하면 다음과 같다. 부숙이 경과함에 따라 3주부터 모든구에서 지렁이의 생존이 가능하였다. 처리구별 탄질비는 왕겨 혼합구가 CRH0구 보다 유의하게 높게 나타났으며, 왕겨 혼합구에서도 왕겨 수준을 30~40%를 혼합한 구(CRH30, CRH40)가 부숙기간 중 가장 높게 나타났다(p<0.05). 지렁이가 생존하기 시작한 3주째의 탄질비는 23.26~34.44로 나타났다. 부숙이 경과함에 따라 pH와 전기전도도는 CRH0구에서 가장 높았고(p<0.05), 왕겨 혼합 비율이 높을수록 pH와 전기전도도는 낮은 경향이었다. 지렁이가 생존하기 시작한 pH와 전기전도도는 각각 7.58~7.74와 0.41~1.17 mS/cm으로 나타났다. 이상의 결과를 종합해 보면 우분에 왕겨를 수준별로 혼합하여 부숙 시켰을 경우 이화학적 성상 변화는 모두 지렁이 생존이 가능한 범위로 나타났으나, 우분의 효율적인 지렁이 퇴비화를 위해서는 왕겨 혼합 비율에 따른 지렁이의 생육과 증식 조사를 위한 시험이 필요하다고 사료되어 진다.

  • The composting practice has been recognized as the most popular way of controlling food waste and many attempt have been made in the field to establish more efficient and economical process. Some of the efforts are mixing cured compost with sawdust as alternative bulking agent, seeding commercially produced microorganism and/or combination of above. However, verification of such efforts is often restricted because of either the lack of engineering consideration on the limitation of composting facility scales. In this study, the effect of mixing materials in food waste composting was investigated by controlling the combination and the mixing ratio of them. When the cured compost was mixed with saw dust. the decomposition of organic material was proven to be more active by observing the compost temperature, the oxygen (O$_2$) consumption, and the cumulative carbon dioxide ($CO_2$) profile. However, the quantity of compost mix-ing seemed not to influence the reaction as long as the minimum required amount was mixed. The feeding of com-mercially produced microorganism had a tendency to prolong the thermophilic stage, which helped to increase the decomposition but it resulted in composting period. Regardless of the composting condition, bacteria and actinomycetes increased in population as the reaction approached to the end. The population of bacteria and actinomycetes were rel-atively higher than those of fungi and yeast throughout the reaction.

Enzymatic Production of D-Tagatose, a Sugar-substituting Sweetener, from D-Galactose

  • D-Tagatose is a potential bulking agent in food as a non-calorific sweetener. To produce D-tagatose from cheaper resources, plasmids harboring the L-arabinose isomerase gene (araA) from Escherichia coli was constructed because L-arabinose isomerase was previously suggested as an enzyme that mediates the bioconversion of galactose to tagatose as well as that of arabinose to ribulose. In the cultures of recombinant E.coli with pTC101, which harboring araA of E.coli, tagatose was produced from galactose in 9.9 % yield. The enzyme extract of E.coli containing pTC101 also converted galactose into tagatose in 96.4 % yield. For the economic production of D-tagatose, an L-arabinose isomerase of E.coli was immobilized using covalent binding on agarose. While the free L-arabinose isomerase produced tagatose with the rate of 0.48 mg/U$.$day, the immobilized one stably converted galactose into average 7.5 g/l$.$day of tagatose during 7 days with higher productivity of 0.87 mg/U$.$day. In the scaled up immobilized enzyme system, 99.9 g/l of tagatose was produced from galactose with 20 % equilibrium in 48 hrs. The process was stably repeated additional 2 times with tagatose production of 104.1 and 103.5 g/l.

Effects of Leachate during Vegetable Waste Composting using Rotary Drum Composter

  • In India, disposal of vegetable market waste along with municipal solid waste in landfills or dumpsites is creating much nuisance in terms of odor nuisance, leachate production, and greenhouse gas emission into the atmosphere. Therefore, vegetable waste with high biodegradable and nutrient content is composted in a 550-L batch scale rotary drum composter to study the degradation process and its compost properties for its potential reuse as high quality compost. A total 150 kg of working volume was fixed for composting studies with two different ratios, trial A (6:3:1) of C/N 24 and trial B (8:1:1) of C/N 30, respectively. A maximum of $63.5^{\circ}C$ and $61.2^{\circ}C$ was observed in trials A and B; an average of $55^{\circ}C$ for more than 5 days, which helped in the degradation of organic matter and reduction of total and fecal coliform. The temperature dropped suddenly after the thermophilic stage in trial B, and leachate was observed due to insufficient amount of bulking agent. Mesophilic bacteria dominated during the initial stages of composting, and reduced considerably during the thermophilic stage. During the thermophilic stage, the rise in spore-forming organisms, including spore-forming bacteria, fungi, actinomycetes and streptomycetes, increased and these were predominant until the end of the composting process. By examination, it was observed that moisture and leachate production had adverse effects on the compost parameters with higher loss of micronutrients and heavy metals.

가축 분뇨의 퇴비화 수분 조절제로서 Kenaf의 유용성 평가 (Studies on the Evaluation of Kenaf as a Bulking Agent in Livestock Composting)

  • 본 시험은 무궁화과에 속하는 초본 식물인 Kenaf를 경상대학교 시험농장에서 2010년 6월 1일 파종하여 같은 해 11월 18일 수확하여 건조 한 후, 축산분뇨의 수분조절제로서의 가치를 평가하였다. 건조된 Kenaf를 다시 bast, core, leaf으로 분류하여 분쇄를 하였으며, 각각의 분쇄물에 대한 수분 흡수율, 수분 증발률, pH변화, 용적중, C/N비를 측정하였다. 분석 결과 수분흡수율은 대조군으로 사용된 왕겨 및 톱밥과 비하여 높은 수분 흡수율과 증발률을 보였고, pH는 Kenaf의 시료 별 증류수와 72시간 반응 후 측정해 본 결과 pH $2.8{\pm}0.01$ - $4.34{\pm}0.02$ 범위로 톱밥 (pH $5.28{\pm}0.01$) 및 왕겨 (pH $6.3{\pm}0.02$)에 비해 다소 낮게 나타났다. C/N비 분석결과 Kenaf의 core가 649, bast 204, leaf 70 순으로 나타나 대조군으로 사용된 톱밥 (789.1) 보다는 다소 낮았으나 왕겨 (132) 보다는 높은 비율을 보였다. 용적중 시험에서는 Kenaf는 각 시료의 입경에 따라 A(${\geq}4cm$), B(${\leq}4cm$, ${\geq}0.25cm$) 및 C 군(${\leq}0.25cm$) 으로 구분하였고, 시험 결과 bast, core 및 leaf의 경우 입경이 큰 A 군과 B 군은 톱밥 및 왕겨와 비슷한 용적중을 보였으며, 입경의 크기가 작은 C 군의 경우 다소 높은 용적중을 보였고, 특히 leaf C 군의 경우 톱밥 및 왕겨에 비해 5배 이상의 높은 용적중을 보여 입경이 작을수록 용적중이 높은 것으로 나타났다. 본 연구는 축산분뇨의 퇴비화를 위해 Kenaf를 이용 수분조절제로서 활용 가능성을 제시하였고, 이러한 결과는 농가의 비용감소, 축산분뇨로 인한 환경오염 예방은 물론 친환경적 처리가 가능 할 것으로 사료된다.

음식쓰레기의 퇴비화공정의 적정운전조건 검토 (The study of Efficient Treatment Conditions on the Composting of Foodwaste)

  • 음식물쓰레기 이용 퇴비화의 운전 및 염분저감을 위한 적정조건 검토을 위한 연구를 수행하여 이하의 결과를 얻었다. 음식쓰레기와 톱밥을 7:3, 6:4, 5:5, 4:6, 3:7 비율로 혼합하여 실험한 결과 6:4에서 가장 안정화된 퇴비가 생산될 수 있었다. 한편 음식물쓰레기 : 톱밥의 혼합비가 7:3 및 음식물쓰레기 : 왕겨의 혼합비가 6.5 : 3.5 와 6:4의 경우는 초기 C/N비 자체가 낮게 나타나 적정 퇴비화를 이룩하기 어려웠다. Agent로 톱밥을 혼합한 경우와 왕겨를 혼합한 경우 모두 pH가 급격히 상승하다가 안정화되어 7.5~8.5를 유지하였다. 수분함량은 왕겨를 사용한 경우가 톱밥을 사용한 경우보다 수분감량율이 높게 나타나 톱밥보다 공극율 및 통기성이 원활함을 알 수 있었다. 따라서 톱밥대체제로 이용가능 했다. 탄소화합물 함량은 톱밥의 경우 음식물쓰레기의 혼합 비율이 낮을수록, 왕겨의 경우 음식물쓰레기의 혼합 비율이 높을수록 높았다. 유해성분(납외7종)은 비료기준치 이하를 나타내 안전한 것으로 판별되었다. 염분의 경우 0.43-0.46%을 나타내어 비료기준치(1%)의 1/2로 저감되었다. 이상의 결과를 종합해 보면 음식물쓰레기 퇴비화에 있어 음식물쓰레기와 Agent의 혼합비가 6:4 일 때, Agent로는 왕겨보다 톱밥을 혼합할 때 효율적인 것으로 나타났다.

난지도 퇴비화시범시설에서의 퇴비화과정 검토 (Performance of the Nanjido Composting Facility)

  • 일일 처리용량이 10톤인 난지도 음식쓰레기 퇴비화시범시설을 대상으로 하여 퇴비화과정동안의 물리 화학적 특성변화에 관한 연구를 수행하였다. 투입되는 음식쓰레기의 수분함량은 79% 내외였으며 공극 개량제로 폐목재를 샤용하였다. 퇴비화가 진행되는 동안 수분은 최종 31%로 감소하였으며 밀도에 상당한 영향을 주었다. 또한, 전기전도도는 퇴비화가 진행되는 동안 증가하는 경향을 보였다. 퇴비화가 진행되는 동안 VS와 TOC의 감소율은 각각 40%와 28%를 보였으며 VS와 TOC와의 상관관계는 유의수준에서 1.00으로 매우 상관성이 높았다. 고형물 C/N비와 물용출 C/N비 모두 숙성퇴비의 값을 보였다. 또한, $CO_2$, $O_2$ 등의 가스조성을 분석한 결과 퇴비화는 호기성조건하에서 정상적으로 진행되고 있었다.

Effect of Extenders with TCG and DMSO on the Viability of Rabbit Sperm

  • The purpose of this study was to evaluate the effect of addition of ethylene glycol, glycerol and sucrose to TCG (Tris, Citric Acid, Glucose, Egg Yolk) and DMSO Frozen. The extender containing Egg yolk concentration (10%, 20%) affects viability and acrosome morphology of rabbit sperm. Sperm viability was then assessed for the freezing extenders TCGD (Tris + Citricacid + Glucose + DMSO), TCGED (Tris + Citricacid + Glucose + Egg yolk + DMSO), TCGGD (Tris + Citricacid + Glucose + Glycerol + DMSO) and TCGSD Tris + Citricacid + Glucose + Sucrose + DMSO) during thawing at 38℃. for 20 seconds, respectively. TCG + 10% egg yolk (viability: 77.0 ± 0.8, NAI: 73.3 ± 0.9) was significantly (sperm viability and normal acrosome interaction (NAI)) higher than TCG + 20% egg yolk (70.7 ± 1.1, 70.0 ± 0.9) in the sperm normalcy analysis according to the yolk concentration. TCGGD (53.4 ± 0.1, 62.3 ± 0.4), TCGSD (61.3 ± 0.0, 67.1 ± 0.1) sperm viability and normal acrosome interaction (NAI) in frozen spermatozoa are TCGD (46.4 ± 2.8 and 56.3 ± 1. 4) and TCGED (23.0 ± 1.1 and 54.6 ± 1.4) extenders was thawed at 38℃ for 20 seconds. According to the results from each frozen bulking agent, sperm membrane integrity by hypotonic swelling test (HOST) analysis in TCGGD (59.8 ± 0.7), TCGSD (59.3 ± 0.5) was significantly high compared to other experimental groups (p < 0.05). In conclusion, these results suggested that TCGGD and TCGSD extenders enhance survivability of rabbit sperm after frozen-thawing.

Solid State Fermentation Reactor를 이용한 유기성 폐기물의 발효 (Composting of Organic Wastes by solid State Fermentation Reactor)

  • Leaves of Aloe vera Linne and bloods of domestic animal were composted in a soild state fermentation reactor (SSFR) by using microbial additive including a bulking and moisture controlling agent. From solid-culture of microbial additive, 10 species of bacteria and 10 species of fungi were isolated and, their enzyme activities including amylase, carboxy methyl cellulase CMCase, lipase and protease were detected. Optimum fermentation conditions of Aloe leaves and domestic animal bloods in SSFR were obtained from the studies of response surface analysis employing microbial additive content, initial moisture content, and fermentation temperature as the independent variables. The optimum conditions for SSFR using Aloe leaves were obtained at 9.45$\pm$73%(w/w) of microbial additives, 62.73$\pm$4.54%(w/w) of initial moisture content and 55.32$\pm$3.14$^{\circ}C$ of fermentation temperature while those for SSFR using domestic animal bloods were obtained at 10.25$\pm$2.04%, 58.68$\pm$4.97% and 57.85$\pm$5.$65^{\circ}C$, respectively. Composting process in SSFR was initially proceeded through fermentation and solid materials were decomposed within 24 hours by maintaining higher moisture level, and maturing and drying steps are followed later. After the fermentation step, the concentrations of solid phase inorganic components were increased while that of organic components were decreased. Also, concentrations of total organic carbon(TOC), peptides, amino acids, polysaccharides, and low fatty acids in water extracts were increased. As fermentation in composting process depends on initial C/N ratios in water extracts of two samples were increased because of increased water-soluble TOC. From these results, it was revealed that solid state fermentation reactor using microbial additives can be used in composting process of organic wastes with broad C/N ratio.

갑오징어(Sepia esculenta)갑 칼슘으로 회수한 surimi 가공폐수 단백질의 어묵소재로서 이용 (Utilization of a Soluble Protein Recovered from Surimi Wastewater by Calcium Powder of Cuttle, Sepia esculents Bone)

  • Utilization of soluble protein recovered from surimi wastewater using calcium powder of cuttle bone were examined. The crude ash content of the heat-induced surimi gel was increased linearly by increasing substitution ratio of recovered protein-ATC toward commercial surimi. Moisture (approximately $76\%$) and lipid $(0.2\%)$ contents were not change, but their protein contents were decreased 15.7 to $14.3\%$ depend on increasing of substitution ratio. The white index of the heat-induced surimi gel by color meter was increased up to $10\%$ of substitution ratio. There were no difference between $0\%\;and\;5\%$ substituted surimi gel in the gel strength. The sensory score on white index and texture of the heat-induced surimi gel did not change in 0 to $10\%$ as a substitution ratio of recovered protein-ATC toward commercial surimi, while decreased in more $15\%.$ The optimal substitution ratio of recovered protein-ATC as a bulking agent was $10\%.$ The heat-induced surimi gel prepared with $10\%$ substitution of recovered protein-ATC was similar to the content and composition of total amino. acids, and superior to calcium content and the ratio of calcium and phosphorus toward those of commercial surimi.
