Effects of Different Dietary Protein Sources on Apparent Digestibility and Growth in Juvenile River Puffer Takifugu obscurus (단백질원료의 종류에 따른 황복(Takifugu obscurus) 치어의 소화율 및 성장 평가)

  • An 8-week feeding trial was conducted to determine the apparent dry matter and protein digestibility of seven different dietary protein sources by juvenile river puffer Takifugu obscurus. Eight diets containing white fish meal (WFM), brown fish meal (BFM), squid liver powder (SLP), krill meal (KM), leather meal (LM), soybean meal (SM), or fermented soybean meal (FSM) were prepared by mixing a basal diet (BD) with one of the seven test ingredients at a ratio of 7 to 3. Fish averaging $10.8{\pm}0.04g$ were fed the experimental diets in triplicate groups. The apparent dry matter digestibilities of BFM, WFM, FSM, SLP, LM, SM, and KM were 80, 78, 72, 67, 56, 55, and 54%, respectively, while the corresponding apparent protein digestibilities were 96, 96, 93, 92, 89, 88, and 86. The weight gain of fish fed BFM was significantly greater than that of fish fed SM or LM (P<0.05), while it was not significantly different from that of fish fed the BD, WFM, FSM, BFM, SLP, or KM. These results indicated that WFM, BFM, FSM, and SLP are potentially good protein sources for replacing fish meal in river puffer feed.

The Detection of Irradiated Agricultural Commodities by Origins with Photostimulated Luminescence (PSL) Analysis (Photostimulated Luminescence (PSL) 분석에 의한 원산지별 농산물의 방사선 조사여부 검지)

  • Photostimulated luminescence (PSL) analysis was applied to determine whether some agricultural commodities of Korean and Chinese origins have been irradiated or not. Cereals (brown rice and buckwheat) and legumes (soybean, peanut, fed bean and mung bean) were exposed to gamma irradiation at 0.5 ∼4 kGy The Korean garlic was irradiated at 0.05 ∼0.5 kGy. Cereals and mung bean were tool applicable to the PSL measurement, showing the intermediate values between the lower (700 photon counts, negative) and the upper threshold values (5,000, positive), however PSL values determined for soybean, peanut, red bean and garlic were suitable for being discriminated between nonirradiated and irradiated samples. There was no significant difference in PSL properties accgrding to the sample origins.

Multiplex PCR Assay for the Simultaneous Detection of Major Pathogenic Bacteria in Soybean (콩에 발생하는 주요 병원세균의 동시검출을 위한 다중 PCR 방법)

  • Bacterial diseases in soybean are bacterial pustule by Xanthomonas axonopodis pv. glycines, wildfire by Pseudomonas syringae pv. tabaci, bacterial blight by Pseudomonas savastanoi pv. glycines and bacterial brown spot by Pseudomonas syringae pv. syringae in Korea. It is difficult to identify each disease by early symptoms in fields, because the initial symptoms of these diseases are very similar to each other. In this study, we developed multiplex PCR detection method for rapid and accurate diagnosis of bacterial diseases. The glycinecin A of X. axonopodis pv. glycines, the tabtoxin of P. syringae pv. tabaci, the coronatine of P. savastanoi pv. glycines and the syringopeptin of P. syringae pv. syringae have been reported previously. These bacteriocin or phytotoxin producing genes were targeted to design the specific diagnostic primers. The primer pairs for diagnosis of each bacterial diseases were selected without nonspecific reactions. The studies on simultaneous diagnosis method were also conducted with primarily selected 21 primers. As a result, we selected PCR primer sets for multiplex PCR. Sizes of the amplified PCR products using the multiplex PCR primer set consist of 280, 355, 563 and 815 bp, respectively. This multiplex PCR method provides a efficient, sensitive and rapid tool for the diagnosis of the bacterial diseases in soybean.

Growth Condition of Liquid Culture by Pleurotus ostreatus (액체배양에서 느타리버섯균의 적합한 생장조건 구명)

  • For the practical using of liquid spawn we carried out selection test of nutrient sources, cultural methods and cultural apparatus for liquid spawn production of oyster mushroom(ASTI 2001, ASTI 2018, ASTI 2072, ASTI 2016, ASTI 2070, ASTI 2180, ASTI 2183, ASTI 2042). The optimal temperature and pH range for mycelial growth of Pleurotus species were $25^{\circ}C\;to\;30^{\circ}C$ and 5.5 to 6.5, respectively. The effect of carbon sources, nitrogen sources and mineral salts on the mycelial growth was studied using petridish culture. Generally, the disaccharides and polysaccharides showed good effect for mycelial growth of Pleurotus species, and the polysaccharides were superior to the other classes of carbon sources for mycelial growth. For the mycelial growth of the 8 oyster mushroom stains, soybean flour was superior to the other kinds of nitrogen sources. On the other hand, addition of mineral salts did not affect, and even poor under certain mineral salts, the mycelial growth of the 8 oyster mushroom stains. The brown sugar selected out the carbon source of the agricultural medium. Also the soybean flour selected out the nitrogen source of agricultural medium. In the medium selection, we selected out agricultural optimum medium composed of brown sugar 3%, soybean flour 0.3%, potassium phosphate 0.05%, magnesium sulfate 0.05%. Under the 250 ml erlenmeyer culture, the effects of such factors as the inoculum rate, the working volume, cultural method and flask shapes on the mycelial growth were examined. The optimal inoculum rate and working volume on the mycelial growth of oyster mushroom was 2 mycelial disk (diameter 6 mm) and 100 ml, respectively. The shake flask culture was enhanced the mycelial growth than at the erlenmeyer flask. Pulp form growth of mycelium in the erlenmeyer flask culture was obtained in the culture with glass rod of length 50 mm, diameter 10 mm.

Evaluation of Rheological and Functional Properties of Roasted Soybean Flour and Mixed Cereals Fermented by Bacillus sp. (고초균을 이용한 볶은 콩과 곡류 혼합 발효물의 물성 및 기능성 평가)

  • Roasted soybean flour (RSF) and mixed cereals were fermented by the solid state fermentation using Bacillus subtilis HA to optimize the production of biologically active compounds. The RFS fermented with 52.7% moisture content showed higher production with protease activity of 42.6 unit/g and 10% mucilage content after fermentation for 24 hr. Tyrosine content and protease activity after 48 hr fermentation time were the highest values with 445.5 mg% and 55.1 unit/g, respectively. However, the wholesome fermented RSF (FRSF) was obtained by fermentation for 24 hr because of the production of unpleasant flavors after fermentation for 48 hr. The RSF fermented with various types and contents of cereals has no effects on tyrosine content and protease activity. However, the addition of brown rice significantly increased mucilage content, especially indicating 24.55% at the addition of 80% (w/w). For addition of barley, fibrinolytic activity was increased to 11.82 unit/g by the fortification of 60% barley. It is concluded that biologically active compounds including fibrinolytic activity and mucilage content in FRSF were dependent upon the type and content of various cereals.

Soybean Sprout Rot Caused by Colletotrichum species (Colletotrichum species에 의한 콩나물 부패)

  • Two novel casual agents of soybean sprout rot occurred at soybean sprouts cultivated under structure in Suwon area in 1997 were isolated and their pathogenicity was tested in vivo. An isolate formed crowed, black acervuli which were oval to elongated with numerous black, needlelike, intermixed long and short setae, 65~110$\times$3.5~6.6 ${\mu}{\textrm}{m}$. Conidia were curved, lunate, unicellular and hyliane and measured 21.5~22.5$\times$3.5~4.0 ${\mu}{\textrm}{m}$. The other isolate produced conidia with straight and cylindrical, and measured 14.0~17.5$\times$3.5~4.5 ${\mu}{\textrm}{m}$. Apressorium size was measured 6.3~8.5$\times$4.5~5.0 ${\mu}{\textrm}{m}$. The agents were identified as Colletotrichum truncatum and C. gloeosporioides based on their morphological characteristics. There was a large difference in pathogenicity between two isolates. C. gloeosporioides caused dark brownish discoloration of whole plants. It showed high pathogenicity with severe disease development. Meanwhile C. gloeosporiodes caused light brown spots on cotyledon and its pathogenicity was not strong. The soybean sprout rot occurred by the two Colletotrichum species was firstly reported in soy-bean sprout in Korea, and we suggest it as “Colletotrichum rot of soybean sprout”.

Effect of Deodorizing Conditions on Color in Soybean Oil (탈취조건이 대두유의 색상에 미치는 영향)

  • Deodorized soybean oils(DSO) were obtained to 2 types. Sample 1 was carried out a general refining process, which was degumming-alkali refining-bleaching-deodorizing. Sample 2 and 3 were not general refining process, its were carried out only both decantation of insoluble impurities and batch type deodorizing(BTD). At this time, BTD was composed of 3 stages, operating differences were vacuum degree, maximum temperature, stripping steam and retention time etc. DSO obtained were appeared original physicochemical characteristics. Sample 1 had acid value 0.034, Lovibond color, 9.1Y/0.9R, peroxide value 0 and thin yellow color's soybean oil. But sample 2 had acid value, 0.078, Lovibond color, 65.0Y/18.39R/4.2B/0.1N, peroxide value, 0.7 and bright green color's soybean oil. Sample 3 had acid value, 0.072, Lovibond color, 37.3Y/3.8R/0.1B/0.1N, peroxide value 1.6 and dark brown color's soybean oil. These colorful DSO were expects as raw-materials of various seasoning oils as like pine needle and/or perilla leaf seasoning oil.

Physiological and Genetic Characteristics of Cultivated Mushroom, Hypsizygus marmoreus

  • A edible mushroom, Hypsizygus marmoreus is commercially cultivated in Northeast Asia. Japan's annual production is 110,000ton or more. Since 2002, cultivation is expanded in Korea. To investigate the morphological, cultural and microscopic characteristics of Hypsizygus marmoreus, 109 isolates were collected from Korea and other countries. Clamp connection, chlamydospore and arthrospore were present in all tested isolates of H. marmoreus except HYM-002 and HYM-004. Also pilealtrama, gilltrama, basidia, basidiospore and cystidia of fruiting body were no difference among the isolates in the present investigation. Morphological characteristics of fruiting body was that color of pileus was brown and white, irregular as marble, the average size 12~22mm and stipes was $46{\sim}91{\times}6{\sim}10mm$. Isolates HYM-031, HYM-047 and HYM-109 formed grayish-brown pileus with a faint pattern. Molecular analysis with RAPD and ITS rDNA sequence analysis were also performed to check the genetic relationships among H. marmoreus isolates. Based on the RAPD analysis using the URP-PCR, all isolates of H. marmoreus were clustered into large 3 groups but more than 90% showed high similarity. In addition, morphological and geographical differences have been classified as an independent cluster. The brown and white strains enclosed in same cluster. So genetically no significance difference was observed between these two strains. ITS gene sequences of 16 selected isolates which were 640 bp long, were aligned and compared. The similarity in ITS sequence was 94.8 to 99.1% among tested isolates and the H. marmoreus isolates in GeneBank. In conclusion the tested isolates were H. marmoreus. Morphological and molecular observations proved that all tested isolates were belonging to H. marmoreus. For the stable artificial cultivation, composition of optimum media, mature period and light condition were established. Optimal formula of artificial cultivation medium was Douglas sawdust: corn cob: soybean meal: wheat bran = 40:30:15:15. In addition, 7% rice bran and 3% yellow sucrose was the most effective composition for spawn's liquid medium. For the maturation of the isolates was favorable for growing for 20 to 30 days at $25^{\circ}C$ and the LED lights in mixture of white and blue was good for growth period. For effective growth, the temperature, humidity and aeration control in every step was important.

Strategies to Reduce Phytate Content in the Korean Diet

  • High dietary phytate is a known factor in reducing the bioavailability of minerals such as zinc and calcium which are already chronically low in the Korean diet. This study was conducted to develop methods for reducing dietary phytate through the addition of phytate and/or the substitution of high phytate foods with low phytate foods. Ten units of phytase per 100g of uncooked brown rice were added to brown rice gruel resulted in a 16.2% phytate reduction after a 3-hour incubation period; an 18.2% reduction was produced after a 6-hour incubation period. The addition of ten units of phytase per 100g of soybean curd residue at 45$^{\circ}C$, followed by refrigeration for 3 hours, resulted in a 19.1% phytate reduction. The addition of 20 units of phytase under the same conditions reduced phytate content by 24.6%. In this study, two typical Korean meals consisting of legumes and unrefined cereals were prepared as high phytate meals; these were then compared to low phytate meals that had been prepared by treating the foods with phytase and substituting unrefined with refined cereals (i.e., brown rice with white rice, whole wheat bread with white bread). The phytate content of the two high phytate meals was 1878.2mg and 1811.8mg. After the addition of phytase and the food substitution, the phytate content of the low phytate meals was reduced to 788.9mg and 606.0mg. The phytate to zinc molar ratio of high phytate diets was 22.4 and 21.3 and 9.4 and 7.9 for the low phytate meals. These results indicate that the nutritional status of Koreans in terms zinc and other minerals can be improved by phytate reduction. This can be accomplished through the change of milling process for some cereals and/or the enzyme treatment of some high phytate food items.

Prevalence and Thermal Stability of Enterobacter sakazakii from Unprocessed Ready-to-Eat Agricultural Products and Powdered Infant Formulas

  • Enterobacter sakazakii, designated as an unique microbial species in 1980, may cause bacteremia, necrotizing enterocolitis and infant meningitis. The distribution and the thermostability of E. sakazakii in unprocessed ready-to-eat (RTE) agricultural products of 252 and in 25 powdered infant formulas (PIF) were analyzed. Eighty one, 50, 43, and 47% of brown rice, pumpkin, potato, and carrot samples, respectively, had aerobic plate counts (ARC) in the range of 5 log CFU/g or more. Almost all the other products sampled had APC of approximately 2 log CFU/g. Fifty three, 75, 67, and 68% of banana, pumpkin, soybean, and carrot had Enterobacteriaceae counts approximating 3 log CFU/g. Sixty six percent of the brown rice tested had Enterobacteriaceae counts approximating 5-6 log CFU/g. E. sakazakii was isolated from 3/25(12%), 4/23(17%), 1/24(4%), and 1/27(4%) of PIF, brown rice, laver, and tomato samples, respectively. D-values were 3.52-4.79 min at 60 and $D_{60}-values$ were similar as the isolates reported. Thermal inactivation of four thermovariant E. sakazakii strains during the rehydration of PIF with hot water were investigated. At $50^{\circ}C$, the levels of E. sakazakii decreased one log CFU/g for 4-6 min and thereafter the levels remained stable for 20 min. At $60^{\circ}C$, inactivation by about 2 log CFU/g occurred for 20 min. Therefore, the unprocessed agricultural products might be a source of contamination for PIF when used as an ingredient after drying and pulverization. Rehydration of PIF for infant feeding with a water temperature of $60^{\circ}C$ rather than $50^{\circ}C$, as recommended by the manufacturers, may be helpful in the reduction of potential E. sakazakii risk.
