Trichloroethylene(TCE)의 분해를 위한 메탄자화균총의 분리 및 배양 (Isolation and Cultivation of Methanotrophic Consortium for Trichloroethylene Degradation)

  • Two unidentified methanotrophic strains (MM-white and MM-red) secreting soluble methane monooxygenase (sMMO) involved in thrichloroethylene biodegradation have been isolated from mixed methanotrophic consortium (MM) around Taejon area. Subsequently four methanotrophic strains were isolated from MM and named according to their color; white (MS-white), yellow (MS-yellow), pink (MS-pink) and reddish brown (MS-rbrown). All strains except MS-yellow which can take glucose as well as methane, metabolized methane as a sole carbon source. They all showed symbiotic behavior when methane was used as the sole carbon source. Optimum conditions of cell growth for MM were pH of 6.8 - 7.2, temperature of 29 - 32$^{\circ}C$, and gas flow rate of 6 (for methane), 40 (for air), and 4 ml/min (for carbon dioxide). The sMMO activity was expressed as naphthalene oxidation rate (${\mu}$mol/ mg protein/ hr). The sMMO activity for MM grown in flask culture with 1 ${\mu}$M of CuSO4 was 36, while it was 61 without copper. The activity for MM grown in the fermentor without CuSO4 was 1077, but is was 197 after reaction with 5 ppm of TCE. The methanotrophs showed significantly high sMMO activity despite the presence of 1 ${\mu}$M of CuSO4, although most of other strains already known could not express sMMO activity under this condition.

절임 고추장 종류가 다른 박대장아찌의 저온 저장 중 품질특성 (Quality Characteristics of Tongue Sole (Cynoglossus semilaevis) Jangajji with Different Types of Gochujang during Low Temperature Storage)

  • In this study, pH, $^{\circ}Brix$, salt concentration, sodium content, color, hardness, and sensory evaluations were measured during storage at low temperatures for 21 days after making tongue sole Jangajji with different types of Gochujang sauces. The pH was higher than those of samples not containing Maesil fermented liquor at all storage periods. The $^{\circ}Brix$ of the Maesil fermented liquor added samples was higher than that of the samples not added at the early stages of storage. On the other hand, there was no significant difference between the samples with and without Maesil fermented liquor at the later stages of storage. The sodium content was lower than that of the samples not containing Maesil fermented liquor. The lightness (L) was lower in the early stages of storage than those of the Maesil fermented liquor. In the late stage of storage, however, the lightness was higher than those of the samples not containing Maesil fermented liquor. The redness (a) of the GRWO sample group showed the highest value at all storage periods compared to the other sample groups. The yellowness (b) of the GRWO sample group was high at the early stages of storage. On the $21^{st}$ day of storage, however, the GRW sample group was significantly higher (p<0.01). The hardness results showed that the value of the later storage period was lower than that at the initial storage stage. The sensory evaluation showed that the Maesil fermented liquor reduced the spicy, salty, bitter, and unpleasant taste. As a result of the acceptance test, the GRWO and GRW samples were evaluated as the acceptance group with color, aroma, texture and overall acceptability compared to the brown rice Gochujang sample group.

Amylase와 Protease의 활성이 높은 현미 발효 미생물의 선별 (Screening for Fermentative Microorganisms that Grow on Brown Rice with High Amylase and Pretense Activities)

  • 현미는 백미에 비하여 식품영양학적 가치 및 식이섬유의 함유량이 높은 쌀겨 및 배아를 포함하고 있으나 소화가 잘되지 않는 단점 이 있다. 따라서 발효현미는 소화력을 높임과 동시에 좋은 영양 공급원이 될 수 있으므로 높은 amylase와 pretense의 활성으로 현미를 발효할 수 있는 미생물을 선별하였다. 2.5%(w/v)의 현미분말을 유일한 영양원으로 한 액체배지를 생장배지로 사용하여 생장능력과 효소 생산능력을 조사하였다. 조사한 8종의 Bacillus 와 11종의 유산균 중에서 모든 Bacillus 균주와 두 종의 유산균이 생장과 효소활성을 보였다. 생균수는 $10^7CFU/mL$을 초과하였으며 Bacillus sp. Bacillus sp. B2, Bacillus sp. B11, Leuconostoc gelidum, Pediococcus pentosaceus 가 생산하는 최고 amylase 활성은 각각 17.85, 17.50, 17.10, 17.10, 3.24 U/ml이었고, 최고 pretense 활성은 각각 22.48, 22.04, 23.76, 12.13, 3.80 U/ml이었다. 따라서 이 균주들은 발효 현미 제조를 위한 접종균주로서 이용이 가능하리라 사료된다.

Evaluation of energy and amino acids of brown rice and its effects on laying performance and egg quality of layers

  • Two experiments were conducted to determine apparent metabolizable energy (AME), nitrogen-corrected AME (AMEn), and ileal digestible amino acid (AA) content of brown rice (BR) and to investigate the effect of dietary supplementation of BR on laying performance and egg quality of laying hens. In Exp. 1, 72 Hy-line Brown layers (49-week-old) were allocated to two treatments using a completely randomized block design, and each treatment included six cages per treatment and six hens per cage. A semi-purified diet was formulated to include BR as the sole source of AA and energy and an N-free diet was used to determine basal endogenous loss of AA. The hens were fed a commercial layer diet for adaptation to the experimental environment and diet for 7 days from d 0, and then fed experimental diets for 5 days from d 7. Excreta were collected from d 10 to 11 and ileal digesta were collected on d 12. On a dry matter (DM) basis, the AME and AMEn of BR was determined at 3,773 and 3,729 kcal/kg, respectively. The apparent ileal digestibility (AID) of BR ranged from 32.7% for Thr to 73.7% for Arg. The range of the standardized ileal digestibility (SID) value was between 79.4% for Met and 96.6% for Lys. In Exp. 2, 252 Hy-line Brown layers (44-week-old) were divided into four groups, comprising seven replicates of nine birds each and assigned to four experimental diets containing 0 (Control), 5%, 10%, or 15% BR for 5 weeks. The BR-containing diets were formulated to be equal in the content of AMEn and digestible AA to those of the diet without BR. No significant differences were observed in laying performances. Egg quality and blood profiles were not linearly or quadratically affected by dietary treatments. These results suggest that up to 15% BR can be included into layer feed without any adverse effects on laying performance and egg quality, if its energy and digestible AA values are well evaluated.

한국 동해안 참가자미, Pleuronectes herzensteini (Jordan et Snyder)의 성숙과 산란 (Maturity and Spawning of Brown Sole, Pleuronectes herzensteini (Jordan et Snyder) in the East Sea of Korea)

  • 2003년 4월부터 2004년 3월까지 우리나라 동해안 강원도 양양군 현남면 남애리 연안에서 어획된 참가자미를 대상으로 성숙과 산란에 관한 연구를 수행하였다. 암컷의 생식소 발달과정 및 생식소중량지수의 월 변화로부터 참가자미의 산란기는 2~5월, 주 산란기는 3~4월로 추정된다. 전장 (TL)과 포란수 (F)간의 관계식은 $F=11.307TL^{2.628}$으로 체장이 커질수록 포란수도 증가하는 경향을 보였다. 산란에 참여하는 군성숙체장은 전장 17.8 cm이었고, 이때의 연령은 3세이었다. 전 조사기간에 대한 성비 (암 수)는 60.2 : 39.8로 암컷이 우세하였고, 체급별 성비는 전장 17 cm 이하에서는 수컷의 비율이 높았으나 체장이 커질수록 암컷의 비율이 증가하여 27 cm 이상에서는 전 출현개체가 암컷이었다.

어육의 가열조건에 따른 몇가지 정미성분 함량의 변화 (Changes in Contents of Some Taste Compounds of Fish Meat by Heating Conditions)

  • 가열조건을 달리 하였을 때의 고등어, 꽁치, 조기 및 가자미의 핵산관련물질 및 유기염류(TMAO, TMA 및 총 creatinine)의 함량 변화를 조사한 결과는 다음과 같다. 각 시료에 있어서 공통적으로 inosine의 함량이 높았으며, 적색육어 보다는 백색육어에서 그 함량이 더 높게 나타났다. 그리고 가열을 함으로서 hypoxanthine의 함량은 증가하였으나, 그 외의 다른 성분들은 감소하였다. TMAO의 함량은 각 시료에서 생시료일 때에 31.1, 26.2, 49.4 및 58.5 mg%로 가자미에 있어서가 가장 많은 함량을 보였으며, 가열을 함에 따라서 그 함량이 감소하였다 그리고 TMA의 함량은 각 시료에서 3.7, 5.8, 22.9 및 16.9 mg%로 나타났으며, 조기의 경우가 가장 많았고, 가열처리를 함에 따라서 TMAO가 환원되어 그 함량이 증가하였다. 그리고 총 creatinine의 함량은 341.2, 469.8, 52.3 및 87.6 mg%로 백색육어 보다는 적색육어에서 더 많은 함량을 보였으며, 꽁치에서 가장 많았다. 가열을 함에 따라서 모든 시료에서 정미성분의 함량이 감소하였다.

서식지에 따른 다소비 어류의 총수은 오염 및 위해 평가 (Mercury Contamination and Risk Evaluation in Commonly Consumed Fishes as Affected by Habitat)

  • This study investigated the concentration of total mercury in 17 species of commonly consumed fishes and assessed the risk to human health from provisional tolerable weekly intakes PTWI% as affected by migratory characterization, such as migration and vagility. The measured mean concentrations of total mercury in these 17 species of commonly consumed fishes suggest that mean concentrations of total mercury in 10 species of migratory fishes (largehead hairtail, chub mackerel, Pacific saury, skipjack tuna, Pacific cod, anchovy, Alaska pollack, brown croaker, Japanese Spanish mackerel, yellow croaker and Pacific herring) were low compared to those in 7 species of demersal fishes (red stingray, brown sole, bastard halibut, conger eel, blackmouth angler, rockfish and filefish). Based on the mean concentrations, the PTWI% of total mercury among commonly consumed migratory fishes was 3.393%, which was higher than that of commonly consumed demersal fishes (2.710%).

  • This study determined the concentrations of lead and cadmium in 18 species of commonly consumed fish and assessed the risk based on provisional tolerable weekly (monthly) intakes [PTW(M)I] % as affected by behavioral characteristics, such as migration and settlement. In the 18 species, the mean concentrations of lead and cadmium were higher in the 11 species of migratory fish (llargehead hairtail Trichiurus lepturus, chub mackerel Scomber japonicus, Pacific saury Cololabis saira, skipjack tuna Katsuwonus pelamis, Pacific cod Gadus macrocephalus, anchovy Engraulis japonicus, Alaska pollack Theragra chalcogramm, brown croaker Miichthys miiuy, Japanese Spanish mackerel Scomberomorus niphonius, yellow croaker Larimichthys polyactis, and Pacific herring Clupea pallasii) than in the seven demersal species (red stingray Dasyatis akajei, brown sole Pleuronectes herzensteini, bastard halibut Paralichthys olivaceus, conger eel Conger myriaster, blackmouth angler Lophiomus setigerus, rockfish Sebastes schlegelii, and filefish Stephanolepis cirrhifer). Based on the mean concentrations, the PTWI % of lead and cadmium in commonly consumed migratory fish were 1.900 and 2.986%, respectively, which were higher than the values for lead and cadmium in the commonly consumed demersal fishes (0.257 and 0.318%, respectively). The estimation of weekly (monthly) intakes and target hazard quotients for the toxic elements lead and cadmium revealed that the commonly consumed migratory and demersal fish do not pose any health risks for consumers.

측경부 낭종으로 발현된 유두상 갑상선암 (Lateral Neck Cyst as the Initial Presentation of Papillary Thyroid Carcinoma)

  • We have experienced 3 cases of papillary carcinoma of the thyroid gland seen as lateral neck cyst, Usually cystic lesions of the neck have been considered as benign lesions from clinical viewpoint. The usual differential diagnosis includes branchial cleft cyst, cystic hygroma, dermoid cyst, cold abscess(tuberculous lymphadenitis) and cavitating squamous cell carcinoma. A lateral neck cyst as the sole presenting complaint of the papillary thyroid carcinoma is very rare. Preoperative diagnosis of lateral neck cyst is often diagnostic dilemma. Fine-needle aspiration cytology can be helpful in detecting the cancer cells and in demonstrating the nature of the fluid component of the lateral neck cyst. Presence of brown murky fluid from the aspirated fluid is highly suggestive of thyroid carcinoma. The thyroid scans and B-mode ultrasonography mayor may not be helpful to detect the primary focus of the thyroid gland. We suggest that a patient with a lateral neck cyst in adult age group should be considered to be a possibility of underlying thyroid carcinoma presanting as lateral neck cyst.

동해안에 서식하는 참가자미(Pseudopleuronectes herzensteini)의 해역별 산란특성 비교 (Comparison of the Spawning Characteristics of the Yellow Striped Flounder Pseudopleuronectes herzensteini in the Coastal Waters off Gangwon and Gyeongbuk, Korea)

  • We investigated the maturity and spawning of the brown sole Pseudopleuronectes herzensteini, in the coastal waters of the East Sea off Gangwon and Gyeongbuk, South Korea, using samples collected by gill net and longline fishery from January 2018 to December 2019. We analyzed oocyte development, monthly maturity stage, gonadosomatic index (GSI) and total length at maturity. Histological analysis of ovarian development in P. herzensteini revealed that the development was group-synchronous. The spawning peak in Gangwon and Gyeongbuk occurred from April to May and from February to March, respectively. Thus, the spawning season in Gangwon was 1-2 months later than that in Gyeongbuk. The length at 50% maturity for females was estimated as 24.2 cm and 19.0 cm in Gangwon and Gyeongbuk, respectively. Similarly, the length at 50% maturity for males was estimated as 19.8 cm and 16.5 cm in Gangwon and Gyeongbuk, respectively.
