Abundance and Breeding Migration of the Asian Toad (Bufo gargarizans)

  • We monitored a breeding population of the Asian toad (Bufo gargarizans), in the Wonheunge pond at Sannamdong, Chungju, from 5 March to 11 April, 2006 and 14 February to 31 March, 2007 to investigate their movement patterns, breeding population sizes, and physical characteristics. Terrestrial migration to the pond started on 5 March in 2006 and 14 February in 2007. We captured a total of 266 immigrating individuals (213 males, 53 females) in 2006 and 307 (222 males, 85 females) in 2007, and found 50 adults apparently killed by motor vehicles while migrating to the pond in 2007. Emigration from the pond to terrestrial sites started on 15 March 2006 and 5 March 2007. We captured a total of 245 emigrating toads (181 males, 65 females) in 2006 and 99 (92 males, 7 females) in 2007. An additional 10 emigrating adults were found dead on the road. During both the immigration and emigration periods, two peaks in capture frequency appeared for each sex in each breeding season. The immigration peaks corresponded with higher temperatures, while the emigration peaks corresponded with high humidity. Migrating Asian toads showed sexual size dimorphism and a male-biased sex ratio. Body weights and SVL (snout-vent length) of immigrating and emigrating individuals were negatively related with migration dates.

Development of a Sequence Characteristic Amplified Region Marker linked to the L4 Locus Conferring Broad Spectrum Resistance to Tobamoviruses in Pepper Plants

  • To develop molecular markers linked to the $L^4$ locus conferring resistance to tobamovirus pathotypes in pepper plants, we performed AFLP with 512 primer combinations for susceptible (S pool) and resistant (R pool) DNA bulks against pathotype 1.2 of pepper mild mottle virus. Each bulk was made by pooling the DNA of five homozygous individuals from a T10 population, which was a near-isogenic $BC_4F_2$ generation for the $L^4$ locus. A total of 19 primer pairs produced scorable bands in the R pool. Further screening with these primer pairs was done on DNA bulks from T102, a $BC_{10}F_2$ derived from T10 by back crossing. Three AFLP markers were finally selected and designated L4-a, L4-b and L4-c. L4-a and L4-c each underwent one recombination event, whereas no recombination for L4-b was seen in 20 individuals of each DNA bulk. Linkage analysis of these markers in 112 $F_2$ T102 individuals showed that they were each within 2.5 cM of the $L^4$ locus. L4-b was successfully converted into a simple 340-bp SCAR marker, designated L4SC340, which mapped 1.8 cM from the $L^4$ locus in T102 and 0.9 cM in another $BC_{10}F_2$ population, T101. We believe that this newly characterized marker will improve selection of tobamovirus resistance in pepper plants by reducing breeding cost and time.

  • Objective: Genome-wide association study and two meta-analysis based on GWAS performed to explore the genetic mechanism underlying variation in pig number born alive (NBA) and total number born (TNB). Methods: Single trait GWAS and two meta-analysis (single-trait meta analysis and multi-trait meta analysis) were used in our study for NBA and TNB on 3,121 Yorkshires from 4 populations, including three different American Yorkshire populations (n = 2,247) and one British Yorkshire populations (n = 874). Results: The result of single trait GWAS showed that no significant associated single nucleotide polymorphisms (SNPs) were identified. Using single-trait meta analysis and multi-trait meta analysis within populations, 11 significant loci were identified associated with target traits. Spindlin 1, vascular endothelial growth factor A, forkhead box Q1, msh homeobox 1, and LHFPL tetraspan submily member 3 are five functionally plausible candidate genes for NBA and TNB. Compared to the single population GWAS, single-trait Meta analysis can improve the detection power to identify SNPs by integrating information of multiple populations. The multiple-trait analysis reduced the power to detect trait-specific loci but enhanced the power to identify the common loci across traits. Conclusion: In total, our findings identified novel genes to be validated as candidates for NBA and TNB in pigs. Also, it enabled us to enlarge population size by including multiple populations with different genetic backgrounds and increase the power of GWAS by using meta analysis.

Agronomic Characteristics of Rice Recombinant Inbred Lines (RILs) Developed from a Cross of Nonganbyeo and BG 279

  • A set of rice recombinant inbred lines was developed from a cross between a Tongil type variety, Nonganbyeo, and an indica variety, BG276, by the single seed descent method. The number of the lines in the population was 272. All the agronomic characters studied except ADV (alkali-digestion value) showed continuous variation among the RILs, implying that their inheritance mode should be quantitative. The patterns of the variation in the RILs were either normal or skewed distribution. ADVs of RILs were segregated into two groups with 1:1 ratio, indicating that ADVs in this KIL population might be controlled by one major gene. Transgressive variations were also observed in all characters. Heritability values of the characters varied from 0.488 in brown/rough rice ratio to 0.895 in alkali-digestion value. In the analysis of genotypic and phenotypic correlations, the character of yield was positively correlated with 8 different agronomic characters. The number of panicles per hill was negatively correlated with culm length, panicle length, and number of spikelets per panicle. Grain length was positively correlated with grain width, grain thickness, grain length/width ratio, white belly, ADV, and amylose. However, grain length/width ratio was negatively correlated with grain width. White core was also negatively correlated with white belly and ADV.

Investigation of Cytoplasmic Male Sterility and Development of Maintainer and Restorer Lines in Rye (Secale cereale L.)

  • Rye has been a major winter forage crop in Korea. Varietal improvement of rye has been practiced either by hybrid or population breeding systems. Hybrid breeding offers important advantages over population breeding since it is normally a cross-pollinated crop. The hybrid breeding in rye has been possible since cytoplasmically inherited forms of male sterility (CMS) and corresponding nuclear restorer genes were found. The objectives of this research were to develop the maintainer and restorer lines of Korean inbred lines and to estimate the effect of 'Pampa' type of CMS cytoplasm on yield and its related characteristics. For easy discrimination of male-sterile status of plants, anther scoring and the restore index system in which seed-setting and pollen quantity of viability were taken into account were established. High significant correlation between pollen quantity and pollen viability was found. For "Pampa" cytoplasm, four of 14 Korean inbred lines tested turned out to be a maintainer but no restorer was found. But for "235b" CMS cytoplasm, seven inbred lines acted as complete restorers. The Korean inbred rye lines acted mainly as maintainers in "Pampa" cytoplasm but acted mainly as restorer in "235b" cytoplasm. The 'Pampa' cytoplasm inducing male sterility reduced cohn length and plant height and increased the number of tiller, so forage yield and grain yield were enhanced. Effects of using Umbilical Hernia Animals as Breeding Pigs on the Reproductive Traits (배꼽탈장(umbilical hernia)돼지의 종돈 활용이 번식형질에 미치는 영향)

  • This study investigated the incidence rate of umbilical hernia (UH) in pig farms and examined the effect of using them as breeding pigs on reproductive traits. The occurrence of UH ranged from 0.1% to 3.0% in pig farms investigated. UH pigs were found in almost all farms except for a single farm in Jeju Island. Spontaneously occurring UH pigs were selected and used for cross breeding tests. UH-related crosses and their progeny showed significant (P < 0.05) differences in gestation period, the numbers of piglets born and alive, and body weights at birth and $21^{st}$ day comparing to those of the control population. UH-related crosses showed longer gestation period, reduced numbers of piglets, and lighter body weights than those from the control population. Interestingly, reduced number of piglets was about one fourth, suggesting that UH inheritance might play a critical role as a lethal gene during embryogenesis. In addition, UH incidence rate in UH-related crosses was significantly (P < 0.05) higher than that in the control except for UH-cross3. However, in the progeny of control cross, a pig also had UH appearance, indicating that porcine UH might be inherited in recessive inheritance mode. Taken together, the results of this study indicate that UH is one of recessively inherited genetic defect that occurs at ordinary times in pig farms, suggesting that the use of UH animals as sire and/or dam may lead to economic losses due to increased gestation period, reduced numbers of piglets born and alive, and lower growth rates after birth of pigs.

Comparison of Selection Efficiency between Marker-Assisted Selection and Phenotypic Selection for Development of Brown Planthopper Resistance Lines in Rice (벼멸구 저항성 계통선발을 위한 MAS의 선발효율 비교)

  • This study presents a case study designed to compare the selection efficiency between phenotypic selection (PS) and marker-assisted selection (MAS) in breeding of resistance lines to brown planthopper (BPH). The efficiency between PS and MAS were compared with four population such as the $F_2$, RILs ($F_6$), DH, and backcrosse ($BC_6F_5$) population, derived from a cross 'Samgang / Nagdong'. The resistance lines were selected using two markers, RM28493 and BpE18-3, related to BPH resistance were screened as resistance lines over 95% in PS. The costs required for BPH screening in the MAS system account for approximately 32% of the total costs of PS. The period needed to select the resistance plants was 30 days in PS and 7 days in MAS. Based on the results, we could establish the breeding system for selection of BPH resistance lines by MAS.

The Accuracy of Genomic Estimated Breeding Value Using a Hanwoo SNP Chip and the Pedigree Data of Hanwoo Cows in Gyeonggi Province (한우 SNP Chip 및 혈통 데이터를 이용한 경기 한우 암소의 유전능력평가 정확도 분석)

  • This study was conducted to establish a genetic evaluation system applicable to general farms for improving cows raised on farms. The analysis used Best Linear Unbiased Prediction (BLUP) and Genomic Best Linear Unbiased Prediction (GBLUP) for 619 cows raised in Gyeonggi-do Province and compared and analyzed the accuracy of the estimated breeding value according to four traits (carcass weight, loineye muscle area, back fat thickness, and marbling). In the case of the GBLUP method, the size of the reference population was divided into different four groups and analyzed. The analysis results confirmed that the accuracy of the breeding value of each trait increased as the size of the GBLUP reference population increased. Comparing the accuracy of the breeding values estimated using the BLUP and GBLUP methods, it was confirmed that when the breeding values were estimated using the GBLUP method, they increased by 0.10, 0.09, 0.09, and 0.11 for carcass weight, eye muscle area, back fat thickness, and marbling scores, respectively. Applying the GBLUP method to the evaluation and selection of cows can enable precise and accurate individual selection, while increasing the size of the reference population can make even more accurate individual selection possible, thus increasing selection efficiency.

Effects of Climate-Changes on Patterns of Seasonal Changes in Bird Population in Rice Fields using a Prey-Predator Model (포식자-피식자 모델을 이용하여 기후변화가 논습지를 이용하는 조류 개체군 동태에 미치는 영향 예측)

  • BACKGROUND: It is well known that rice-fields can provide excellent foraging places for birds including seasonal migrants, wintering, and breeding and hence the high biodiversity of rice-fields may be expected. However, how environmental change including climate-changes on life-history and population dynamics in birds on rice-fields has not been fully understood. In order to investigate how climate-change affects population migratory patterns and migration timing, I modeled a population dynamics of birds in rice-fields over a whole year. METHODS AND RESULTS: I applied the Lotka-Volterra equation to model the population dynamics of birds that have been foraging/visiting rice-fields in Korea. The simple model involves the number of interspecific individuals and temperature, and the model parameters are periodic in time as the biological activities related to the migration, wintering and reproduction are seasonal. As results, firstly there was a positive relationship between the variation of seasonal population sizes and temperature change. Secondly, the reduced lengths of season were negatively related to the population size. Overall, the effects of the difference of lengths of season on seasonal population dynamics were higher than the effects of seasonal temperature change. CONCLUSION(S): Climate change can alter population dynamics of birds in rice-fields and hence the variation may affect the fitness, such as reproduction, survival and migration. The unstable balances of population dynamics in birds using paddy rice field as affected by climate change can reduce the population growth and species diversity in rice fields. The results suggest that the agricultural production is partly affected by the unstable balance of population in birds using rice-fields.

Comparison of Nucleic Acid Levels, Ratio and Ecophysiological Aspects among Three Populations of the Fleshy Prawn Fenneropenaeus chinensis in Korea

  • Using biochemical methods, we determined the potential of local female shrimp populations as breeding stock to select the best adult prawns for improving larval production. As condition indexes, we selected total RNA, DNA, their ratio, and trypsin activity. The DNA content in the pleopods of each local population was similar, i.e., between $0.90{\pm}0.06\;and\;1.02{\pm}0.04(SE){\mu}g/mg$. In comparison, the RNA contents differed markedly between $2.00{\pm}0.09$ and $0.96{\pm}0.08\;{\mu}g/mg$. Therefore, the RNA/DNA (R/D) ratio in the pleopod could be used as a condition index because it represents a biochemical characteristic of the population. The mean pleopodal R/D ratio of the Goheung population was the highest at $2.52{\pm}0.19$, which indicated the best condition. Trypsin activity was influenced little by shrimp condition and more by the amount of food ingested. The gonadosomatic index (GSI) and R/D ratio in the gonads provided offsetting information about the instantaneous gonad maturity. The Goheung population had the highest instantaneous GSI, despite some spawning. Based on the condition indexes and time of gonad maturation, the Goheung shrimp population is suitable for use as breeding stock.
