경사도 각도에 따른 트레드밀 보행훈련 시 뇌졸중 환자의 뇌파에 미치는 영향 (Effect of Change in Degrees of Inclination during Treadmill Gait Training on EEG of Stroke Patients)

  • Purpose: This study aimed to investigate the effects of gradually increasing treadmill inclination on the electroencephalogram (EEG) of stroke patients during gait training. Methods: Three stroke patients who were diagnosed with stroke within six months and capable of walking on a treadmill were selected as subjects. EEG electrodes were attached at Fp1, Fp2, F3, F4, C3, C4, P3, and P4 positions of the cerebral hemispheres using the International 10-20 system. The intervention involved walking for 2 minutes each at 0 degrees, 15 degrees, and 30 degrees inclination on the treadmill while focusing on a target point located in front during the treadmill gait training. The EEG (Smartingmobi, Serbia) generated when the treadmill gradient gradually increased was measured. In addition, relative alpha and relative beta waves were visualized through the Brain mapping program in the TeleScan program to assess the changes in each brain region for the activity of the EEG. Results: The relative alpha wave value decreased as treadmill inclination increased, while the relative beta wave value increased. Conclusion: Gradually increasing the inclination during treadmill gait training appears to be a crucial parameter for increasing the brain activity levels of stroke patients.

가상현실에서 아바타를 통한 정보전달 시 뇌의 활성화와 현존감의 관계 (The Relationship between Brain Activities and Presence on Communication using an Avatar in Virtual Reality)

  • 가상현실은 감각기관에 전달하는 정보를 조작하여 가상의 경험을 피험자에게 제공하고, 사람은 가상현실에서 제공하는 감각적인 정보를 뇌에서 통합하여 상황을 판단한다. 그러한 판단을 통해 가상현실이 얼마나 실제와 같은지 아닌지 지각하고 판단하게 된다. 이렇게 어떤 특정한 또는 이해할 수 있는 장소에 존재한다고 생각하는 인간의 지각상태를 나타내는 요소를 '현존감'이라고 한다. 가상의 환경을 사용자가 실제처럼 느끼고 그 내용을 받아들이도록 하는 것은 중요한 목표이다. 또한 현존감은 가상현실에서 경험하게 되는 여러 가지 감각적인 정보들을 통하여 느끼는 종합적인 느낌을 반영한다. 따라서 여러 가지 감각적인 정보들을 통합하는 과정에서 현존감과 관련된 뇌의 메커니즘이 존재할 것이다. 현존감은 매체를 통한 정보전달 시, 매체에서 전달되는 여러 가지 정보나 자극에 대한 사용자의 지각 및 인식과 중요한 관련이 있기 때문에 현존감의 정도는 개인이 지각하는 정보의 명확성에 영향을 준다. 이를 알아보기 위해서 사회적인 상황에서의 명확성의 차이를 가지는 내용을 아바타를 통해 전달하고 피험자가 그 내용을 바탕으로 추론해보는 가상현실을 구성하였다. 그리고 피험자가 전달받은 내용을 바탕으로 추론하는 동안 뇌 영역 활성화의 차이와 가상현실에 대한 현존감 점수가 관련된 뇌 영역을 알아보았다. 실험 결과 우측 혀이랑, 우측 쐐기소엽, 좌측 혀이랑, 우측 방추상이랑, 좌측 하측두이랑, 전대상피질 그리고 우측 후대상피질에서 유의미한 양의 상관관계가 있었다. 본 연구는 가상현실을 통한 정보의 전달 시 현존감의 영향을 뇌 기능영상 촬영을 통해서 내부 메커니즘의 측면에서 살펴보았다는 것에 의의가 있다.

GRIM-19 Expression and Function in Human Gliomas

  • Objective : We determined whether the expression of GRIM-19 is correlated with pathologic types and malignant grades in gliomas, and determined the function of GRIM-19 in human gliomas. Methods : Tumor tissues were isolated and frozen at $-80^{\circ}C$ just after surgery. The tissues consisted of normal brain tissue (4), astrocytomas (2), anaplastic astrocytomas (2), oligodendrogliomas (13), anaplastic oligodendrogliomas (11), and glioblastomas (16). To profile tumor-related genes, we applied RNA differential display using a $Genefishing^{TM}$ DEG kit, and validated the tumor-related genes by reverse transcription polymerase chain reaction (RT-PCR). A human glioblastoma cell line (U343MG-A) was used for the GRIM-19 functional studies. The morphologic and cytoskeletal changes were examined via light and confocal microscopy. The migratory and invasive abilities were investigated by the simple scratch technique and Matrigel assay. The antiproliferative activity was determined by thiazolyl blue Tetrazolium bromide (MTT) assay and FACS analysis. Results : Based on RT-PCR analysis, the expression of GRIM-19 was higher in astrocytic tumors than oligodendroglial tumors. The expression of GRIM-19 was higher in high-grade tumors than low-grade tumors or normal brain tissue; glioblastomas showed the highest expression. After transfection of GRIM-19 into U343MG-A, the morphology of the sense-transfection cells became larger and more spindly. The antisensetransfection cells became smaller and rounder compared with wild type U343MG-A. The MTT assay showed that the sense-transfection cells were more sensitive to the combination of interferon-$\beta$ and retinoic acid than U343MG-A cells or antisense-transfection cells; the antiproliferative activity was related to apoptosis. Conclusion : GRIM-19 may be one of the gene profiles which regulate cell death via apoptosis in human gliomas.

과학적 창의력과 정서적 감수성의 관계에 대한 뇌영상 연구 (An fMRI Study of Relationship between Scientific Creativity and Emotional Susceptibility)

  • 본 연구에서는 과학적 창의력과 정서적 감수성의 관계를 알아보고자 고등학교 과학발명반집단(n=13)과 일반 고등학생집단(n=13)을 대상으로 정서를 유발하는 시각자극을 제시하고 뇌활동성을 기능성자기공명영상(fMRI)을 이용하여 분석하였다. 국내외 창의력 경진대회 수상자들을 중심으로 구성된 과학발명반집단은 창의적 성격검사(WKOPAY, SAM) 점수가 상위 96%이내였으며 일반집단은 보통 수준이었다. 정서를 유발하는 시각자극(IAPS사진) 제시 시 뇌활동성을 분석한 결과 과학발명반집단은 일반집단에 비해 중간측두이랑(MTG)과 상측두이랑(STG) 영역 등에서 뇌활동성이 높게 나타났다. 창의적 성격검사점수와 뇌활동성 간의 상관도를 분석한 결과 MTG, STG 영역 등에서 유의적인 상관도(p<0.005)를 나타내었다. 특히 흥분의 측면보다는 기분(유쾌/불쾌)의 측면에서 유의적인 상관도를 보이는 영역이 많았다. 이러한 결과는 과학적 창의력이 높은 학생들이 일반 학생들에 비해 정서적 감수성이 발달되어 있다는 점을 보여주는 것으로 과학영재교육 대상자의 선발과 교육프로그램에서 정서적 측면이 고려되어야 함을 시사한다.

단일조사 저출력 $CO_2$ Laser가 치주인대 섬유아세포의 세포 증식능과 세포 분화에 미치는 영향 (The Effect of single Low-Power $CO_2$ Laser irradiation on human PDL fibroblast Proliferation & Differentiation)

  • 이 논문은 단일조사 저출력 $CO_2$ Laser조사가 치주인대 섬유아세포의 증식과 분화에 미치는 영향을 살펴보고 가장 효과적인 에너지와 파워밀도(power density)를 알아보기 위해 다음과 같이 실험하였다. 0.5W 출력, 10.6 ${\mu}m$ 파장, 50 Hz 연속형 $CO_2$ Laser를 사용하여, 실험군은 laser tip과 배양된 세포 사이의 거리를 2cm, 3cm으로 나누고, 조사시간을 1초, 3초로 나누어 4개의 군으로 설정하였고 대조군은 laser를 조사하지 않은 군으로 하였다. 치주인대 섬유아세포의 증삭정도와 골모세포로의 분화정도를 보기 위하여 각각 MTT 실험과 ALP activity 실험을 시행하여 다음과 같은 결과를 얻었다. 1. Laser를 조사하고 난 후 5일째에, 모든 군에서 유의하게 세포가 증식되는 것을 확인할 수 있었고 조사방법간에 유의한 차이가 없었다. 2. 대조군과 살험군에서 0일째에 비하여 3일째, 5일째, 7일째, 10일째에 통계적으로 유의 하게 ALP activity가 증가하였고, 이중 2cm,1sec 군을 제외하면 3일째에서 가장 높은 ALP activity 값을 보였다. 특징적으로 2cm,1sec 군은 3일째부터 10일까지 통계적으로 유의하지는 않지만 시간이 지남에 따라서 ALP activity가 증가함을 보였다. 7일과 10일째에는 2cm,1sec, 3cm,3sec군에서 다른 군에 비하여 큰 activity값을 보였다. 이번실험에서 저출력 $CO_2$ Laser 조사는 세포의 증식보다는 분화에 더 큰 영향을 끼쳤고, 2cm, 1sec, 3cm, 3sec 군이 치주인대 섬유아세포의 분화에 가장 효과적인 laser 조사방법으로 분석되었다.

A Novel Thrombolytic and Anticoagulant Serine Protease from Polychaeta, Diopatra sugokai

  • Ischemic stroke can result from blockage of blood vessels, forming fibrin clots in the body and causing irreparable brain damage. Remedial thrombolytic agents or anticoagulants have been studied; however, because the FDA-approved tissue plasminogen activator has low efficacy and side effects, it is necessary to develop safer and more effective treatment candidates. This study aimed at assessing the fibrinolytic and anticoagulation features of a novel serine protease extracted and purified from Diopatra sugokai, a polychaeta that inhabits tidal flats. The purified serine protease was obtained through ammonium sulfate precipitation, affinity chromatography, and ion-exchange chromatography. Its molecular size was identified via SDS-PAGE. To characterize its enzymatic activities, the protease activity at various pH and temperatures, and in the presence of various inhibitors, was measured via azocasein assay. Its fibrinolytic activity and anticoagulant effect were assessed by fibrin zymography, fibrin plate assay, and fibrinogenolytic activity assays. The novel 38 kDa serine protease had strong indirect thrombolytic activity rather than direct activity over broad pH (4-10) and temperature ($37^{\circ}C-70^{\circ}C$) ranges. In addition, the novel serine protease exhibited anticoagulant activity by degrading the ${\alpha}$-, ${\beta}$-, and ${\gamma}$-chains of fibrinogen. In addition, it did not produce cytotoxicity in endothelial cells. Therefore, this newly isolated serine protease is worthy of further investigation as a novel alkaline serine protease for thrombolytic therapy against brain ischemia.

중추신경흥분제 및 Adenosine이 마우스의 자발운동에 미치는 영향 (Effects of Adenosine and CNS Stimulants on Motor Activity in Mice)

  • The behavioral pattern of an animal is influenced by endogenous and endogenous stimuli such as humoral secretion, neurohumoral transmitters, drugs, light and environmental change. It has teen known that adenosine is a normal constituent of brain, and has sedative or hypnotic effects and anticonvulsant effects, inhibiting the spontaneous firing of cells in the brain via membrane adenosine receptors. Recent studies suggest that the excitatory responses to xanthines in the CNS might be related to the competitive antagonism of xanthines to adenosine. This study was undertaken to Investigate the effects of adenosine and the CNS stimulants such as picrotoxin, strychnine and caffeine on the spontaneous activity of mire, and to examine the influence of adenosine on the seizures induced by large doses of CNS stimulants. Subjects were $20{\sim}30\;g$ adult mice, and the spontaneous activity was measured using the Selective Activity Meter after intraperitoneal injection of adenosine (10 mg/kg), caffeine (100 mg/kg), strychnine(0.2 mg/kg) or picrotoxin(0.5 mg/kg) with or without adenosine pretreatment. The seizures were induced with caffeine(200, 250 and 300 mg/kg), strychnine(1.25 and 1.5 mg/kg) or picrotoxin(10 and 15 mg/kg). The results are summarized as follows : 1) The spontaneous activity in mite was significantly inhibited between 10 and 20 minutes after adenosine treatment. 2) Caffeine and picrotoxin increased the motor activity significantly while strychnine had no effect on the activity. 3) The ambulatory activity in the caffeine, strychnine and picrotoxin treated groups was significantly inhibited by adenosine pretreatment. 4) The seizures were observed with caffeine(200, 250 and 300 m9/kg), strychnine(1.25 and 1.5 mg/kg) or picrotoxin(10 and 15 mg/kg). The caffeine induced seizures were inhibited by adenosine pretreatment, but the strychnine or picrotoxin induced seizures were not affected.

THe Effect of Chronic Ehronic Treatment and Cold stress on Catecholaminergic Enzyme activity and mRNA in Rat Brain and Adrenals

  • Sprague-Dawley male rats (150 g) were chronically treated with 5 v/v % ethanol admixed with nutritionally complete liquid diet and fed ad libitum for 3 weeks. One half of each group was exposed to cold stress at 4 ^{\circ}C either for 24 h (for determination of mRNA by in situ hybridization) or for 48 h (for determination of enzyme activity). Chronic ethanol treatment (ethanol) did not affect tyrosine hydroxylase(TH) mRNA level in locus coeruleus(LC) of brain and adrenal medulla(AM) compared to controls. Cold stress showed strong increase of TH mRNA level in LC and AM compared to controls. Pretreated ethanol reduced the increased TH mRNA level by cold stress in LC and AM. Ethanol did not affect TH activity in LC and adenal glands(adrenals). Cold stress increased TH activity in LC but not in adrenals. Pretreated ethanol did not reduce the increased TH activity by cold stress in LC but this result was not shown in adrenals. Phenylethanolamine-N-methyltransferase(PNMT) activity in $C_{1}$$C_{2}$ and adrenals increased only in ethanol treated group. THese results suggest that ethanol does not affect TH mRNA level and activity in LC and adrenals, but increases PNMT activity in $C_{1}$$C_{2}$ and adrenals in normal rat. It is also suggested that pretreated ethanol reduces the magnitude of cold stress response, that is induction of TH mRNA in LC and AM, and does not reduce the protein activation of TH that is also cold stress response in LC.

  • A sexual dimorphism in terbufos (S-t-butylthiomethyl-O,O-diethyl phosphorodithioate: TBF) toxicity was not reported and remains unclear. Previously. we reported that TBF treatment showed sexual dimorphic effects on acetylcholinesterase (AchE) activity.   • Body weight is maintained at a relatively constant level over days and months despite variability in food intake and physical activity. To achieve energy homeostasis, the hypothalamus receives information related to energy surplus or shortage from the periphery and controls food intake and energy expenditure. Leptin, an adipocyte derived hormone, is a principal mediator that signals the brain about the stored energy status. Increased leptin signaling in the brain prevents excess energy stores by suppressing food intake and increasing energy expenditure. In addition, insulin and nutrients themselves, such as glucose and free fatty acids, also regulate food intake.
